The complete chloroplast genome of leguminous forage Onobrychis viciifolia.

ABSTRACT: Onobrychis viciifolia is mainly distributed in Europe and has been widely cultivated in North and Northwest of China. The complete chloroplast genome was sequenced using the Illumina Hiseq X-Ten platform. The genome lacks an inverted repeat (IR) region, containing 76 protein-coding genes, 30 tRNAs genes and 4 rRNAs. The overall GC content is 34.6%. A phylogenetic tree based on the whole chloroplast genomes of 14 species indicated that Onobrychis viciifolia belonged to the tribe Hydysareae in IRLC group of the subfamily Papilionoideae (Leguminosae), and it was sister to the genus Hedysarum.

Project description:Sainfoin (Onobrychis viciifolia) is one of the dominant legume forages distributed in Northern China. In our study, we assembled and annotated the structure of the complete chloroplast genome of sainfoin. The length of the circular genome is 122,102 bp. It contains 115 genes, including 79 protein-coding genes (68.7%), 31 tRNA genes (26.96%) and 5 rRNA genes (4.35%). The GC content of the total chloroplast genome of sainfoin is 34.58%. We construct the phylogenetic relationships between the chloroplast genome of sainfoin and the other 16 species by the Maximum likelihood (ML), and found sainfoin is most closely related to Hedysarum petrovii and Hedysarum taipeicum.

The complete chloroplast genome sequence of Astragalus melilotoides (Fabaceae), a leguminous forage in Northern China.

Project description:Astragalus melilotoides Pall. 1776 is a perennial leguminous forage, widely distributed in northern China, with cold, drought and disease resistance characteristics. Here, we determined the complete chloroplast (cp) genome sequence of A. melilotoides. It was 123,827 bp in length and 36.97% GC content with IR loss. The cp genome contained 110 complete genes, including 76 protein-coding genes, 30 tRNA genes, and four rRNA genes. A maximum likelihood (ML) phylogenetic tree revealed that A. melilotoides was related to A. americanus, A. gummifer, A. mongholicus, A. nakaianus, A. mongholicus var. nakaianus, and A. membranaceus var. membranaceus. The cp genome analysis of A. melilotoides will provide a reference for the phylogenetic study of Astragalus in the future.

Characterization of the complete chloroplast genome of Arachis pintoi Krapov. & W.C.Greg., a perennial leguminous forage.

Project description:Arachis pintoi Krapov. & W.C.Greg. is an important leguminous forage grass species that have extremely wide ranges of distribution in the tropical and sub-tropical regions. It has high feeding value and horticultural value. In this study, we sequenced and assembled the complete chloroplast genome of A. pintoi. The chloroplast genome is 156,185 bp in length, containing a pair of inverted repeated (IR) regions of 25,820 bp that are separated by a large single copy (LSC) region of 85,637 bp, and a small single copy (SSC) region of 18,908 bp. The complete chloroplast genome contains 112 unique genes, including 80 protein-coding genes, 28 transfer RNA genes (tRNAs), and four ribosomal RNA genes (rRNAs). The overall GC content was 36.4%. The phylogenetic analysis demonstrated that A. pintoi formed a single branch among genus Arachis. The whole chloroplast genome of A. pintoi will be a useful resource for future studies on phylogeny and conservation in Arachis.

Large Variability of Proanthocyanidin Content and Composition in Sainfoin (Onobrychis viciifolia).

Project description:Proanthocyanidins (PAs) in sainfoin (Onobrychis viciifolia Scop.) are of interest to ameliorate the sustainability of livestock production. However, sainfoin forage yield and PA concentrations, as well as their composition, require optimization. Individual plants of 27 sainfoin accessions from four continents were analyzed with LC-ESI-QqQ-MS/MS for PA concentrations and simple phenolic compounds. Large variability existed in PA concentrations (23.0-47.5 mg g(-1) leaf dry matter (DM)), share of prodelphinidins (79-96%), and mean degree of polymerization (11-14) among, but also within, accessions. PAs were mainly located in leaves (26.8 mg g(-1) DM), whereas stems had less PAs (7.8 mg g(-1) DM). Overall, high-yielding plants had lower PA leaf concentrations (R(2) = 0.16, P < 0.001) and fewer leaves (R(2) = 0.66, P < 0.001). However, the results show that these two trade-offs between yield and bioactive PAs can be overcome.

A chromosome-level genome assembly for Onobrychis viciifolia reveals gene copy number gain underlying enhanced proanthocyanidin biosynthesis.

Project description:Sainfoin (Onobrychis viciifolia), which belongs to subfamily Papilionoideae of Leguminosae, is a vital perennial forage known as "holy hay" due to its high contents of crude proteins and proanthocyanidins (PAs, also called condensed tannins) that have various pharmacological properties in animal feed, such as alleviating rumen tympanic disease in ruminants. In this study, we select an autotetraploid common sainfoin (2n = 4x = 28) and report its high-quality chromosome-level genome assembly with 28 pseudochromosomes and four haplotypes (~1950.14 Mb, contig N50 = 10.91 Mb). The copy numbers of genes involved in PA biosynthesis in sainfoin are significantly greater than those in four selected Fabales species, namely, autotetraploid Medicago sativa and three other diploid species, Lotus japonicus, Medicago truncatula, and Glycine max. Furthermore, gene expansion is confirmed to be the key contributor to the increased expression of these genes and subsequent PA enhancement in sainfoin. Transcriptomic analyses reveal that the expression of genes involved in the PA biosynthesis pathway is significantly increased in the lines with high PA content compared to the lines with medium and low PA content. The sainfoin genome assembly will improve our understanding of leguminous genome evolution and biosynthesis of secondary metabolites in sainfoin.
