Natural depolymerization of waste poly(ethylene terephthalate) by neutral hydrolysis in marine water.

ABSTRACT: Polyethylene terephthalate (PET) is one of the most widely used materials for food packaging and fishing nets. After use it become waste and, due to poor collection, most will be found floating in marine waters. This paper presents the results of a study of PET depolymerization by hydrolysis. We observed that marine water is a perfect reactant because it contains a multitude of metal ions that act as catalysts. A first-order kinetic model was developed and experimental data fitted to it. An activation energy of 73.5 kJ/mole and a pre-exponential factor of 5.33?×?10⁷ h^-1 were obtained. Considering that the global ocean is a huge batch reactor operating under isothermal conditions, the solution of the mathematical model shows that in tropical regions only 72 years is needed for total and only 4.5 years for 50% PET conversion.

Synergistic chemo-enzymatic hydrolysis of poly(ethylene terephthalate) from textile waste.

Project description:Due to the rising global environment protection awareness, recycling strategies that comply with the circular economy principles are needed. Polyesters are among the most used materials in the textile industry; therefore, achieving a complete poly(ethylene terephthalate) (PET) hydrolysis in an environmentally friendly way is a current challenge. In this work, a chemo-enzymatic treatment was developed to recover the PET building blocks, namely terephthalic acid (TA) and ethylene glycol. To monitor the monomer and oligomer content in solid samples, a Fourier-transformed Raman method was successfully developed. A shift of the free carboxylic groups (1632 cm-1 ) of TA into the deprotonated state (1604 and 1398 cm-1 ) was observed and bands at 1728 and 1398 cm-1 were used to assess purity of TA after the chemo-enzymatic PET hydrolysis. The chemical treatment, performed under neutral conditions (T = 250 °C, P = 40 bar), led to conversion of PET into 85% TA and small oligomers. The latter were hydrolysed in a second step using the Humicola insolens cutinase (HiC) yielding 97% pure TA, therefore comparable with the commercial synthesis-grade TA (98%).

Microbial synthesis of vanillin from waste poly(ethylene terephthalate).

Project description:Poly(ethylene terephthalate) (PET) is an abundant and extremely useful material, with widespread applications across society. However, there is an urgent need to develop technologies to valorise post-consumer PET waste to tackle plastic pollution and move towards a circular economy. Whilst PET degradation and recycling technologies have been reported, examples focus on repurposing the resultant monomers to produce more PET or other second-generation materials. Herein, we report a novel pathway in engineered Escherichia coli for the direct upcycling of PET derived monomer terephthalic acid into the value-added small molecule vanillin, a flavour compound ubiquitous in the food and cosmetic industries, and an important bulk chemical. After process optimisation, 79% conversion to vanillin from TA was achieved, a 157-fold improvement over our initial conditions. Parameters such as temperature, cell permeabilisation and in situ product removal were key to maximising vanillin titres. Finally, we demonstrate the conversion of post-consumer PET from a plastic bottle into vanillin by coupling the pathway with enzyme-catalysed PET hydrolysis. This work demonstrates the first biological upcycling of post-consumer plastic waste into vanillin using an engineered microorganism.

Fungal and enzymatic bio-depolymerization of waste post-consumer poly(ethylene terephthalate) (PET) bottles using <i>Penicillium</i> species.

Project description:Poly(ethylene terephthalate) (PET) is a petroleum-based plastic that is massively produced and used worldwide. A promising PET recycling process to circumvent petroleum feedstock consumption and help to reduce environmental pollution is microbial or enzymatic biodegradation of post-consumer (PC) PET packages to its monomers-terephthalic acid (TPA) and ethylene glycol (EG)-or to key intermediates in PET synthesis-such as mono- and bis-(2-hydroxyethyl) terephthalate (MHET and BHET). Two species of filamentous fungi previously characterized as lipase producers (Penicillium restrictum and P. simplicissimum) were evaluated in submerged fermentation for induction of lipase production by two inducers (BHET and amorphous PET), and for biodegradation of two substrates (BHET and PC-PET). BHET induced lipase production in P. simplicissimum, achieving a peak of 606.4 U/L at 49 h (12.38 U/L.h), representing an almost twofold increase in comparison to the highest peak in the control (without inducers). Microbial biodegradation by P. simplicissimum after 28 days led to a 3.09% mass loss on PC-PET fragments. In contrast, enzymatic PC-PET depolymerization by cell-free filtrates from a P. simplicissimum culture resulted in low concentrations of BHET, MHET and TPA (up to 9.51 µmol/L), suggesting that there are mechanisms at the organism level that enhance biodegradation. Enzymatic BHET hydrolysis revealed that P. simplicissimum extracellular enzymes catalyze the release of MHET as the predominant product. Our results show that P. simplicissimum is a promising biodegrader of PC-PET that can be further explored for monomer recovery in the context of feedstock recycling processes.

Hydrocarbon-associated substrates reveal promising fungi for poly (ethylene terephthalate) (PET) depolymerization.

Project description:Recalcitrant characteristics and insolubility in water make the disposal of synthetic polymers a great environmental problem to be faced by modern society. Strategies towards the recycling of post-consumer polymers, like poly (ethylene terephthalate, PET) degradation/depolymerization have been studied but still need improvement. To contribute with this purpose, 100 fungal strains from hydrocarbon-associated environments were screened for lipase and esterase activities by plate assays and high-throughput screening (HTS), using short- and long-chain fluorogenic probes. Nine isolates were selected for their outstanding hydrolytic activity, comprising the genera Microsphaeropsis, Mucor, Trichoderma, Westerdykella, and Pycnidiophora. Two strains of Microsphaeropsis arundinis were able to convert 2-3% of PET nanoparticle into terephthalic acid, and when cultured with two kinds of commercial PET bottle fragments, they also promoted weight loss, surface and chemical changes, increased lipase and esterase activities, and led to PET depolymerization with release of terephthalic acid at concentrations above 20.0 ppm and other oligomers over 0.6 ppm. The results corroborate that hydrocarbon-associated areas are important source of microorganisms for application in environmental technologies, and the sources investigated revealed important strains with potential for PET depolymerization.

Glycolysis of Poly(Ethylene Terephthalate) Using Biomass-Waste Derived Recyclable Heterogeneous Catalyst.

Project description:Plastic production has increased by almost 200-fold annually from 2 million metric tons per year in 1950s to 359 million metric tons in 2018. With this rapidly increasing production, plastic pollution has become one of the most demanding environmental issues and tremendous efforts have been initiated by the research community for its disposal. In this present study, we reported for the first time, a biomass-waste-derived heterogeneous catalyst prepared from waste orange peel for the depolymerisation of poly(ethylene terephthalate) (PET) to its monomer, bis(2-hydroxyethyl terephthalate) (BHET). The prepared orange peel ash (OPA) catalyst was well-characterised using techniques such as IR, inductively coupled plasma (ICP)-OES (Optical Emission Spectrometry), XRD, X-ray fluorescence (XRF), SEM, energy-dispersive X-ray spectroscopy (EDX), TEM, BET (Brunauer-Emmett-Teller) and TGA. The catalyst was found to be composed of basic sites, high surface area, and a notable type-IV N2 adsorption-desorption isotherm indicating the mesoporous nature of the catalyst, which might have eventually enhanced the rate of the reaction as well as the yield of the product. The catalyst completely depolymerises PET within 90 min, producing 79% of recrystallised BHET. The ability of reusing the catalysts for 5 consecutive runs without significant depreciation in the catalytic activity and its eco- and environmental-friendliness endorses this protocol as a greener route for PET recycling.

Sourcing thermotolerant poly(ethylene terephthalate) hydrolase scaffolds from natural diversity.

Project description:Enzymatic deconstruction of poly(ethylene terephthalate) (PET) is under intense investigation, given the ability of hydrolase enzymes to depolymerize PET to its constituent monomers near the polymer glass transition temperature. To date, reported PET hydrolases have been sourced from a relatively narrow sequence space. Here, we identify additional PET-active biocatalysts from natural diversity by using bioinformatics and machine learning to mine 74 putative thermotolerant PET hydrolases. We successfully express, purify, and assay 51 enzymes from seven distinct phylogenetic groups; observing PET hydrolysis activity on amorphous PET film from 37 enzymes in reactions spanning pH from 4.5-9.0 and temperatures from 30-70 °C. We conduct PET hydrolysis time-course reactions with the best-performing enzymes, where we observe differences in substrate selectivity as function of PET morphology. We employed X-ray crystallography and AlphaFold to examine the enzyme architectures of all 74 candidates, revealing protein folds and accessory domains not previously associated with PET deconstruction. Overall, this study expands the number and diversity of thermotolerant scaffolds for enzymatic PET deconstruction.

Improved polymerization and depolymerization kinetics of poly(ethylene terephthalate) by co-polymerization with 2,5-furandicarboxylic acid.

Project description:Poly(ethylene terephthalate) (PET), known for its clarity, food safety, toughness, and barrier properties, is a preferred polymer for rigid packaging applications. PET is also one of the most recycled polymers worldwide. In light of climate change, significant efforts are underway to improve the carbon footprint of PET by synthesizing it from bio-based feedstocks. Often times, specific applications demand PET to be copolymerized with other monomers. This work focuses on copolymerization of PET with a bio-based co-monomer, 2,5-furandicarboxylic acid (FDCA) to produce the copolyester (PETF). We report the multifunction of FDCA to influence the esterification reaction kinetics and the depolymerization kinetics (via alkaline hydrolysis) of the copolyester PETF. NMR spectroscopy and titrimetric studies revealed that copolymerization of PET with different levels of FDCA improved the esterification reaction kinetics by enhancing the solubility of monomers. During the alkaline hydrolysis, the presence of FDCA units in the backbone almost doubled the PET conversion and monomer yield. Based on these findings, it is demonstrated that the FDCA facilitates the esterification, as well as depolymerization of PET, and potentially enables reduction of reaction temperatures or shortened reaction times to improve the carbon footprint of the PET synthesis and depolymerization process.

A tough and sustainable fiber-forming material from lignin and waste poly(ethylene terephthalate).

Project description:In this report we describe repurposing of recycled polyesters as a matrix for lignin-a biorefinery coproduct that is used as a solid fuel and needs to find higher value-to make sustainable high-performance thermoplastic materials. Brittle lignin oligomers, isolated from plant biomass, require a low-melting host polymer matrix to form a rigid and tough renewable material. We demonstrate controlled lignin dispersion and interfacial interactions in softened recycled polyethylene terephthalate (PET) using a simple solvent-free, melt-blending technique. To avoid lignin degradation and devolatilization during melt processing, it was thermally treated. Tall oil fatty acid was used to enable PET processability at low enough temperature to accommodate lignin without charring. Chemical analysis reveals reduction of aliphatic hydroxyl content from 2 mmol g-1 to 1.63 mmol g-1 and an increase of total phenolic hydroxyl moieties from 5.86 to 6.64 mmol g-1 and cleavage of β-O-4 ether linkages due to thermal treatment. Structural transformation of lignin macromolecules during heat treatment was further confirmed by an increase in molar mass and improved thermal stability. Interfacial interactions between lignin and PET were assessed from mechanical properties and thermal analyses. Thermal treatment not only helps to improve the stability of lignin but also slightly reduces the size of the dispersed lignin domains via favored interfacial interactions with the PET matrix. These methods improve mechanical properties of the material. Further, incorporation of lignin in the plasticized PET matrix increases the ductility in the blended products. The method we discuss here utilizes industrial wastes and co-products, and it does not require solvent or toxic chemicals during the reactive extrusion process that yields complete conversion to products.

Parallel DNA Synthesis on Poly(ethylene terephthalate).

Project description:The fabrication of DNA arrays directly on aminolyzed sheets of poly(ethylene terephthalate) (PET) is described. Array surfaces typically employ bifunctional linkers or layers of covalently attached polymers to provide substrate hydroxy groups as synthesis attachment points. An amine treatment is used here to expose hydroxy groups on films of PET. These hydroxy groups can then be used to couple phosphoramidites and initiate the array synthesis without further functionalization steps. Arrays fabricated on these substrates with a maskless array synthesizer are tolerant of the high number of chemical exposure steps required to synthesize relatively long oligonucleotides. The results might be of the greatest use to the synthetic biology community, for whom a flexible and robust substrate could enable new strategies to enhance the throughput of oligonucleotide synthesis.

Progressing Ultragreen, Energy-Efficient Biobased Depolymerization of Poly(ethylene terephthalate) via Microwave-Assisted Green Deep Eutectic Solvent and Enzymatic Treatment.

Project description:Effective interfacing of energy-efficient and biobased technologies presents an all-green route to achieving continuous circular production, utilization, and reproduction of plastics. Here, we show combined ultragreen chemical and biocatalytic depolymerization of polyethylene terephthalate (PET) using deep eutectic solvent (DES)-based low-energy microwave (MW) treatment followed by enzymatic hydrolysis. DESs are emerging as attractive sustainable catalysts due to their low toxicity, biodegradability, and unique biological compatibility. A green DES with triplet composition of choline chloride, glycerol, and urea was selected for PET depolymerization under MW irradiation without the use of additional depolymerization agents. Treatment conditions were studied using Box-Behnken design (BBD) with respect to MW irradiation time, MW power, and volume of DES. Under the optimized conditions of 20 mL DES volume, 260 W MW power, and 3 min MW time, a significant increase in the carbonyl index and PET percentage weight loss was observed. The combined MW-assisted DES depolymerization and enzymatic hydrolysis of the treated PET residue using LCC variant ICCG resulted in a total monomer conversion of ≈16% (w/w) in the form of terephthalic acid, mono-(2-hydroxyethyl) terephthalate, and bis-(2-hydroxyethyl) terephthalate. Such high monomer conversion in comparison to enzymatically hydrolyzed virgin PET (1.56% (w/w)) could be attributed to the recognized depolymerization effect of the selected DES MW treatment process. Hence, MW-assisted DES technology proved itself as an efficient process for boosting the biodepolymerization of PET in an ultrafast and eco-friendly manner.
