Draft Genome Sequence of the Sattazolin-Producing Strain Pseudonocardia sp. C8, Isolated from a Mud Dauber Wasp Nest in Nepal.

ABSTRACT: Here, we report a 6.2-Mbp draft genome sequence of the bacterium Pseudonocardia sp. strain C8, which gave insight into the complete secondary metabolite production capacity of the strain and hinted that the strain possibly represents a new species.

Oxandrastins: Antibacterial Meroterpenes from an Australian Mud Dauber Wasp Nest-Associated Fungus, Penicillium sp. CMB-MD14.

Project description:The ethyl acetate extract of an ISP-2 agar cultivation of the wasp nest-associated fungus Penicillium sp. CMB-MD14 exhibited promising antibacterial activity against vancomycin-resistant enterococci (VRE), with a bioassay guided chemical investigation yielding the new meroterpene, oxandrastin A (1), the first andrastin-like metabolite with an extra oxygenation at C-2. A culture media optimisation strategy informed a scaled-up rice cultivation that yielded 1, together with three new oxandrastins B-D (2-4), two known andrastins C (5) and F (6), and a new meroterpene of the austalide family, isoaustalide F (7). Structures of 1-7 were assigned based on detailed spectroscopic analysis and chemical interconversion. A GNPS molecular networking analysis of the rice cultivation extract detected the known austalides B (8), H (9), and H acid (10), tentatively identified based on molecular formulae and co-clustering with 7. That the anti-VRE properties of the CMB-MD14 extract were exclusively attributed to 1 (IC50 6.0 µM, MIC99 13.9 µM), highlights the importance of the 2-OAc and 3-OAc moieties to the oxandrastin anti-VRE pharmacophore.

Mechanical and thermal properties of mud dauber nests under atmospheric drying.

Project description:Mud dauber wasps construct soil nests to protect their offspring from predators, extreme temperatures, and rainwater. The mechanical and thermal properties of these nests are important for the reproductive success of mud daubers. The previous study showed that the high densities and strengths of mud dauber nests were due to the repetitive tapping and atmospheric drying used by mud daubers during nest construction. This study investigated the effect of atmospheric drying on the mechanical and thermal properties of mud dauber nests. The soil shrinkage curve, elastic modulus, suction stress characteristic curve, soil water retention curve, shear strength, and thermal conductivity function of mud dauber nest soils were measured by performing drying cake tests, direct shear tests, unconfined compression tests, and thermal conductivity measurements. This study showed atmospheric drying could increase Young's moduli (from hundreds to thousands of kPa), the magnitudes of suction stress (up to 64 kPa), and shear strengths (e.g., unconfined compressive strength increased up to 2100 kPa) of mud dauber nests. The thermal conductivity was reduced by 47% due to atmospheric drying. Investigation of mud dauber nests under atmospheric drying could provide insights and inspiration to improve human manufacturing and manipulation of soils for earthen building construction.

Specialists and generalists coexist within a population of spider-hunting mud dauber wasps.

Project description:Individual foraging specialization describes the phenomenon where conspecifics within a population of generalists exhibit differences in foraging behavior, each specializing on different prey types. Individual specialization is widespread in animals, yet is understudied in invertebrates, despite potential impacts to food web and population dynamics. Sceliphron caementarium (Hymenoptera: Sphecidae) is an excellent system to examine individual specialization. Females of these mud dauber wasps capture and paralyze spiders which they store in mud nests to provision their offspring. Individuals may make hundreds of prey choices in their short lifespan and fully intact prey items can be easily excavated from their mud nests, where each distinct nest cell represents a discrete foraging bout. Using data collected from a single population of S. caementarium (where all individuals had access to the same resources), we found evidence of strong individual specialization; individuals utilized different resources (with respect to prey taxa, prey ecological guild, and prey size) to provision their nests. The extent of individual specialization differed widely within the population with some females displaying extreme specialization (taking only prey from a single species) while others were generalists (taking prey from up to 6 spider families). We also found evidence of temporal consistency in individual specialization over multiple foraging events. We discuss these findings broadly in the context of search images, responses to changing prey availability, and intraspecific competition pressure.

Avian mud nest architecture by self-secreted saliva.

Project description:Mud nests built by swallows (Hirundinidae) and phoebes (Sayornis) are stable granular piles attached to cliffs, walls, or ceilings. Although these birds have been observed to mix saliva with incohesive mud granules, how such biopolymer solutions provide the nest with sufficient strength to support the weight of the residents as well as its own remains elusive. Here, we elucidate the mechanism of strong granular cohesion by the viscoelastic paste of bird saliva through a combination of theoretical analysis and experimental measurements in both natural and artificial nests. Our mathematical model considering the mechanics of mud nest construction allows us to explain the biological observation that all mud-nesting bird species should be lightweight.

Draft genome sequence data of the facultative, thermophilic, xylanolytic bacterium <i>Paenibacillus</i> sp. strain DA-C8.

Project description:Thermophilic, facultatively anaerobic, xylanolytic bacterial strain DA-C8 (=JCM34211 =DSM111723), newly isolated from compost, shows strong beechwood xylan degradation ability. Whole-genome sequencing of strain DA-C8 on the Ion GeneStudio S5 system yielded 69 contigs with a total size of 3,110,565 bp, 2,877 protein-coding sequences, and a G+C content of 52.3 mol%. Genome annotation revealed that strain DA-C8 possesses debranching enzymes, such as β-L-arabinofuranosidase and polygalacturonase, that are important for efficient degradation of xylan. As inferred from 16S rRNA sequences and average nucleotide identity values, the closest relatives of strain DA-C8 are Paenibacillus cisolokensis and P. chitinolyticus. An actinomycete isolate from solitary wasp mud nest having strong antibacterial activity and kills the Candida cells due to the shrinkage and the cytosolic loss.

Project description:An actinomycetes strain designated as MN 2(6) was isolated from the solitary wasp mud nest. The isolate was identified using polyphasic taxonomy. It produced the extensive branched brown substrate and white aerial hyphae that changed into grayish black. The aerial mycelia produced the spiral spore chains with rugose spore surface. The growth was observed between temperature range of 27-37°C, pH 8-10 and below salt concentration of 6% (w/v). The comparative analysis of 16S rRNA gene sequence and phylogenetic relationship showed that strain MN 2(6) lies in clade with Streptomyces hygroscopicus subsp. hygroscopicus NRRL 2387(T), Streptomyces sporocinereus NBRC 100766(T) and Streptomyces demainii NRRL B-1478(T) with which it shares a 16S rRNA gene sequence similarity of 99.3%. The strain MN 2(6) can be differentiated from type strains based on phenotypic characteristics. The strain MN 2(6) showed most promising activity against Gram-positive, Gram-negative bacteria, acid-fast bacilli and Candida species suggesting broad-spectrum characteristics of the active metabolite. Evaluation of anti-candidal activity of the metabolite of strain MN 2(6) by scanning electron microscopy (SEM) revealed changed external morphology of yeast. It kills the Candida cells due to the shrinkage and the cytosolic loss. However, further studies are required to elucidate the structure of the active metabolite produced by the isolate MN 2(6).

The key step involved a sp2–sp3 Suzuki–Miyaura reaction between a common boronic ester lateral chain and various functionalised quinolone cores. The quinolones slowed growth of E. coli and S. aureus by inducing extended lag phases.
