Fluorescence Switchable Conjugated Polymer Microdisk Arrays by Cosolvent Vapor Annealing.

ABSTRACT: Depositing minute light emitters into a regular array is a basic but essential technique in display technology. However, conventional lithographic methodologies involve multistep and energy-consuming processes. Here, we develop a facile method in which organic and polymeric fluorescent dyes spontaneously aggregate to form a patterned microarray. We find that a thin film of fluorescent ?-conjugated polymer transforms into micrometer-sized aggregates when exposed to binary organic vapor at ambient temperature. The arrayed microaggregates can be formed over the whole substrate surface when using a quartz substrate that is prepatterned with regular hydrophilic boxes and hydrophobic grids. The resultant microarray is applicable to optical memories and displays when photoswitchable fluorophores are doped into the polymer matrix.

The Effect of Solvent Vapor Annealing on Drug-Loaded Electrospun Polymer Fibers.

Project description:Electrospinning has emerged as a powerful strategy to develop controlled release drug delivery systems but the effects of post-fabrication solvent vapor annealing on drug-loaded electrospun fibers have not been explored to date. In this work, electrospun poly(ԑ-caprolactone) (PCL) fibers loaded with the hydrophobic small-molecule spironolactone (SPL) were explored. Immediately after fabrication, the fibers are smooth and cylindrical. However, during storage the PCL crystallinity in the fibers is observed to increase, demonstrating a lack of stability. When freshly-prepared fibers are annealed with acetone vapor, the amorphous PCL chains recrystallize, resulting in the fiber surfaces becoming wrinkled and yielding shish-kebab like structures. This effect does not arise after the fibers have been aged. SPL is found to be amorphously dispersed in the PCL matrix both immediately after electrospinning and after annealing. In vitro dissolution studies revealed that while the fresh fibers show a rapid burst of SPL release, after annealing more extended release profiles are observed. Both the rate and extent of release can be varied through changing the annealing time. Further, the annealed formulations are shown to be stable upon storage.

Morphological, Chemical, and Electronic Changes of the Conjugated Polymer PTB7 with Thermal Annealing.

Project description:There is considerable interest in improving the performance of organic optoelectronic devices through processing techniques. Here, we study the effect of high-temperature annealing on the properties of the semiconducting polymer PTB7 and PTB7:fullerene blends, of interest as efficient organic photovoltaic (OPV) devices. Annealing to moderate temperature improves the PTB7 morphology and optoelectronic properties. High-temperature annealing also improves morphology but results in poorer optoelectronic properties. This is a result of side chain cleavage that creates by-products that act as trap states, increasing electronic disorder and decreasing mobility. We further observe changes to the PTB7 chemical structure after thermal cleavage that are similar to those following solar irradiation. This implies that side chain cleavage is an important mechanism in device photodegradation, which is a major "burn-in" loss mechanism in OPV. These results lend insight into side chain cleavage as a method of improving optoelectronic properties and suggest strategies for improvement in device photostability.

Conjugated polymer-based fluorescence turn-on sensor for nitric oxide.

Project description:A turn-on fluorescent sensor for NO (g) in solution was synthesized using a bipyridyl-substituted poly(p-phenylene vinylene) derivative (CP1) as the sensory scaffold. The action of NO (g) upon the CP1-Cu(II) complex reduces it to the CP1-Cu(I) complex with a concomitant 2.8-fold increase in emission intensity. The reagent is selective for NO (g) versus other biological reactive nitrogen species, except for nitroxyl, and has a detection sensitivity limit of 6.3 nM. [structure: see text]

Fluorescence Modulation of Conjugated Polymer Nanoparticles Embedded in Poly(N-Isopropylacrylamide) Hydrogel.

Project description:A series of conjugated polymers (CPs) emitting red, green, and blue (RGB) fluorescence were synthesized via the Suzuki coupling polymerization. Polymer dots (Pdots) were fabricated by the reprecipitation method from corresponding CPs, in which the Pdot surface was functionalized to have an allyl moiety. The CP backbones were based on the phenylene group, causing the Pdots to show identical ultraviolet-visible absorption at 350 nm, indicating that the same excitation wavelength could be used. The Pdots were covalently embedded in poly(N-isopropylacrylamide) (PNIPAM) hydrogel for further use as a thermoresponsive moiety in the polymer hydrogel. The polymer hydrogel with RGB emission colors could provide thermally reversible fluorescence changes. The size of the hydrogel varied with temperature change because of the PNIPAM's shrinking and swelling. The swollen and contracted conformations of the Pdot-embedded PNIPAM enabled on-and-off fluorescence, respectively. Fluorescence modulation with 20 to 80% of the hydrogel was possible via thermoreversibility. The fluorescent hydrogel could be a new fluorescence-tuning hybrid material that changes with temperature.

Bio-inspired polymer array vapor sensor with dual signals of fluorescence intensity and wavelength shift.

Project description:Organic vapor sensors based on polymer owing to their tunable molecular structures and designable functions have attracted considerable research interest. However, detecting multiple organic vapors with high accuracy and a low detection limit is still challenging. Herein, inspired by the mammalian olfactory recognition system, organic vapor sensors based on one-dimensional microfilament array structures with a wide range of sensing gases are demonstrated. By introducing aggregation-induced emission (AIE) molecules, sensors possess dual-optical sensing mechanisms of variation in fluorescence intensity and wavelength. By virtue of the synergistic effects of dual signals, superb accuracy and incredibly low detection limit are achieved for identifying analytes. In particular, the polymer/AIE microfilament array can detect acetone vapor down to 0.03% of saturated vapor pressure. In the saturated vapor of acetone, the fluorescence intensity of the sensor arrays was reduced by 53.7%, while the fluorescence wavelength was red-shifted by 21 nm. Combined with the principal component analysis (PCA) algorithm, the polymer/AIE molecular sensor arrays accomplished the classification and identification of acetone, ethanol, methylene chloride, toluene, and benzene. This bioinspired approach with dual sensing signals may broaden practical applications to high-performance gas sensors for precise molecular detection.

Enhanced Two-Photon Fluorescence and Fluorescence Imaging of Novel Probe for Calcium Ion by Self-Assembly with Conjugated Polymer.

Project description:The calcium ion (Ca2+) isa highly versatile intracellular signal messenger regulating many different cellular functions. It is important to design probes with good fluorescence and two-photon (TP) active cross-sections (??) to explore the concentration distribution of Ca2+. In this manuscript, a novel TP fluorescence calcium probe (BAPTAVP) with positive charges, based on the classical Ca2+ indicator of BAPTA (1,2-bis(2-aminophenoxy)-ethane-N,N,N',N'-tetra acetic acid), and a conjugated polymer (PCBMB) with negative charges were designed and synthesized. The results from transmission electron microscopy (TEM), atomic force microscopy (AFM), dynamic light scattering (DLS), and the zeta potential (ZP) showed that nanoparticles were obtained by the self-assembly of PCBMB and BAPTAVP. Moreover, the fluorescence properties of BAPTAVP were effectively improved by fluorescence resonance energy transfer (FRET) with PCBMB and attenuating the intramolecular charge transfer (ICT) after the addition of Ca2+. The quantum yield and ?? of PCBMB-BAPTAVP increased by about four and six times in comparison to those of BAPTAVP, respectively. The TP fluorescence imaging experiments indicated that the PCBMB-BAPTAVP system could effectively detect Ca2+ in living cells with high sensitivity.

Cyclic voltammograms at coplanar and shallow recessed microdisk electrode arrays: guidelines for design and experiment.

Project description:Although microdisk electrode arrays (MEAs) have been extensively used for more than three decades, the existing rules do not provide an unambiguous formula for the calculation of the minimum interelectrode distance (d) necessary for steady-state current response. With the aim of formulating generally applicable guidelines for design and experiment with MEAs, cyclic voltammograms were simulated for coplanar and shallow recessed microdisk electrode arrays with various interelectrode distances and dimensionless scan rates (V). The dimensionless scan rate (V) is a function of the radius (a) of the individual electrodes in the array, the diffusion coefficient (D) of the analyte, and the potential scan rate (v). The cyclic voltammograms at microdisk electrode arrays are grouped into five categories corresponding to the contributions of linear and radial diffusion to the overall responses. These categories are illustrated in a zone diagram based on the effect of V and d on the shape of cyclic voltammograms. The zone diagram reveals the minimum d and a cluster of linked d and V values that are incident to sigmoidal wave responses. For shallow recessed microdisk electrode arrays, the zones representing hemispherical diffusion are larger than that for coplanar arrays. The minimum d necessary for hemispherical diffusion becomes smaller as recess depth increases. With the zone diagram, one can predict the type of the cyclic voltammograms that can be expected for different microelectrode array geometries and experimental conditions. The fitting between simulation and experimental data validates our conclusions.

In vivo ultrasound-switchable fluorescence imaging.

Project description:The conventional fluorescence imaging has limited spatial resolution in centimeter-deep tissue because of the tissue's high scattering property. Ultrasound-switchable fluorescence (USF) imaging, a new imaging technique, was recently proposed to realize high-resolution fluorescence imaging in centimeter-deep tissue. However, in vivo USF imaging has not been achieved so far because of the lack of stable near-infrared contrast agents in a biological environment and the lack of data about their biodistributions. In this study, for the first time, we achieved in vivo USF imaging successfully in mice with high resolution. USF imaging in porcine heart tissue and mouse breast tumor via local injections were studied and demonstrated. In vivo and ex vivo USF imaging of the mouse spleen via intravenous injections was also successfully achieved. The results showed that the USF contrast agent adopted in this study was very stable in a biological environment, and it was mainly accumulated into the spleen of the mice. By comparing the results of CT imaging and the results of USF imaging, the accuracy of USF imaging was proved.

Near infrared conjugated polymer nanoparticles (CPN™) for tracking cells using fluorescence and optoacoustic imaging.

Project description:Tracking the biodistribution of cell therapies is crucial for understanding their safety and efficacy. Optical imaging techniques are particularly useful for tracking cells due to their clinical translatability and potential for intra-operative use to validate cell delivery. However, there is a lack of appropriate optical probes for cell tracking. The only FDA-approved material for clinical use is indocyanine green (ICG). ICG can be used for both fluorescence and photoacoustic imaging, but is prone to photodegradation, and at higher concentrations, undergoes quenching and can adversely affect cell health. We have developed novel near-infrared imaging probes comprising conjugated polymer nanoparticles (CPNs™) that can be fine-tuned to absorb and emit light at specific wavelengths. To compare the performance of the CPNs™ with ICG for in vivo cell tracking, labelled mesenchymal stromal cells (MSCs) were injected subcutaneously in mice and detected using fluorescence imaging (FI) and a form of photoacoustic imaging called multispectral optoacoustic tomography (MSOT). MSCs labelled with either ICG or CPN™ 770 could be detected with FI, but only CPN™ 770-labelled MSCs could be detected with MSOT. These results show that CPNs™ show great promise for tracking cells in vivo using optical imaging techniques, and for some applications, out-perform ICG.
