Project description:The COVID-19 pandemic has led to considerable morbidity and mortality, and consumed enormous resources (e.g. energy) to control and prevent the disease. It is crucial to balance infection risk and energy consumption when reducing the spread of infection. In this study, a quantitative human, behavior-based, infection risk-energy consumption model for different indoor environments was developed. An optimal balance point for each indoor environment can be obtained using the anti-problem method. For this study we selected Wangjing Block, one of the most densely populated places in Beijing, as an example. Under the current ventilation standard (30 m3/h/person), prevention and control of the COVID-19 pandemic would be insufficient because the basic reproduction number (R0 ) for students, workers and elders are greater than 1. The optimal required fresh air ventilation rates in most indoor environments are near or below 60 m3/h/person, after considering the combined effects of multiple mitigation measures. In residences, sports buildings and restaurants, the demand for fresh air ventilation rate is relatively high. After our global optimization of infection risk control (R0 ≤ 1), energy consumption can be reduced by 13.7% and 45.1% on weekdays and weekends, respectively, in contrast to a strategy of strict control (R0 = 1 for each indoor environment).

Project description:The role of aerosolized SARS-CoV-2 viruses in airborne transmission of COVID-19 has been debated. The aerosols are transmitted through breathing and vocalization by infectious subjects. Some authors state that this represents the dominant route of spreading, while others dismiss the option. Here we present an adjustable algorithm to estimate the infection risk for different indoor environments, constrained by published data of human aerosol emissions, SARS-CoV-2 viral loads, infective dose and other parameters. We evaluate typical indoor settings such as an office, a classroom, choir practice, and a reception/party. Our results suggest that aerosols from highly infective subjects can effectively transmit COVID-19 in indoor environments. This "highly infective" category represents approximately 20% of the patients who tested positive for SARS-CoV-2. We find that "super infective" subjects, representing the top 5-10% of subjects with a positive test, plus an unknown fraction of less-but still highly infective, high aerosol-emitting subjects-may cause COVID-19 clusters (>10 infections). In general, active room ventilation and the ubiquitous wearing of face masks (i.e., by all subjects) may reduce the individual infection risk by a factor of five to ten, similar to high-volume, high-efficiency particulate air (HEPA) filtering. A particularly effective mitigation measure is the use of high-quality masks, which can drastically reduce the indoor infection risk through aerosols.

Project description:COVID-19 transmits by droplets generated from surfaces of airway mucus during processes of respiration within hosts infected by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) virus. We studied respiratory droplet generation and exhalation in human and nonhuman primate subjects with and without COVID-19 infection to explore whether SARS-CoV-2 infection, and other changes in physiological state, translate into observable evolution of numbers and sizes of exhaled respiratory droplets in healthy and diseased subjects. In our observational cohort study of the exhaled breath particles of 194 healthy human subjects, and in our experimental infection study of eight nonhuman primates infected, by aerosol, with SARS-CoV-2, we found that exhaled aerosol particles vary between subjects by three orders of magnitude, with exhaled respiratory droplet number increasing with degree of COVID-19 infection and elevated BMI-years. We observed that 18% of human subjects (35) accounted for 80% of the exhaled bioaerosol of the group (194), reflecting a superspreader distribution of bioaerosol analogous to a classical 20:80 superspreader of infection distribution. These findings suggest that quantitative assessment and control of exhaled aerosol may be critical to slowing the airborne spread of COVID-19 in the absence of an effective and widely disseminated vaccine.

Project description:This paper describes the in situ monitoring of indoor air quality (IAQ) in two dwellings, using low-cost IAQ sensors to provide high-density temporal and spatial data. IAQ measurements were conducted over 2-week periods in the kitchen and bedroom of each home during the winter, spring, and summer seasons, characterized by different outside parameters, that were simultaneously measured. The mean indoor PM2.5 concentrations were about 15 μg m-3 in winter, they dropped to values close to 10 μg m-3 in spring and increased to levels of about 13 μg m-3 in summer. During the winter campaign, indoor PM2.5 was found mainly associated with particle penetration inside the rooms from outdoors, because of the high outdoor PM2.5 levels in the season. Such pollution winter episodes occur frequently in the study region, due to the combined contributions of strong anthropogenic emissions and stable atmospheric conditions. The concentrations of indoor volatile organic compounds (VOCs) and CO2 increased with the number of occupants (humans and pets), as likely associated with consequent higher emissions through breathing and metabolic processes. They also varied with occupants' daily activities, like cooking and cleaning. Critic CO2 levels above the limit of 1000 ppm were observed in spring campaign, in the weeks close to the end of the COVID-19 quarantine, likely associated with the increased time that the occupants spent at home.

Project description:How to mitigate the spread of infectious diseases like COVID-19 in indoor environments remains an important research question. In this study, we propose an agent-based modeling framework to evaluate facility usage policies that aim to lower the probability of outbreaks. The proposed framework is individual-based, spatially-resolved with time resolution of up to 1 s, and takes into detailed account specific floor layouts, occupant schedules and movement. It enables decision makers to compute realistic contact networks and generate risk profiles of their facilities without relying on wearable devices, smartphone tagging or surveillance cameras. Our demonstrative modeling results indicate that not all facility occupants present the same risk of starting an outbreak, where the driver of outbreaks varies with facility layouts as well as individual occupant schedules. Therefore, generic mitigation strategies applied across all facilities should be considered inferior to tailored policies that take into account individual characteristics of the facilities of interest. The proposed modeling framework, implemented in Python and now available to the public in an open-source platform, enables such strategy evaluation.

Project description:Environmental contamination caused by COVID-19 patients could be a medium of transmission. Previous reports of SARS-CoV-2 in environmental surfaces were about short-term contamination. This study investigated SARS-CoV-2 RNA existence in room-temperature and low-temperature environments long after exposure (>28 days). A department store, where a COVID-19 outbreak was occurred in January 2020 (the epicenter of 43 COVID-19 patients), and a patient's apartment were included as room-temperature environments after being blocked for 57 days and 48 days, respectively. Seven cold storages and imported frozen foods inside were included as low-temperature environments (under -18 °C). Twenty food markets with potential contamination of imported frozen foods were also included to study the consecutive contamination. Information about temperature, relative humidity, and the number of days of environmental samples since the last exposure was collected and analyzed. In sum, 11,808 swab samples were collected before disinfection, of which 35 samples were positive. Persistent contamination of SARS-CoV-2 RNA was identified in the apartment (6/19), the department store (3/50), food packages in cold storages (23/1360), environmental surfaces of cold storages (2/345), and a package in the food market (1/10,034). Two positive samples were isolated from the bathroom of the apartment (66.7 %, 2/3), and doorknobs were proved with contamination in the apartment (40 %, 2/5) and cold storage (33.3 %, 1/3). The epidemiology information and environmental contamination results of an imported frozen food related COVID-19 case (138th COVID-19 patient in Tianjin) were analyzed. Based on the Ct values, the number of copies of two target genes was calculated by standard curves and linear regressions. In conclusion, SARS-CoV-2 RNA can be detected in room-temperature environments at least 57 days after the last exposure, much longer than previous reports. Based on the results of this study and previous studies, infectious SARS-CoV-2 could exist for at least 60 days on the surface of cold-chain food packages. Doorknobs and toilets (bathrooms) were important positions in COVID-19 control. High-risk populations of cold-chain-related logistic operations, such as porters, require strict prevention and high-level personal protection.

Project description:The SARS-CoV-2 Delta (B.1.617.2) variant is capable of infecting vaccinated persons. An open question remains as to whether deficiencies in specific vaccine-elicited immune responses result in susceptibility to vaccine breakthrough infection. We investigated 55 vaccine breakthrough infection cases (mostly Delta) in Singapore, comparing them against 86 vaccinated close contacts who did not contract infection. Vaccine breakthrough cases showed lower memory B cell frequencies against SARS-CoV-2 receptor binding domain (RBD). Compared to plasma antibodies, antibodies secreted by memory B cells retained a higher fraction of neutralizing properties against the Delta variant. Inflammatory cytokines including IL-1β and TNF were lower in vaccine breakthrough infections than primary infection of similar disease severity, underscoring the usefulness of vaccination in preventing inflammation. This report highlights the importance of memory B cells against vaccine breakthrough, and suggests that lower memory B cell levels may be a correlate of risk for Delta vaccine breakthrough infection.

Project description:During the normalized phase of COVID-19, droplets or aerosol particles produced by infected personnel are considered as the potential source of infection with uncertain exposure risk. As such, in densely populated open spaces, it is necessary to adopt strategies to mitigate the risk of infection disease transmission while providing sufficient ventilation air. An example of such strategies is use of physical barriers. In this study, the impact of barrier heights on the spread of aerosol particles is investigated in an open office environment with the well-designed ventilation mode and supply air rate. The risk of infection disease transmission is evaluated using simulation of particle concentration in different locations and subject to a number of source scenarios. It was found that a barrier height of at least 60 cm above the desk surface is needed to effectively prevent the transmission of viruses. For workstations within 4 m from the outlet, a 70 cm height is considered, and with a proper ventilation mode, it is shown that the barriers can reduce the risk of infection by 72%. However, for the workstations further away from the outlet (beyond 4 m), the effect of physical barrier cannot be that significant. In summary, this study provides a theoretical analysis for implementing physical barriers, as a low-cost mitigation strategy, subject to various height scenarios and investigation of their effectiveness in reducing the infection transmission probability.

Project description:As part of a worldwide survey of the indoor mycobiota about 520 new Cladosporium isolates from indoor environments mainly collected in China, Europe, New Zealand, North America and South Africa were investigated by using a polyphasic approach to determine their species identity. All Cladosporium species occurring in indoor environments are fully described and illustrated. Fourty-six Cladosporium species are treated of which 16 species are introduced as new. A key for the most common Cladosporium species isolated from indoor environments is provided. Cladosporium halotolerans proved to be the most frequently isolated Cladosporium species indoors.
