One Pot Synthesis of Large Gold Nanoparticles with Triple Functional Ferrocene Ligands.

ABSTRACT: In biomedical, toxicological, and optoelectronic applications, the size of nanoparticles is one of the decisive factors. Therefore, synthesis of nanoparticles with controlled sizes is required. The current methods for synthesis of larger gold nanoparticles (GNPs, ~200 nm) are complex and tedious, producing nanoparticles with a lower yield and more irregular shapes. Using ferrocene as a primary reducing agent and stabilizer, sodium citrate as a dispersant, and sodium borohydride as an accessory reducing agent, GNPs of 200 nm were synthesized in a one pot reaction. Besides the roles of reducing agent and GNP stabilizer, ferrocene also served a role of quantitative marker for ligand loading, allowing an accurate determinate of surface ligands.

Here, we present for the first time, a one pot direct route to synthesize gold nanoparticles (AuNPs) using natural cacao extract as both a reducing and stabilizing agent. The nanoparticles were characterized by UV-visible spectroscopy (UV-VIS), dynamic light scattering (DLS), and transmission electron microscopy (TEM); and have excellent biocompatibility with human primary dermal fibroblasts.

One-Pot Synthesis of Nucleoside-Templated Fluorescent Silver Nanoparticles and Gold Nanoparticles.

Project description:In this study, a simple one-pot method was proposed to synthesize water-soluble nucleoside-templated fluorescent silver nanoparticles (Ag NPs) and gold nanoparticles (Au NPs). The nucleoside-templated fluorescent Ag NPs and Au NPs were further characterized by transmission electron microscopy (TEM), Fourier transform infrared spectroscopy (FTIR), and fluorescence spectroscopy (FLS). The effects of the molar ratio of reactants, reaction environment, and nucleotides on the synthesis of Ag NPs and Au NPs were also discussed. The results showed that nucleoside and ascorbic acid acted as a stabilizer and reductant, respectively, in the synthesis of Ag NPs and Au NPs, while citrate buffer acted as both a pH regulator and reductant. The synthesized nucleoside-templated fluorescent Ag NPs and Au NPs have good fluorescence stability and easy water solubility. In this study, a simple one-pot method was proposed to synthesize water-soluble nucleoside-templated fluorescent silver nanoparticles (Ag NPs) and gold nanoparticles (Au NPs).

One-Pot Synthesis of Multi-Branch Gold Nanoparticles and Investigation of Their SERS Performance.

Project description:Gold nanoparticles with multiple branches have attracted intensive studies for their application in sensing of low trace molecules. A large number of the merits found on the gold nanoparticles for the above applications are attributed to the strong localized surface plasmon resonance excited by the incident radiation. However, a facile and flexible way of synthesizing the multi-branch gold nanoparticles with tunable localized surface plasmon resonance frequency is still a challenge for the plasmonic research field. Herein, we report an efficient one-pot synthesis of multi-branch gold nanoparticles method that resembles a seed-medicated approach while using no further chemicals except chloroauric acid, ascorbic acid and 4-(2-Hydroxyethyl)-1-piperazinyl]-ethanesulfonic acid. By controlling the amounts of ascorbic acid volumes in the reaction mixture, the morphology and the localized surface plasmon resonance frequency of the synthesized multi-branch gold nanoparticles can be manipulated conveniently. Moreover, using the 4-Mercaptobenzoic acid as the Raman reporter, the multi-branch gold nanoparticles show superior surface-enhanced Raman spectroscopy characteristics that can be potentially used in chemical and biological sensing.

Biomimetic one-pot synthesis of gold nanoclusters/nanoparticles for targeted tumor cellular dual-modality imaging.

Project description:Biomimetic synthesis has become a promising green pathway to prepare nanomaterials. In this study, bovine serum albumin (BSA)-conjugated gold nanoclusters/nanoparticles were successfully synthesized in water at room temperature by a protein-directed, solution-phase, green synthetic method. The synthesized BSA-Au nanocomplexes have fluorescence emission (588 nm) of gold nanoclusters and surface plasmon resonance of gold nanoparticles. The BSA-Au nanocomplexes display non-cytotoxicity and excellent biocompatibility on MGC803 gastric cancer cells. After conjugation of folic acid molecules, the obtained BSA-Au nanocomplexes showed highly selective targeting for MGC803 cells and dual-modality dark-field and fluorescence imaging.

In this review, the characteristics and limitations of various one-pot syntheses of biologically active molecules are explained, primarily involving organocatalytic methods as key tactics. Besides catalysis, the pot-economy concepts presented herein are also applicable to organometallic and organic reaction methods in general.

One-pot green synthesis of gold and silver nanoparticles using <i>Rosa canina</i> L. extract.

Project description:This study reports a green, simple, and fast method for the synthesis of gold and silver nanoparticles using natural antioxidant compounds. The aqueous extract from dried rosehips (pseudofruit of Rosa canina L.) was used as a reducing and capping agent of HAuCl4 and AgNO3 during the noble metal colloid synthesis at room temperature and no other chemical reagent was used. The high antioxidant activity of the plant extract was proven by 2,2-diphenyl-1-picrylhydrazyl assay by a spectrophotometric method. The formation of stable gold and silver nanoparticles was observed by UV-visible spectroscopy and the evolution of their characteristic surface plasmon resonance band was followed over several days. Transmission electron microscopy confirmed the formation of quasi-spherical nanoparticles with mean diameters 26 and 34 nm, for gold and silver nanoparticles, respectively; XRD revealed an FCC crystalline structure for both gold and silver NPs. The effects of concentrations of noble metal precursor and plant extract solution on the formation, stabilization and size of nanoparticles are discussed, as well as some applications of these colloids.

One-pot synthesis of a new generation of hybrid bisphosphonate polyoxometalate gold nanoparticles as antibiofilm agents.

Project description:A reduced polyoxovanadate functionalized with bisphosphonate molecules was synthesized and used to prepare in one step hybrid organic-inorganic polyoxometalate decorated gold nanoparticles. These new composites were shown to strongly inhibit P. aeruginosa and S. epidermidis biofilm growth, with the three components constituting the nanoparticles (Au0 core, vanadium and alendronate) acting synergistically.

Versatile one-pot synthesis of gold nanoclusters and nanoparticles using 3,6-(dipyridin-2-yl)-(1,2,4,5)-tetrazine.

Project description:A one-pot synthesis of gold nano-objects is described by simply mixing a gold salt (HAuCl4), dodecanethiol and 3,6-di-2-pyridyl-1,2,4,5-tetrazine. When a large excess of thiol is used, gold nanoclusters of 2 nm are obtained in a large amount and with a narrow size distribution. The reaction mechanism was investigated by absorption and emission spectroscopies and shows the in situ formation of dihydrotetrazine acting as the reductant of Au(iii) to make Au(0). Au nanoclusters were isolated from the molecular precursors by HPLC. The nature of the ligands stabilizing Au nanoclusters was investigated by various techniques such as mass spectrometry, SEM-EDS, XPS and NMR. Thiol and tetrazine are shown to play both the role of ligand stabilizing the clusters. Finally, when a much smaller amount of thiol is used, a mixture of Au nanoclusters and Au nanoparticles of 10-15 nm, sometimes aggregated into clusters of 50 nm is obtained. The formation of larger nanoobjects is explained by the lower amount of thiol available to block the growth at the early stage as shown by UV-vis absorption monitoring.

One-Pot Green Synthesis of Graphene Nanosheets Encapsulated Gold Nanoparticles for Sensitive and Selective Detection of Dopamine.

Project description:We report a simple new approach for green preparation of gallic acid supported reduced graphene oxide encapsulated gold nanoparticles (GA-RGO/AuNPs) via one-pot hydrothermal method. The as-prepared composites were successfully characterized by using Fourier transform infrared spectroscopy (FTIR), Raman spectroscopy, X-ray powder diffraction techniques (XRD), scanning electron microscope (SEM), high resolution transmission electron microscopy (HRTEM) and elemental analysis. The GA-RGO/AuNPs modified electrode behaves as a hybrid electrode material for sensitive and selective detection of dopamine (DA) in presence of ascorbic acid (AA) and uric acid (UA). The GA-RGO/AuNPs modified electrode displays an excellent electrocatalytic activity towards the oxidation of DA and exhibits a wide linear response range over the DA concentrations from 0.01-100.3 μM with a detection limit (LOD) of 2.6 nM based on S/N = 3. In addition, the proposed sensor could be applied for the determination of DA in human serum and urine samples for practical analysis.

Multifaceted peptide assisted one-pot synthesis of gold nanoparticles for plectin-1 targeted gemcitabine delivery in pancreatic cancer.

Project description:An astute modification of the plectin-1-targeting peptide KTLLPTP by introducing a C-terminal cysteine preceded by a tyrosine residue imparted a reducing property to the peptide. This novel property is then exploited to fabricate gold nanoparticles (GNP) via an in situ reduction of gold(iii) chloride in a one-pot, green synthesis. The modified peptide KTLLPTPYC also acts as a template to generate highly monodispersed, spherical GNPs with a narrow size distribution and improved stability. Plectin-1 is known to be aberrantly expressed in the surface of pancreatic ductal adenocarcinoma (PDAC) cells while showing cytoplasmic expression in normal cells. The synthesized GNPs are thus in situ surface modified with the peptides via the cysteine residue leaving the N-terminal KTLLPTP sequence free for targeting plectin-1. The visual molecular dynamics based simulations support the experimental observations like particle size, gemcitabine conjugation and architecture of the peptide-grafted nanoassembly. Additionally, GNPs conjugated to gemcitabine demonstrate significantly higher cytotoxicity in vitro in two established PDAC cell lines (AsPC-1 and PANC-1) and an admirable in vivo antitumor efficacy in a PANC-1 orthotopic xenograft model through selective uptake in PDAC tumor tissues. Altogether, this strategy represents a unique method for the fabrication of a GNP based targeted drug delivery platform using a multifaceted peptide that acts as reducing agent, template for GNP synthesis and targeting agent to display remarkable selectivity towards PDAC.
