Chromium stress induced oxidative burst in Vigna mungo (L.) Hepper: physio-molecular and antioxidative enzymes regulation in cellular homeostasis.

ABSTRACT: Vigna mungo (L.) Hepper commonly known as blackgram is an important legume crop with good quality dietary proteins and vitamins. Low production of blackgram in the chromium rich soil of Odisha is a serious concern against its demand. Chromium (VI) was tested on V. mungo var. B3-8-8 at 100, 150, 200, 250 and 300 µM concentration on growth, anti-oxidative enzymes and chromium content at 15, 30 and 45 d of treatments. Seed germination and growth decreased with increase dose and duration. Cr uptake induced oxidative burst with significant increase of osmolytes was observed in cell at lower doses but failed to adjust homeostasis at higher dose. Increase of GPX and SOD and decrease of CAT was observed as dose dependent. Increased protein content was detected in < 200 µM Cr concentration whereas, significant decrease of protein was noted thereafter. Down regulation of proteins (29.2 kDa and 32.6 kDa) was observed at > 250 µM of Cr. Total Cr uptake was greater in root than in shoot which might be due to poor translocation of heavy metal or detoxification. Thus, blackgram was able to maintain homeostasis at lower concentrations of Cr by activating the cascade of enzymes following cellular detoxification mechanism.

Project description:Black gram [V. mungo (L.) Hepper] is an important legume crop extensively grown in south and south-east Asia, where it is a major source of dietary protein for its predominantly vegetarian population. However, lack of genomic information and markers has become a limitation for genetic improvement of this crop. Here, we report the transcriptome sequencing of the immature seeds of black gram cv. TU94-2, by Illumina paired end sequencing technology to generate transcriptome sequences for gene discovery and genic-SSR marker development. A total of 17.2 million paired-end reads were generated and 48,291 transcript contigs (TCS) were assembled with an average length of 443 bp. Based on sequence similarity search, 33,766 TCS showed significant similarity to known proteins. Among these, only 29,564 TCS were annotated with gene ontology (GO) functional categories. A total number of 138 unique KEGG (Kyoto Encyclopedia of Genes and Genomes) pathways were identified, of which majority of TCS are grouped into purine metabolism (678) followed by pyrimidine metabolism (263). A total of 48,291 TCS were searched for SSRs and 1,840 SSRs were identified in 1,572 TCS with an average frequency of one SSR per 11.9 kb. The tri-nucleotide repeats were most abundant (35%) followed by di-nucleotide repeats (32%). PCR primer pairs were successfully designed for 933 SSR loci. Sequences analyses indicate that about 64.4% and 35.6% of the SSR motifs were present in the coding sequences (CDS) and untranslated regions (UTRs) respectively. Tri-nucleotide repeats (57.3%) were preferentially present in the CDS. The rate of successful amplification and polymorphism were investigated using selected primers among 18 black gram accessions. Genic-SSR markers developed from the Illumina paired end sequencing of black gram immature seed transcriptome will provide a valuable resource for genetic diversity, evolution, linkage mapping, comparative genomics and marker-assisted selection in black gram.

Project description:Multidrug-resistant bacterial infections, helminthiasis, thrombosis, anxiety and insomnia are some of the major global health concerns. Vigna mungo (L.) Hepper (VM) has been used traditionally to treat microbial infection, helminthic disorder, schizophrenia, memory loss, and blood circulatory problem. This research aims to discover antibacterial, anthelmintic, thrombolytic and neuropharmacological effects of the methanol extract of Vigna mungo seeds (MESVM), and also in-silico prediction of relevant lead compounds by molecular docking and ADME/T analysis. The crude extracts and subsequent fractions of MESVM were investigated for antibacterial activity by disc diffusion method, anthelmintic activity by paralysis and death test on earthworms, and thrombolytic activity by in vitro blood clot dissolution test. Open-field test and elevated plus maze test were performed for evaluating anxiolytic activity of the extracts. Using molecular docking, ligand poses of selected VM seeds' phytoconstituents were predicted targeting tubulin, GlcN-6-P synthase, and human tissue plasminogen activator proteins for anthelmintic, antibacterial, and thrombolytic activity, respectively. In the antibacterial activity test, the MESVM at 10000 μg/mL concentration created highest and significant (P < 0.001) zone of inhibition against Staphylococcus aureus (15.42 mm) and Escherichia coli (12 mm) compared with tetracycline. The MESVM exhibited remarkable anthelmintic activity at 50 mg/mL concentration with 35.4 min paralysis time, 75.2 min death time and were closer to the durations of standard drug albendazole. No test extract showed anxiolytic activity. In thrombolytic activity test, all concentrations of MESVM produced clot lytic activity with high significance (P < 0.001) in comparison with the blank. In docking, 2'-hydroxygenistein, cyclokievitone hydrate, and aureol displayed maximum affinity to the target proteins for anthelmintic, antibacterial, and thrombolytic activity, respectively. This research revealed that the MESVM demonstrated potential anthelmintic, antibacterial and thrombolytic effects that confirmed the folkloric uses of VM and the found relevant lead compounds might be further optimized in future drug development.

Project description:Organ size and architecture of plants are important traits affecting crop yield and agronomic practices. An induced mutant, multiple-organ gigantism (MOG), of black gram (Vigna mungo) has been obtained, which shows gigantic leaves, fruit, seed, and architecture (plant height) but lower number of pods per plant. These traits are a pleiotropic effect of a single recessive gene, mog. In this study, we investigated variation of 16 agronomic and adaptive traits in a recombinant inbred line (RIL) population derived from a cross between the MOG mutant (V. mungo var. mungo) and wild black gram (V. mungo var. silvestris) accession TC2210 and identified quantitative trait loci (QTLs) controlling those traits to gain a better understanding of the effect of the mog gene on breeding. The results showed that most of the traits (100-seed weight, leaf size, and plant height) showed moderate narrow-sense heritability (h 2) (45-65%), while pod size and seed length (SDL) showed high h 2 (>75%) and pod dehiscence (shattering), and seed width (SDW) and days to flowering showed low h 2 (<35%). The QTLs for the traits were mapped onto a high-density linkage map developed for the RIL population. Inclusive composite interval mapping identified 42 QTLs in total for the 16 traits with number of QTLs per trait ranging from one to six. Major QTLs for the MOG phenotypes were clustered on linkage group (LG) 6, confirming the pleiotropic effect of the mog gene. Effect of the mog gene/QTL for the MOG phenotypic variations was not high, ranging from about 15% in plant height to 40% in leaf size. For 100-seed weight, which is the most interesting trait, the mog gene/QTL contributed about 30% of the total trait variation and showed an additive effect of only 0.51 g, which is only about 1.5-fold higher than that of the other five QTLs detected for this trait. These results indicated that mog gene expression is highly affected by environment and the effect of the gene toward organ size and plant height is not extraordinarily high. Implications of the findings of this study and exploiting of the MOG mutant in breeding were also discussed.
