Feline morbillivirus infections have gained increased attention due to repeated reports of their association with urinary tract disease in cats. In the present study, 112 serum samples from free-roaming domestic cats in Chile were tested for antibodies against feline morbillivirus genotypes 1 and 2 (FeMV-1 and FeMV-2) using an indirect immunofluorescence assay. Project description:Owned, free-roaming domestic cats are abundant in the Chilean countryside, having high probability of contact with wildlife and potentially participating as reservoirs of zoonotic pathogens. In the present study, 131 cats from two remote study areas (Valdivia and Chiloe Island) in southern Chile were analyzed for infection/exposure to eight pathogens. Serum samples from 112 cats were tested for antigens against feline leukemia virus (FeLV antigen-ELISA) and antibodies against feline immunodeficiency virus (FIV-ELISA) and canine distemper virus (CDV-serum neutralization), yielded occurrence of 8.9, 1.7 and 0.8% respectively. The presence of DNA of five vector-borne pathogens, piroplasmids, Ehrlichia spp., Anaplasma spp., Rickettsia spp. and Bartonella spp. was investigated in thirty cats. Overall observed occurrence was 6.6% (2/30) for both Anaplasma platys, and B. henselae, and 3.3% (1/30) for both Bartonella sp. and Theileria equi. Observed occurrence for all vector-borne pathogens in Valdivia area was significantly higher than in Chiloe Island (5/15 vs 0/15; P=0.04). Our results represent the first description of exposure to CDV and DNA detection of T. equi and A. platys in domestic cats in Chile. The results highlight the importance of performing pathogen screening in owned, free-roaming rural cats to evaluate their potential role as reservoirs of infection and vectors for disease transmission to wildlife.

Project description:The "trap, spay/neuter, and release" programs to manage unowned free-roaming cat populations are diffused worldwide and they are largely advised even in countries where the suppression of unowned cats is enforced by law. Despite the massive neutering campaigns in the world, there is little information on the influence of neutering on individual cat behaviour, as well as on the social structure of cat colonies. The aim of this study is to verify such effects. Before neutering, the group consisted of 17 free-roaming domestic cats, who were totally unrestrained. After neutering it consisted of 16 individuals. Data on the outcomes of aggressive, submissive, affiliative, and territorial behaviour were collected, using classic ethological methods ("focal animal", "all occurrences", and "1/0" sampling methods) (735 h of observation before and 537 h after neutering). The dominance hierarchy did not change after neutering. On the other hand, the frequency of aggressive, territorial behaviour as well as proximity among individuals decreased significantly. Affiliative behaviour has been observed among neutered adult males that never performed it before neutering. The results of this study suggest that, after neutering: (i) The group of free-ranging cats was stable over time; (ii) the hierarchical structure of the cat social group did not change; (iii) the general level of activity, decreased; (iv) urine spraying marking behaviour almost disappeared; (v) the level of social proximity among group members decreased, although it tended to increase in some male-male dyads. The most obvious effect of neutering, detected on individual behaviour, was that cats were less active.

Project description:Despite public concern on the role of free-roaming cats as reservoirs of zoonotic agents, little is known about the influence of urban and peri-urban landscapes on the exposure risk. We evaluated the seroprevalence of three zoonotic agents (Chlamydia felis, Coxiella burnetii and Toxoplasma gondii) in domestic cats (Felis catus). Two hundred and ninety-one free-roaming cats were trapped in Murcia municipality (Southeast Spain), and their sera were tested for specific antibodies against T. gondii using a modified agglutination test (MAT), and for C. felis, C. burnetii and feline immunodeficiency virus (FIV) antibodies with ELISA technique. Pathogen seroprevalence at 95% CI was calculated for each sex and age category (up to and over 12 months) and compared with a chi-squared test. The role of human population density and urban landscape characteristics on the risk of pathogen exposure in the cat population was explored using generalized linear models. Seropositivity against a single pathogen was found in 60% of the cats, while 19% was seropositive for two or three pathogens. Seroprevalence of C. felis was 8% (CI95% : 5-11), 37% (CI95% : 31-42) for C. burnetii and 42% (CI95% : 36-47) for T. gondii. In addition to these three pathogens, FIV seropositivity was low (1%, CI95% : -0.1 to 2) and adult cats were more likely to be seropositive to C. burnetii than young individuals (OR: 2.3, CI95% : 1.2-4.2). No sex or age class differences in seroprevalence were observed for the rest of the pathogens. Seropositivity was correlated with water surface areas for C. felis, and not with crop areas. Coxiella burnetii seropositivity was correlated with the percentage of urban areas (continuous with only buildings and discontinuous, that include buildings, parks, and pedestrian and urban green areas), human population size and peri-urban areas with shrubs, and not correlated with other agricultural landscapes (orchards and crop areas). However, the seroprevalence of T. gondii was only associated with agricultural landscapes such as orchards. The detection of hotspot areas of high pathogen exposure risk is the basis for municipal services to implement surveillance and risk factor control campaigns in specific-risk areas, including (a) efficient health management of urban cat colonies by geographical location, population census and health status monitoring of the components of each cat colony, (b) improvement of hygiene and sanitary conditions at the feeding points of the cat colony and (c) free-roaming cat trapping for health monitoring and, in the long term, to know the evolution of the health status of their populations.

Project description:Objectives:Feline immunodeficiency virus (FIV) and feline leukaemia virus (FeLV) are retroviruses affecting cats worldwide. The objectives of the study were to estimate the prevalence of these retroviruses in domestic cats in Hungary and to characterise the phylogenetic relationships of FIV strains. Methods:A total of 335 anticoagulated whole-blood samples obtained from both a healthy and ill cat population were examined for the presence of FIV and FeLV with two methods: ELISA and PCR. Statistical analysis was carried out to analyse the data obtained. Sequencing and phylogenetic analysis of partial polymerase (pol) gene sequences was performed to describe circulating FIV subtypes. Results:Statistical analysis showed 11.8% and 9.9% true prevalence of FeLV and FIV, respectively, with ELISA. The apparent prevalence calculated from the PCR results were 17.3% for FeLV and 13.1% for FIV. Phylogenetic analysis of partial pol gene sequences obtained from 22 FIV strains showed that all observed Hungarian strains belonged to FIV subtype B. The strains were grouped into several monophyletic subgroups reflecting the geographic locations of the origin of the samples. The overall mean genetic similarity between the analysed strains was 98.2%. Conclusions and relevance:We report the first thorough overview of the prevalence of FeLV and FIV in Hungary, which is relatively high, and give insight into the genetic diversity of Hungarian strains of FIV.

Project description:Feline astrovirus (FeAstV) which belonged to the genus Mamastrovirus was first identified in the feces of kittens with diarrhea in the USA in 1981 by electron microscopy, and had been reported in many countries. Presently, there are no any reports of the circulation of FeAstV in mainland China. We performed this study to investigate the apparent prevalence and genetic variability of FeAstV infected in cats in mainland China for the first time. We tested fecal samples of 105 cats with diarrhea and 92 asymptomatic cats in five cities in northeast China by RT-PCR targeting RNA-dependent RNA polymerase (RdRp) gene of FeAstV, and analyzed sequences variability and phylogenetic evolution based on the complete capsid gene of FeAstV strains obtained from positive samples. The overall prevalence of FeAstV was 23.4% (46/197) of which 38 were tested in cats with diarrhea (36.2%, 38/105) and 8 were in asymptomatic cats (8.7%, 8/92). Mixed infection with other enteroviruses including feline parvovirus (FPV), feline bocavirus (FBoV) and feline kobuvirus (FeKoV) was found in 38 FeAstV-positive samples. Phylogenetic analysis based on the complete capsid gene revealed all FeAstV strains were divided into two different groups with a 0.454±0.016 of mean amino acid genetic distance between two groups, suggesting that FeAstVs should be classified into two different genotype species. This study provided the first molecular evidence that FeAstV with considerable genetic diversity was circulating in northeast China, and analyzed genetic variability and classification of FeAstVs for the first time.

Project description:BACKGROUND:Recombination is a relatively common phenomenon in retroviruses. We investigated recombination in Feline Immunodeficiency Virus from naturally-infected New Zealand domestic cats (Felis catus) by sequencing regions of the gag, pol and env genes. RESULTS:The occurrence of intragenic recombination was highest in env, with evidence of recombination in 6.4% (n = 156) of all cats. A further recombinant was identified in each of the gag (n = 48) and pol (n = 91) genes. Comparisons of phylogenetic trees across genes identified cases of incongruence, indicating intergenic recombination. Three (7.7%, n = 39) of these incongruencies were found to be significantly different using the Shimodaira-Hasegawa test.Surprisingly, our phylogenies from the gag and pol genes showed that no New Zealand sequences group with reference subtype C sequences within intrasubtype pairwise distances. Indeed, we find one and two distinct unknown subtype groups in gag and pol, respectively. These observations cause us to speculate that these New Zealand FIV strains have undergone several recombination events between subtype A parent strains and undefined unknown subtype strains, similar to the evolutionary history hypothesised for HIV-1 "subtype E".Endpoint dilution sequencing was used to confirm the consensus sequences of the putative recombinants and unknown subtype groups, providing evidence for the authenticity of these sequences. Endpoint dilution sequencing also resulted in the identification of a dual infection event in the env gene. In addition, an intrahost recombination event between variants of the same subtype in the pol gene was established. This is the first known example of naturally-occurring recombination in a cat with infection of the parent strains. CONCLUSION:Evidence of intragenic recombination in the gag, pol and env regions, and complex intergenic recombination, of FIV from naturally-infected domestic cats in New Zealand was found. Strains of unknown subtype were identified in all three gene regions. These results have implications for the use of the current FIV vaccine in New Zealand.

Project description:The prevalence of fleas and gastrointestinal parasites in free-roaming and domestic cats in central Mexico was evaluated. Three hundred and fifty eight cats captured in the street or brought in by owners to the Animal Control Center Unit, a unit of State Government, from June 2010 to May 2011, were included in the study. All cats were examined for the presence of fleas and gastrointestinal worms. One-hundred and ninety (53%) cats were infested with at least one flea species. Single infestations were observed in 106 (30%) cats and mixed infestations in 84 (23%) cats. Four species of fleas were recovered: Ctenocephalides felis in 53% of the cats, C. canis in 18%, Echidnophaga gallinacea in 7% and Pulex irritans in 1%. One-hundred and sixty three (45%) cats were infected with one or more species of gastrointestinal parasites: 48 (13%) with nematodes, 145 (40%) with cestodes, and one animal presented Moniliformis moniliformis. Prevalences and mean intensity of infection were: Physaloptera praeputialis 7 and 18; T. cati 3 and 2; Ancylostoma tubaeforme 2.5 and 2; Toxascaris leonina 0.5 and 2; Dipylidium caninum 36 and 32; Taenia taeniformis 4 and 3 and Moniliformis moniliformis 0.3 and 106, respectively. There was significant association (P<0.01), between season and ectoparasites load, more fleas were obtained in the summer and autumn than in the winter and spring; however, no statistical difference was observed for endoparasites load (P>0.05). The correlation between the total number of ectoparasites and endoparasites was not significant (r = 0.089, P = 0.094).

Project description:Feline morbillivirus (FeMV), a novel virus from the family of Paramyxoviridae, was first identified in stray cat populations. The objectives of the current study were to (i) determine the molecular prevalence of FeMV in Malaysia; (ii) identify risk factors associated with FeMV infection; and (iii) characterise any FeMV isolates by phylogenetic analyses. Molecular analysis utilising nested RT-PCR assay targeting the L gene of FeMV performed on either urine, blood and/or kidney samples collected from 208 cats in this study revealed 82 (39.4%) positive cats. FeMV-positive samples were obtained from 63/124 (50.8%) urine and 20/25 (80.0%) kidneys while all blood samples were negative for FeMV. In addition, from the 35 cats that had more than one type of samples collected (blood and urine; blood and kidney; blood, urine and kidney), only one cat had FeMV RNA in the urine and kidney samples. Risk factors such as gender, presence of kidney-associated symptoms and cat source were also investigated. Male cats had a higher risk (p = 0.031) of FeMV infection than females. In addition, no significant association (p = 0.083) was observed between the presence of kidney-associated symptoms with FeMV status. From the 82 positive samples, FeMV RNA was detected from 48/82 (58.5%) pet cats and 34/126 (27.0%) shelter cats (p < 0.0001). Partial L and N gene sequencing of the RT-PCR-positive samples showed 85-99% identity to the published FeMV sequences and it was significantly different from all other morbilliviruses. A phylogenetic analysis of the identified Malaysian FeMVs was performed with isolates from Japan, Thailand and China. Molecular characterisation revealed high relatedness of the Malaysian isolates with other Asian FeMVs, indicating that the virus had been circulating only within the region. Therefore, this study confirmed the existence of FeMV among domestic cats in Malaysia. The findings suggest further characterisation of the local isolates, including the whole genome sequencing and that studies at determining the direct consequences of FeMV infection in domestic cats are needed.

Project description:Three known feline hemoplasmas are Mycoplsama haemofelis, 'CandidatusMycoplasma haemominutum' and 'CandidatusMycoplasma turicensis'. They are described as cause of feline infectious anemia in domestic and wild felids. Other blood parasites or blood-related pathogens like concurrent retroviral infections may deteriorate the clinical condition and severity of anemia. The aims of this study were molecular characterization and phylogenetic analysis of hemoplasmas in domestic cats in Iran for the first time. Blood samples were collected from 185 healthy and diseased domestic cats. Blood smears were prepared and hematological parameters were measured to determine possible anemia. Using 16S rRNA gene universal and species specific polymerase chain reactions with the following sequencing, 47 (25.40%) of cats were hemoplasma positive. Also, 17.02%, 72.50% and 40.40% of total positive samples were M.haemofelis, 'Ca. M. haemominutum' and 'Ca. M. turicensis' infected, respectively. 10 (21.20%) of hemoplasma positive cats had anemic blood profiles (HCT < 24.00%). All M. haemofelis infected cases were included. Partial 16S rRNA gene phylogenetic analysis revealed a high identity between the hemoplasma species found in this study and domestic cat sequences existing in GenBank. Phylogenetic analysis revealed 94.00% to 100% sequence identity between sequences of this study and existing sequences in Genbank. All hemoplasma isolates in this study were grouped within a single clade and additionally subdivided into two groups; haemofelis group including M.haemofelis and 'Ca. M. turicensis' and haemominutum group including 'Ca. M. haemominutum'.
