Complete Genome Sequences of Staphylococcus argenteus Tokyo13064 and Tokyo13069, Isolated from Specimens Obtained during a Food Poisoning Outbreak in Tokyo, Japan.

ABSTRACT: The complete genome sequences of two Staphylococcus argenteus strains, Tokyo13064 and Tokyo13069, isolated from human feces and suspected causative foods during a staphylococcal food poisoning outbreak, consist of 2,750,811-bp and 2,751,556-bp circular chromosomes and 2,543 and 2,548 genome annotation-predicted coding DNA sequences, respectively, with 19 rRNAs, 61 tRNAs, and 1 CRISPR each.

OUTBREAK OF SALMONELLA FOOD POISONING AT HIGH ALTITUDE.

Project description:An outbreak of food poisoning occurred among soldiers of an army unit located at high altitude (3300m) in western Himalayas. A total of 78 of the 103 soldiers who had consumed the incriminated meal suffered from the illness. Sixteen of the 78 had symptoms severe enough to warrant admission to hospital. The symptoms consisted of diarrhoea, vomiting, fever, headache, vertigo and abdominal cramps of varying intensity. Salmonella enteritidis was isolated from 6 stool samples and in one blood sample. Epidemiologically, frozen fowl was traced as the probable incriminating food agent responsible for the outbreak. This is probably the first such outbreak being reported from high altitude. It is of significance for the Indian Armed Forces, as the cause of the food poisoning was traced to an inherent shortcoming in the supply of food items to the troops located in far flung and isolated regions and in the cooking practices being followed.

Molecular epidemiology and identification of a Staphylococcus aureus clone causing food poisoning outbreaks in Japan.

Project description:Molecular characterization of isolates from staphylococcal food poisoning (SFP) outbreaks in Japan showed that the dominant lineage causing SFP outbreaks is clonal complex 81 (CC81), a single-locus variant of sequence type 1, coagulase type VII, positive for sea and/or seb, and positive for seh. Among various CC lineages producing staphylococcal enterotoxin A, CC81 showed the highest toxin productivity.

Project description:Staphylococcus argenteus TWCC 58113 was isolated from a specimen from a 12-year-old boy with purulent lymphadenitis. The S. argenteus TWCC 58113 genome was completely sequenced. The TWCC 58113 chromosome was 2,761,442?bp in size with a GC content of 32.44%. S. argenteus TWCC 58113 was found to harbor two plasmids.

Here, we report the complete annotated genome sequence of the chromosome and a plasmid.

Staphylococcus argenteus from rabbits in Thailand.

Project description:Staphylococcus argenteus, a novel species of the genus Staphylococcus or a member of the S. aureus complex, is closely related to S. aureus and is usually misidentified. In this study, the presence of S. argenteus in isolated S. aureus was investigated in 67 rabbits with abscess lesions during 2014-2016. Among 19 S. aureus complex isolates, three were confirmed to be S. argenteus by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry, nonribosomal peptide synthetase gene amplification, and multilocus sequence type. All S. aureus complex isolates, including the S. aureus isolates, were examined for their antimicrobial resistance phenotype by disk diffusion and for their resistance genotype by PCR assays. Among the S. argenteus isolates, one was susceptible to all antimicrobial drugs and the other two were resistant to penicillin and doxycycline. In contrast, most S. aureus isolates were resistant to penicillin (37.5%), and gentamicin (12.5%). Moreover, S. aureus isolates harbored the blaZ, mecA, aacA-aphD, and mrs(A) as well as mutations of gyrA and grlA, but S. argenteus isolates carried solely the blaZ. S. argenteus isolates were investigated for enterotoxin (sea-sed) and virulence genes by PCR. One isolate carried sea, sec, and sed, whereas the other two isolates carried only sea or sed. No isolate carried seb and see. All three S. argenteus isolates carried hla, hlb, and clfA, followed by pvl, whereas coa, spa (IgG-binding region), and spa (x region) were not detected in the three isolates. This paper presents the first identification of S. argenteus from rabbits in Thailand. S. argenteus might be pathogenic because the isolates carried virulence genes. Moreover, antimicrobial resistance was observed. Investigations of this new bacterial species should be conducted in other animal species as well as in humans.

APPLICATION OF HAZARD ANALYSIS CRITICAL CONTROL POINT SYSTEM : ANALYSIS OF A FOOD POISONING OUTBREAK.

Project description:The World Health Organization as well as several experts have recently recommended the use of hazard analysis critical control point system for prevention of food-borne illnesses. The present study proceeded to identify the hazardous operations and their critical control points by investigating, through a case-control design, an outbreak of food poisoning. The predominant presenting symptoms were watery diarrhoea and abdominal cramps. The cumulative incidence during this episode was 28.6 per cent, with a median incubation of 13 hours. Evaluation of food histories indicated a very high and significant odds ratio for mutton dish (OR = 6.45; p < 0.001), which persisted even after adjusting for the consumption of other food items through stratified analysis using the Mantel-Haenszel procedure. Environmental assessment revealed a strong possibility of contamination of raw mutton at the source with animal excreta and prolonged storage at room temperature of cooked mutton dish. Based on these findings, hazardous operations were identified and critical control points and monitoring criteria for prevention of food poisoning have been suggested.

APPLICATION OF STRATIFIED ANALYSIS IN EPIDEMIC INVESTIGATIONS : ANALYSIS OF AN OUTBREAK OF FOOD POISONING.

Project description:The present study demonstrates the use of stratified analysis when high (or low) risk is observed for two or more food items during the investigation of food poisoning, rather than simply implicating the item showing the highest risk. In this study an outbreak of food poisoning, which exhibited a clinico-epidemiological profile of Clostridium perfringens as the possible agent, was investigated using a case-control typology. Evaluation of food histories showed a high and significant odds ratio (OR) for a mutton dish (OR = 6.45, p < 0.001) and peas dish (OR = 5.6, p < 0.001), while a legume dish showed a protective effect (OR = 0.13, p < 0.001) which proved erroneous. Stratified analysis using Mantel-Haenszel procedure indicated that the risk due to mutton persisted even after adjusting for consumption of peas (adjusted OR = 4.22) or legume dish (adjusted OR = 4.25). The use of stratified analysis in investigating epidemics has been recommended.

Dengue Virus Infection in Aedes albopictus during the 2014 Autochthonous Dengue Outbreak in Tokyo Metropolis, Japan.

Project description:In 2014 in Japan, 162 autochthonous dengue cases were reported for the first time in nearly 70 years. Here, we report the results of the detection and isolation of dengue virus (DENV) from mosquitoes collected in Tokyo Metropolis in 2014 and 2015. The phylogenetic relationship among DENV isolates from mosquitoes and from patients based on both the entire envelope gene and whole coding sequences was evaluated. Herein, 2,298 female and 956 male Aedes albopictus mosquitoes were collected at six suspected locations of DENV infection in Tokyo Metropolis from August to October in 2014 and grouped into 124 and 35 pools, respectively, for viral genome detection and DENV isolation. Dengue virus RNA was detected using reverse transcription polymerase chain reaction and TaqMan assays from 49 female pools; 16 isolates were obtained using C6/36 and Vero cells. High minimum infection rates (11.2-66.7) persisted until mid-September. All DENV isolates belonged to the genotype I in serotype 1 (DENV-1), and its sequences demonstrated > 99% homology to the sequence of the DENV isolated from a patient in the vicinity of Tokyo Metropolis in 2014. Therefore, Ae. albopictus was a major DENV vector, and a single DENV-1 strain circulated in Tokyo Metropolis in 2014. Dengue virus was not detected from male mosquitoes in 2014 and wild larvae in April 2015. Thus, the possibility of both vertical transmission and overwintering of DENV was extremely low, even in dengue-epidemic areas. This study reports the first entomological information on a dengue outbreak in a temperate region, where no Aedes aegypti mosquitoes are distributed.

Autochthonous dengue fever, Tokyo, Japan, 2014.

Project description:After 70 years with no confirmed autochthonous cases of dengue fever in Japan, 19 cases were reported during August-September 2014. Dengue virus serotype 1 was detected in 18 patients. Phylogenetic analysis of the envelope protein genome sequence from 3 patients revealed 100% identity with the strain from the first patient (2014) in Japan.
