ABSTRACT: In complex structures such as flowers, organ-organ interactions are critical for morphogenesis. The corolla plays a central role in attracting pollinators: thus, its proper development is important in nature, agriculture, and horticulture. Although the intraorgan mechanism of corolla development has been studied, the importance of organ-organ interactions during development remains unknown. Here, using corolla mutants of morning glory described approximately 200 years ago, we show that glandular secretory trichomes (GSTs) regulate floral organ interactions needed for corolla morphogenesis. Defects in GST development in perianth organs result in folding of the corolla tube, and release of mechanical stress by sepal removal restores corolla elongation. Computational modeling shows that the folding occurs because of buckling caused by mechanical stress from friction at the distal side of the corolla. Our results suggest a novel function of GSTs in regulating the physical interaction of floral organs for macroscopic morphogenesis of the corolla.

Project description:BackgroundThe Convolvulaceae is a large family containing species exhibiting a range of breeding systems and pollinated by diverse animal taxa. We studied the pollination ecology of 15 Convolvulaceae species, representing seven genera (Argyreia Lour., Camonea Raf., Evolvulus L., Hewittia Wight & Arn., Ipomoea L., Merremia Dennst. ex Endl., and Operculina Silva Manso), in northeastern Thailand, a family that is highly diverse yet understudied in the paleotropics. Specifically, we studied their compatibility systems and degrees of pollinator dependency using pollination experiments, examined pollinator composition and visitation rates using video observation, and determined if there is an association between pollinator visitation rates and degree of pollinator dependence.ResultsOur results showed that most species are self-compatible, but the degree of pollinator dependence varies. Six species were found to be highly dependent on pollinators, as two are self-incompatible and four are self-compatible but had reduced seed set when pollinators were excluded, possibly due to herkogamy. Seven species showed low dependence on pollinators and seed set remained high when pollinators were excluded. Pollinator dependence was inconclusive for two species as seed set was low in all pollination treatments. We also found an association between pollinator visitation rates and degree of pollinator dependence. Specifically, species exhibiting high pollinator dependence received frequent visits from pollinators, while species exhibiting low pollinator dependence either received frequent visits from pollinators (and received high amounts of xenogamous pollen) or infrequent visits from pollinators (and received significantly lower amounts of xenogamous pollen). Most of our study species were primarily visited by bees (e.g., Lasioglossum, Amegilla, Apis, and meliponines), with the exception of one night-blooming species that was visited primarily by crepuscular butterflies and hawkmoths.ConclusionsThe cumulative findings of this study demonstrate how pollinator dependence is influenced by breeding system, and suggest that pollinator visitation is consistently high for species exhibiting high pollinator dependence but varies across species exhibiting low pollinator dependence. Our findings are also important for assessing the conservation risks of paleotropical Convolvulaceae.

Project description:A common evolutionary trend in highly selfing plants is the evolution of the "selfing syndrome", in which traits associated with pollinator attraction are lost or greatly reduced. Limited information is available on whether these trait reductions are favored by natural selection or result from reduced purifying selection coupled with genetic drift. This study attempted to distinguish between these two possibilities for the evolutionary loss of floral pigmentation in the highly selfing species Ipomoea lacunosa. This study also tested the hypothesis that loss of floral pigmentation is caused by downregulation or loss of function in a tissue-specific anthocyanin transcription factor, as has been found in other plants. F2 individuals of a cross between white and pigmented individuals revealed segregation at two epistatically acting loci: one affecting pigmentation in both corolla throat and limbs (Anl1) and one affecting limb pigmentation (Anl2). Individuals that are homozygous for the "white" allele at Anl1 have white throats and limbs regardless of genotype at Anl2. In individuals with pigmented throats, homozygosity of the "white" allele at Anl2 produces white limbs. Flower color variation at Anl1 cosegregates with an R2R3-Myb anthocyanin transcription factor, which is down-regulated in white-flowers but not in pigmented flowers. Differential expression of the two alleles of this gene indicates that down regulation is caused by a cis-regulatory change. Finally, allele-frequency differences at Anl1 were substantially and significantly greater than differences in allele frequencies at four microsatellite loci. These results are consistent with the hypotheses that the identified R2R3-Myb gene corresponds to Anl1 and that evolutionary loss of pigmentation in I. lacunosa was caused by selection. They are also consistent with previous studies demonstrating that loss of floral pigmentation is usually caused by down-regulation or functional inactivation of an R2R3-Myb gene.

Project description:The aquaporin (AQP) family, also called water channels or major intrinsic proteins, facilitate water transport. AQPs also transport low-molecular-weight solutes, including boric acid, glycerol, urea, and ammonia. Since plants are sessile, water homeostasis is crucial. Therefore, plants have developed diverse AQP variants at higher expression levels than animals. For example, 35 and 33 AQPs have been identified in Arabidopsis and rice, respectively. In the present study, we identified AQPs in morning glory (Ipomoea nil), which has been widely used as a model plant in research on flowering and floral morphology. The importance of AQPs in the opening of morning glory flowers has been reported. In the morning glory genome, 44 AQPs were identified, and their characteristics were analyzed. A phylogenetic analysis revealed five AQP subfamilies in morning glory: plasma membrane-intrinsic proteins (PIPs), tonoplast-intrinsic proteins (TIPs), nodulin 26-like intrinsic proteins (NIPs), small basic intrinsic proteins (SIPs), and X-intrinsic proteins (XIPs). Further, transport substrates of morning glory AQPs were estimated based on their homology to the known AQPs in other plant species and their corresponding amino acid motifs that possess permeability pores. It was expected that PIPs are likely to transport water, carbon dioxide, and hydrogen peroxide; TIPs are likely transport water, hydrogen peroxide, ammonia, urea, and boric acid; NIPs are likely transport water, boric acid, ammonia, glycerol, and formamide; and XIPs are likely to transport water, hydrogen peroxide, and glycerol. Overall, these results suggest that AQPs are involved in water and nutrient transport in Japanese morning glory. An in silico gene expression analysis suggested the importance of AQPs in flower opening, water or nutrient uptakes from the soil to roots, and photosynthesis in morning glory. Our findings provide fundamental information that enables further study into the importance of AQPs in morning glory, including their roles in flower opening and other physiological events.
