Project description:Semi-natural dry grassland sites are of great importance for nature conservation because they support high species diversity and the abundance of "Red-List" species. Grazing has proved to be a successful management tool in terms of maintenance and restoration of biodiversity. For a deeper understanding of the effects of different grazers on species biodiversity in dry grasslands, it is necessary to study the long-term effects of major changes in grazing management. In a semi-natural dry grassland habitat, which was formerly grazed by cattle, we investigated the changes in plant species composition due to long term grazing by sheep and goats. Specifically we asked: a) How does long-term grazing by sheep and goats change the composition of all plant species and particularly those that are on the Red-List? Are changes caused mainly by species turnover? b) How does long-term grazing by sheep and goats influence the fertility and acidity of the soil? To address these questions, we compared the composition and diversity of plants as well deriving Ellenberg indicator values of the species. Long-term grazing by sheep and goats subsequent to a year-round cattle grazing changed the plant species composition of the dry grasslands resulting in a high species turnover rate. It did not, however, lead to an increase in plant species diversity even though Red-List species were considerably more abundant in 2013. Overall, the grazing regime studied positively influenced vegetation composition. The effects on local species composition due to species turnover might further be influenced by local factors like soil nitrogen availability.

Project description:Restoration is important for biodiversity conservation worldwide, but surprisingly little is known about its efficiency in a long-term perspective. In this study, we re-examined Swedish semi-natural grasslands 12-20 years after the restoration, by comparing field inventories of vascular plant species diversity made in 2001 with follow-up inventories in 2012. We also analysed restoration effect in relation to six environmental factors and used continuously managed semi-natural grasslands as references of desired state after restoration. We found that total species richness increased over time but not to reference levels, while there were no significant changes in species density or number of grassland specialists. However, the overall species composition in the restored sites, as well as grassland specialist composition, now largely resembled reference conditions. Fertilisation and time between abandonment and restoration were the only environmental variables that affected total species composition change, while site area affected change in grassland specialist composition. Our results show that restoration of semi-natural grasslands can contribute to conservation of semi-natural habitats and their associated biodiversity. Yet, due to the vague restoration goals for these sites, it is difficult to evaluate the restoration success, which emphasise the general need for clear and measurable goals.

Project description:Soil organic carbon (SOC) is integral to soil health and agroecosystem resilience. Despite much research, understanding of temperature sensitivity of SOC under long-term agricultural management is very limited. The main objective of this study was to evaluate SOC and nitrogen (N) dynamics under grasslands and winter wheat (Triticum aestivum L)-based crop rotations in the inland Pacific Northwest (IPNW), USA, and measure SOC mineralization under ambient and elevated incubation temperatures. Soil samples were collected from 0-10 and 10-20 cm depths from an undisturbed grassland (GP), winter wheat-pea (Pisum sativum L) rotations under conventional tillage (WP-CT) and no-tillage (WP-NT), and winter wheat-fallow rotation under conventional tillage (WF-CT) and analyzed for SOC and N pools. Soil samples were incubated at 20 °C and 30 °C for 10 weeks, and SOC mineralization rates were estimated using the first order kinetic model. The GP had the greatest amounts of SOC, total N (TN), and microbial biomass carbon (MBC) and WP rotations had higher inorganic N content than other treatments. The SOC mineralization at elevated incubation temperature was 72-177% more than at the ambient temperature, and the greatest effect was observed in GP. The SOC storage under a given management did not have consistent effects on soil carbon (C) and N mineralization under elevated temperature. However, soil disturbance under WP-CT and WF-CT accelerated SOC mineralization leading to soil C loss. Reducing tillage, integrating legumes into crop rotations, and growing perennial grasses could minimize SOC loss and have the potential to improve soil health and agroecosystem resilience under projected climate warming.

Project description:Semi-natural grassland areas expanded worldwide several thousand years ago following an increase in anthropogenic activities. However, semi-natural grassland habitat areas have been declining in recent decades due to changes in landuse, which have caused a loss of grassland biodiversity. Reconstructing historical and recent demographic changes in semi-natural grassland species will help clarify the factors affecting their population decline. Here we quantified past and recent demographic histories of Melitaea ambigua (Lepidoptera; Nymphalidae), an endangered grassland butterfly species in Japan. We examined changes in demography over the past 10,000 years based on 1378 bp of mitochondrial COI gene. We then examined changes in its genetic diversity and structure during the last 30 years using nine microsatellite DNA markers. The effective population size of M. ambigua increased about 3000-6000 years ago. In contrast, the genetic diversity and effective population sizes of many populations significantly declined from the 1980s to 2010s, which is consistent with a recent decline in the species population size. Our data suggest that the M. ambigua demography can be traced to changes in area covered by semi-natural grasslands throughout the Holocene.

Project description:We studied β diversity of grasses in a subtropical grassland over 60 years in South Africa. We examined the effects of burning and mowing on 132 large plots. We sought to determine the effects of burning and mowing, and mowing frequency, on the replacement of species and the species richness. We conducted the study at Ukulinga, research farm of the University of KwaZulu-Natal, Pietermaritzburg, South Africa (29°24'E, 30°24'S) from 1950-2010. Plots were burned annually, biennially, triennially, and a control (unburned). Plots were mowed in spring, late summer, spring plus late summer, and a control (unmowed). We calculated β diversity, with a focus on replacement and richness differences. We also used distance-based redundancy analyses to examine the relative effects of replacement and richness differences on mowing and burning. We used beta regressions to test for the effect of soil depth and its interactions with mowing and burning. There was no significant change in grass beta diversity until 1995. Thereafter, there were changes in β diversity that demonstrated the primary effects of summer mowing frequency. There was no significant effect of richness differences but a strong effect of replacement post-1995. There was a significant interaction between mowing frequency and soil depth in one of the analyses. Changes in grassland composition took a long time to manifest themselves and were unapparent prior to 1988. However, there was a change in sampling strategy prior to 1988, from point hits to nearest plants, that may also have influenced the rates of changes in replacement and richness differences. Using β-diversity indices, we found that mowing was more important than burning that burning frequency was unimportant, and there was a significant interaction effect between mowing and soil depth in one of the analyses.

Project description:North American grassland birds have widely declined over the past 50 years, largely due to anthropogenic-driven loss of native prairie habitat. In response to these declines, many conservation programs have been implemented to help secure wildlife habitat on private and public lands. The Grasslands Coalition is one such initiative established to advance the conservation of grassland birds in Missouri. The Missouri Department of Conservation conducted annual point count surveys for comparison of grassland bird relative abundance between focal grassland areas and nearby paired (i.e., containing no targeted management) sites. We analyzed 17 years of point count data with a generalized linear mixed model in a Bayesian framework to estimate relative abundance and trends across focal or paired sites for nine bird species of management interest that rely on grasslands: barn swallow (Hirundo rustica), brown-headed cowbird (Molothrus ater), dickcissel (Spiza americana), eastern meadowlark (Sturnella magna), grasshopper sparrow (Ammodramus savannarum), Henslow's sparrow (A. henslowii), horned lark (Eremophila alpestris), northern bobwhite (Colinus virginianus), and red-winged blackbird (Agelaius phoeniceus). Relative abundance of all species except eastern meadowlarks declined regionally. Relative abundance of barn swallows, brown-headed cowbirds, dickcissels, eastern meadowlarks, Henslow's sparrows, and northern bobwhites was higher in focal than paired sites, though relative abundance trends were only improved in focal vs. paired areas for dickcissels and Henslow's sparrows. Relative abundance increased with increasing grassland cover at the local (250-m radius) scale for all species except horned larks and red-winged blackbirds and at the landscape (2,500-m radius) scale for all species except dickcissels, eastern meadowlarks, and northern bobwhites. Our results suggest focal areas contained greater relative abundances of several grassland species of concern, likely due to increased availability of grassland habitat at local and landscape scales. Further efforts to decrease landscape-scale fragmentation and improve habitat quality may be needed to achieve conservation goals.

Project description:Intensive land use is a driving force for biodiversity decline in many ecosystems. In semi-natural grasslands, land-use activities such as mowing, grazing and fertilization affect the diversity of plants and arthropods, but the combined effects of different drivers and the chain of effects are largely unknown. In this study we used structural equation modelling to analyse how the arthropod communities in managed grasslands respond to land use and whether these responses are mediated through changes in resource diversity or resource quantity (biomass). Plants were considered resources for herbivores which themselves were considered resources for predators. Plant and arthropod (herbivores and predators) communities were sampled on 141 meadows, pastures and mown pastures within three regions in Germany in 2008 and 2009. Increasing land-use intensity generally increased plant biomass and decreased plant diversity, mainly through increasing fertilization. Herbivore diversity decreased together with plant diversity but showed no response to changes in plant biomass. Hence, land-use effects on herbivore diversity were mediated through resource diversity rather than quantity. Land-use effects on predator diversity were mediated by both herbivore diversity (resource diversity) and herbivore quantity (herbivore biomass), but indirect effects through resource quantity were stronger. Our findings highlight the importance of assessing both direct and indirect effects of land-use intensity and mode on different trophic levels. In addition to the overall effects, there were subtle differences between the different regions, pointing to the importance of regional land-use specificities. Our study underlines the commonly observed strong effect of grassland land use on biodiversity. It also highlights that mechanistic approaches help us to understand how different land-use modes affect biodiversity.

Project description:Glioblastoma (GBM) is a highly aggressive type of glioma with poor prognosis. However, a small number of patients live much longer than the median survival. A better understanding of these long-term survivors (LTS) may provide important insight into the biology of GBM. We identified 7 patients with GBM treated at Memorial Sloan-Kettering Cancer Center (MSKCC) with survival greater than 48 months. We characterized the transcriptome of each patient and determined rates of MGMT promoter methylation and IDH1 and IDH2 mutational status. We identified LTS in two independent cohorts (TCGA and REMBRANDT) and analyzed the transcriptomal characteristics of these LTS. The median overall survival of our cohort was 62.5 months. LTS were distributed between the proneural (n=2), neural (n=2), classical (n=2) and mesenchymal (n=1) subtypes. Similarly, LTS in the TCGA and REMBRANDT cohorts demonstrated diverse transcriptomal subclassification identity. The majority of the MSKCC LTS (71%) were found to have methylation of the MGMT promoter. None of the patients had an IDH1 or IDH2 mutations, and IDH mutation occurred in a minority of the TCGA LTS as well. A set of 42 genes was found to be differentially expressed in the MSKCC and TCGA LTS. While IDH mutant proneural tumors impart a better prognosis in the short-term, survival beyond 4 years does not require IDH mutation and is not dictated by a single transcriptional subclass. In contrast, MGMT methylation continues to have strong prognostic value for survival beyond 4 years. These findings have substantial impact for understanding GBM biology and progression. All tumors (n = 7) were obtained following surgical resection at the MSKCC as part of routine clinical care, and snap frozen. Tumors were obtained in accordance with Institutional Review Board policies at the MSKCC. Each sample was examined histologically by 3 independent neuropathologists and confirmed to be World Health Organization (WHO) grade IV glioma (GBM). Before analysis, tumors were sectioned and microdissected. Genomic DNA or RNA was extracted using the DNeasy kit (Qiagen) or RNeasy Lipid Tissue Mini Kit (Qiagen) as per the manufacturer’s instructions. Expression analysis of tumors was performed using the Affymetrics U133 2.0 microarray (Affymetrix). Affymetrix CEL files were imported into the Partek Genomics Suite (Partek). The TCGA gene expression data (HT-HG-U133A) was accessed from the TCGA data repositories (http://cancergenome.nih.gov, date of download 12/2013).

Project description:We measured 34 years of plant community change in a degraded oak woodland undergoing ecological management. Management included regular prescribed fire, control of white-tailed deer populations, repeated sowing of a diverse seed mix, and removal of invasive plants. We tracked change with several conservation metrics. Time series analysis showed no significant changes over time in either plant species richness or the Shannon-Weiner diversity index. Floristic Quality Assessment measures-the Floristic Quality Index (FQI), Cover-weighted FQI, and the Mean Coefficient of Conservatism (Mean C)-all increased dramatically over time, such that their values now surpass those of the highest quality representative of this habitat in the region. Cover-weighted FQI had the added benefit of being quick to respond (negatively and positively) to short-term management changes during the study. This sensitivity highlights its utility for adaptive management, enabling timely, data-driven changes to ongoing management regimes. Plant community composition showed striking changes during the study period, as species of high conservation value replaced weedier species. As a group, conservative woodland species are notoriously slow to recover from degradation, making this flora's recovery particularly notable. A mid-study cessation of management immediately stalled the woodland's recovery according to Floristic Quality metrics, but the restoration quickly returned to its positive trajectory with the resumption of management treatments. These results illustrate that impressive plant biodiversity restoration can be achieved, even in highly degraded contemporary oak ecosystems, if ecological management is comprehensive and if it is sustained over time.

Project description:The introduction of rhizoma peanut (RP Arachis glabrata Benth) into bahiagrass (Paspalum notatum Flüggé) may require time to develop stable plant-soil microbe interactions as the microbial legacy of the previous plant community may be long-lasting. A previous study showed that <2 years of introducing rhizoma peanut into bahiagrass pastures minimally affected soil bacterial diversity and community composition. In this study, we compared the effects of the long-term inclusion of rhizoma peanut (>8 years) into bahiagrass on soil bacterial diversity and community composition against their monocultures at 0 to 15 and 15 to 30 cm soil depths using next-generation sequencing to target bacterial 16S V3-V4 regions. We observed that a well-established RP-bahiagrass mixed stand led to a 36% increase in bacterial alpha diversity compared to the bahiagrass monoculture. There was a shift from a soil bacterial community dominated by Proteobacteria (~26%) reported in other bahiagrass and rhizoma peanut studies to a soil bacterial community dominated by Firmicutes (39%) in our study. The relative abundance of the bacterial genus Crossiella, known for its antimicrobial traits, was enhanced in the presence of RP. Differences in soil bacterial diversity and community composition were substantial between 0 to 15 and 15 to 30 cm soil layers, with N2-fixing bacteria belonging to the phylum Proteobacteria concentrated in 0 to 15 cm. Introducing RP into bahiagrass pastures is a highly sustainable alternative to mineral N fertilizer inputs. Our results provide evidence that this system also promotes greater soil microbial diversity and is associated with unique taxa that require further study to better understand their contributions to healthy pastures.
