ABSTRACT: Nowadays, macroporous polymer monoliths represent widely used stationary phases for a number of dynamic interphase mass exchange processes such as high-performance liquid chromatography, gas chromatography, electrochromatography, solid-phase extraction, and flow-through solid-state biocatalysis. This review represents the first summary in the field of current achievements on the preparation of macroporous polymer monolithic layers, as well as their application as solid phases for thin-layer chromatography and different kinds of microarray.

Self-standing zeolite foam monoliths with hierarchical micro-meso-macroporous structures.

Project description:The zeolite monoliths were synthesized by a facile polymer scaffold template assisted hydrothermal method. The selected foam-shaped template of a polyurethane (PU) foam monolith, was used to prepare the self-standing zeolite foam (ZF) monolithic materials. The obtained ZF products can preserve the same size, shape and macroporous network structure of the original PU foam scaffold template, although the zeolite nano-crystallites had been fully substituted for the PU template to form the new skeleton struts and walls. The as-synthesized ZF products demonstrated abundant hierarchical porosity (involving triple micro-, meso- and macropores). Meanwhile, compared with the conventional zeolite powders, the self-standing ZF monolithic materials exhibited greater total pore volume and nearly three times higher mesopore volume, suggesting wider applications as catalysts, catalyst supports and adsorbents in industry.

Methacrylate Polymer Monoliths for Separation Applications.

Project description:This review summarizes the development of methacrylate-based polymer monoliths for separation science applications. An introduction to monoliths is presented, followed by the preparation methods and characteristics specific to methacrylate monoliths. Both traditional chemical based syntheses and emerging additive manufacturing methods are presented along with an analysis of the different types of functional groups, which have been utilized with methacrylate monoliths. The role of methacrylate based porous materials in separation science in industrially important chemical and biological separations are discussed, with particular attention given to the most recent developments and challenges associated with these materials. While these monoliths have been shown to be useful for a wide variety of applications, there is still scope for exerting better control over the porous architectures and chemistries obtained from the different fabrication routes. Conclusions regarding this previous work are drawn and an outlook towards future challenges and potential developments in this vibrant research area are presented. Discussed in particular are the potential of additive manufacturing for the preparation of monolithic structures with pre-defined multi-scale porous morphologies and for the optimization of surface reactive chemistries.

Preparation of macroporous transition metal hydroxide monoliths via a sol-gel process accompanied by phase separation.

Project description:Three-dimensional transition metal hydroxide monoliths were facilely fabricated by a sol-gel process accompanied by phase separation in the presence of polyacrylic acid (PAA) and propylene oxide (PO). In the typical ZnCl2-PAA-PO system, PAA is used as a phase separation inducer as well as a framework former to control the phase separation and the formation of macrostructures, whereas PO works as a proton scavenger to initiate the gelation of the system and freeze the macrostructures. Appropriate amount of PAA, PO and solvents allow the formation of zinc (Zn) hydroxide monolith with cocontinuous skeletons and interconnected macropores, and the construction mechanism and characteristics of macrostructure are also investigated. The resultant dried gels are amorphous Zn hydroxide monolith with a narrow macropore size distribution (~1 μm). This approach is further used to successfully prepare macroporous single or binary composite transition metal hydroxide monoliths.

Nanostructured Polymer Thin Films Fabricated with Brush-based Layer-by-Layer Self-assembly for Site-selective Construction and Drug release.

Project description:Layer-by-Layer (LbL) self-assembly has been investigated for several decades. However, the conventional LbL method has performance problems on the chair-side caused by its cumbersome and time-consuming process. Thus, we investigate a new LbL self-assembly technique for the fast and high efficient preparation process based on the brush. The multilayer films fabricated by simple sequential brushing of polyelectrolyte solutions are compared to the classical dipping method. We characterize the multilayer films by characteristics such as their morphology and thickness, and compare them against those of the classic method by profilometry, atomic force microscopy. We prepare multilayer films with biocompatible polyelectrolytes, chitosan, and alginate incorporated with a hydrophobic drug carrier. For the drug carrier, a poly(ethylene glycol)-block-poly(ε-caprolactone) (PEG-b-PCL) block copolymer is introduced to construct micelles containing dexamethasone, which is a well-known osteogenesis-inducing drug. The hydrogen bonding behavior between adjacent layers and micelles is investigated by Fourier transform infrared spectroscopy. Additionally, we analyze the release profiles, degradation profiles and toxicity of the multilayer films for biomedical applications. From these results, we can identify the brushing LbL method as a reliable and more efficient multilayer film-construction process compared to conventional dipping LbL, especially for practical applications in dental and clinical situations.

An advanced structural characterization of templated meso-macroporous carbon monoliths by small- and wide-angle scattering techniques.

Project description:This study is dedicated to link the nanoscale pore space of carbon materials, prepared by hard-templating of meso-macroporous SiO2 monoliths, to the corresponding nanoscale polyaromatic microstructure using two different carbon precursors wthat generally exhibit markedly different carbonization properties, i.e., a graphitizable pitch and a non-graphitizable resin. The micro- and mesoporosity of these monolithic carbon materials was studied by the sorption behavior of a relatively large organic molecule (p-xylene) in comparison to typical gas adsorbates (Ar). In addition, to obtain a detailed view on the nanopore space small-angle neutron scattering (SANS) combined with in situ physisorption was applied, using deuterated p-xylene (DPX) as a contrast-matching agent in the neutron scattering process. The impact of the carbon precursor on the structural order on an atomic scale in terms of size and disorder of the carbon microstructure, on the nanopore structure, and on the template process is analyzed by special evaluation approaches for SANS and wide-angle X-ray scattering (WAXS). The WAXS analysis shows that the pitch-based monolithic material exhibits a more ordered microstructure consisting of larger graphene stacks and similar graphene layer sizes compared to the monolithic resin. Another major finding is the discrepancy in the accessible micro/mesoporosity between Ar and deuterated p-xylene that found for the two different carbon precursors, pitch and resin, which can be regarded as representative carbon precursors in general. These differences essentially indicate that physisorption using probe gases such as Ar or N2 can provide misleading parameters if to be used to appraise the accessibility of the nanoscale pore space.

Macroporous epoxy-based monoliths for rapid quantification of Pseudomonas aeruginosa by adsorption elution method optimized for qPCR.

Project description:Pseudomonas aeruginosa contaminations in tap water systems have caused severe health problems in both hospital and household settings. To ensure fast and reliable detection, culture-independent methods are recommendable. However, the typically low cell number in water samples requires sample enrichment prior to analysis. Therefore, we developed and optimized an adsorption elution method using monolithic adsorption filtration and subsequent centrifugal ultrafiltration that can be combined with culture-independent detection methods. The principle of adsorption of Pseudomonas aeruginosa by hydrophobic and ionic interactions was studied in modified epoxy-based monoliths. Optimized conditions (5-L initial sample volume at pH 3 filtered for 30 min through hydrolyzed monoliths (MAF-OH) and eluted with beef extract glycine buffer at pH 9.5) achieved a recovery of 67.1 ± 1.2% and a concentration factor of 103. For the first time, we therefore present a culture-independent approach for rapid enrichment and subsequent molecular biological quantification of P. aeruginosa by qPCR from tap water samples by monolithic adsorption filtration. The total enrichment and quantification process takes 4 h. This work further stresses the versatility of the monolithic adsorption filtration and its possibilities as a concentration tool for culture-independent analytics of pathogenic bacteria in the environment.Graphical abstract.

Macroporous Epoxy-Based Monoliths Functionalized with Anti-CD63 Nanobodies for Effective Isolation of Extracellular Vesicles in Urine.

Project description:Extracellular vesicles (EVs) have enormous potential for the implementation of liquid biopsy and as effective drug delivery means, but the fulfilment of these expectations requires overcoming at least two bottlenecks relative to their purification, namely the finalization of reliable and affordable protocols for: (i) EV sub-population selective isolation and (ii) the scalability of their production/isolation from complex biological fluids. In this work, we demonstrated that these objectives can be achieved by a conceptually new affinity chromatography platform composed of a macroporous epoxy monolith matrix functionalized with anti-CD63 nanobodies with afflux of samples and buffers regulated through a pump. Such a system successfully captured and released integral EVs from urine samples and showed negligible unspecific binding for circulating proteins. Additionally, size discrimination of eluted EVs was achieved by different elution approaches (competitive versus pH-dependent). The physical characteristics of monolith material and the inexpensive production of recombinant nanobodies make scaling-up the capture unit feasible and affordable. Additionally, the availability of nanobodies for further specific EV biomarkers will allow for the preparation of monolithic affinity filters selective for different EV subclasses.

Room temperature rubbing for few-layer two-dimensional thin flakes directly on flexible polymer substrates.

Project description:The functional layers of few-layer two-dimensional (2-D) thin flakes on flexible polymers for stretchable applications have attracted much interest. However, most fabrication methods are "indirect" processes that require transfer steps. Moreover, previously reported "transfer-free" methods are only suitable for graphene and not for other few-layer 2-D thin flakes. Here, a friction based room temperature rubbing method is proposed for fabricating different types of few-layer 2-D thin flakes (graphene, hexagonal boron nitride (h-BN), molybdenum disulphide (MoS₂), and tungsten disulphide (WS₂)) on flexible polymer substrates. Commercial 2-D raw materials (graphite, h-BN, MoS₂, and WS₂) that contain thousands of atom layers were used. After several minutes, different types of few-layer 2-D thin flakes were fabricated directly on the flexible polymer substrates by rubbing procedures at room temperature and without any transfer step. These few-layer 2-D thin flakes strongly adhere to the flexible polymer substrates. This strong adhesion is beneficial for future applications.

Optimal acceleration voltage for near-atomic resolution imaging of layer-stacked 2D polymer thin films.

Project description:Despite superb instrumental resolution in modern transmission electron microscopes (TEM), high-resolution imaging of organic two-dimensional (2D) materials is a formidable task. Here, we present that the appropriate selection of the incident electron energy plays a crucial role in reducing the gap between achievable resolution in the image and the instrumental limit. Among a broad range of electron acceleration voltages (300 kV, 200 kV, 120 kV, and 80 kV) tested, we found that the highest resolution in the HRTEM image is achieved at 120 kV, which is 1.9 Å. In two imine-based 2D polymer thin films, unexpected molecular interstitial defects were unraveled. Their structural nature is identified with the aid of quantum mechanical calculations. Furthermore, the increased image resolution and enhanced image contrast at 120 kV enabled the detection of functional groups at the pore interfaces. The experimental setup has also been employed for an amorphous organic 2D material.

Thin-layer polymer wrapped enzymes encapsulated in hierarchically mesoporous silica with high activity and enhanced stability.

Project description:A novel soft-hard cooperative approach was developed to synthesize bioactive mesoporous composite by pre-wrapping Penicillin G amidase with poly(acrylaimde) nanogel skin and subsequently incorporating such Penicillin G amidase nanocapsules into hierarchically mesoporous silica. The as-received bioactive mesoporous composite exhibited comparable activity and extraordinarily high stability in comparison with native Penicillin G amidase and could be used repetitively in the water-medium hydrolysis of penicillin G potassium salt. Furthermore, this strategy could be extended to the synthesis of multifunctional bioactive mesoporous composite by simultaneously introducing glucose oxidase nanocapsules and horseradish peroxidase nanocapsules into hierarchically mesoporous silica, which demonstrated a synergic effect in one-pot tandem oxidation reaction. Improvements in the catalytic performances were attributed to the combinational unique structure from soft polymer skin and hard inorganic mesoporous silica shell, which cooperatively helped enzyme molecules to retain their appropriate geometry and simultaneously decreased the enzyme-support negative interaction and mass transfer limitation under heterogeneous conditions.
