ABSTRACT: The high costs of N fertilizers in the coffee production emphasizes the need to optimize fertilization practices and improve nitrogen use efficiency. Urea is widespread in nature, characterizing itself as a significant source of nitrogen for the growth and development of several organisms. Thus, the characterization of genes involved in urea transport in coffee plants is an important research topic for the sustainable production of this valuable cash crop. In the current study, we evaluated the expression of the DUR3 gene under abiotic and biotic stresses in coffee plants. Here, we show that the expression of a high-affinity urea transporter gene (CaDUR3) was up-regulated by N starvation in leaves and roots of two out of three C. arabica cultivars examined. Moreover, the CaDUR3 gene was differentially expressed in coffee plants under different abiotic and biotic stresses. In plants of cv. IAPAR59, CaDUR3 showed an increased expression in leaves after exposure to water deficit and heat stress, while it was downregulated in plants under salinity. Upon infection with H. vastatrix (coffee rust), the CaDUR3 was markedly up-regulated at the beginning of the infection process in the disease susceptible Catuaí Vermelho 99 in comparison with the resistant cultivar. These results indicate that besides urea acquisition and N-remobilization, CaDUR3 gene may be closely involved in the response to various stresses.

Project description:BackgroundBy 2050, the world population will increase to 10 billion which urged global demand for food production to double. Plant disease and land drought will make the situation more dire, and safer and environment-friendly materials are thus considered as a new countermeasure. The rice blast fungus, Magnaporthe oryzae, causes one of the most destructive diseases of cultivated rice worldwide that seriously threatens rice production. Unfortunately, traditional breeding nor chemical approaches along control it well. Nowadays, nanotechnology stands as a new weapon against these mounting challenges and silica nanoparticles (SiO2 NPs) have been considered as potential new safer agrochemicals recently but the systematically studies remain limited, especially in rice.ResultsSalicylic acid (SA) is a key plant hormone essential for establishing plant resistance to several pathogens and its further affected a special form of induced resistance, the systemic acquired resistance (SAR), which considered as an important aspect of plant innate immunity from the locally induced disease resistance to the whole plant. Here we showed that SiO2 NPs could stimulate plant immunity to protect rice against M. oryzae through foliar treatment that significantly decreased disease severity by nearly 70% within an appropriate concentration range. Excessive concentration of foliar treatment led to disordered intake and abnormal SA responsive genes expressions which weaken the plant resistance and even aggravated the disease. Importantly, this SA-dependent fungal resistance could achieve better results with root treatment through a SAR manner with no phytotoxicity since the orderly and moderate absorption. What's more, root treatment with SiO2 NPs could also promote root development which was better to deal with drought.ConclusionsTaken together, our findings not only revealed SiO2 NPs as a potential effective and safe strategy to protect rice against biotic and abiotic stresses, but also identify root treatment for the appropriate application method since it seems not causing negative effects and even have promotion on root development.

Project description:BackgroundSmall RNAs (19-24 nt) are key regulators of gene expression that guide both transcriptional and post-transcriptional silencing mechanisms in eukaryotes. Current studies have demonstrated that microRNAs (miRNAs) act in several plant pathways associated with tissue proliferation, differentiation, and development and in response to abiotic and biotic stresses. In order to identify new miRNAs in soybean and to verify those that are possibly water deficit and rust-stress regulated, eight libraries of small RNAs were constructed and submitted to Solexa sequencing.ResultsThe libraries were developed from drought-sensitive and tolerant seedlings and rust-susceptible and resistant soybeans with or without stressors. Sequencing the library and subsequent analyses detected 256 miRNAs. From this total, we identified 24 families of novel miRNAs that had not been reported before, six families of conserved miRNAs that exist in other plants species, and 22 families previously reported in soybean. We also observed the presence of several isomiRNAs during our analyses. To validate novel miRNAs, we performed RT-qPCR across the eight different libraries. Among the 11 miRNAs analyzed, all showed different expression profiles during biotic and abiotic stresses to soybean. The majority of miRNAs were up-regulated during water deficit stress in the sensitive plants. However, for the tolerant genotype, most of the miRNAs were down regulated. The pattern of miRNAs expression was also different for the distinct genotypes submitted to the pathogen stress. Most miRNAs were down regulated during the fungus infection in the susceptible genotype; however, in the resistant genotype, most miRNAs did not vary during rust attack. A prediction of the putative targets was carried out for conserved and novel miRNAs families.ConclusionsValidation of our results with quantitative RT-qPCR revealed that Solexa sequencing is a powerful tool for miRNA discovery. The identification of differentially expressed plant miRNAs provides molecular evidence for the possible involvement of miRNAs in the process of water deficit- and rust-stress responses.
