HTRA1 Variants and the Interaction with Smoking Confer the Genetic Susceptibility to Ischemic Stroke.

ABSTRACT: High temperature requirement protein A1 (HtrA1) was identified as the causative gene of autosomal recessive arteriopathy and associated with lacunar ischemic stroke (IS) in European. This study aimed at evaluating the association of HTRA1 with IS and four tagging single-nucleotide polymorphisms (SNPs) were genotyped in a cohort of 4,098 Chinese. The mRNA level of HTRA1 in 72 IS cases and 72 hypertension controls were measured and compared. In whole population, SNP rs2268350 (C>T) was significantly associated with IS incidence (P=0.034). Stratification analysis observed significant association of rs2268350 in male, smoking and drinking populations, rs2672587 (C>G) in smoking and nonsmoking populations and rs3793917 (C>G) in smoking, nonsmoking and nondrinking populations with stroke respectively (P<0.05). The additive interaction and multiplicative interaction between rs2268350 and smoking were both of significant (P<0.05) after adjustment for the covariates. There was a cumulated risk of IS among genotypes of rs3793917 (P=0.009) and rs2672587 (P=0.047) in smoking population. The mRNA level of HTRA1 in non-smokers with rs2268350 CC was significantly higher than smokers with rs2268350 CT/TT (P=0.046) in IS cases. Our findings support that HTRA1 confers the genetic susceptibility to IS and smoking might modify the genetic effect of HTRA1 on IS by suppressing HTRA1 mRNA expression.

Project description:BackgroundComplement system plays an important role in innate immunity which involved in the changes tumor immune microenvironment by mediating the inflammatory response. This study aims to explore the relationship between complement component 7 (C7) polymorphisms and the risk of gastric cancer (GC).Materials and methodsAll selected SNPs of C7 were genotyped in 471 patients and 471 controls using the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Odds ratios (ORs) and 95% confidence intervals (CIs) were calculated by unconditional Logistic regression to analyze the relationship between each genotype and the genetic susceptibility to gastric cancer. The level of C7 expression in GC was analyzed by Gene Expression Profiling Interactive Analysis (GEPIA) and detected by Enzyme Linked Immunosorbent Assay. Kaplan-Meier plotter were used to reveal C7 of prognostic value in GC. We examined SNPs associated with the expression of C7 using the GTEx database. The effect of C7 polymorphisms on the regulatory activity of C7 was detected by luciferase reporter assay.ResultsUnconditional logistic regression showed that individuals with C7 rs1376178 AA or CA genotype had a higher risk of GC with OR (95% CI) of 2.09 (1.43-3.03) and 1.88 (1.35-2.63), respectively. For C7 rs1061429 C > A polymorphism, AA genotype was associated with the elevated risk for developing gastric cancer (OR = 2.16, 95% CI [1.37-3.38]). In stratified analysis, C7 rs1376178 AA genotype increased the risk of GC among males (OR = 2.88, 95% CI [1.81-4.58]), but not among females (OR = 1.06, 95% CI [0.55-2.06]). Individuals carrying rs1061429 AA significantly increased the risk of gastric cancer among youngers (OR = 2.84, 95% CI [1.39-5.80]) and non-smokers (OR = 2.79, 95% CI [1.63-4.77]). C7 was overexpressed in gastric cancer tissues and serum of cancer patients and was significantly associated with the prognosis. C7 rs1061429 C > A variant contributed to reduced protein level of C7 (P = 0.029), but rs1376178 didn't. Luciferase reporter assay showed that rs1376178C-containing plasmid exhibited 2.86-fold higher luciferase activity than rs1376178 A-containing plasmid (P < 0.001). We also found that rs1061429A allele contributed 1.34-fold increased luciferase activity than rs1061429C allele when co-transfected with miR-591 (P = 0.0012).ConclusionsThese findings highlight the role of C7 in the development of gastric cancer.

Project description:BackgroundMinor ischemic strokes, despite their initial mild symptoms, pose a significant risk of recurrence, potentially leading to severe disability. The role of C-reactive protein (CRP) gene variants in predicting the recurrent minor stroke, particularly their interaction with lifestyle factors such as smoking, remains unclear. We aimed to investigate the relationship of single-nucleotide polymorphisms (SNPs) in CRP gene with minor stroke recurrence, focusing on gene-environment interactions.MethodsA retrospective cohort study was conducted using data from the stroke registry at Linfen People's Hospital, including 2032 first-time minor stroke patients (NIHSS score ≤ 5) admitted within 7 days of symptom onset from January 2019 to December 2022. Follow-up assessments were conducted every 3 months for one year. Based on recurrence during follow-up, participants were classified into Recurrence and Non-recurrence group. Genomic DNA was extracted for genotyping four CRP gene SNPs: rs1130864, rs1800947, rs2808632, and rs3093059. Genetic associations with the stroke recurrence were analyzed using additive, dominant, recessive genetic and allelic models. Generalized Multifactor Dimensionality Reduction (GMDR) was employed to explore the complex interaction of these SNPs with smoking status. Multivariate logistic regression was used to estimate the strength of these associations.ResultsOur study recruited 261 participants who experienced recurrent minor strokes and 264 age- and sex-matched controls without recurrence. The A allele of rs2808632 (P = 0.002) and C allele of rs3093059 (P = 0.001) were significantly associated with an increased risk of stroke recurrence. Participants with the combined genotypes rs2808632 CA + AA and rs3093059 TC + CC had a 1.325-fold higher risk of recurrence when compared to those with the genotypes rs2808632 CC and rs3093059 TT (P = 0.003). Additionally, among smokers, the rs3093059 TC + CC genotypes were associated with a 2.467-fold increased risk of recurrence compared to non-smokers with TT genotype (P < 0.001).ConclusionThe rs2808632 and rs3093059 polymorphisms independently and interactively contribute to an increased risk of recurrent minor stroke. Furthermore, the interaction between rs3093059 SNP and smoking status significantly influenced stroke recurrence, highlighting the importance of considering both genetic and environmental factors in predicting the risk of minor stroke recurrence.
