Electrochemical DNA Sensor Based on Poly(Azure A) Obtained from the Buffer Saturated with Chloroform.

ABSTRACT: Electropolymerized redox polymers offer broad opportunities in detection of biospecific interactions of DNA. In this work, Azure A was electrochemically polymerized by multiple cycling of the potential in phosphate buffer saturated with chloroform and applied for discrimination of the DNA damage. The influence of organic solvent on electrochemical properties of the coating was quantified and conditions for implementation of DNA in the growing polymer film were assessed using cyclic voltammetry, quartz crystal microbalance, and electrochemical impedance spectroscopy. As shown, both chloroform and DNA affected the morphology of the polymer surface and electropolymerization efficiency. The electrochemical DNA sensor developed made it possible to distinguish native and thermally and chemically damaged DNA by changes in the charge transfer resistance and capacitance.

Electrochemical DNA Sensor for Valrubicin Detection Based on Poly(Azure C) Films Deposited from Deep Eutectic Solvent.

Project description:A novel electrochemical DNA sensor was developed for the detection of the anthracycline drug, valrubicin, on the base of poly(Azure C) electropolymerized from the deep eutectic solvent reline and covered with adsorbed DNA from calf thymus. Biosensor assembling was performed by multiple scanning of the potential in one drop (100 µL) of the dye dissolved in reline and placed on the surface of a screen-printed carbon electrode. Stabilization of the coating was achieved by its polarization in the phosphate buffer. The electrochemical characteristics of the electron transfer were determined and compared with a similar coating obtained from phosphate buffer. The use of deep eutectic solvent made it possible to increase the monomer concentration and avoid using organic solvents on the stage of electrode modification. After the contact of the DNA sensor with valrubicin, two signals related to the intrinsic redox activity of the coating and the drug redox conversion were found on voltammogram. Their synchronous changes with the analyte concentration increased the reliability of the detection. In the square-wave mode, the DNA sensor made it possible to determine from 3 µM to 1 mM (limit of detection, 1 µM) in optimal conditions. The DNA sensor was successfully tested in the voltammetric determination of valrubicin in spiked artificial urine, Ringer-Locke solution mimicking plasma electrolytes and biological samples (urine and saliva) with a recovery of 90-110%. After further testing on clinical samples, it can find application in the pharmacokinetics studies and screening of new drugs' interaction with DNA.

Electrochemical DNA Sensor Based on the Copolymer of Proflavine and Azure B for Doxorubicin Determination.

Project description:A DNA sensor has been developed for the determination of doxorubicin by consecutive electropolymerization of an equimolar mixture of Azure B and proflavine and adsorption of native DNA from salmon sperm on a polymer film. Electrochemical investigation showed a difference in the behavior of individual drugs polymerized and their mixture. The use of the copolymer offered some advantages, i.e., a higher roughness of the surface, a wider range of the pH sensitivity of the response, a denser and more robust film, etc. The formation of the polymer film and its redox properties were studied using scanning electron microscopy and electrochemical impedance spectroscopy. For the doxorubicin determination, its solution was mixed with DNA and applied on the polymer surface. After that, charge transfer resistance was assessed in the presence of [Fe(CN)6]3-/4- as the redox probe. Its value regularly grew with the doxorubicin concentration in the range from 0.03 to 10 nM (limit of detection 0.01 nM). The DNA sensor was tested on the doxorubicin preparations and spiked samples mimicking blood serum. The recovery was found to be 98-106%. The DNA sensor developed can find application for the determination of drug residues in blood and for the pharmacokinetics studies.

Electrochemical DNA Sensor for Sensitive BRCA1 Detection Based on DNA Tetrahedral-Structured Probe and Poly-Adenine Mediated Gold Nanoparticles.

Project description:BRCA1 is the biomarker for the early diagnosis of breast cancer. Detection of BRCA1 has great significance for the genetic analysis, early diagnosis and clinical treatment of breast cancer. In this work, we developed a simple electrochemical DNA sensor based on a DNA tetrahedral-structured probe (TSP) and poly-adenine (polyA) mediated gold nanoparticles (AuNPs) for the sensitive detection of BRCA1. A thiol-modified TSP was used as the scaffold on the surface of the screen-printed AuNPs electrode. The capture DNA (TSP) and reporter DNA were hybridized to the target DNA (BRCA1), respectively, to form the typical sandwich system. The nanocomposites of reporter DNA (polyA at the 5' end) combined with AuNPs were employed for signal amplification which can capture multiple enzymes by the specificity between biotin and streptavidin. Measurements were completed in the electrochemical workstation by cyclic voltammetry and amperometry and we obtained the low limit of detection of 0.1 fM with the linear range from 1 fM to 1 nM. High sensitivity and good specificity of the proposed electrochemical DNA sensor showed potential applications in clinical early diagnosis for breast cancer.

Electrochemical DNA Sensor Based on Acridine Yellow Adsorbed on Glassy Carbon Electrode.

Project description:Electrochemical DNA sensors offer unique opportunities for the sensitive detection of specific DNA interactions. In this work, a voltametric DNA sensor is proposed on the base of glassy carbon electrode modified with carbon black, adsorbed acridine yellow and DNA for highly sensitive determination of doxorubicin antitumor drug. The signal recorded by cyclic voltammetry was attributed to irreversible oxidation of the dye. Its value was altered by aggregation of the hydrophobic dye molecules on the carbon black particles. DNA molecules promote disaggregation of the dye and increased the signal. This effect was partially suppressed by doxorubicin compensate for the charge of DNA in the intercalation. Sensitivity of the signal toward DNA and doxorubicin was additionally increased by treatment of the layer with dimethylformamide. In optimal conditions, the linear range of doxorubicin concentrations determined was 0.1 pM-1.0 nM, and the detection limit was 0.07 pM. No influence of sulfonamide medicines and plasma electrolytes on the doxorubicin determination was shown. The DNA sensor was tested on two medications (doxorubicin-TEVA and doxorubicin-LANS) and showed recoveries of 102-105%. The DNA sensor developed can find applications in the determination of drug residues in blood and for the pharmacokinetics studies.

An Exfoliated Graphite-Based Bisphenol A Electrochemical Sensor.

Project description:The use of an exfoliated graphite (EG) electrode in the square wave voltammetric detection of bisphenol A (a model phenolic pollutant) in water, whereby the phenolic electrode fouling challenge is mitigated, is described. The oxidation peak of BPA was observed at about 0.45 V in phosphate buffer solution at pH 10. The current response exhibited a linear relationship with the concentration over a range from 1.56 μM–50 μM. The detection limit was calculated to be 0.76 μM. The EG electrode surface was renewed after each measurement with excellent reproducibility. A real sample application was also investigated.

Water Electrolysis in Saturated Phosphate Buffer at Neutral pH.

Project description:Hydrogen production from renewable energy and ubiquitous water has a potential to achieve sustainability, although current water electrolyzers cannot compete economically with the fossil fuel-based technology. Here, we evaluate water electrolysis at pH 7 that is milder than acidic and alkaline pH counterparts and may overcome this issue. The physicochemical properties of concentrated buffer electrolytes were assessed at various temperatures and molalities for quantitative determination of losses associated with mass-transport during the water electrolysis. Subsequently, in saturated K-phosphate solutions at 80 °C and 100 °C that were found to be optimal to minimize the losses originating from mass-transport at the neutral pH, the water electrolysis performance over model electrodes of IrOx and Pt as an anode and a cathode, respectively, was reasonably comparable with those of the extreme pH. Remarkably, this concentrated buffer solution also achieved enhanced stability, adding another merit of this electrolyte for water electrolysis.

Role of Green Chemistry in Antipsychotics' Electrochemical Investigations Using a Nontoxic Modified Sensor in McIlvaine Buffer Solution.

Project description:A new low-cost green electrochemical sensor based on nontoxic polyethylene glycol (PEG) and silver nanoparticles was used to improve the sensitivity of the carbon paste electrode for the investigation of olanzapine (OLZ) in dosage arrangements and in the existence of its coadministered drug fluoxetine and in the drug formulation. Scanning electron microscopy measurements were carried out to emphasize the morphology of the electrode surface. Cyclic voltammetry and differential pulse voltammetry were used to explore the diffusion and linearity behaviors of OLZ. Impedance spectroscopy measurements were determined to investigate the ac behavior of OLZ and then an ideal electrical circuit was modeled. A linear calibration was obtained from 1.0 × 10-8 to 1.25 × 10-4 M. The limit of detection was 1.5 × 10-9 M, whereas the limit of quantification was 5 × 10-9 M. The way has been wholly authenticated concerning linearity, precision, accuracy, reproducibility, sensitivity, and selectivity.

Heterogeneous Electrochemical Aptamer-Based Sensor Surfaces for Controlled Sensor Response.

Project description:Structure-switching sensors utilize recognition elements that undergo a conformation change upon target binding that is converted into a quantitative signal. Electrochemical, aptamer-based sensors achieve detection of analytes through a conformation change in an electrode-bound, oligonucleotide aptamer by measuring changes in electron transfer efficiencies. The analytical performance of these sensors is related to the magnitude of the conformation change of the aptamer. The goal of the present work is to develop a general method to predictably tune the analytical performance (sensitivity and linear range) of electrochemical, aptamer-based sensors by utilizing a mixture of rationally designed aptamer sequences that are specific for the same target but with different affinities on the same electrode surface. To demonstrate control over sensor performance, we developed heterogeneous sensors for two representative small molecule targets (adenosine triphosphate and tobramycin). We demonstrate that mixtures of modified sequences can be used to tune the affinity, dynamic range, and sensitivity of the resulting sensors predicted by a bi-Langmuir-type isotherm.

An electrochemical sensor based on poly(procaterol hydrochloride)/carboxyl multi-walled carbon nanotube for the determination of bromhexine hydrochloride.

Project description:Poly(procaterol hydrochloride) (p-ProH) polymeric film was successfully deposited onto the carboxyl multi-walled carbon nanotube (CMWCNT) modified glass carbon electrode (GCE) to construct a p-ProH/CMWCNT composite modified GCE. Due to the synergistic effect of p-ProH and CMWCNT in the composite, the developed sensor can enormously enhance the oxidation peak current of bromhexine hydrochloride (BrH) at ca. + 0.90 V. Based on this appearance, an electrochemical method was established for the sensitive and selective determination of BrH with differential pulse voltammetry (DPV). Various conditions affecting the peak current response of BrH were studied and optimized. Under the best conditions, the oxidation peak current of BrH is linear to its concentration in two linear dynamic ranges of 0.2-1.0 μmol L-1 (R = 0.9948) and 1.0-8.0 μmol L-1 (R = 0.9956), with a detection limit of 0.1 μmol L-1 (S/N = 3). Interference experiment indicated that the as-prepared electrochemical sensor showed wonderful selectivity to the recognition of BrH and was free from disturbance of many other electro-active substances such as dopamine, ascorbic and uric acid. Finally, the practicability of the BrH sensor was verified by the satisfactory results acquired from the BrH determination in pharmaceutical preparation and human serum.

Glucose Biosensor Based on Disposable Activated Carbon Electrodes Modified with Platinum Nanoparticles Electrodeposited on Poly(Azure A).

Project description:Herein, a novel electrochemical glucose biosensor based on glucose oxidase (GOx) immobilized on a surface containing platinum nanoparticles (PtNPs) electrodeposited on poly(Azure A) (PAA) previously electropolymerized on activated screen-printed carbon electrodes (GOx-PtNPs-PAA-aSPCEs) is reported. The resulting electrochemical biosensor was validated towards glucose oxidation in real samples and further electrochemical measurement associated with the generated H2O2. The electrochemical biosensor showed an excellent sensitivity (42.7 μA mM-1 cm-2), limit of detection (7.6 μM), linear range (20 μM-2.3 mM), and good selectivity towards glucose determination. Furthermore, and most importantly, the detection of glucose was performed at a low potential (0.2 V vs. Ag). The high performance of the electrochemical biosensor was explained through surface exploration using field emission SEM, XPS, and impedance measurements. The electrochemical biosensor was successfully applied to glucose quantification in several real samples (commercial juices and a plant cell culture medium), exhibiting a high accuracy when compared with a classical spectrophotometric method. This electrochemical biosensor can be easily prepared and opens up a good alternative in the development of new sensitive glucose sensors.
