Complete mitochondrial genome of the mulberry psyllid Paurocephala sauteri Enderlein (Hemiptera: Psyllidae) and phylogenetic analysis.

ABSTRACT: Paurocephala sauteri (Enderlein, 1914) (Hemiptera: Psyllidae) is a species of a psyllid distributed in Asia. Mulberry is the only known host for P. sauteri until now. The complete mitogenome of P. sauteri (accession number: MT759765) 14,963 bp in size, including 13 protein-coding genes, 22 transfer RNAs, and two ribosomal RNAs genes. The base composition of the whole P. sauteri mitogenome is 40.26% for A, 7.86% for G, 34.07% for T, and 11.81% for C, with a high AT bias of 80.33%. The mitochondrial genome of P. sauteri was sequenced and annotated as the first representative of family Paurocephalidae. The present data could contribute to a detailed phylogeographic analysis of this valuable economic insect for further study in differentiating closely related species.

The complete mitochondrial genome sequence of Diaphorina citri (Hemiptera: Psyllidae).

Project description:The first complete mitochondrial genome (mitogenome) sequence of Asian citrus psyllid, Diaphorina citri (Hemiptera: Psyllidae) from Guangzhou, China, is presented. The circular mitogenome is 14,996?bp in length with an A?+?T content of 74.5%, and contains 13 protein-coding genes (PCGs), 22 tRNA genes (tRNAs), two rRNA genes and an AT-rich control region. Synteny was identical to the ancestral insect mitogenomes. All PCGs start with 'ATN' codons. Eleven PCGs use the complete termination codons ('TAA' or 'TAG'), while the remaining two (cox2 and nad5) use a single 'T' as the stop codon. All tRNAs have the typical cloverleaf structure, except for trnTrp which lacks the variable arm and trnSer (AGN) which lacks the dihydrouridine arm. A phylogenetic tree, including 12 members of Sternorrhyncha, was constructed based on the 13 PCGs sequences. The resulting phylogenetic tree matched the established taxonomic scheme based on morphology.

The complete mitochondrial genome of Palomena viridissima (Hemiptera: Pentatomidae) and phylogenetic analysis.

Project description:Here, we sequenced and annotated the complete mitochondrial genome (mitogenome) of Palomena viridissima (Hemiptera: Pentatomidae). This mitogenome was 15,118 bp long, comprising of 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), 2 ribosomal RNA genes (rrnL and rrnS) and a large non-coding control region. The P. viridissima mitogenome with an A + T content of 76.0%, presented a positive AT-skew (0.11) and a negative GC-skew (-0.13). Ten PCGs started with a typical ATN codon, two PCGs started with TTG (atp8, nad1), whereas the remaining one used AAC (cox1). All tRNAs had a typical secondary cloverleaf structure, except for trnS1 which lacked the dihydrouridine arm. The Bayesian phylogenetic analysis based on mitogenomic data supported a sister relationship of P. viridissima and Nezara viridula from the same tribe Nezarini and recovered a phylogeny of Pentatominae: (Menidini + (Strachiini + (Pentatomini + ((Cappaeini + Halyini) + (Eysarcorini + (Nezarini + Carpocori)))))).

The Complete Mitochondrial Genome of Corizus tetraspilus (Hemiptera: Rhopalidae) and Phylogenetic Analysis of Pentatomomorpha.

Project description:Insect mitochondrial genome (mitogenome) are the most extensively used genetic information for molecular evolution, phylogenetics and population genetics. Pentatomomorpha (>14,000 species) is the second largest infraorder of Heteroptera and of great economic importance. To better understand the diversity and phylogeny within Pentatomomorpha, we sequenced and annotated the complete mitogenome of Corizus tetraspilus (Hemiptera: Rhopalidae), an important pest of alfalfa in China. We analyzed the main features of the C. tetraspilus mitogenome, and provided a comparative analysis with four other Coreoidea species. Our results reveal that gene content, gene arrangement, nucleotide composition, codon usage, rRNA structures and sequences of mitochondrial transcription termination factor are conserved in Coreoidea. Comparative analysis shows that different protein-coding genes have been subject to different evolutionary rates correlated with the G+C content. All the transfer RNA genes found in Coreoidea have the typical clover leaf secondary structure, except for trnS1 (AGN) which lacks the dihydrouridine (DHU) arm and possesses a unusual anticodon stem (9 bp vs. the normal 5 bp). The control regions (CRs) among Coreoidea are highly variable in size, of which the CR of C. tetraspilus is the smallest (440 bp), making the C. tetraspilus mitogenome the smallest (14,989 bp) within all completely sequenced Coreoidea mitogenomes. No conserved motifs are found in the CRs of Coreoidea. In addition, the A+T content (60.68%) of the CR of C. tetraspilus is much lower than that of the entire mitogenome (74.88%), and is lowest among Coreoidea. Phylogenetic analyses based on mitogenomic data support the monophyly of each superfamily within Pentatomomorpha, and recognize a phylogenetic relationship of (Aradoidea + (Pentatomoidea + (Lygaeoidea + (Pyrrhocoroidea + Coreoidea)))).

Complete Mitochondrial Genome of Dinorhynchus dybowskyi (Hemiptera: Pentatomidae: Asopinae) and Phylogenetic Analysis of Pentatomomorpha Species.

Project description:Dinorhynchus dybowskyi (Hemiptera: Pentatomidae: Asopinae) is used as a biological control agent against various insect pests for its predatory. In the present study, the complete mitochondrial genome (mitogenome) of the species was sequenced using the next-generation sequencing technology. The results showed that the mitogenome is 15,952 bp long, including 13 protein-coding genes (PCGs), 22 transfer RNAs (tRNAs), two ribosomal RNAs (rRNAs), and a control region. Furthermore, the gene order and orientation of this mitogenome are identical to those of most heteropterans. There are 21 intergenic spacers (of length 1-28 bp) and 13 overlapping regions (of length 1-23 bp) throughout the genome. The control region is 1,291 bp long. The start codon of the PCGs is ATN, except cox1 (TTG), and stop codon is TAA, except nad1 (TAG). The 22 tRNAs exhibit a typical cloverleaf secondary structure, except trnS1, which lacks a dihydrouridine (DHU) arm and trnV, where the DHU arm forms a simple loop. The analyses based on nucleotide sequences of the 13 PCGs by Bayesian Inference and maximum likelihood methods. The results support the monophyly of five superfamilies Aradoidea, Pentatomoidea, Pyrrhocoroidea, Lygaeoidea, and Coreoidea. Within Pentatomoidea, the relationship observed is as follows: (Plataspidae + Urostylididae) + (Pentatomidae + (Acanthosomatidae + (Cydnidae + (Scutelleridae + (Dinidoridae + Tessaratomidae))))), and D. dybowskyi was placed in Pentatomidae and close to Eurydema gebleri.

Characterization and phylogenetic analysis of the complete mitochondrial genome of Aphis spiraecola (Hemiptera: Aphididae).

Project description:Aphis spiraecola is an important pest of citrus and transmits a number of plant viruses. Here, we sequenced and annotated the complete mitochondrial genome (mitogenome) of A. spiraecola. This mitogenome was 15,465?bp long and encoded 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), and 2 ribosomal RNA unit genes (rRNAs). Gene order was conserved and identical to that of Drosophila yakuba and to most other previously sequenced Aphididae. The whole mitogenome exhibited heavy AT nucleotide bias (81.8%). All 13 PCGs were initiated by the ATN (ATG, ATT and ATA) codon. Except for cox1 and nad4 which end with the incomplete codon T-, all PCGs terminated with the stop codon TAA. Phylogenetic analysis showed that A. spiraecola got together with the same genus species Aphis gossypii and Aphis craccivora with high support value, and Aphis had a close relationship with Schizaphis and Rhopalosiphum.

Complete mitochondrial genome of Mukaria splendida Distant (Hemiptera: Cicadellidae: Deltocephalinae: Mukariini) and phylogenetic analysis.

Project description:The complete mitochondrial genome (mitogenome) of Mukaria splendida Distant, 1908 (Hemiptera: Cicadellidae: Deltocephalinae) was first reported in Mukariini. The length of this mitogenome is 16,711 bp, which has an A + T content of 79% (A = 44.5%, T = 34.5%, G = 8.8%, C = 12.2%). A total of 37 genes were annotated [13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNA), and 2 ribosomal RNA genes (rRNA)]. Among the 37 genes, 4 protein coding genes (ND1, ND4, ND4L, ND5), 8 tRNA genes (trnQ, trnC, trnY, trnF, trnH, trnP, trnL2, trnV), and 2 rRNA (12S rRNA, 16S rRNA) were encoded by N chain, and the remaining genes were encoded by J chain. Overall, there were 14 gene overlaps and 9 gene gaps in the mitochondrial genome of this species. All PCGs were started with ATD (ATA/ATT/ATG), and stopped with TAR, except ATP6, which ends with single T. The phylogenetic analysis confirms that M. splendida clustered with other Deltocephalinae species.

Complete mitochondrial genome of Neotoxoptera formosana (Takahashi, 1921) (Hemiptera: Aphididae), with the phylogenetic analysis.

Project description:In this study, we sequenced and annotated the complete mitochondrial genome (mitogenome) of Neotoxoptera formosana (Takahashi) (Hemiptera: Aphididae). The complete mitogenome of N. formosana is 15,642 bp in length, and includes 13 protein-coding genes (PCGs), 2 ribosome RNA (rRNA) genes, 22 transfer RNA (tRNA) genes, and one control region. The overall base composition was as follows: 45.2% of A, 5.8% of G, 10.5% of C, and 38.4% of T, with a total of A + T content of 83.6%. The phylogenetic tree showed that N. formosana and Myzus persicae were clustered into one branch. This result will enrich the mitogenome of family Aphididae.

Two Complete Mitochondrial Genomes of Mileewinae (Hemiptera: Cicadellidae) and a Phylogenetic Analysis.

Project description:More studies are using mitochondrial genomes of insects to explore the sequence variability, evolutionary traits, monophyly of groups and phylogenetic relationships. Controversies remain on the classification of the Mileewinae and the phylogenetic relationships between Mileewinae and other subfamilies remain ambiguous. In this study, we present two newly completed mitogenomes of Mileewinae (Mileewa rufivena Cai and Kuoh 1997 and Ujna puerana Yang and Meng 2010) and conduct comparative mitogenomic analyses based on several different factors. These species have quite similar features, including their nucleotide content, codon usage of protein genes and the secondary structure of tRNA. Gene arrangement is identical and conserved, the same as the putative ancestral pattern of insects. All protein-coding genes of U. puerana began with the start codon ATN, while 5 Mileewa species had the abnormal initiation codon TTG in ND5 and ATP8. Moreover, M. rufivena had an intergenic spacer of 17 bp that could not be found in other mileewine species. Phylogenetic analysis based on three datasets (PCG123, PCG12 and AA) with two methods (maximum likelihood and Bayesian inference) recovered the Mileewinae as a monophyletic group with strong support values. All results in our study indicate that Mileewinae has a closer phylogenetic relationship to Typhlocybinae compared to Cicadellinae. Additionally, six species within Mileewini revealed the relationship (U. puerana + (M. ponta + (M. rufivena + M. alara) + (M. albovittata + M. margheritae))) in most of our phylogenetic trees. These results contribute to the study of the taxonomic status and phylogenetic relationships of Mileewinae.

De Novo Assembly of the Asian Citrus Psyllid Diaphorina citri (Hemiptera: Psyllidae) Transcriptome across Developmental Stages.

Project description:Asian citrus psyllid Diaphorina citri Kuwayama is an important economic pest of citrus, as it transmits Candidatus Liberibacter asiaticus, the causative agent of huanglongbing. In this study, we used RNA-seq to identify novel genes and provide the first high-resolution view of the of D. citri transcriptome throughout development. The transcriptomes of D. citri during eight developmental stages, including the egg, five instars, and male and female adults were sequenced. In total, 115 million clean reads were obtained and assembled into 354,726 unigenes with an average length of 925.65 bp and an N50 length of 1733 bp. Clusters of Orthologous Groups, Gene Ontology, and Kyoto Encyclopedia of Genes and Genomes analyses were conducted to functionally annotate the genes. Differential expression analysis highlighted developmental stage-specific expression patterns. Furthermore, two trehalase genes were characterized with lower expression in adults compared to that in the other stages. The RNA interference (RNAi)-mediated suppression of the two trehalase genes resulted in significantly high D. citri mortality. This study enriched the genomic information regarding D. citri. Importantly, these data represent the most comprehensive transcriptomic resource currently available for D. citri and will facilitate functional genomics studies of this notorious pest.

The complete mitochondrial genome of Bothrogonia qiongana (Hemiptera: Cicadellidae) with phylogenetic analyses.

Project description:The species of Bothrogonia qiongana was reported by Yang and Li in 1980. The mitogenome is 15,788 bp in length, including 37 genes and a control region, with an overall A + T content of 76.9%. Most of the PCGs start with ATN (ATA, ATC, ATT, ATG), but ND5 starts with TTG. All PCGs used TAA or TAG as stop codon except COX2 which is using incomplete single T--. The phylogenetic tree was reconstructed based on B. qiongana and 24 reference species, which is consistent with traditional taxonomy.
