The complete mitochondrial genome of Anomaloglossus baeobatrachus (Amphibia: Anura: Aromobatidae).

ABSTRACT: The complete mitogenome of the rocket frog Anomaloglossus baeobatrachus was sequenced using a shotgun approach on an Illumina HiSeq 2500 platform (Illumina Inc., San Diego, CA), providing the first mitogenome for this genus. The genome was 17,572?bp long and presents the typical organization found in other neobatrachian anurans. A phylogenetic analysis including A. baeobatrachus and all other available mitogenomes of Hyloidea provided relationships in accordance with previous phylogenetic studies.

Project description:The complete mitochondrial genome was determined from a Microhyla butleri, Microhylidae, Microhyla, which was collected from Shenzhen, China. The mitogenome was 16?714?bp in length, containing 13 protein-coding genes, two rRNA genes, 22 tRNA genes and a control region (D-loop). The base composition was 28.7% A, 29.5% T, 27.2% C and 14.6% G. The gene order and contents were identical to most amphibian mitogenome. Except ND1 gene beginning with GTG and COI gene beginning with ATA, all other protein-coding genes began with ATG as start codon. Six protein-coding genes (ND1, COII, ATP6, COIII, ND3 and ND4) ended with incomplete stop codon T. The 22 tRNA genes with the size ranging from 65?bp to 74?bp were interspersed along the whole genome. The D-loop region containing tandem repetition was 1334?bp in length and heavily biased to A?+?T nucleotides.

It is a circular molecule of 17?702 bp in size and consists of 13 protein-coding genes, 23 tRNA genes, two rRNA genes and a control region (D-loop). The base composition on light strand was 28.0% A, 32.2% T, 24.9% C and 14.9% G. Compared with most other vertebrates, this mitogenome appear a tandem duplication of tRNA-Met gene. This study will facilitate the further research of the population genetics of this species and systematic study of the genus Oreolalax.

Complete mitochondrial genome sequence of the Asiatic Toad Bufo Gargarizans (Amphibia, Anura, Bufonidae).

Project description:The Asiatic toad Bufo gargarizans belongs to Bufonidae. This species is known from the Russian Far East, central, northern and north-eastern China, the Democratic People's Republic of Korea and Japan. In this study, the complete mitochondrial genome of B. gargarizans was sequenced. The mitogenome was 17,407?bp in length, consisting of 13 protein-coding genes, 22 transfer RNA (tRNA) genes, two ribosomal RNA (rRNA) genes, and a non-coding control region. As in other vertebrates, most mitochondrial genes are encoded on the heavy strand, except for ND6 and eight tRNA genes which are encoded on the light strand. The overall base composition of the B. gargarizans is 28.9% A, 28.2% T, 27.5% C, and 15.3% G. Phylogenetic analysis showed B. gargarizans was closely related to B. bankorensis and B. tibetanus. The complete mitogenome of B. gargarizans can provide an important data for the studies on phylogenetic relationship and population genetics to further explore the taxonomic status of this species.

Project description:We determined the complete mitochondrial genome of Rana uenoi (Anura: Ranidae) for the first time. The whole sequences were 17,370 bp and included 13 protein-coding genes, 2 ribosomal RNA genes, and 22 transfer RNA genes. The gene arrangement was completely identical to those observed from other Ranidae species. We used 11 protein-coding genes to examine the phylogenetic placement of this species in the genus Rana. Rana dybowskii was the closest sister species to R. uenoi. The clade of R. uenoi and R. dybowskii formed a cluster with Rana huarensis, which had a sister relationship with the group of Rana amurensis, Rana coreana, and Rana kunyuensis.

The first complete mitochondrial genome sequence of Nanorana parkeri and Nanorana ventripunctata (Amphibia: Anura: Dicroglossidae), with related phylogenetic analyses.

Project description:Members of the Nanorana genus (family Dicroglossidae) are often referred to as excellent model species with which to study amphibian adaptations to extreme environments and also as excellent keystone taxa for providing insights into the evolution of the Dicroglossidae. However, a complete mitochondrial genome is currently only available for Nanorana pleskei. Thus, we analyzed the complete mitochondrial genomes of Nanorana parkeri and Nanorana ventripunctata to investigate their evolutionary relationships within Nanorana and their phylogenetic position in the family Dicroglossidae. Our results showed that the genomes of N. parkeri (17,837 bp) and N. ventripunctata (18,373 bp) encode 13 protein-coding genes (PCGs), two ribosomal RNA genes, 23 transfer RNA (tRNA) genes, and a noncoding control region. Overall sequences and genome structure of the two species showed high degree of similarity with N. pleskei, although the motif structures and repeat sequences of the putative control region showed clear differences among these three Nanorana species. In addition, a tandem repeat of the tRNA-Met gene was found located between the tRNA-Gln and ND2 genes. On both the 5' and 3'-sides, the control region possessed distinct repeat regions; however, the CSB-2 motif was not found in N. pleskei. Based on the nucleotide sequences of 13 PCGs, our phylogenetic analyses, using Bayesian inference and maximum-likelihood methods, illustrate the taxonomic status of Nanorana with robust support showing that N. ventripunctata and N. pleskei are more closely related than they are to N. parkeri. In conclusion, our analyses provide a more robust and reliable perspective on the evolutionary history of Dicroglossidae than earlier analyses, which used only a single species (N. pleskei).

We described 17,448?bp of D. suweonensis mtDNA, which had a shorter D-loop than other closely related species. The variation in nucleotide composition was similar to that of Hyla tsinlingensis but was larger than the one of its sister clade, D. japonicus.

Project description:The complete mitochondrial genome of Salamandra salamandra was reconstructed using bycatch sequences from an RNAseq library generated from muscle tissue. This study shows the potential of using data originally produced for transcriptome assembly to additionally generate complete mitochondrial genomes. The resulting mitogenome was circular, consisted of 16,331?bp and followed the standard vertebrate gene order. Subsequent analysis of the current mitochondrial genome sequence for the species as listed on GenBank highlighted a species misidentification.

The total length of mitogenome is 17,189?bp, and contains 13 protein-coding genes, 22 tRNA genes, two ribosome RNA genes, and one D-loop region. All gene conformations were consistent with the gene arrangement in vertebrates. Most of the genes of B. zamdaensis were distributed on the H-strand, except for the ND6 subunit gene and eight tRNA genes which were encoded on the L-strand. Phylogenetic analysis of B. zamdaensis and 12 other closely amphibian species was reconstructed using Maximum-likelihood methods. The sequences of B. zamdaensis were clustered in genus Bufotes. The phylogenetic analyses based on these mitogenomes presented here will be useful to further insights on the evolutionary relationships of Bufonidae.

As the first report of the mitogenome sequence from the genus Oreolalax, it will provide fundamental data for further research of phylogeny and biogeography with this genus.

This mitogenome with 17,046 bp long contains 13 protein-coding genes (PCGs), 22 transfer RNA genes, two ribosomal RNA genes, and one non-coding regions. The overall AT content of 58.6%. The data using Bayesian phylogenetic inference supports the close relationship with Q. spinosa from the genus Quasipaa. Our results will be helpful for detail study on phylogenetic relationships of the related taxa.
