Morphological and phylogenetic analyses reveal three new species of Diaporthe from Yunnan, China.

ABSTRACT: Species of Diaporthe have often been reported as plant pathogens, endophytes or saprobes, commonly isolated from a wide range of plant hosts. Sixteen strains isolated from species of ten host genera in Yunnan Province, China, represented three new species of Diaporthe, D. chrysalidocarpi, D. machili and D. pometiae as well as five known species D. arecae, D. hongkongensis, D. middletonii, D. osmanthi and D. pandanicola. Morphological comparisons with known species and DNA-based phylogenies based on the analysis of a multigene (ITS, TUB, TEF, CAL and HIS) dataset support the establishment of the new species. This study reveals that a high species diversity of Diaporthe with wide host ranges occur in tropical rainforest in Yunnan Province, China.

Project description:Three new species of Distoseptispora, viz. D. mengsongensis, D. nabanheensis, and D. sinensis, are described and illustrated from specimens collected on dead branches of unidentified plants in Yunnan Province, China. Phylogenetic analyses of LSU, ITS, and TEF1 sequence data, using maximum-likelihood (ML) and Bayesian inference (BI), reveal the taxonomic placement of D. mengsongensis, D. nabanheensis, and D. sinensis within Distoseptispora. Both morphological observations and molecular phylogenetic analyses supported D. mengsongensis, D. nabanheensis, and D. sinensis as three new taxa. To extend our knowledge of the diversity of Distoseptispora-like taxa, a list of recognized species of Distoseptispora with major morphological features, habitat, host, and locality is also provided.

Phylogenetic analyses and morphological characters reveal two new species of Ganoderma from Yunnan province, China.

Project description:Ganodermadianzhongense sp. nov. and G.esculentum sp. nov. are proposed as two new species based on both phenotypic and genotypic evidences. Ganodermadianzhongense is characterized by the stipitate basidiomata, laccate and oxblood red pileus, gray white pore surface, duplex context and broadly ellipsoid basidiospores (9.0-12.5 × 6.5-9.0 μm) with coarse interwall pillars. Ganodermaesculentum is characterized by its basidiomata with slender stipe, white pore surface, homogeneous pileus context, and slightly truncate, narrow basidiospores (8.0-12.5 × 5.0-8.0 µm). Phylogenetic analyses were carried out based on the internal transcribed spacer (ITS), translation elongation factor 1-α (TEF1-α) and the second subunit of RNA polymerase II (RPB2) sequence data. The illustrations and descriptions for the new taxa are provided.

Project description:Species of Sporocadaceae have often been reported as plant pathogens, endophytes or saprophytes and are commonly isolated from a wide range of plant hosts. The isolated fungi were studied through a complete examination, based on multilocus phylogenies from combined datasets of ITS/tub2/tef1, in conjunction with morphological characteristics. Nine strains were isolated from Ficusmicrocarpa, Ilexchinensis and Schimasuperba in China which represented four species, viz., Monochaetiaschimae sp. nov., Neopestalotiopsishaikouensis sp. nov., Neopestalotiopsispiceana and Pestalotiopsislicualicola. Neopestalotiopsispiceana was a new country record for China and first host record from Ficusmacrocarpa. Pestalotiopsislicualicola was first report from Ilexchinensis in China.

Project description:Four new fungi, viz. Clavulinopsiswumengshanensis (Clavariaceae, Agaricales), Henningsomycesbambusae (Porotheleaceae, Agaricales), Xenasmabisterigmatae, and X.guttulata (Xenasmataceae, Russulales), from Yunnan Province in China, are proposed, based on a combination of morphological features and molecular evidence. Phylogenetic analyses were conducted using a combined dataset of internal transcribed space and nuclear ribosomal RNA large subunit sequences. The ITS+LSU analysis showed that Clavulinopsiswumengshanensis sp. nov. groups with C.aurantiocinnabarina, Henningsomycesbambusae sp. nov. forms a sister group with H.candidus, Xenasmabisterigmatae sp. nov. is clustered with X.rimicola, and X.guttulata sp. nov. is clustered with X.pruinosum. The morphology and multi-gene phylogenetic analyses confirmed the placement of the four new taxa. Clavulinopsiswumengshanensis is distinguished by buff-yellow to straw-yellow basidiomata, clavate to subcylindrical basidia with four sterigmata, and subglobose basidiospores with several guttules (7-8.5 × 6-7.5 µm); Henningsomycesbambusae is characterized by white to cream basidiomata with short cylindric to tubular colonies, cylindrical to subcylindrical basidia with two sterigmata and globose to subglobose basidiospores (6.5-8.5 × 6.5-8.5 µm); Xenasmabisterigmatae can be characterized by its membranaceous and ash-grey basidiomata, clavate to subcylindrical basidia with two sterigmata and ellipsoid to subglobose basidiospores (10-12.5 × 8-10.5 µm); X.guttulata is distinguished by membranaceous and white to cream basidiomata, clavate to subcylindrical basidia with two sterigmata and ellipsoid to narrowly ellipsoid basidiospores (7-9 × 5.5-7.5 µm).

Morphological and molecular analyses reveal two new species of Grifola (Polyporales) from Yunnan, China.

Project description:Species of Grifola are famous edible mushrooms and are deeply loved by consumers around the world. Most species of this genus have been described and recorded in Oceania, Europe and South America, with only Grifolafrondosa being recorded in Asia. In this study, two novel species of Grifola from southwestern China (Asia) are introduced. Macro and micromorphological characters are described. Grifolaedulissp. nov. present medium-size basidiomata with gray to gray-brown lobes upper surface, mostly tibiiform or narrowly clavate, rarely narrowly lageniform or ellipsoid chlamydospores, cuticle hyphae terminal segments slightly enlarged. Grifolasinensissp. nov. has white to grayish white lobes upper surface, mostly ellipsoid, rarely narrowly utriform chlamydospores, and broadly ellipsoid to ellipsoid basidiospores (4.6-7.9 × 3.0-5.9 μm). The two new species are supported by phylogenetic analyses of combined nuclear rDNA internal transcribed spacer ITS1-5.8S-ITS2 rDNA (ITS) and β-tubulin (TUBB). Moreover, the genetic distance between TUBB sequences of those specimen from GenBank was 1.76-1.9%. Thus, the conspecificity relationship of our specimens remains uncertain, and further specimens are required to conclusively confirm its identity.

Over the course of our mycological surveys of plant pathogens from terrestrial plants in Jiangxi Province, China, eight strains isolated from diseased leaves of four host genera represented three new species of Didymella, D. bischofiae sp. nov., D. clerodendri sp. nov., and D. pittospori sp. nov. Phylogenetic analyses of combined ITS, LSU, RPB2, and TUB2 sequence data, using maximum-likelihood (ML) and Bayesian inference (BI), revealed their taxonomic placement within Didymella. Both morphological examinations and molecular phylogenetic analyses supported D. bischofiae, D. clerodendri, and D. pittospori as three new taxa within Didymella. Illustrations and descriptions of these three taxa were provided, along with comparisons with closely related taxa in the genus.

Project description:During an investigation of helicosporous fungi in China, a total of seven helicosporous hyphomycetes were obtained from decaying wood in the southern region of the country. Based on phylogenetic analyses using a combined LSU, ITS, tef1α, and rpb2 sequence matrix, in conjunction with morphological comparisons, these taxa were classified within Tubeufia (Tubeufiaceae, Tubeufiales) and were recognized as three new species, viz. Tubeufiaguttulata, T.hainanensis, and T.muriformis, as well as one new distribution record, viz. T.cocois. Evidence for these new taxa and the new record, descriptions, illustrations, notes, and phylogenetic evidence are provided for the newly collected helicosporous species.

Morphological and Phylogenetic Analyses Reveal Five New Species in Chaetosphaeriaceae.

Project description:Chaetosphaeriaceae is a genera-rich and highly diverse group of fungi with a worldwide distribution in terrestrial and aquatic habitats. Eight fresh collections of Chaetosphaeriaceae were obtained during investigations of hyaline-spored hyphomycetes in China and Thailand. Based on morphological characteristics and phylogenetic analysis of a combined LSU and ITS sequence dataset, Chaetosphaeria obovoidea, Codinaea aseptata, Codinaeella hyalina, Dictyochaeta guizhouensis and Paragaeumannomyces guttulatus were introduced as new species, Codinaea terminalis was reported as new host record, and Codinaea dwaya and Phialosporostilbe scutiformis were introduced as new collections. Phylogenetic analysis in this study revealed that Chaetosphaeria was polyphyletic. Detailed descriptions and illustrations of new taxa and identified species are provided, as well as an updated phylogenetic tree to confirm the placements of these eight new collections.
