Project description:In this study, native chemical ligation (NCL) was used as a selective cross-linking method to form core-cross-linked thermosensitive polymeric micelles for drug delivery applications. To this end, two complementary ABA triblock copolymers having polyethylene glycol (PEG) as midblock were synthesized by atom transfer radical polymerization (ATRP). The thermosensitive poly isopropylacrylamide (PNIPAM) outer blocks of the polymers were copolymerized with either N-(2-hydroxypropyl)methacrylamide-cysteine (HPMA-Cys), P(NIPAM- co-HPMA-Cys)-PEG-P(NIPAM- co-HPMA-Cys) (PNC) or N-(2-hydroxypropyl)methacrylamide-ethylthioglycolate succinic acid (HPMA-ETSA), P(NIPAM- co-HPMA-ETSA)-PEG-P(NIPAM- co-HPMA-ETSA) (PNE). Mixing of these polymers in aqueous solution followed by heating to 50 °C resulted in the formation of thermosensitive flower-like micelles. Subsequently, native chemical ligation in the core of micelles resulted in stabilization of the micelles with a Z-average of 65 nm at body temperature. Decreasing the temperature to 10 °C only affected the size of the micelles (increased to 90 nm) but hardly affected the polydispersity index (PDI) and aggregation number ( Nagg) confirming covalent stabilization of the micelles by NCL. CryoTEM images showed micelles with an uniform spherical shape and dark patches close to the corona of micelles were observed in the tomographic view. The dark patches represent more dense areas in the micelles which coincide with the higher content of HPMA-Cys/ETSA close to the PEG chain revealed by the polymerization kinetics study. Notably, this cross-linking method provides the possibility for conjugation of functional molecules either by using the thiol moieties still present after NCL or by simply adjusting the molar ratio between the polymers (resulting in excess cysteine or thioester moieties) during micelle formation. Furthermore, in vitro cell experiments demonstrated that fluorescently labeled micelles were successfully taken up by HeLa cells while cell viability remained high even at high micelle concentrations. These results demonstrate the potential of these micelles for drug delivery applications.

Project description:Plants can show long-term effects of environmental stresses and in some cases a stress "memory" has been reported to persist across generations, potentially mediated by epigenetic mechanisms. However, few documented cases exist of transgenerational effects that persist for multiple generations and it remains unclear if or how epigenetic mechanisms are involved. Here, we show that the composition of small regulatory RNAs in apomictic dandelion lineages reveals a footprint of drought stress and salicylic acid treatment experienced two generations ago. Overall proportions of 21 and 24?nt RNA pools were shifted due to grandparental treatments. While individual genes did not show strong up- or downregulation of associated sRNAs, the subset of genes that showed the strongest shifts in sRNA abundance was significantly enriched for several GO terms including stress-specific functions. This suggests that a stress-induced signal was transmitted across multiple unexposed generations leading to persistent changes in epigenetic gene regulation.

Project description:Tumor-associated macrophages (TAMs) constitute over 50% of the number of cells within the tumor, playing a major role in tumor progression and invasion. Remodeling the tumor immune microenvironment by modulating TAM polarization has been emerging as a new and promising therapeutic strategy. However, the high interstitial fluid pressure and dense extracellular matrix lead to insufficient penetration of nanosized therapies. To overcome this dilemma, an acid-triggered size-changeable nanoparticle (aptamer/acid sensitive linker crosslinked DGL/zoledronic acid, i.e., Apt@(DGL-ZA) n NPs) with effective tumor distribution, extravasation, and penetration is designed. Dendrigraft poly-L-lysines (DGLs) which can induce tumor autophagy as mimics of natural abnormal proteins are crosslinked via a mild-acid-responsive linker (1,6-bis(4-formylbenzoyloxy) hexane). Long circulation property and tumor penetration are achieved simultaneously by catching DGLs in neutral pH while releasing them in the tumor's pH, like dandelion seeds in midair. The macrophage conditioning agent zoledronic acid (ZA) is loaded on DGLs by the charge attraction. A Tenascin-C targeting aptamer (GBI-10) is modified onto (DGL-ZA) n NPs for a tumor-homing effect. Apt@(DGL-ZA) n NPs show both enhanced penetration in in vitro 3D triple negative breast cancer spheroids and in vivo tumor tissues. Effective macrophage regulation, enhanced tumor autophagy, and excellent in vivo antitumor efficacy are achieved, suggesting this tactic as a significant antitumor strategy.

Project description:The edible and medicinal perennial herb dandelion is known to have antitumor, antioxidant, and anticomplement properties. However, the structural characterization and biological effects of its polysaccharides are not well understood. Here, we aimed to extract and investigate a novel polysaccharide from dandelion. A water-soluble polysaccharide, PD1-1, was successfully obtained from dandelion through ultrasonic-assisted extraction and purification using diethylaminoethyl (DEAE)-Sepharose fast flow and Sephadex G-75 columns. The results showed that PD1-1 is an inulin-type polysaccharide with a molecular weight of 2.6 kDa and is composed of glucose (52.39%), and mannose (45.41%). Glycosidic linkage analysis demonstrated that PD1-1 contains terminal ?-d-Man/Glcp-(1? and ?1)-?-d-Man/Glcf-(2? glycosidic linkage conformations. A physicochemical analysis indicated that PD1-1 has a triple helix structure and exhibits important properties, including good swelling, water-holding, and oil-holding capacities. Furthermore, PD1-1 showed good antioxidant activities in DPPH and hydroxyl free radical scavenging abilities, with IC50 values of 0.23 mg/mL and 0.25 mg/mL, respectively, and good hypoglycemic activities in ?-amylase and ?-glucosidase inhibition, with IC50 values of 0.53 mg/mL and 0.40 mg/mL, respectively, in a concentration-dependent manner. Results suggest that PD1-1 possesses efficacious antioxidant and hypoglycemic properties and has potential applications as a functional food ingredient.

Three beta-fructofuranosidases were separated by chromatography on a DEAE-cellulose column from the soluble protein extracted from dandelion (Taraxacum officinale Weber) roots. 2. One enzyme, which acted on sucrose, was characterized as an invertase, with a K(m) of 2.00x10(-2)m and pH optimum of 7.5. 3. The other two enzymes are hydrolases (A and B), which act on the inulin series of oligosaccharides [general formula glucose-fructose-(fructose)(n)]. They both have a pH optimum of 4.0 and K(m) of 1.54x10(-2)m but differ in their chromatographic behaviour on DEAE-cellulose. Neither of the hydrolases is inhibited by sucrose. 4. The physiological role of these three hydrolytic enzymes is discussed.

Project description:Urban environments expose species to contrasting selection pressures relative to rural areas due to altered microclimatic conditions, habitat fragmentation, and changes in species interactions. To improve our understanding on how urbanization impacts selection through biotic interactions, we assessed differences in plant defense and tolerance, dispersal, and flowering phenology of a common plant species (Taraxacum officinale) along an urbanization gradient and their reaction norms in response to a biotic stressor (i.e., herbivory). We raised plants from 45 lines collected along an urbanization gradient under common garden conditions and assessed the impact of herbivory on plant growth (i.e., aboveground biomass), dispersal capacity (i.e., seed morphology), and plant phenology (i.e., early seed production) by exposing half of our plants to two events of herbivory (i.e., grazing by locusts). Independent from their genetic background, all plants consistently increased their resistance to herbivores by which the second exposure to locusts resulted in lower levels of damage suffered. Herbivory had consistent effects on seed pappus length, with seeds showing a longer pappus (and, hence, increased dispersal capacities) regardless of urbanization level. Aboveground plant biomass was neither affected by urbanization nor herbivore presence. In contrast to consistent responses in plant defenses and pappus length, plant fitness did vary between lines. Urban lines had a reduced early seed production following herbivory while rural and suburban lines did not show any plastic response. Our results show that herbivory affects plant phenotypes but more importantly that differences in herbivory reaction norms exist between urban and rural populations.

Project description:Many sexual-asexual complexes show a distinct pattern where the asexuals have larger and more northerly ranges than closely related sexuals. A prime candidate to explain this so-called "geographical parthenogenesis" is ecological niche divergence between the sexuals and asexuals. Modern niche modelling techniques allow testing niche divergence by directly comparing the niches of sexuals and asexuals. In this study, I use such techniques to perform range-wide tests of whether nine bioclimatic variables, including annual mean temperature and annual precipitation, contribute to geographical parthenogenesis in two dandelion taxa: Taraxacum section Ruderalia and Taraxacum section Erythrosperma, which are both comprised of sexual diploids and asexual triploids. For both sections, I found evidence of niche divergence, though the exact nature of this divergence was different for the two sections. In section Ruderalia, the sexuals preferred warmer and wetter conditions, whereas in section Erythrosperma, the sexuals preferred dryer conditions. Using Species Distribution Modelling, consistent differences between the sexuals and asexuals were found when looking at the niche determinants: the variables that are most important for modelling the distribution. Furthermore, and in contrast with theoretical expectations that predict that the sexuals should have a wider niche, in section Erythrosperma the asexuals were found to have a wider niche than the sexuals. In conclusion, differences in niche optima, niche determinants, and niche width all contribute to the pattern of geographical parthenogenesis of these two dandelion taxa. However, the results also indicate that the exact causation of geographical parthenogenesis is not uniform across taxa.

Project description:Flowers are the most complex structures of plants. Studies of Arabidopsis thaliana, which has typical eudicot flowers, have been fundamental in advancing the structural and molecular understanding of flower development. The main processes and stages of Arabidopsis flower development are summarized to provide a framework in which to interpret the detailed molecular genetic studies of genes assigned functions during flower development and is extended to recent genomics studies uncovering the key regulatory modules involved. Computational models have been used to study the concerted action and dynamics of the gene regulatory module that underlies patterning of the Arabidopsis inflorescence meristem and specification of the primordial cell types during early stages of flower development. This includes the gene combinations that specify sepal, petal, stamen and carpel identity, and genes that interact with them. As a dynamic gene regulatory network this module has been shown to converge to stable multigenic profiles that depend upon the overall network topology and are thus robust, which can explain the canalization of flower organ determination and the overall conservation of the basic flower plan among eudicots. Comparative and evolutionary approaches derived from Arabidopsis studies pave the way to studying the molecular basis of diverse floral morphologies.

Project description:The Russian dandelion, Taraxacum kok-saghyz (TKS), is a perennial species native to Central Asia that produces high quality, natural rubber. Despite its potential to help maintain a stable worldwide rubber supply, little is known about genetic variation in this species. To facilitate future germplasm improvement efforts, we developed simple-sequence repeat (SSR) markers from available expressed-sequence tag (EST) data and used them to investigate patterns of population genetic diversity in this nascent crop species. We identified numerous SSRs (1,510 total) in 1,248 unigenes from a larger set of 6,960 unigenes (derived from 16,441 ESTs) and designed PCR primers targeting 767 of these loci. Screening of a subset of 192 of these primer pairs resulted in the identification of 48 pairs that appeared to produce single-locus polymorphisms. We then used the most reliable 17 of these primer pairs to genotype 176 individuals from 17 natural TKS populations. We observed an average of 4.8 alleles per locus with population-level expected heterozygosities ranging from 0.28 to 0.50. An average pairwise FST of 0.11 indicated moderate but statistically significant levels of genetic differentiation, though there was no clear geographic patterning to this differentiation. We also tested these 17 primer pairs in the widespread common dandelion, T. officinale, and a majority successfully produced apparently single-locus amplicons. This result demonstrates the potential utility of these markers for genetic analyses in other species in the genus.

Project description:The phenolic compounds in dandelion were extracted using different ethanol percentage solutions, identified with HPLC-MS, and their scavenging capabilities of DPPH, ABTS and OH radicals were determined. Then the excitation-emission matrix fluorescence spectroscopy coupled with parallel factor analysis (PARAFAC) was conveyed to analyze the relationship between phenolics, components scores of PARAFAC model and antioxidant capacities, based on linear regression method. The results showed that the relative content of chicoric acid, esculetin, caffeic acid, gallic acid monohydrate, eupatilin, caffeic acid-3-glucoside, corchorifatty acid F, and luteolin was higher than 0.5%, and the extraction solutions with 100% and 75% water had a better scavenging capacity of DPPH, ABTS and OH radicals. Two components PARAFAC model was identified with the comparatively higher sum of squares, core consistency values, and lower interactions numbers, and the established equations indicated the component scores had a linear regression relationship with antioxidant capacities of DPPH, and ABTS. The paper was proposed for the first time that the component scores of PARAFAC model might be treated as a useful indication for antioxidant capacity evaluation.
