Complete mitochondrial genome of Bombus consobrinus (Hymenoptera: Apidae).

ABSTRACT: The complete mitochondrial genome of Bombus consobrinus was sequenced. This circular mitogenome is 17,966 bp (86.7% of AT) in length, which contains 22 tRNA genes, 13 protein-coding genes and two rRNA genes. All protein-coding genes are initiated with the typical ATN codon and terminated by TAA codon except an incomplete stop codon T for ND4 gene. tRNA-Ser (AGN) lacks a dihydrouridine (DHU) arm while other tRNA genes form a typical cloverleaf structure. The complete mitochondrial genome of B. consobrinus will be useful to understand the inter-species relationship and genetic diversity of bumblebees.

Project description:The complete mitochondrial genome of Bombus breviceps, which belongs to the subgenus of Alpigenobombus in Bombus genus, was sequenced. This circular mitogenome is 16,743 bp (83.5% AT) in length, with two rRNA genes, 22 tRNA genes, and 13 protein-coding genes. All protein-coding genes are initiated with the common invertebrate initiation codon except TTA for ND2, and terminated by the typical stop codon TAA or TAG. tRNA-Ser (TCT) lacks a dihydrouridine (DHU) arm while other tRNA genes form a cloverleaf structure. Phylogenetic analysis shows that genus Bombus has a closer relationship with genus Melipona than any other genera in Apidae family, and B. breviceps clusters closer to B. lapidarius than any other bumblebees with sequenced mitogenome.

The complete mitochondrial genome of Bombus filchnerae (Hymenoptera: Apidae) and phylogenetic analysis.

Project description:Here, we report the complete mitochondrial genome (mitogenome) of Bombus filchnerae (Hymenoptera: Apidae). The genome size of B. filchnerae was 18,553 bp with 88.7% A + T content, containing 13 protein-coding genes (PCGs), 22 tRNA genes, two rRNA genes, and an AT-rich control region (D-loop). tRNA rearrangement was observed in this mitochondrial genome when compared to those of other bumblebee species (e.g. Bombus breviceps and Bombus asiaticus). All the 13 PCGs initiated with typical ATN codons. Among them, 11 PCGs terminated with TAA, only nad4 and nad5 with incomplete stop codon TA and T, respectively. All the 22 tRNAs can be folded into typical cloverleaf structure, except for trnS1, whose dihydrouridine (DHU) arm forms a simple loop. The phylogenetic analysis based on the concatenated nucleotide sequences of all 13 PCGs indicated that B. filchnerae showed the closest relationship with Bombus pascuorum, forming a mono clade of the subgenus Thoracobombus, with well-resolved relationships among nine Bombus subgenera.

Project description:In the present report, we describe the complete mitochondrial genome of the common bumblebee, Bombus hypocrita hypocrita, from the Otome Plateau, in Yamanashi Prefecture, Japan. The mitochondrial genome of B. h. hypocrita is a circular molecule of 15,795?bp. It contains 13 protein-coding, 22 tRNA and two rDNA genes. The protein-coding genes had ATA, ATG or ATT as the initiation codon and were terminated by the typical stop codon TAA, except for ND4 and Cytb. All the tRNA genes typically formed a cloverleaf secondary structure, except for trnE and trnS1.

Next-generation sequencing of the mitochondrial genome of Bombus longipennis Friese, 1918 (Hymenoptera: Apidae).

Project description:Bombus longipennis is the species of Bumblebees (Hymenoptera: Apidae), which are important pollinators for wild plants and greenhouses crops. The complete mitochondrial genome (mitogenome) of B. longipennis was determined by next-generation sequencing. The mitogenome was 18,458 bp in size with 87.2% A + T content, containing 13 protein-coding genes (PCGs), 22 tRNA genes, two rRNA genes, and an AT-rich control region (D-loop). Gene arrangement was found to be identical to those of other mitogenomes of bumblebees (e.g. Bombus terrestris and Bombus ignitus). All 13 PCGs initiated with typical ATN codons. Among them, 11 PCGs terminated with TAA or TAG; only cox2 and nad4 have incomplete stop codon T. All 22 tRNAs can be folded into typical cloverleaf structure. Phylogenetic analysis based on the concatenated nucleotide sequences of all 13 PCGs indicated that B. longipennis was more closely related to other species of subgenus Bombus, which clustered into a monophyletic group.

The complete mitochondrial genome of Apis mellifera meda (Insecta: Hymenoptera: Apidae).

Project description:The complete mitochondrial genome of the western honey bee subspecies Apis mellifera meda was sequenced. This mitochondrial genome is 16,248?bp in length, with 37 classical eukaryotic mitochondrial genes and an A?+?T-rich region. Gene direction and arrangement are similar to those of other Apis mitogenomes. All genes initiate with ATT (six genes), ATG (four genes), ATA (two genes), and ATC (one gene) start codons and terminate with a TAA stop codon. Four genes are encoded on the heavy and nine on the light strands, respectively. All of the 22 tRNA genes, ranging from 66 to 78?bp, have a typical cloverleaf structure. The complete mitogenome of A.m. meda provides information on the biogeography and evolution of A. mellifera subspecies.

The complete mitochondrial genome of the Asiatic cavity-nesting honeybee Apis cerana (Hymenoptera: Apidae).

Project description:In the present study, we determined the complete mitochondrial DNA (mtDNA) sequence of Apis cerana, the Asiatic cavity-nesting honeybee. We present here an analysis of features of its gene content and genome organization in comparison with Apis mellifera to assess the variation within the genus Apis and among main groups of Hymenoptera. The size of the entire mt genome of A. cerana is 15,895 bp, containing 2 ribosomal RNA genes, 13 protein-coding genes, 22 transfer RNA (tRNA) genes and one control region. These genes are transcribed from both strands and have a nucleotide composition high in A and T. The contents of A+T of the complete genomes are 83.96% for A. cerana. The AT bias had a significant effect on both the codon usage pattern and amino acid composition of proteins. There are a total of 3672 codons in all 13 protein-coding genes, excluding termination codons. The most frequently used amino acid is Leu (15.52%), followed by Ile (12.85%), Phe (10.10%), Ser (9.15%) and Met (8.96%). Intergenic regions in the mt genome of A. cerana are 705 bp in total. The order and orientation of the gene arrangement pattern is identical to that of A. mellifera, except for the position of the tRNA-Ser(AGN) gene. Phylogenetic analyses using concatenated amino acid sequences of 13 protein-coding genes, with three different computational algorithms (NJ, MP and ML), all revealed two distinct groups with high statistical support, indicating that A. cerana and A. mellifera are two separate species, consistent with results of previous morphological and molecular studies. The complete mtDNA sequence of A. cerana provides additional genetic markers for studying population genetics, systematics and phylogeographics of honeybees.

Characteristics of the Two Asian Bumblebee Species Bombus friseanus and Bombus breviceps (Hymenoptera: Apidae).

Project description:This study compared the food plants, life cycle, colony development, and mating behaviour of the two Asian bumblebee species Bombus friseanus and B. breviceps, which are very important pollinators for many wild flowers and crops in local ecosystems. Both species were shown to be highly polylectic. Differences were observed in their life cycles and colony development patterns. The colony foundation rate of the field-collected queens was high in both species, 95.5% in B. friseanus and 86.5% in B. breviceps. The intervals from colony initiation to colony sizes of 30, 60, and 80 workers and to the first male and gyne emergence were significantly shorter in B. friseanus than in B. breviceps (p < 0.01). The development period of the first batch of workers showed no significant difference between the two species (p > 0.05). Compared with B. friseanus, B. breviceps produced remarkably higher numbers of workers (135 ± 30 workers/colony in B. friseanus and 318 ± 123 workers/colony in B. breviceps) and males (199 ± 46 males/colony in B. friseanus and 355 ± 166 males/colony in B. breviceps) (p < 0.01), with notable variation was found among the colonies in both species. With no significant difference in the mating rate between these two species, the copulation duration of B. breviceps (1.54 ± 0.63 min) was strikingly shorter than that of B. friseanus (27.44 ± 11.16 min) (p < 0.001). This study highlights the characteristics of the two Asian bumblebee species and will aid further studies on their conservation and agricultural pollination use.

Impact of Diflubenzuron on Bombus impatiens (Hymenoptera: Apidae) Microcolony Development.

Project description:Reliance on the honey bee as a surrogate organism for risk assessment performed on other bees is widely challenged due to differences in phenology, life history, and sensitivity to pesticides between bee species. Consequently, there is a need to develop validated methods for assessing toxicity in non-Apis bees including bumble bees. The usefulness of small-scale, queenless colonies, termed microcolonies, has not been fully investigated for hazard assessment. Using the insect growth regulator diflubenzuron as a reference toxicant, we monitored microcolony development from egg laying to drone emergence using the Eastern bumble bee Bombus impatiens (C.), a non-Apis species native to North America. Microcolonies were monitored following dietary exposure to diflubenzuron (nominal concentrations: 0.1, 1, 10, 100, and 1,000 µg/liter). Microcolony syrup and pollen consumption was significantly reduced by diflubenzuron exposure. Pupal cell production was also significantly decreased at the highest diflubenzuron concentration assessed. Ultimately, diflubenzuron inhibited drone production in a concentration-dependent manner and a 42-d 50% inhibitory concentration (IC50) was determined. None of the dietary concentrations of diflubenzuron tested affected adult worker survival, or average drone weight. These data strengthen the foundation for use of this methodology, and provide valuable information for B. impatiens; however, more work is required to better understand the utility of the bumble bee microcolony model for pesticide hazard assessment.
