Project description:Bradysia impatiens (Johannsen, 1912) is an important pest of various mushrooms and plant seedlings, while it also damages plants as vectors transmitting pathogens. The complete B. impatiens mitogenome is 16,479 bp in length, with an A + T content of 80.6%. The mitogenome had three overlapped regions; the longest overlap is 7 bp. Phylogenetic analysis reveals that B. impatiens is closely related to Bradysia amoena. The study provided a basis for future studies of species divergence in Bradysia.

Project description:A new species of black fungus gnat from Canada, Peyerimhoffiajaschhoforum sp. n., is presented with a description, illustrations, biotope information and a brief discussion of the placement and concept of the genus Peyerimhoffia. P.jaschhoforum is characterized by a unique gonostylar structure where the apex is hollowed but not enclosed and contains a mass of mega setae housed within. P.jaschhoforum was reared from decomposing jack pine (Pinusbanksiana Lamb.) deadwood using both in-situ and ex-situ photoeclectors. We documented three additional specimens originating from Fennoscandia that resemble P.jaschhoforum but differ based on a broader tegmen, placement of setigerous papillae behind the tegmen and the fused intercoxal area. Based on this, these specimens are assigned to a new subspecies, Peyerimhoffiajaschhoforumfennoscandica ssp. n.

Project description:A new species of the genus Ctenosciara Tuomikoski, 1960 is here described based upon a single specimen, obtained from collectings in the garden at Museum Alexander Koenig in Bonn. Ctenosciara alexanderkoenigi sp. n. differs from all other congeneric European species by its striking coloration and distinct male genitalia. However, DNA barcoding reveals associations with two specimens from New Zealand. Therefore a recent migration of Ctenosciara species from the Australasian Region, the likely center of origin of the genus, is discussed. A key to the European species of Ctenosciara is provided. Barcoding results reveale that Ctenosciara exigua is not clearly distinguished from Ctenosciara hyalipennis by its COI sequence (both share the same BIN BOLD:AAH3983) and that its species status may be questionable.

Project description:Micronucleoli are among the structures composing the peculiar scenario of the nucleolus in salivary gland nuclei of dipterans representative of Sciaridae. Micronucleolar bodies contain ribosomal DNA and RNA, are transcriptionally active and may appear free in the nucleoplasm or associated with specific chromosome regions in salivary gland nuclei. This report deals with an extreme case of nucleolar fragmentation/dispersion detected in the salivary gland of Schwenkfeldina sp. Such a phenomenon in this species was found to be restricted to cell types undergoing polyteny and seems to be differentially controlled according to the cell type. Furthermore, transcriptional activity was detected in virtually all the micronucleolar bodies generated in the salivary gland. The relative proportion of the rDNA in polytene and diploid tissues showed that rDNA under-replication did not occur in polytene nuclei suggesting that the nucleolar and concomitant rDNA dispersion in Schwenkfeldina sp. may reflect a previously hypothesised process in order to counterbalance the rDNA loss due to the under-replication. The chromosomal distribution of epigenetic markers for the heterochromatin agreed with early cytological observations in this species suggesting that heterochromatin is spread throughout the chromosome length of Schwenkfeldina sp. A comparison made with results from another sciarid species argues for a role played by the heterochromatin in the establishment of the rDNA topology in polytene nuclei of Sciaridae.

Project description:In order to investigate the effects of artificial diets on the population growth of root maggot Bradysia impatiens, its population growth parameters were assayed on eight artificial diets (Diet 1, D2, D3, D4, D5, D6, D7, and D8). Results showed that developmental duration from egg to pupa was successfully completed on all eight artificial diets. However, the egg to pupal duration was shortest, while the survival rate of four insect stages was lowest when B. impatiens was reared on D1. When B. impatiens was reared on D7 and D8, the survival rate, female longevity, and female oviposition were higher than those reared on other diets. When B. impatiens was reared on D7, the intrinsic rate of increase (rm = 0.19/d), net reproductive rate (R0 = 39.88 offspring per individual), and finite rate of increase (λ = 1.21/d) were higher for its population growth with shorter generation time (T = 19.49 d) and doubling time (Dt = 3.67 d). The findings indicate that the D7 artificial diet is more appropriate for the biological parameters of B. impatiens and can be used an indoor breeding food for population expansion as well as further research. We propose that vitamin C supplement added to the D7 is critical for the improvement of the B. impatiens growth.

Project description:The chive gnat, Bradysia odoriphaga, is a notorious pest of Allium species in China. Colour trapping is an established method for monitoring and control of Bradysia species. In order to clarify the effect of colour preference of B. odoriphaga for the perched substrate, multiple-choice tests were used to assess the response of the chive gnat to different colour hues and brightness levels under different intensities of white illumination and two spectrally different illuminations. Given the choice among four colours differing in hue under different intensities of white illumination and two spectrally different illuminations, chive gnat adults significant preferred the black substrate, a lesser preference to brown and green substrates, and the least preference to orange substrate irrespective of illumination. Given the choice among four levels of brightness under the same illumination conditions as those in the previous experiment (different intensities of white illumination and two spectrally different illuminations), chive gnats preferred black substrate over dark grey, light grey and white substrates. Meanwhile, both virgin and mated adults significantly preferred black over other colour hues and brightness. Based on our results, we conclude that the chive gnat adults significantly prefer black substrates irrespective of colour hues and brightness. This behaviour does not alter with ambient light condition changes. No difference observed between choices of female and male adults. Our results provide new insight for understanding the colour choice behaviour in chive gnat and pave a way to improve monitoring and control of chive gnats and management.

Project description:Seven species of Keilbachia Mohrig are recognized, and among them, two new species, Keilbachia subacumina Wu & Zhang, sp. n. and Keilbachia fengyangensis Wu & Zhang, sp. n. are described and illustrated. Five species, Keilbachia orthonema, Keilbachia flagrispina, Keilbachia demssia, Keilbachia oligonema and Keilbachia acumina are reported for the first time from China. A key to the 15 Chinese species of this genus is also provided.

Project description:Throughout China, the dipteran pest Bradysia odoriphaga significantly reduces Chinese chive production; therefore, identifying conditions that influence its growth and development is crucial for developing ecological regulation strategies. In this study, different non-lethal high temperatures and treatment durations were used to stress the third-instar larvae of B. odoriphaga, and the effects of this treatment on their growth and offspring were recorded and analyzed. The results showed that the average larval mortality increased with increased temperature and prolonged exposure times. After stress treatment at 40 °C for 2 h, 100% of larvae died within 5 days, which was not significantly different from the 5-day average larval mortality (90.66%) after stress at 37 °C for 4 h, but significantly higher than the 5-day average larval mortality (72.00%) after stress at 40 °C for 1 h. After 5 days, all still-living larvae could pupate, and there was no significant difference in average pupal period after pupation. However, the eclosion rate of subsequent pupae decreased with increased temperature and prolonged exposure times, and were only 43.00% and 42.73% after larvae were stressed at 37 °C for 4 h and 40 °C for 1 h, respectively. After eclosion into adults, there was no significant difference in the lifespan of unmated female adults, while the lifespan of unmated male adults was significantly reduced to 1.67 d and 2 d after larvae were stressed at 37 °C for 4 h and 40 °C for 1 h, respectively. However, there was no significant difference in male and female adult longevity after mating. There was no significant difference in oviposition or egg hatchability. This indicates that non-lethal high temperature at 37 °C for 4 h can hinder development and allow control of B. odoriphaga. There is great potential for non-lethal high temperature to be applied in the field to control agricultural pests.

Project description:The dance fly Oreogeton sp. belongs to the subfamily Oreogetoninae of Empididae. The mitogenome of Oreogeton sp. (GenBank accession number: MK639348) was sequenced, the first representative of the mitogenome of the subfamily. The nearly complete mitogenome is 15,718?bp totally, consisting of 13 protein-coding genes, two rRNAs, and 22 transfer RNAs. All genes have the similar locations and strands with that of other published species of Empididae. The nucleotide composition biases toward A and T, which together made up 77.2?of the entirety. Bayesian inference analysis strongly supported the monophyly of Empidoidea, Empididae, and Dolichopodidae. This result also suggested that Oreogetoninae was assigned to the sister group to the clade that consists of Trichopezinae and Clinocerinae, and then Empidinae is the sister to the clade that contains Oreogetoninae, Trichopezinae, and Clinocerinae.
