The complete mitochondrial genome of Linguatula serrata (tongue worm) isolated from a dog and phylogenetic analysis.

ABSTRACT: The complete mitogenome of Linguatula serrata isolated from nasal cavity of a dog was characterized for the first time. The total size of the circular mitogenome was 15,328?bp consisting of 37 genes including 13 protein coding genes, 22 tRNA genes, two rRNA genes and two control regions. Phylogenetic tree was constructed based on 17 closely related species and their genetic relationship with Linguatula serrata was analysed.

Characterisation of the tongue worm, Linguatula serrata (Pentastomida: Linguatulidae), in Australia.

Project description:We describe adult males and females and a nymph belonging to Linguatula serrata in Australia, based on light and scanning electron microscopies. In addition, 18S and Cox1 sequence data have also been provided and were compared with similar sequences in GenBank. Our specimens had identical 18S sequences and limited genetic distance in Cox1 region which fell within the intra-specific range observed for Linguatula spp. suggesting that they all belong to one species. Phylogenetic analyses showed that Australian specimens were grouped with L. serrata in Europe where the species was first found and described. A number of L. serrata from Iran and Bangladesh formed a distinct group. The genetic distance between these Linguatula and Australian/European L. serrata ranged from 0.46% to 2.21% which is larger than the genetic distance observed between L. arctica and Australian/European L. serrata (0.12%) suggesting that they belong to a different species. As pointed out previously by several other authors, L. serrata comprises more than one species and those from the Palearctic region (including Iran and Bangladesh) should not be automatically named L. serrata unless there is enough evidence for the identification. To accurately address the complex taxonomy of Linguatula spp. a detailed morphological and genetic characterisation of numerous developmental stages of the parasite is necessary, to ensure morphological differences are not due to development. This however may not be achievable in the near future due to significant reduction in expertise as well as research funding awarded in this area of research to understand the basics of our planet.

Complete Mitochondrial Genome of a Tongue Worm Armillifer agkistrodontis.

Project description:Armillifer agkistrodontis (Ichthyostraca: Pantastomida) is a parasitic pathogen, only reported in China, which can cause a zoonotic disease, pentastomiasis. A complete mitochondrial (mt) genome was 16,521 bp comprising 13 protein-coding genes (PCGs), 22 tRNA genes, 2 rRNA genes, and 1 non-coding region (NCR). A phylogenetic tree drawn with the concatenated amino acid sequences of the 6 conserved PCGs (atp6, cox1-3, and nad2) showed that A. agkistrodontis and Armillifer armillatus constituted a clade Pentastomida which was a sister group of the Branchiura. The complete mt genome sequence of A. agkistrodontis provides important genetic markers for both phylogenetic and epidemiological studies of pentastomids.

Project description:BackgroundGongylonema pulchrum (Nematoda: Gongylonematidae), a thread-like spirurid gullet worm, infects a range of mammalian definitive hosts, including cattle, pigs, equines, goats, primates and humans, and can cause gongylonemiasis.MethodsIn the present study, the complete mitochondrial (mt) genome of G. pulchrum was obtained using Long-range PCR and subsequent primer walking. The phylogenetic position of G. pulchrum within the Spiruromorpha was established using Bayesian analyses of the protein-coding genes at the amino acid level.ResultsThe length of this AT-rich (75.94%) mt genome is 13,798 bp. It contains 12 protein-coding genes, two ribosomal RNA genes, 22 transfer RNA genes and one non-coding region. The gene arrangement is the same as those of Thelazia callipaeda (Thelaziidae) and Setaria digitata (Onchocercidae), but distinct from that of Heliconema longissimum (Physalopteridae). Phylogenetic analyses, based on the concatenated amino acid sequence data for all 12 protein-coding genes using Bayesian inference (BI) method, showed that G. pulchrum (Gongylonematidae) was more closely related to Spirocerca lupi (Spiruroidea) than other members of the infraorder Spiruromorpha.ConclusionsThe present study represents the first mt genome sequence for the family Gongylonematidae, which provides the opportunity to develop novel genetic markers for studies of epidemiology, population genetics and systematics of this nematode of human and animal health significance.

Complete mitochondrial genome of yellow meal worm (Tenebrio molitor).

Project description:The yellow meal worm (Tenebrio molitor L.) is an important resource insect typically used as animal feed additive. It is also widely used for biological research. The first complete mitochondrial genome of T. molitor was determined for the first time by long PCR and conserved primer walking approaches. The results showed that the entire mitogenome of T. molitor was 15 785 bp long, with 72.35% A+T content [deposited in GenBank with accession number KF418153]. The gene order and orientation were the same as the most common type suggested as ancestral for insects. Two protein-coding genes used atypical start codons (CTA in ND2 and AAT in COX1), and the remaining 11 protein-coding genes started with a typical insect initiation codon ATN. All tRNAs showed standard clover-leaf structure, except for tRNA(Ser) (AGN), which lacked a dihydrouridine (DHU) arm. The newly added T. molitor mitogenome could provide information for future studies on yellow meal worm.

The complete chloroplast genome of Zelkova serrata and its phylogenetic position within Ulmaceae.

Project description:The complete chloroplast genome sequence of the Tertiary relict tree Zelkova serrata was reported in this study. The chloroplast genome is 158,875 bp in length with a typical angiosperm quantitative structure consisting of a large single copy (87,412 bp) and a small single copy (18,683 bp) separated by a pair of inverted repeat (26,390 bp). Genome annotation revealed a total of 129 genes comprising 84 protein-coding genes, 37 tRNA genes, and eight rRNA genes. Phylogenomic analysis based on the whole plastomes indicated that Z. serrata and Z. schneideriana formed a well-supported monophyletic clade sister to genus Ulmus in Ulmaceae.

Project description:In this study, we describe the complete mitochondrial genome of Diopatra cuprea (Bosc, 1802). The mitogenome was found to contain 14,990 base pairs (67.53% A + T content), with a total of 37 genes (13 protein coding, 22 transfer RNAs, and 2 ribosomal RNAs). This study also examined mitogenome phylogenetics relationships of closely related species and recovered that D. cuprea is closely related to eunicids. This work has added to the genetic resources for furthering evolutionary studies of Annelida.

Project description:Here, we present complete mitochondrial genome of the Eastern Slow Worm, Anguis colchica (Nordmann, 1840). Mitogenome complete sequence is 17,097?bp long and consists of 13 protein-coding genes, 22 tRNA genes, two rRNA genes and one control region. Anguis colchica mitochondrial genome has the same gene order as other mitogenomes of Anguis spp. Their analyzed genome has base composition as: A (30.4%), T (24.6%), C (30.4%), G (14.6%), with an A?+?T bias (55%). Length of the all 22 tRNA genes varies from 65 to 73?bp with an average of 69?bp. Presented mitogenome will provide new data for phylogenetic analysis within the genus Anguis.

Complete mitochondrial genome of the composting worm Dendrobaena veneta (Clitellata: Oligochaeta, Lumbricidae).

Project description:Dendrobaena veneta (Rosa, 1886) is widely distributed all over Europe due to its use as compost worm. The specimen presented here was collected in Tiranë district, Albania. Currently, only two species' complete or nearly complete mitochondrial genome (mitogenome) sequences have been reported in the genus Dendrobaena; D. octaedra (Savigny, 1826) and D. tellermanica Perel, 1966. In this study, the complete mitogenome of D. veneta was sequenced, assembled, and annotated. The mitogenome of D. veneta is a circular DNA molecule, consisting of 15,475 bp with an A + T content of 61.2%. It contains 13 protein-coding genes, 2 ribosomal RNA genes, 22 transfer RNA genes, and 1 non-coding region (control region). Phylogenetic analysis showed that D. veneta is clustered with the other two Dendrobaena species in the well-supported family Lumbricidae.

Molecular characterization and phylogeny of Linguatula serrata (Pentastomida: Linguatulidae) based on the nuclear 18S rDNA and mitochondrial cytochrome c oxidase I gene.

Project description:Linguatula serrata, a cosmopolitan parasite, is commonly known as tongue worm belonging to the subclass Pentastomida.We collected the nymphal stage of the worm from mesenteric lymph nodes of cattle and identified these as L. serrata based on morphology and morphometry. The 18S rDNA sequences showed no intraspecific variation, although cox1 sequences showed 99.7-99.9% homology. In the phylogenies inferred from both gene loci, members of the genus Linguatula (order Porocephalida) were closer to those of the order Cephalobaenida than to those of Porocephalida, reflecting a mismatch with the corresponding morphology-based taxonomy. Accordingly, analyses of additional gene loci using a larger number of taxa across the Pentastomida should be undertaken to determine an accurate phylogenetic position within the Arthropoda.
