The mitochondrial genome of Ephestia elutella (Insecta: Lepidoptera: Pyralidae).

ABSTRACT: In this study, the complete mitochondrial genome of the tobacco moth Ephestia elutella was sequenced and analyzed. The mitochondrial genome is 15345 bp long and contains 13 protein-coding genes, two rRNA genes, 22 tRNA genes, and one control region. Twenty-three genes were found to be encoded by the majority strand and the other 14 genes by minority strand, those is similar to that of other insects. The nucleotide compositing of the majority strand are 38.6% of A, 11.77% of C, 42.05% of T and 7.58% of G. The phylogenetic analysis by Maximum-likelihood (ML) method revealed that the E. elutella was close to the same genus insect Ephestia kuehniella.

A Chromosome-Level Genome Assembly of Ephestia elutella (Hubner, 1796) (Lepidoptera: Pyralidae).

Project description:The moth Ephestia elutella (Hübner), is a storage pest that feeds on tobacco, cacao beans, cereals, dried fruits, and nuts. We generated a chromosome-level genome assembly containing 576.94 Mb using Nanopore long reads (approximately 130×) and Hi-C data (approximately 134×). The final assembly contained 804 scaffolds, with an N50 length of 19.00 Mb, and 94.96% (547.89 Mb) of the assembly was anchored into 31 pseudochromosomes. We masked 58.12% (335.32 Mb) of the genome as repetitive elements, identified 727 noncoding RNAs, and predicted 15,637 protein-coding genes. Gene family evolution and functional enrichment analyses revealed significantly expanded gene families primarily involved in digestion, detoxification, and chemosensation. Strong chromosomal syntenic relationships were also observed among E. elutella, silkworm, and tobacco cutworm. This study could provide a valuable genomic basis for better understanding the biology of E. elutella.

The complete mitochondrial genome of the lesser aspen webworm moth Meroptera pravella (Insecta: Lepidoptera: Pyralidae).

Project description:The lesser aspen webworm moth, Meroptera pravella, is a small pyralid that uses quaking aspen (Populus tremuloides) and related tree species as larval hosts. Whole-genome Illumina sequencing allowed the assembly of a complete circular mitochondrial genome of 15,260 bp consisting of 80.7% AT nucleotides, 22 tRNAs, 13 protein-coding genes, 2 rRNAs and a control region. Mitogenome structure maintains complete synteny with other sequenced pyralid mitogenomes. Parsimony and maximum-likelihood phylogenetic reconstruction places M. pravella within monophyletic subfamily Phycitinae and monophyletic family Pyralidae. The Pyralidae, with monophyletic sister family Crambidae, constitute monophyletic superfamily Pyraloidea, which is consistent with conventional taxonomy.

The complete mitogenome of D. yiai was described in this study, which is typical circular duplex molecules and 15,430 bp in length, containing the standard metazoan set of 13 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes, and an A + T-rich region. The gene order is same with other lepidopterans. Except for cox1 started with CGA, all other PCGs started with the standard ATN codons. Most of the PCGs terminated with the stop codon TAA, whereas cox2 has the incomplete stop codon T. The phylogenetic tree showed that D. yiai and other six species belong to Phycitinae, are clustered into a clade.

The complete mitochondrial genome of Orthaga achatina (Lepidoptera: Pyralidae).

Project description:Orthaga achatina Butler is an important pest of camphor trees in Asia. The complete mitochondrial genome of O. achatina was sequenced in this study, which was 15,150 bp in size and comprised of 13 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes, and a control region. Besides, we used a phylogenomic approach to infer evolutionary relationships of O. achatina and 23 Lepidoptera species based on 13 conserved protein sequences of the mitochondrial genome. Our results underline the potential importance of mitochondrial genomes in comparative genomic analyses of Lepidoptera species and provide a robust evolutionary insight across the tree of Lepidoptera insects.

Diapause, signal and molecular characteristics of overwintering Chilo suppressalis (Insecta: Lepidoptera: Pyralidae).

Project description:Diapause is a complex and dynamic process. Chilo suppressalis, an important rice pest in Asia enters facultative diapause as larvae. Our results demonstrated in Yangzhou, China, diapause was initiated between September 4 and 12, 2010. After diapause termination, C. suppressalis remained in quiescence in the field for as long as three months. The average time between collection of field larvae of C. suppressalis and their pupation decreased as the season progressed from fall to next spring. Unexpectedly, the pupated ratio of female to male in the initiation of diapause was 0.22. The abundance of hsp90, hsp70, hsp60 and CsAQP1 all peaked on January 8 or 15, 2011. Nitric oxide (NO) is a secondary messenger that is positively correlated with the diapause of C. suppressalis. Among several geographically separated populations of C. suppressalis, there are no significant differences in the mRNA levels of hsp70, hsp60 or CsAQP1.

The complete mitochondrial genome sequence of Aglossa dimidiata (Haworth, 1809) (Lepidoptera: Pyralidae).

Project description:The complete mitochondrial genome (mitogenome) of Aglossa dimidiata (Lepidoptera: Pyralidae) was sequenced using high-throughput sequencing. The mitogenome of A. dimidiata was 15,225 bp in length. It comprised 37 typical genes and one control region. All protein-coding genes (PCGs) were initiated with ATN, except for COX1 (TTG). All PCGs used TAN as stop codon, except for ND5 and ND4 terminated with incomplete T. Twenty-two tRNA genes ranged from 61 to 71 bp in size. The monophyly of family Pyralidae and the sister relationship between A. dimidiata and Orthopygia glanucinalis are both supported by maximum likelihood method using the nucleotide sequences of 13 PCGs.

Effect of Different Wheat-Based Diets and Corn Meal Addition on Development Parameters of Ephestia kuehniella (Zeller, 1879) (Lepidoptera: Pyralidae).

Project description:The expansion of Integrated Pest Management (IPM), including biological control, has had several positive consequences for the agricultural environment and participants in the production chain. To enable successful operation and applications of biological control, production of insects used for rearing natural enemies (parasitoids and predators) must be optimized to reduce time and costs and improve production both qualitatively and quantitatively. The present study evaluated the effect of wheat varieties, the main component of artificial diets for Ephestia kuehniella, on the reproductive performance and biological parameters of this flour moth, which is used for mass production of Trichogramma spp. (Hymenoptera: Trichogrammatidae) and other parasitoids and predators. Four varieties of wheat were compared: BRS 327, BRS Marcante, BRS Parrudo, and KBR, with and without the addition of corn E. kuehniella reared on 97% BRS 327 wheat flour + 3% nutritional yeast had the best biological parameters and substitution of corn for about half of the wheat increased the number of eggs per female.

The complete mitochondrial genome analysis of Locastra muscosalis (Walker, 1866) (Lepidoptera: Pyralidae) and phylogenetic implications.

Project description:The complete mitochondrial genome of Locastra muscosalis (Walker, 1866) was sequenced and characterized in this study, which was the first reported complete mitogenome of the genus Locastra. The mitogenome of L. muscosalis has a total length of 15,177 bp, encompassing 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), 2 ribosomal RNA genes (rRNAs), and an A-T rich region. Phylogenetic analysis revealed that L. muscosalis is closely associated with Orthaga euadrusalis. These data will serve as a valuable foundation for future investigations into the Epipaschiinae and Pyralidae evolutionary history.

Characterization of the complete mitochondrial genome of Orthaga olivacea Warre (Lepidoptera Pyralidae) and comparison with other Lepidopteran insects.

Project description:Orthaga olivacea Warre (Lepidoptera: Pyralidae) is an important agricultural pest of camphor trees (Cinnamomum camphora). To further supplement the known genome-level features of related species, the complete mitochondrial genome of Orthaga olivacea is amplified, sequenced, annotated, analyzed, and compared with 58 other species of Lepidopteran. The complete sequence is 15,174 bp, containing 13 protein-coding genes (PCGs), 22 transfer RNA (tRNA) genes, 2 ribosomal RNA (rRNA) genes, and a putative control region. Base composition is biased toward adenine and thymine (79.02% A+T) and A+T skew are slightly negative. Twelve of the 13 PCGs use typical ATN start codons. The exception is cytochrome oxidase 1 (cox1) that utilizes a CGA initiation codon. Nine PCGs have standard termination codon (TAA); others have incomplete stop codons, a single T or TA nucleotide. All the tRNA genes have the typical clover-leaf secondary structure, except for trnS(AGN), in which dihydrouridine (DHU) arm fails to form a stable stem-loop structure. The A+T-rich region (293 bp) contains a typical Lepidopter motifs 'ATAGA' followed by a 17 bp poly-T stretch, and a microsatellite-like (AT)13 repeat. Codon usage analysis revealed that Asn, Ile, Leu2, Lys, Tyr and Phe were the most frequently used amino acids, while Cys was the least utilized. Phylogenetic analysis suggested that among sequenced lepidopteran mitochondrial genomes, Orthaga olivacea Warre was most closely related to Hypsopygia regina, and confirmed that Orthaga olivacea Warre belongs to the Pyralidae family.
