Project description:By adapting microfabrication techniques originally developed in the microelectronics industry novel devices for drug delivery, tissue engineering and biosensing have been engineered for in vivo use. Implant microfabrication uses a broad range of techniques including photolithography, and micromachining to create devices with features ranging from 0.1 to hundreds of microns with high aspect ratios and precise features. Microfabrication offers device feature scale that is relevant to the tissues and cells to which they are applied, as well as offering ease of en masse fabrication, small device size, and facile incorporation of integrated circuit technology. Utilizing these methods, drug delivery applications have been developed for in vivo use through many delivery routes including intravenous, oral, and transdermal. Additionally, novel microfabricated tissue engineering approaches propose therapies for the cardiovascular, orthopedic, and ocular systems, among others. Biosensing devices have been designed to detect a variety of analytes and conditions in vivo through both enzymatic-electrochemical reactions and sensor displacement through mechanical loading. Overall, the impact of microfabricated devices has had an impact over a broad range of therapies and tissues. This review addresses many of these devices and highlights their fabrication as well as discusses materials relevant to microfabrication techniques.

Project description:Hydrogels containing colorimetric nanoparticles have been used for ion sensing, glucose detection, and microbial metabolite analyses. In particular, the rapid chemical reaction owing to both the hydrogel form of water retention and the sensitive color change of nanoparticles enables the rapid detection of target substances. Despite this advantage, the poor dispersibility of nanoparticles and the mechanical strength of nanoparticle-hydrogel complexes have limited their application. In this study, we demonstrate a milliliter agarose gel containing homogeneously synthesized polyaniline nanoparticles (PAni-NPs), referred to as PAni-NP-hydrogel complexes (PNHCs). To fabricate the optimal PNHC, we tested various pH solvents based on distilled water and phosphate-buffered saline and studied the colorimetric response of the PNHC with thickness. The colorimetric response of the prepared PNHC to the changes in the pH of the solution demonstrated excellent linearity, suggesting the possibility of using PNHC as a pH sensor. In addition, it was verified that the PNHC could detect minute pH changes caused by the cancer cell metabolites without cytotoxicity. Furthermore, the PNHC can be stably maintained outside water for approximately 12 h without deformation, indicating that it can be used as a disposable patch-type wearable biosensing platform.

Project description:Hydrogels commonly used in tissue engineering are mechanically soft, thus often display structural weakness. Herein, we introduce a strategy for enhancing the structural integrity and fracture toughness of cell-laden hydrogels by incorporating a three-dimensional (3D) microfabricated scaffold as a structural element. A digital micromirror device projection printing (DMD-PP) system, a rapid prototyping technology which employs a layer-by-layer stereolithographic approach, was utilized to efficiently fabricate 3D scaffolds made from photocrosslinkable poly(ethylene glycol) diacrylate (PEGDA). The scaffold was incorporated into a photocrosslinkable gelatin hydrogel by placing it in a pre-gel solution, and inducing in situ hydrogel formation. The resulting scaffold-reinforced hydrogels demonstrated significant increase in ultimate stress and provided structural support for weak hydrogels. In addition, the scaffold did not affect the rigidity of hydrogels, as it was not involved in the crosslinking reaction to form the hydrogel. Therefore, the presented approach could avoid inadvertent and undesired changes in the hydrogel rigidity which is a known regulator of cellular activities. Furthermore, the biocompatibility of scaffold-reinforced hydrogels was confirmed by evaluating the viability and proliferation of encapsulated fibroblasts. Overall, the strategy of incorporating 3D scaffolds into hydrogels as structural reinforcements presented in this study will be highly useful for enhancing the mechanical toughness of hydrogels for various tissue engineering applications.

Project description:Natural materials exhibit anisotropy with variations in soluble factors, cell distribution, and matrix properties. The ability to recreate the heterogeneity of the natural materials is a major challenge for investigating cell-material interactions and for developing biomimetic materials. Here we present a generic fluidic approach using convection and alternating flow to rapidly generate multi-centimeter gradients of biomolecules, polymers, beads and cells and cross-gradients of two species in a microchannel. Accompanying theoretical estimates and simulations of gradient growth provide design criteria over a range of material properties. A poly(ethylene-glycol) hydrogel gradient, a porous collagen gradient and a composite material with a hyaluronic acid/gelatin cross-gradient were generated with continuous variations in material properties and in their ability to regulate cellular response. This simple yet generic fluidic platform should prove useful for creating anisotropic biomimetic materials and high-throughput platforms for investigating cell-microenvironment interactions.

Project description:Laser speckle imaging (LSI) is an interferometric technique that provides information about the relative speed of moving scatterers in a sample. Photothermal LSI overcomes limitations in depth resolution faced by conventional LSI by incorporating an excitation pulse to target absorption by hemoglobin within the vascular network. Here we present results from experiments designed to determine the mechanism by which photothermal LSI decreases speckle contrast. We measured the impact of mechanical properties on speckle contrast, as well as the spatiotemporal temperature dynamics and bulk convective motion occurring during photothermal LSI. Our collective data strongly support the hypothesis that photothermal LSI achieves a transient reduction in speckle contrast due to bulk motion associated with thermally driven convection. The ability of photothermal LSI to image structures below a scattering medium may have important preclinical and clinical applications.

Project description:Marangoni effect plays an important role in many industrial applications where a surface tension gradient induces fluid flow, e.g., the cleaning process of silicon wafers and the welding process of melted metal. Surface tension gradient can also be caused by a spatially varying temperature field which, in the absence of gravity, is solely responsible for driving a large scale convective flow. NASA STDC-1 (Surface Tension Driven Convection) experiments performed on USML-1 Spacelab missions in 1992 were designed to study thermocapillary flows in microgravity. Since then these experiments have become a benchmark in thermocapillary studies in the absence of gravity. However, interpretation of results of the original STDC-1 experiments remains challenging due to the low resolution of the available data. Analysis of the velocity field in those experiments was limited to a single tracking method without systematic and comparative studies. In the present study, we utilize multiple state-of-the-art Particle Image Velocimetry and Particle Tracking Velocimetry tools to extract the flow field from NASA STDCE-1 videos and compare the experimental data to the numerical results from COMSOL Multiphysics® v5.6. Finally, we discuss how our findings of temperature-driven Marangoni flow in the microgravity setting can improve future experiments and analysis.

Project description:Microfluidic quartz crystal microbalances (QCM) can be used as powerful biosensors that not only allow quantifying a target analyte, but also provide kinetic information about the surface processes of binding and release. Nevertheless, their practical use as point-of-care devices is restricted by a limit of detection (LoD) of some ng/cm². It prohibits the measurement of small molecules in low concentrations within the initial sample. Here, two concepts based on superparamagnetic particles are presented that allow enhancing the LoD of a QCM. First, a particle-enhanced C-reactive protein (CRP) measurement on a QCM is shown. The signal response could be increased by a factor of up to five by utilizing the particles for mass amplification. Further, a scheme for sample pre-preparation utilizing convective up-concentration involving magnetic bead manipulation is investigated. These experiments are carried out with a glass device that is fabricated by utilizing a femtosecond laser. Operation regimes for the magnetic manipulation of particles within the microfluidic channel with integrated pole pieces that are activated by external permanent magnets are described. Finally, the potential combination of the concepts of mass amplification and up-concentration within an integrated lab-on-a chip device is discussed.

Project description:The surfaces of Jupiter and Saturn have magnificent vortical storms that help shape the dynamic nature of their atmospheres. Land- and space-based observational campaigns have established several properties of these vortices, with some being similar between the two planets, while others are different. Shallow-water hydrodynamics, where the vortices are treated as shallow weather-layer phenomenon, is commonly evoked for explaining their formation and properties. Here, we report novel formation mechanisms for vortices where the primary driving mechanism is the deep planetary convection occurring in these planets. Using three-dimensional simulations of turbulent convection in rotating spherical shells, we propose two ideas: (i) Rotating turbulent convection generates deep axially aligned cyclones and anticyclones; (ii) a deep planetary dynamo acts to promote additional anticyclones, some as large as Jupiter's Great Red Spot, in an overlying atmospheric layer. We use these ideas to interpret several observational properties of vortices on Jupiter and Saturn.

Project description:Synthetic materials that are capable of autonomous healing upon damage are being developed at a rapid pace because of their many potential applications. Despite these advancements, achieving self-healing in permanently cross-linked hydrogels has remained elusive because of the presence of water and irreversible cross-links. Here, we demonstrate that permanently cross-linked hydrogels can be engineered to exhibit self-healing in an aqueous environment. We achieve this feature by arming the hydrogel network with flexible-pendant side chains carrying an optimal balance of hydrophilic and hydrophobic moieties that allows the side chains to mediate hydrogen bonds across the hydrogel interfaces with minimal steric hindrance and hydrophobic collapse. The self-healing reported here is rapid, occurring within seconds of the insertion of a crack into the hydrogel or juxtaposition of two separate hydrogel pieces. The healing is reversible and can be switched on and off via changes in pH, allowing external control over the healing process. Moreover, the hydrogels can sustain multiple cycles of healing and separation without compromising their mechanical properties and healing kinetics. Beyond revealing how secondary interactions could be harnessed to introduce new functions to chemically cross-linked polymeric systems, we also demonstrate various potential applications of such easy-to-synthesize, smart, self-healing hydrogels.

Project description:Hydrogels are of great interest in the field of biosensing for their good biocompatibility, plasticity, and capability of providing 3D scaffolds. Nevertheless, the application of hydrogels has not been linked with broad surface biosensing systems yet. To overcome the limitations, here for the first time, surface-immobilized pure DNA hydrogels were synthesized using a surficial primer-induced strategy and adopted for biosensing applications. The DNA hydrogel 3D scaffold is successfully constructed on a transparent ITO electrode, which facilitates both colourimetric and electrochemical measurements. Results show that the hydrogel is able to wrap enzymes solidly and exhibits favourable stability under different conditions. Owing to the free diffusion of the micromolecular targets throughout the hydrogel, while isolating the enzymes from the macromolecular interferences outside the hydrogel, the direct colourimetric and electrochemical detection of hydrogen peroxide and bilirubin in serum is achieved. The detection limit of hydrogen peroxide in serum is 22 nM by colourimetric analysis and 13 nM by electrochemical measurement. The detection limit of bilirubin is 32 nM, a favourable limit that could be used in jaundice diagnosis. In addition, the enzyme@hydrogel can be easily regenerated and the catalytic activity is retained for a few cycles, thus allowing the recycling of the hydrogel-based biosensing system. The successful integration of DNA hydrogels with surface biosensing systems will greatly expand the applications of hydrogels for diagnostic and environmental monitoring purposes.
