Stepwise Self-Assembly and Dynamic Exchange of Supramolecular Snowflakes.

ABSTRACT: Snowflake, a highly symmetrical hexagram figure, is challenging to be expressed by chemistry/supramolecular chemistry due to the complex structure. Herein, we have constructed super snowflake supramolecules using terpyridine (tpy)-based metal-organic building blocks with and connectivities through stepwise strategies in high yield. The structures were characterized by multi-dimensional mass spectrometry and multi-dimensional NMR spectrometry. In order to address the stability/tolerance of our designed super snowflake structures, ligand exchange behaviors between different supramolecules with various arm length were fully investigated by mass spectrometry. The study revealed that three modes could exist in such binary systems, including full exchange, partial exchange and self-sorting (no exchange) depending on the length difference of ligands.

Stepwise Self-Assembly and Dynamic Exchange of Supramolecular Nanocages Based on Terpridine Building Blocks.

Project description:Coordination-driven self-assembly as a powerful bottom-up approach has been extensively used to construct multifarious supramolecular architectures with increasing complexity and functionality. Due to the unique cavity structures and precisely controllable dimensions, 3D supramolecules display unprecedented properties and functions in catalysis, sensing, gas storage, and smart materials. Herein, we have built two 3D nanocages with different sizes by changing the length of the organic ligand arms. The structures were characterized by 1D and 2D NMR spectroscopy, electrospray ionization-mass spectrometry (ESI-MS), traveling wave ion mobility-mass spectrometry (TWIM-MS), gradient tandem-mass spectrometry (gMS2 ), and transmission electron microscopy (TEM). Furthermore, the intermolecular dynamic exchange of two 3D nanocages was conducted to construct a series of hybrid 3D structures as evidenced by mass spectrometry.

Dynamic microfluidic control of supramolecular peptide self-assembly.

Project description:The dynamic nature of supramolecular polymers has a key role in their organization. Yet, the manipulation of their dimensions and polarity remains a challenge. Here, the minimalistic diphenylalanine building block was applied to demonstrate control of nano-assemblies growth and shrinkage using microfluidics. To fine-tune differential local environments, peptide nanotubes were confined by micron-scale pillars and subjected to monomer flows of various saturation levels to control assembly and disassembly. The small-volume device allows the rapid adjustment of conditions within the system. A simplified kinetic model was applied to calculate parameters of the growth mechanism. Direct real-time microscopy analysis revealed that different peptide derivatives show unidirectional or bidirectional axial dimension variation. Atomistic simulations show that unidirectional growth is dictated by the differences in the axial ends, as observed in the crystalline order of symmetry. This work lays foundations for the rational control of nano-materials dimensions for applications in biomedicine and material science.

Dynamic self-assembly of supramolecular catalysts from precision macromolecules.

Project description:We show the emergence of strong catalytic activity at low concentrations in dynamic libraries of complementary sequence-defined oligomeric chains comprising pendant functional catalytic groups and terminal recognition units. In solution, the dynamic constitutional library created from pairs of such complementary oligomers comprises free oligomers, self-assembled di(oligomeric) macrocycles, and a virtually infinite collection of linear poly(oligomeric) chains. We demonstrate, on an exemplary catalytic system requiring the cooperation of no less than five chemical groups, that supramolecular di(oligomeric) macrocycles exhibit a catalytic turnover frequency ca. 20 times larger than the whole collection of linear poly(oligomers) and free chains. Molecular dynamics simulations and network analysis indicate that self-assembled supramolecular di(oligomeric) macrocycles are stabilized by different interactions, among which chain end pairing. We mathematically model the catalytic properties of such complex dynamic libraries with a small set of physically relevant parameters, which provides guidelines for the synthesis of oligomers capable to self-assemble into functionally-active supramolecular macrocycles over a larger range of concentrations.

Dynamic Surface Interactions Enable the Self-Assembly of Perfect Supramolecular Crystals.

Project description:Supramolecular crystals arise from noncovalent interactions between macromonomers and allow for the engineering of dynamic functional materials. For two-dimensional (2D) crystals, the substrate surface can induce the formation of new polymorphs not available in solution, adding a layer of complexity to the supramolecular self-assembly process. Despite extensive studies on the 2D self-assembly of supramolecular crystals, unknowns remain regarding substrate-monomer interactions and the effects on network self-assembly and defect repair. Here, we used a DNA-mica model system to modulate and understand the impact of substrate-monomer interactions on the crystalline order. We controlled the surface interactions by tuning the Mg2+ concentration, varying the divalent cation type, and adjusting the relative concentration of divalent and monovalent cations. The competition between monovalent and divalent cations yielded nearly defect-free crystals with minimal polygon defects. These findings highlight the critical role of surface interactions in achieving high crystalline order, which is essential for optimizing the efficiency and performance of supramolecular functional nanomaterials.

Direct and quantitative characterization of dynamic ligand exchange between coordination-driven self-assembled supramolecular polygons.

Project description:The direct observation of dynamic ligand exchange between Pt-N coordination-driven self-assembled supramolecular polygons (triangles and rectangles) has been achieved using stable (1)H/(2)D isotope labeling of the pyridyl donors and electrospray ionization mass spectrometry combined with NMR spectroscopy. Both the thermodynamic and kinetic aspects of such exchange processes have been established on the basis of quantitative mass spectral results. Further investigation has shown that the exchange is highly dependent on experimental conditions such as temperature, solvent, and the counteranions.

Self-assembly of a supramolecular hexagram and a supramolecular pentagram.

Project description:Five- and six-pointed star structures occur frequently in nature as flowers, snow-flakes, leaves and so on. These star-shaped patterns are also frequently used in both functional and artistic man-made architectures. Here following a stepwise synthesis and self-assembly approach, pentagonal and hexagonal metallosupramolecules possessing star-shaped motifs were prepared based on the careful design of metallo-organic ligands (MOLs). In the MOL design and preparation, robust ruthenium-terpyridyl complexes were employed to construct brominated metallo-organic intermediates, followed by a Suzuki coupling reaction to achieve the required ensemble. Ligand LA (VRu2+X, V=bisterpyridine, X=tetraterpyridine, Ru=Ruthenium) was initially used for the self-assembly of an anticipated hexagram upon reaction with Cd2+ or Fe2+; however, unexpected pentagonal structures were formed, that is, [Cd5LA5]30+ and [Fe5LA5]30+. In our redesign, LB [V(Ru2+X)2] was synthesized and treated with 60° V-shaped bisterpyridine (V) and Cd2+ to create hexagonal hexagram [Cd12V3LB3]36+ along with traces of the triangle [Cd3V3]6+. Finally, a pure supramolecular hexagram [Fe12V3LB3]36+ was successfully isolated in a high yield using Fe2+ with a higher assembly temperature.

Controlling Exchange Pathways in Dynamic Supramolecular Polymers by Controlling Defects.

Project description:Supramolecular fibers composed of monomers that self-assemble directionally via noncovalent interactions are ubiquitous in nature, and of great interest in chemistry. In these structures, the constitutive monomers continuously exchange in-and-out the assembly according to a well-defined supramolecular equilibrium. However, unraveling the exchange pathways and their molecular determinants constitutes a nontrivial challenge. Here, we combine coarse-grained modeling, enhanced sampling, and machine learning to investigate the key factors controlling the monomer exchange pathways in synthetic supramolecular polymers having an intrinsic dynamic behavior. We demonstrate how the competition of directional vs. nondirectional interactions between the monomers controls the creation/annihilation of defects in the supramolecular polymers, from where monomers exchange proceeds. This competition determines the exchange pathway, dictating whether a fiber statistically swaps monomers from the tips or from all along its length. Finally, thanks to their generality, our models allow the investigation of molecular approaches to control the exchange pathways in these dynamic assemblies.

Supramolecular Self-Assembly of Engineered Polyproline Helices.

Project description:The ability to rationally design biomaterials to form desired supramolecular constructs presents an ever-growing research field, with many burgeoning works within recent years providing exciting results; however, there exists a broad expanse of promising avenues of research yet to be investigated. As such we have set out to make use of the polyproline helix as a rigid, tunable, and chiral ligand for the rational design and synthesis of supramolecular constructs. In this investigation, we show how an oligoproline tetramer can be specifically designed and functionalized, allowing predictable tuning of supramolecular interactions, to engineer the formation of supramolecular peptide frameworks with varying properties and, consequently, laying the groundwork for further studies utilizing the polyproline helix, with the ability to design desired supramolecular structures containing these peptide building blocks, having tunable structural features and functionalities.

Multicomponent supramolecular systems: self-organization in coordination-driven self-assembly.

Project description:The self-organization of multicomponent supramolecular systems involving a variety of two-dimensional (2 D) polygons and three-dimensional (3 D) cages is presented. Nine self-organizing systems, SS(1)-SS(9), have been studied. Each involves the simultaneous mixing of organoplatinum acceptors and pyridyl donors of varying geometry and their selective self-assembly into three to four specific 2 D (rectangular, triangular, and rhomboid) and/or 3 D (triangular prism and distorted and nondistorted trigonal bipyramidal) supramolecules. The formation of these discrete structures is characterized using NMR spectroscopy and electrospray ionization mass spectrometry (ESI-MS). In all cases, the self-organization process is directed by: 1) the geometric information encoded within the molecular subunits and 2) a thermodynamically driven dynamic self-correction process. The result is the selective self-assembly of multiple discrete products from a randomly formed complex. The influence of key experimental variables--temperature and solvent--on the self-correction process and the fidelity of the resulting self-organization systems is also described.

Introducing Seven Transition Metal Ions into Terpyridine-Based Supramolecules: Self-Assembly and Dynamic Ligand Exchange Study.

Project description:In coordination-driven self-assembly, 2,2':6',2″-terpyridine (tpy) has gained extensive attention in constructing supramolecular architectures on the basis of ⟨tpy-M-tpy⟩ connectivity. In direct self-assembly of large discrete structures, however, the metal ions were mainly limited to Cd(II), Zn(II), and Fe(II) ions. Herein, we significantly broaden the spectrum of metal ions with seven divalent transition metal ions M(II) (M = Mn, Fe, Co, Ni, Cu, Zn, Cd) to assemble a series of supramolecular fractals. In particular, Mn(II), Co(II), Ni(II), and Cu(II) were reported for the first time to form such large and discrete structures with ⟨tpy-M-tpy⟩ connectivity. In addition, the structural stabilities of those supramolecules in the gas phase and the kinetics of the ligand exchange process in solution were investigated using mass spectrometry. Such a fundamental study gave the relative order of structural stability in the gas phase and revealed the inertness of coordination in solution depending on the metal ions. Those results would guide the future study in tpy-based supramolecular chemistry in terms of self-assembly, characterization, property, and application.
