Characterization of the complete mitochondrial genome of Sogatella kolophon (Hemiptera: Delphacidae).

ABSTRACT: In this study, we sequenced and annotated the complete mitochondrial genome of Sogatella kolophon (Kirkaldy) (Hemiptera: Delphacidae). The mitogenome was 16,708 bp in length, containing 13 protein-coding genes (PCGs), 22 tRNA genes, two rRNA genes and a control region. All PCGs started with ATN, except for atp6 and nad5, which used noncanonical codon GTG, respectively. All tRNAs could fold into typical clover-leaf secondary structures, with the exception of two trnS genes, lacking the dihydrouridine (DHU) stem.

The Complete Mitochondrial Genome of Ugyops sp. (Hemiptera: Delphacidae).

Project description:The complete mitochondrial genome (mitogenome) of Ugyops sp. (Hemiptera: Delphacidae) was sequenced, making it the first determined mitogenome from the subfamily Asiracinae, the basal clade of the family Delphacidae. The mitogenome was 15,259 bp in length with A + T content of 77.65% and contained 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), two ribosomal RNA genes (rRNAs), and a control region. The gene order was identical with that of the ancestral insect. The nucleotide composition analysis indicated that the whole mitogenome was strongly A-skewed (0.288) and highly C-skewed (-0.270). For PCGs on the J-strand, the AT skew was positive, and the GC skew was negative. All PCGs started with canonical ATN codons, except for cox1 and nad5, which used CTG and GTG as start codon, respectively. All tRNAs could fold into typical cloverleaf secondary structures, with the exception of trnS1 (AGN), in which the dihydrouridine arm was reduced to a simple loop. The control region included a poly-T stretch downstream of the small rRNA gene (rrnS), a subregion of higher A + T content and tandemly repeated sequence near trnI. The mitogenome of Ugyops sp. could be very helpful in exploring the diversity and evolution of mitogenomes in Delphacidae.

A New Mitochondrial Genome of Sogatella furcifera (Horvath) (Hemiptera: Delphacidae) and Mitogenome-Wide Investigation on Polymorphisms.

Project description:White-backed planthopper (WBPH), Sogatella furcifera (Horváth), is one of the major sap-sucking rice pests in East Asia. We have determined a new complete mitochondrial genome of WBPH collected in the Korean peninsula using NGS technology. Its length and GC percentages are 16,613 bp and 23.8%, respectively. We observed one polymorphic site, a non-synonymous change, in the COX3 gene with confirmation heteroplasmy phenomenon within individuals of WBPH by PCR amplification and Sanger sequencing, the first report in this species. In addition, this heteroplasmy was not observed in wild WBPH populations, suggesting that it may be uncommon in fields. We analyzed single nucleotide polymorphisms, insertion, and deletions, and simple sequence repeats among the three WBPH mitogenomes from Korea and China and found diverse intraspecific variations, which could be potential candidates for developing markers to distinguish geographical populations. Phylogenetic analysis of 32 mitogenomes of Delphacidae including the three WBPH mitogenomes suggested that Delphacinae seems to be monophyletic and Sogatella species including WBPH are clearly formed as one clade. In the future, it is expected that complete mitogenomes of individuals of geographically dispersed WBPH populations will be used for further population genetic studies to understand the migration pathway of WBPH.

Molecular characterization of Vitellogenin-like1 gene in Sogatella furcifera (Hemiptera: Delphacidae), and its function on reproduction.

Project description:In this study, a vitellogenin like1 gene (SfVg-like1) in Sogatella furcifera was identified. The open reading frame (ORF) encoded 1,321 amino acid sequence. Structure analysis reveals that the amino acid sequence of SfVg-like1 has 3 conserved LPD_N, DUF1943 and VWFD domains. Phylogenetic analyses showed that SfVg-like1 was clustered in the same branch with the Vg-like1 of Nilaparvata lugens (100% bootstrap value) compared with other Hemiptera insects Vgs associated with vitellogenesis. Temporo-spatial expression analyses showed that SfVg-like1 expressed during all stages, and in both genders. The relative expression levels of SfVg-like1 mRNA were higher in adults than in nymph developmental stages. The knockdown of SfVg-like1 gene resulted in the inhibition of the ovarian development in female adults, whereas the morphology of the testis in male adults was not been affected. The silence of SfVg-like1 could decrease the relative expression levels of target of rapamycin (SfTOR, GenBank MW193765) and vitellogenin (SfVg, GenBank MH271114) genes significantly in female adults. However, the knockdown of SfTOR or SfVg genes in female adults did not affect the transcript level of SfVg-like1. Therefore, it demonstrated that SfVg-like1 might locate on the upstream signaling pathways of SfTOR and SfVg. These results demonstrate that SfVg-like1 is essential for S. furcifera reproduction, and it could be the potential target for the control of this pest.

Knockdown of Methoprene-Tolerant Arrests Ovarian Development in the Sogatella furcifera (Hemiptera: Delphacidae).

Project description:Juvenile hormone (JH) is responsible for repressing larval metamorphosis and inducing vitellogenesis and egg production in insects. Methoprene-tolerant (Met) is known to be an intracellular receptor and transducer of JH. We examined the role of Met in ovarian development in the rice pest Sogatella furcifera (Horváth). We first cloned and sequenced S. furcifera Met (SfMet). The SfMet protein belongs to the basic helix-loop-helix/Per-Arnt-Sim (bHLH-PAS) family with a bHLH domain and two PAS domains (PAS-A and PAS-B). SfMet was expressed in all developmental stages and tissues but was most highly expressed in the ovaries of adult females. Furthermore, RNA interference (RNAi) mediated silencing of SfMet substantially reduced the expression of SfVg, decreased yolk protein deposition and blocked oocyte maturation and ovarian development. These results demonstrate that SfMet plays a key role in female reproduction in S. furcifera and suggest that targeting this gene could be an effective way of controlling this pest.

Species-specific markers for Nilaparvata lugens and Sogatella furcifera (Hemiptera: Delphacidae) based on mitochondrial cytochrome oxidase I.

Project description:Brown planthopper (BPH), Nilaparvata lugens (Stål) and white-backed planthopper (WBPH), Sogatella furcifera (Horváth) are the most destructive sucking insect pests of rice in all rice growing parts of the world. For their accurate identification at early stages, we have developed two species-specific markers (SNL4F and SNL4R for BPH; SNF2F and SNF2R for WBPH) based on mitochondrial cytochrome oxidase I (COI) for their easy detection using Polymerase Chain Reaction (PCR). The markers were developed based on nucleotide differences in COI gene and were subjected to various tests based on PCR-based gel images. The designed primers were cross-checked with five other species, which confirmed their specificity. The primers were also found to be efficient in identification of their respective species (BPH and WBPH) in all the individuals sampled from different regions of India. The lowest detection sensitivity of both the primers was up to 1 ng/µl DNA after testing them through a series of varied DNA concentrations. The species-specific primers developed in this study will help in easy and rapid identification of BPH and WBPH in all the stages of their development and in turn facilitate their timely management.Supplementary informationThe online version contains supplementary material available at 10.1007/s13205-023-03693-x.

Identification and RNAi-Based Functional Analysis of Four Chitin Deacetylase Genes in Sogatella furcifera (Hemiptera: Delphacidae).

Project description:Chitin deacetylases (CDAs) are chitin-degrading enzymes that play a key role in insect molting. In this study, we identified and characterized four full-length cDNAs of CDAs from Sogatella furcifera (Horváth). Developmental expression showed that SfCDA1 and SfCDA2 were expressed at all nymph developmental stages, SfCDA3 and SfCDA4 were mainly expressed in the third-instar to fifth-instar nymph stages, whereas tissue-specific analyses indicated that four CDA genes were mainly high expressed in the integument and head during the fifth-instar nymph. RNA interference (RNAi) results revealed that SfCDA1, SfCDA2, and SfCDA4 are associated with molting defect and high mortality with nymph-adult molting. Furthermore, transcripts of chitin synthase 1 variants (SfCHS1, SfCHS1a, and SfCHS1b) were significantly downregulated and causing significant changes in the expression levels of trehalases (TRE1 and TRE2) in the SfCDA1, SfCDA2, and SfCDA4 dsRNA treatment groups. By contrast, no significant phenotypic characteristics were observed after dsSfCDA3 injection. Taken together, our results suggest that SfCDA1, SfCDA2, and SfCDA4 play a vital role in nymph-adult transition, and these genes could regulate chitin biosynthesis expression levels.

Asymmetric Spread of SRBSDV between Rice and Corn Plants by the Vector Sogatella furcifera (Hemiptera: Delphacidae).

Project description:Plant viruses are mostly transmitted by sucking insects via their piercing behaviors, which may differ due to host plant species and their developmental stages. We characterized the transmission of a fijivirus, southern rice black-streaked dwarf virus (SRBSDV), by the planthopper vector Sogatella furcifera Horváth (Hemiptera: Delphacidae), between rice and corn plants of varying developmental stages. SRBSDV was transmitted from infected rice to uninfected corn plants as efficiently as its transmission between rice plants, while was acquired by S. furcifera nymphs at a much lower rate from infected corn plants than from infected rice plants. We also recorded a high mortality of S. furcifera nymphs on corn plants. It is evident that young stages of both the virus donor and recipient plants added to the transmission efficiency of SRBSDV from rice to corn plants. Feeding behaviors of the vector recorded by electrical penetration graph showed that phloem sap ingestion, the behavioral event that is linked with plant virus acquisition, was impaired on corn plants, which accounts for the high mortality of and low virus acquisition by S. furcifera nymphs on corn plants. Our results reveal an asymmetric spread of SRBSDV between its two host plants and the underlying behavioral mechanism, which is of significance for assessing SRBSDV transmission risks and field epidemiology, and for developing integrated management approaches for SRBSDV disease.

Project description:A 16,359 bp mitochondrial genome of Laodelphax striatellus collected in a southern part of Korean peninsula was completed and their intraspecies variations were compared with Korean and Chinese L. striatellus mitogenomes. The circular mitogenome of L. striatellus contains 13 protein-coding genes, two rRNA genes, 22 tRNAs, and a single large non-coding region of 1,972 bp. The base composition was AT-biased (77.3%). It has only one single nucleotide polymorphism (SNP) in AT-rich region compared to other Korean mitogenome, but total 41 and 141 SNPs and 118 and 166 insertions and deletions, respectively, compared to two Chinese mitogenomes, suggesting the possibility of tracing migration based on geographic genetic diversity.

The circular mitogenome of L. striatellus is 16,359 bp long including 13 protein-coding genes, two ribosomal RNA genes, 22 transfer RNAs, and a single large non-coding region of 1,972 bp. The base composition was AT-biased (77.3%). In comparison of the two Chinese L. striatellus mitogenomes with Korean mitogenome, total 140 and 40 single nucleotide polymorphisms and 166 and 118 insertions and deletions were identified, presenting intra-species variations based on geographical distribution.
