Project description:In natural listening situations, speech perception is often impaired by degraded speech sounds arriving at the ear. Contextual speech information can improve the perception of degraded speech and modify neuronal responses elicited by degraded speech. However, most studies on context effects on neural responses to degraded speech confounded lexico-semantic and sublexical cues. Here, we used fMRI to investigate how prior sublexical speech (e.g. pseudowords cues) affects neural responses to degraded sublexical speech and hence its processing and recognition. Each trial consisted of three consecutively presented pseudowords, of which the first and third were identical and degraded. The second pseudoword was always presented in clear form and either matched or did not match the degraded pseudowords. Improved speech processing through sublexical processing was associated with BOLD activation increases in frontal, temporal, and parietal regions, including the primary auditory cortex (PAC), posterior superior temporal cortex, angular gyrus, supramarginal gyrus, middle temporal cortex, and somato-motor cortex. These brain regions are part of a speech processing network and are involved in lexico-semantic processing. To further investigate the adaptive changes in PAC, we conducted a bilateral region of interest analysis on PAC subregions. PAC ROIs showed bilaterally increased activation in the match condition compared with the mismatch condition. Our results show that the perception of unintelligible degraded speech is improved and the neuronal population response is enhanced after exposure to intact sublexical cues. Furthermore, our findings indicate that the processing of clear meaningless sublexical speech preceding degraded speech could enhance the activity in the brain regions that belong to the cortical speech processing network previously reported in studies investigating lexico-semantic speech.

Project description:The human auditory system is capable of processing human speech even in situations when it has been heavily degraded, such as during noise-vocoding, when frequency domain-based cues to phonetic content are strongly reduced. This has contributed to arguments that speech processing is highly specialized and likely a de novo evolved trait in humans. Previous comparative research has demonstrated that a language competent chimpanzee was also capable of recognizing degraded speech, and therefore that the mechanisms underlying speech processing may not be uniquely human. However, to form a robust reconstruction of the evolutionary origins of speech processing, additional data from other closely related ape species is needed. Specifically, such data can help disentangle whether these capabilities evolved independently in humans and chimpanzees, or if they were inherited from our last common ancestor. Here we provide evidence of processing of highly varied (degraded and computer-generated) speech in a language competent bonobo, Kanzi. We took advantage of Kanzi's existing proficiency with touchscreens and his ability to report his understanding of human speech through interacting with arbitrary symbols called lexigrams. Specifically, we asked Kanzi to recognise both human (natural) and computer-generated forms of 40 highly familiar words that had been degraded (noise-vocoded and sinusoidal forms) using a match-to-sample paradigm. Results suggest that-apart from noise-vocoded computer-generated speech-Kanzi recognised both natural and computer-generated voices that had been degraded, at rates significantly above chance. Kanzi performed better with all forms of natural voice speech compared to computer-generated speech. This work provides additional support for the hypothesis that the processing apparatus necessary to deal with highly variable speech, including for the first time in nonhuman animals, computer-generated speech, may be at least as old as the last common ancestor we share with bonobos and chimpanzees.

Project description:Co-speech hand gestures influence language comprehension. The present experiment explored what part of the visual processing system is optimized for processing these gestures. Participants viewed short video clips of speech and gestures (e.g., a person saying "chop" or "twist" while making a chopping gesture) and had to determine whether the two modalities were congruent or incongruent. Gesture videos were designed to stimulate the parvocellular or magnocellular visual pathways by filtering out low or high spatial frequencies (HSF versus LSF) at two levels of degradation severity (moderate and severe). Participants were less accurate and slower at processing gesture and speech at severe versus moderate levels of degradation. In addition, they were slower for LSF versus HSF stimuli, and this difference was most pronounced in the severely degraded condition. However, exploratory item analyses showed that the HSF advantage was modulated by the range of motion and amount of motion energy in each video. The results suggest that hand gestures exploit a wide range of spatial frequencies, and depending on what frequencies carry the most motion energy, parvocellular or magnocellular visual pathways are maximized to quickly and optimally extract meaning.

Project description:The present study aimed to investigate the effects of degraded speech perception and binaural unmasking using functional near-infrared spectroscopy (fNIRS). Normal hearing listeners were tested when attending to unprocessed or vocoded speech, presented to the left ear at two speech-to-noise ratios (SNRs). Additionally, by comparing monaural versus diotic masker noise, we measured binaural unmasking. Our primary research question was whether the prefrontal cortex and temporal cortex responded differently to varying listening configurations. Our a priori regions of interest (ROIs) were located at the left dorsolateral prefrontal cortex (DLPFC) and auditory cortex (AC). The left DLPFC has been reported to be involved in attentional processes when listening to degraded speech and in spatial hearing processing, while the AC has been reported to be sensitive to speech intelligibility. Comparisons of cortical activity between these two ROIs revealed significantly different fNIRS response patterns. Further, we showed a significant and positive correlation between self-reported task difficulty levels and fNIRS responses in the DLPFC, with a negative but non-significant correlation for the left AC, suggesting that the two ROIs played different roles in effortful speech perception. Our secondary question was whether activity within three sub-regions of the lateral PFC (LPFC) including the DLPFC was differentially affected by varying speech-noise configurations. We found significant effects of spectral degradation and SNR, and significant differences in fNIRS response amplitudes between the three regions, but no significant interaction between ROI and speech type, or between ROI and SNR. When attending to speech with monaural and diotic noises, participants reported the latter conditions being easier; however, no significant main effect of masker condition on cortical activity was observed. For cortical responses in the LPFC, a significant interaction between SNR and masker condition was observed. These findings suggest that binaural unmasking affects cortical activity through improving speech reception threshold in noise, rather than by reducing effort exerted.

Project description:Individuals with Rett syndrome have greatly impaired speech and language abilities. Auditory brainstem responses to sounds are normal, but cortical responses are highly abnormal. In this study, we used the novel rat Mecp2 knockout model of Rett syndrome to document the neural and behavioral processing of speech sounds. We hypothesized that both speech discrimination ability and the neural response to speech sounds would be impaired in Mecp2 rats. We expected that extensive speech training would improve speech discrimination ability and the cortical response to speech sounds. Our results reveal that speech responses across all four auditory cortex fields of Mecp2 rats were hyperexcitable, responded slower, and were less able to follow rapidly presented sounds. While Mecp2 rats could accurately perform consonant and vowel discrimination tasks in quiet, they were significantly impaired at speech sound discrimination in background noise. Extensive speech training improved discrimination ability. Training shifted cortical responses in both Mecp2 and control rats to favor the onset of speech sounds. While training increased the response to low frequency sounds in control rats, the opposite occurred in Mecp2 rats. Although neural coding and plasticity are abnormal in the rat model of Rett syndrome, extensive therapy appears to be effective. These findings may help to explain some aspects of communication deficits in Rett syndrome and suggest that extensive rehabilitation therapy might prove beneficial.

Project description:Individuals use semantic expectancy - applying conceptual and linguistic knowledge to speech input - to improve the accuracy and speed of language comprehension. This study tested how adults use semantic expectancy in quiet and in the presence of speech-shaped broadband noise at -7 and -12 dB signal-to-noise ratio. Twenty-four adults (22.1 ± 3.6 years, mean ±SD) were tested on a four-alternative-forced-choice task whereby they listened to sentences and were instructed to select an image matching the sentence-final word. The semantic expectancy of the sentences was unrelated to (neutral), congruent with, or conflicting with the acoustic target. Congruent expectancy improved accuracy and conflicting expectancy decreased accuracy relative to neutral, consistent with a theory where expectancy shifts beliefs toward likely words and away from unlikely words. Additionally, there were no significant interactions of expectancy and noise level when analyzed in log-odds, supporting the predictions of ideal observer models of speech perception.

Project description:ObjectiveMusic and speech are complex signals containing regularities in how they unfold in time. Similarities between music and speech/language in terms of their auditory features, rhythmic structure, and hierarchical structure have led to a large body of literature suggesting connections between the two domains. However, the precise underlying mechanisms behind this connection remain to be elucidated.MethodIn this theoretical review article, we synthesize previous research and present a framework of potentially shared neural mechanisms for music and speech rhythm processing. We outline structural similarities of rhythmic signals in music and speech, synthesize prominent music and speech rhythm theories, discuss impaired timing in developmental speech and language disorders, and discuss music rhythm training as an additional, potentially effective therapeutic tool to enhance speech/language processing in these disorders.ResultsWe propose the processing rhythm in speech and music (PRISM) framework, which outlines three underlying mechanisms that appear to be shared across music and speech/language processing: Precise auditory processing, synchronization/entrainment of neural oscillations to external stimuli, and sensorimotor coupling. The goal of this framework is to inform directions for future research that integrate cognitive and biological evidence for relationships between rhythm processing in music and speech.ConclusionThe current framework can be used as a basis to investigate potential links between observed timing deficits in developmental disorders, impairments in the proposed mechanisms, and pathology-specific deficits which can be targeted in treatment and training supporting speech therapy outcomes. Project description:IntroductionCochlear implants (CIs) are the treatment of choice for severe to profound hearing loss. Variability in CI outcomes remains despite advances in technology and is attributed in part to differences in cortical processing. Studying these differences in CI users is technically challenging. Spectrally degraded stimuli presented to normal-hearing individuals approximate input to the central auditory system in CI users. This study used intracranial electroencephalography (iEEG) to investigate cortical processing of spectrally degraded speech.MethodsParticipants were adult neurosurgical epilepsy patients. Stimuli were utterances /aba/ and /ada/, spectrally degraded using a noise vocoder (1-4 bands) or presented without vocoding. The stimuli were presented in a two-alternative forced choice task. Cortical activity was recorded using depth and subdural iEEG electrodes. Electrode coverage included auditory core in posteromedial Heschl's gyrus (HGPM), superior temporal gyrus (STG), ventral and dorsal auditory-related areas, and prefrontal and sensorimotor cortex. Analysis focused on high gamma (70-150 Hz) power augmentation and alpha (8-14 Hz) suppression.ResultsChance task performance occurred with 1-2 spectral bands and was near-ceiling for clear stimuli. Performance was variable with 3-4 bands, permitting identification of good and poor performers. There was no relationship between task performance and participants demographic, audiometric, neuropsychological, or clinical profiles. Several response patterns were identified based on magnitude and differences between stimulus conditions. HGPM responded strongly to all stimuli. A preference for clear speech emerged within non-core auditory cortex. Good performers typically had strong responses to all stimuli along the dorsal stream, including posterior STG, supramarginal, and precentral gyrus; a minority of sites in STG and supramarginal gyrus had a preference for vocoded stimuli. In poor performers, responses were typically restricted to clear speech. Alpha suppression was more pronounced in good performers. In contrast, poor performers exhibited a greater involvement of posterior middle temporal gyrus when listening to clear speech.DiscussionResponses to noise-vocoded speech provide insights into potential factors underlying CI outcome variability. The results emphasize differences in the balance of neural processing along the dorsal and ventral stream between good and poor performers, identify specific cortical regions that may have diagnostic and prognostic utility, and suggest potential targets for neuromodulation-based CI rehabilitation strategies.

Project description:Cochlear implants provide good speech discrimination ability despite highly limited amount of information they transmit compared with normal cochlea. Noise vocoded speech, simulating cochlear implants in normal hearing listeners, have demonstrated that spectrally and temporally degraded speech contains sufficient cues to provide accurate speech discrimination. We hypothesized that neural activity patterns generated in the primary auditory cortex by spectrally and temporally degraded speech sounds will account for the robust behavioral discrimination of speech. We examined the behavioral discrimination of noise vocoded consonants and vowels by rats and recorded neural activity patterns from rat primary auditory cortex (A1) for the same sounds. We report the first evidence of behavioral discrimination of degraded speech sounds by an animal model. Our results show that rats are able to accurately discriminate both consonant and vowel sounds even after significant spectral and temporal degradation. The degree of degradation that rats can tolerate is comparable to human listeners. We observed that neural discrimination based on spatiotemporal patterns (spike timing) of A1 neurons is highly correlated with behavioral discrimination of consonants and that neural discrimination based on spatial activity patterns (spike count) of A1 neurons is highly correlated with behavioral discrimination of vowels. The results of the current study indicate that speech discrimination is resistant to degradation as long as the degraded sounds generate distinct patterns of neural activity.

Project description:Previous studies have shown that at moderate levels of spectral degradation, semantic predictability facilitates language comprehension. It is argued that when speech is degraded, listeners have narrowed expectations about the sentence endings; i.e., semantic prediction may be limited to only most highly predictable sentence completions. The main objectives of this study were to (i) examine whether listeners form narrowed expectations or whether they form predictions across a wide range of probable sentence endings, (ii) assess whether the facilitatory effect of semantic predictability is modulated by perceptual adaptation to degraded speech, and (iii) use and establish a sensitive metric for the measurement of language comprehension. For this, we created 360 German Subject-Verb-Object sentences that varied in semantic predictability of a sentence-final target word in a graded manner (high, medium, and low) and levels of spectral degradation (1, 4, 6, and 8 channels noise-vocoding). These sentences were presented auditorily to two groups: One group (n =48) performed a listening task in an unpredictable channel context in which the degraded speech levels were randomized, while the other group (n =50) performed the task in a predictable channel context in which the degraded speech levels were blocked. The results showed that at 4 channels noise-vocoding, response accuracy was higher in high-predictability sentences than in the medium-predictability sentences, which in turn was higher than in the low-predictability sentences. This suggests that, in contrast to the narrowed expectations view, comprehension of moderately degraded speech, ranging from low- to high- including medium-predictability sentences, is facilitated in a graded manner; listeners probabilistically preactivate upcoming words from a wide range of semantic space, not limiting only to highly probable sentence endings. Additionally, in both channel contexts, we did not observe learning effects; i.e., response accuracy did not increase over the course of experiment, and response accuracy was higher in the predictable than in the unpredictable channel context. We speculate from these observations that when there is no trial-by-trial variation of the levels of speech degradation, listeners adapt to speech quality at a long timescale; however, when there is a trial-by-trial variation of the high-level semantic feature (e.g., sentence predictability), listeners do not adapt to low-level perceptual property (e.g., speech quality) at a short timescale.
