The complete mitochondrial genome of Reticulitermes ovatilabrum (Isoptera: Rhinotermitidae).

ABSTRACT: The mitochondrial genome of the Reticulitermes ovatilabrum is 15,913 bp in length and encodes 37 genes including 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNA), 2 ribosomal RNA genes (rRNA), and a non-coding control region (D-loop). The percentage of A/T (65.59%) is much higher than that of C/G (34.41%). The phylogenetic tree revealed that R. ovatilabrum was closest to R. kanmonensi and R. periflaviceps. The mitochondrial genome of the R. ovatilabrum provides a resource for evolutional analysis within termites especially Reticulitermes.

Project description:The mitochondrial genome of the Reticulitermes periflaviceps contains 15,925 bp of nucleotide in length including 13 protein-coding genes, 22 transport RNAs, 2 ribosomal RNAs, and a non-coding region. The overall A + T among the genome sequence is 65.64%. The start codons of all protein-coding genes are ATN and the stop codon is TAA except TAG for Nad1 and incomplete T for COII and Nad5. The lengths of 12sRNA and 16sRNA genes are 743 and 1309 bp, respectively, and the control region was 1118 bp in length, with two repeating tandem regions. The phylogenetic tree revealed that R. periflaviceps and Reticulitermes kanmonensis constituted a sister group to Reticulitermes flaviceps. The mitochondrial genome here provides a resource for evolution analysis within termites especially Reticulitermes.

It consists of 13 PCGs, 22 tRNA, 2 rRNA genes and an A + T-rich control region. All the protein-coding genes used ATN as start codon. But the stop codons were TAA, TAG, and an incomplete termination codon (T) abutting an adjacent tRNA gene. The A + T-rich control region was 1680 bp in length with 70.4% A + T content.

All the protein-coding genes used ATN as start codon. But the stop codons were TAA, TAG, and an incomplete termination codon (T) abutting an adjacent tRNA gene. The A + T-rich control region was 1105 bp in length with 67.8% A + T content.

The complete mitochondrial genome of Reticulitermes labralis and implications for Rhinotermitidae taxonomy.

Project description:The complete nucleotide sequence of the mitochondrial genome of Reticulitermes labralis was analyzed. This mitochondrial genome is a circular molecule of 15,914 bp in length and has the same gene content and organization as that found in other Reticulitermes species. It contains 13 protein-coding genes (PCGs), 22 tRNAs, two rRNAs (12S and 16S rRNA) and a non-coding AT-rich region (CR). The total base composition is strongly biased toward A + T nucleotides (65.1%). Most of the genes are encoded on the H strand, except for the other four protein-coding genes and eight tRNA genes on the L strand. The phylogenetic tree based on the nucleotide sequences of 13 mitochondrial PCGs using the Bayesian inference method supports the traditional morphologically analysis.

The Complete Mitochondrial Genome of Coptotermes 'suzhouensis' (syn. Coptotermes formosanus) (Isoptera: Rhinotermitidae) and Molecular Phylogeny Analysis.

Project description:Coptotermes suzhouensis (Isoptera: Rhinotermitidae) is a significant subterranean termite pest of wooden structures and is widely distributed in southeastern China. The complete mitochondrial DNA sequence of C. suzhouensis was analyzed in this study. The mitogenome was a circular molecule of 15,764 bp in length, which contained 13 protein-coding genes (PCGs), 22 transfer RNA genes, two ribosomal RNA genes, and an A+T-rich region with a gene arrangement typical of Isoptera mitogenomes. All PCGs were initiated by ATN codons and terminated by complete termination codons (TAA), except COX2, ND5, and Cytb, which ended with an incomplete termination codon T. All tRNAs displayed a typical clover-leaf structure, except for tRNASer(AGN), which did not contain the stem-loop structure in the DHU arm. The A+T content (69.23%) of the A+T-rich region (949 bp) was higher than that of the entire mitogenome (65.60%), and two different sets of repeat units (A+B) were distributed in this region. Comparison of complete mitogenome sequences with those of Coptotermes formosanus indicated that the two taxa have very high genetic similarity. Forty-one representative termite species were used to construct phylogenetic trees by maximum likelihood, maximum parsimony, and Bayesian inference methods. The phylogenetic analyses also strongly supported (BPP, MLBP, and MPBP = 100%) that all C. suzhouensis and C. formosanus samples gathered into one clade with genetic distances between 0.000 and 0.002. This study provides molecular evidence for a more robust phylogenetic position of C. suzhouensis and inferrs that C. suzhouensis was the synonymy of C. formosanus.

The Antennal Sensilla and Expression Patterns of Olfactory Genes in the Lower Termite Reticulitermes aculabialis (Isoptera: Rhinotermitidae).

Project description:The insect olfactory system plays pivotal roles in insect survival and reproduction through odor detection. Morphological and physiological adaptations are caste-specific and evolved independently in workers, soldiers, and reproductives in termites. However, it is unclear whether the olfactory system is involved in the division of labor in termite colonies. In the present study, the antennal sensilla of alates, workers, soldiers, nymphs, and larvae of the termite Reticulitermes aculabialis Tsai et Hwang ( Isoptera: Rhinotermitidae) were investigated. Transcriptomes were used to detect olfactory genes, and differential expression levels of olfactory genes were confirmed in various castes by qRT-PCR analysis. Nine types of sensilla were identified on the antennae of R. aculabialis, and soldiers possessed all 9 types. In 89,475 assembled unigenes, we found 16 olfactory genes, including 6 chemosensory protein (CSP) and 10 odorant-binding protein (OBP) genes. These OBP genes included 8 general odorant-binding protein genes (GOBPs) and 2 pheromone-binding protein-related protein (PBP) genes. Five CSP genes were more highly expressed in alates than in workers, soldiers, larvae, and nymphs, and the expression levels of CSP6 were significantly higher in nymphs. Seven GOBP and two PBP genes exhibited significantly higher expression levels in alates, and there were no significant differences in the expression levels of GOBP2 among workers, soldiers, alates, and larvae. These results suggest that alates, as primary reproductives, have unique expression patterns of olfactory genes, which play key roles in nuptial flight, mate seeking, and new colony foundation.

Effect on genetic diversity of the absence of intraspecies preference in 2 sympatric Reticulitermes termite species (Isoptera: Rhinotermitidae).

Project description:The recombinant genotypes that can be produced when closely related species mate improve the genetic diversity of the population. Among closely related species, the link between interspecific reproduction behaviors and genetic diversity has barely been studied. Reticulitermes chinensis and R. flaviceps, which live close to each other, were used as research subjects in our study to find out how preferring conspecifics affects reproductive behavior between species. We discovered that neither R. chinensis nor R. flaviceps displayed preference behavior for conspecifics. Males of R. chinensis and R. flaviceps chased and groomed not only intraspecific females but also interspecific females. In a brief period of time, 2 mating behaviors, intra- and interspecific mating, were also observed. There were no significant differences in the duration of each behavior (tandem, grooming, and mating) between interspecies and intraspecies partners. Moreover, genetic analysis showed both interspecific mating and intraspecific mating can produce living offspring when the 2 types of mating occur in a colony. Our findings showed that there was no obvious intraspecific preference between the 2 species of termite Reticulitermes when it came to tandem, grooming, and mating, which not only makes it easier for interspecific hybridization to occur but also sheds light on the genetic diversity.

Maternal determination of soldier proportion and paternal determination of soldier sex ratio in hybrid Reticulitermes (Isoptera: Rhinotermitidae) termite colonies.

Project description:Altruistic caste, including worker and soldier (derived from worker), plays a critical role in the ecological success of social insects. The proportion of soldiers, soldier sex ratios, and the number of workers vary significantly between species, and also within species, depending on colony developmental stage and environmental factors. However, it is unknown whether there are sex-linked effects from parents on controlling the caste fate or not. Here, we compared soldier sex ratios, soldier proportions, and population size among a four mating types of Reticulitermes amamianus (Ra) and R. speratus (Rs) (male × female, mRa × fRa, mRa × fRs, mRs × fRa, mRs × fRs) and demonstrate that the soldier sex ratio and worker population size of hybrid colonies skew to colonies of king's species, while the soldier proportion skew to queen's species. The survival rate of offspring resulting from interspecies hybridization was significantly higher for mRa × fRs than for mRs × fRa. The results of this study demonstrate the asymmetric influence of kings and queens on caste determination and colony growth, which can contribute to our better understanding of parental influence on the colony dynamics of social insects.

Efficient RNA interference method during caste differentiation with hormone treatment in the termite Reticulitermes speratus (Isoptera: Rhinotermitidae).

Project description:Unveiling the proximate mechanism of caste differentiation is crucial for understanding insect social evolution, and gene function analysis is an important tool in this endeavor. The RNA interference (RNAi) technique is useful in termites, but its knockdown effects may differ among species. One of the most important model species in the field of termite sociogenomics is Reticulitermes speratus Kolbe (Isoptera: Rhinotermitidae). Presoldier and worker differentiation of this species can be artificially induced by juvenile hormone and 20-hydroxyecdysone application, respectively. However, appropriate RNAi technique of genes expressed during caste differentiation has never been considered. To clarify this issue, first, we injected nine different volumes of nuclease-free water (NFW, 0-404.8 nL) into workers and found that survival and caste differentiation rates were strongly reduced by the application of the top three largest volumes. Second, we injected double-stranded (ds) RNA of ecdysone receptor homolog (RsEcR) (2.0 µg/151.8 nL NFW) into workers with hormone treatments. The expression levels of RsEcR were significantly reduced at 9 days after dsRNA injection. RsEcR RNAi strongly affected both molting events during presoldier and worker differentiation induced by hormone treatments. The present results highlight the need for caution regarding injection volumes for RNAi experiments using hormone treatments. We suggest that the injection of dsRNA solution (2 µg; approximately 100-200 nL) is suitable for RNAi experiments during caste differentiation induced by hormone application in R. speratus.

The development of adultoid reproductives and brachypterous neotenic reproductives from the last instar nymphs in Reticulitermes labralis (Isoptera: Rhinotermitidae): a comparative study.

Project description:Secondary reproductives develop primarily from nymphs. However, they have been rarely studied; in particular, the development of adultoid reproductives (AR) with floppy wings is still unclear. In this study, the change in juvenile hormone (JH) levels, vitellogenin gene expression, and oogenesis during the development of AR and brachypterous neotenic reproductives (BN) from the last instar nymphs of Reticulitermes labralis are investigated and compared. The results showed that the AR derived from the last instar nymphs by molting, and they were more similar to neotenic reproductives in morphology. In addition, the paired AR were not able to survive in the absence of workers. In R. labralis, the process of the last instar nymphs developing into AR and BN took an increase in JH level as a starting point. The JH level of the last instar nymphs molting into BN was approximately 1.5-fold higher than that of the AR. Additionally, The JHIII level of BN peaked on day 5, and that of AR peaked on day 10, which induced the onset of vitellogenesis in BN and AR, respectively. After molting, the vitellogenin gene expression levels of both BN and AR initially increased and then declined, and the expression levels in the BN were significantly higher than those in the AR. In addition, the oocytes of BN matured earlier than those of the AR, and the number of eggs laid by the BN was higher than the number laid by the AR. Our results demonstrate that, in R. labralis, the last instar nymphs can develop into AR, which are significantly different from BN in their development.
