Cholesterol Content, Fatty Acid Profile and Health Lipid Indices in the Egg Yolk of Eggs from Hens at the End of the Laying Cycle, Following Alpha-Ketoglutarate Supplementation.

ABSTRACT: The current study aimed to assess the effects of dietary alpha-ketoglutarate (AKG) supplementation to laying hens on the fatty acid (FA) profile and cholesterol levels of the egg yolk at the end of production cycle. The experiment was performed on forty-eight Bovans Brown laying hens randomly assigned to either a control group (CONT) or a group supplemented with AKG. The CONT group was fed the basal diet, and the AKG group was fed the basal diet plus 1.0% AKG from the 31st until the 60th week of age, when FA profile, fat and cholesterol content of the egg yolks were determined. No significant changes in the cholesterol and total fat content of the egg yolks were observed. However, there were positive (the decrease in n-6 FA and the increase in MUFA), and negative (decrease in PUFA and n-3 FA, increase in TI and n-6/n-3 ratio) changes in FA profile following AKG supplementation. In conclusion, it was shown that dietary AKG after a 30-week long supplementation influence FA profile in egg yolk and its nutritional value.

Project description:This study was designed to evaluate the effect of dietary Ser on performance, egg quality, serum indices, and ileal mucosal immunity in laying hens fed low crude protein (LCP), essential amino acids (EAA) balanced diets. A total of 480 Hy-Line Brown layers at 24 wk of age were randomly assigned into 5 dietary treatments with 8 replicates of 12 birds each. Treatments included a control diet (16.49% CP), and 4 LCP, EAA balanced diets (14.05% CP) supplemented with 0, 0.114%, 0.306%, 0.498% L-Ser, respectively. Dietary Ser supplementation linearly increased hen-day egg production (HDEP; P < 0.05) and decreased feed-to-egg ratio (P < 0.05) among LCP groups from wk 6 to 10, and the optimal HDEP of layers occurred at Ser level of 0.498%. At the end of wk 10, birds in the control had higher albumen height and thick white proportion than those fed the LCP diet without Ser addition (P < 0.05), and presented a lower yolk color score than all LCP groups (P < 0.05). Among LCP groups, serum total protein and globulin contents were significantly increased by dietary Ser addition at the levels of 0.306% and 0.498% (P < 0.05), and had a linear response to the supplemental Ser levels (P < 0.05). Furthermore, dietary 0.498% Ser supplementation significantly increased serum immunoglobulin G and immunoglobulin M contents (P < 0.05) and up-regulated the expression of mucin 2, secretory immunoglobulin A, and relevant glycosyltransferases of O-glycosylation in ileal mucosa (P < 0.05). The increased expression of proinflammatory cytokines IFN-γ and IL-1β induced by LCP diets (P < 0.05) was reversed following 0.498% Ser addition (P < 0.05). Collectively, dietary CP reduction by 2.44% could maintain the productive performance of layers when it was fortified with certain EAA, though poor albumen quality, and ileal inflammation were occurred. The addition of Ser to LCP diets improved performance probably through enhancing humoral and ileal mucosal immunity and attenuating the ileal inflammation of layers.

Project description:BackgroundThe utilization of mulberry branch fiber (MF), the largest by-product of the sericulture industry, is an important issue. Supplementation with MF as a dietary fiber for poultry may serve as a useful application. However, little is known about the effects of MF on liver lipid metabolism and egg yolk fatty acid composition of laying hens and their underlying mechanisms. In this study, we performed a multi-omics investigation to explore the variations in liver lipid metabolism, egg yolk fatty acid composition, gut microbiota, and the associations among them induced by dietary MF in laying hens.ResultsDietary MF had no harmful effects on the laying performance or egg quality in laying hens. The enzyme activities associated with lipid metabolism in the liver were altered by the addition of 5% MF, resulting in reduced liver fat accumulation. Furthermore, dietary 5% MF induced the variation in the fatty acid profiles of egg yolk, and increased the polyunsaturated fatty acid (PUFA) content. We observed a significant reduction in the diversity of both gut bacteria and changes in their compositions after the addition of MF. Dietary MF significantly increased the abundance of genes involved in fatty acid biodegradation, and short-chain fatty acids biosynthesis in the gut microbiota of laying hens. The significant correlations were observed between the liver lipid metabolism enzyme activities of hepatic lipase, lipoprotein lipase, and total esterase with gut microbiota, including negative correlations with gut microbiota diversity, and multiple correlations with gut bacteria and viruses. Moreover, various correlations between the contents of PUFAs and monounsaturated fatty acids in egg yolk with the gut microbiota were obtained. Based on partial-least-squares path modeling integrated with the multi-omics datasets, we deduced the direct effects of liver enzyme activities and gut bacterial compositions on liver fat content and the roles of liver enzyme activities and gut bacterial diversity on egg yolk fatty acid composition.ConclusionsThe results indicate that dietary MF is beneficial to laying hens as it reduces the liver fat and improves egg yolk fatty acid composition through the enterohepatic axis. Project description:There are an increasing number of studies on the effect of cassava (Manihot esculenta Crantz) on performance of laying hens with inconsistent outcomes. This study, therefore, used a meta-analytic approach to examine the effect of cassava-based diets on feed intake (FI), feed conversion ratio (FCR) end production data such as hen day egg production (HDEP), egg weight (EW), egg mass (EM), Haugh unit (HU), shell weight (SW) and shell thickness (ST) in laying hens. Two hundred and three studies were identified in a search performed in PubMed, Scopus and Google Scholar databases of which thirteen studies were suitable for the meta-analysis. Subgroup and meta-regression analyses were performed to explore the responses of laying hens to dietary cassava and sources of heterogeneity, respectively using the following moderators: study continent, cassava type, cassava processing methods, inclusion level, layer strain, feeding durations, number of layers and hen's age. Data were pooled using a random-effects model and statistical analyses were performed in OpenMEE software. Results show that cassava increased FI (mean difference MD = 0.97 g/d; 95% confidence intervals (CI) 0.05, 1.90) and reduced egg weight (MD = -0.72 g; 95% CI -1.26, -0.19) and SW (MD = -0.11 g; 95% CI -0.18, -0.04) when compared to control. Laying hens fed cassava diets had HDEP, FCR, EM, ST and HU values that compared favourably with the controls. Subgroup analysis demonstrated that inclusion of cassava at ≤ 25% in layer diets had no deleterious effects on measured outcomes taking cognizance of significant heterogeneity. However, meta-regression results showed that most of the sources of heterogeneity were explained by the studied moderators. In conclusion, ≤25% of cassava (CRM and CPM) can be included in layer diets without adverse effects on feed intake, feed conversion ratio and egg production characteristics, indicating that cassava has a very bright future as an energy source in the diets of laying hens.

Project description:This study investigated the effects of multi-enzyme supplementation on various aspects of laying hens, including laying performance, egg quality, intestinal health and digestive function. In total, 384 Jingfen No.6 laying hens at 65-week-age were randomly assigned to four distinct dietary treatments: a basal diet (CON), CON supplemented with 150 g/t multi-enzyme (T1), CON with 300 g/t multi-enzyme (T2), and 600 g/t multi-enzyme (T3). A significant linear (P = 0.044) and quadratic (P = 0.014) increase was observed in the laying rate, while the feed/egg ratio exhibited a linear (P = 0.001) and quadratic (P < 0.001) decrease with increasing multi-enzyme supplementation. Additionally, linear (P < 0.05) and quadratic (P < 0.05) increases were observed in yolk rate and haugh unit with increasing levels of multi-enzyme supplementation. The trypsin activity in the duodenum and crude protein digestibility showed linear (P < 0.05) and quadratic (P < 0.05) increase with the addition of multi-enzyme. Furthermore, lipase and amylase activities in the duodenum increased quadratically (P = 0.041) and linearly (P = 0.040), respectively. Both jejunal and ileal digesta viscosities showed linear (P < 0.05) and quadratic (P < 0.05) decrease with the increasing addition of multi-enzyme. Moreover, multi-enzyme supplementation significantly increased (P < 0.05) the number of goblet cells in the intestinal of the treatment groups. The mRNA expression of Occludin-1, mucin 2 (MUC-2), large neutral amino acids transporter small subunit 1 (LAT-1) in the jejunum were significantly increased (P < 0.05) in the treatment groups (T1, T2 and T3) compared to the CON group. Additionally, the mRNA expression of solute carrier family 6-member 19 (B0AT-1) and large neutral amino acids transporter small subunit 4 (LAT-4) were significantly evaluated (P < 0.05) in the T2 and T3 groups, respectively. In conclusion, multi-enzyme supplementation enhanced digestive enzyme activities and intestinal barrier function, reduced intestinal digesta viscosity, and regulated mRNA expression of intestinal amino acid and lipid transporter genes, thereby improving crude protein digestibility and positively affecting laying performance and egg quality in hens.
