Project description:Native breeds of ducks have been the subject of many studies in the past, yet the relevant knowledge is still incomplete and needs to be further expanded. The objective of this study was to provide information about differences in growth performance, dressing percentage, carcass composition and digestive morphometry among three lines of Pekin ducks from conservation flocks raised in Poland. The study used 180 sexed Pekin ducks-30 males and 30 females of line P33 (ducks of Polish origin), 30 males and 30 females of line P8 (ducks of Danish origin), and 30 males and 30 females of line P9 (ducks of French origin). Throughout the study (49 d), ducks were confined indoors in six pens. Birds were fed complete commercial diets ad libitum and had unrestricted access to water. The compared lines of ducks differed significantly in body weight from 1 to 49 d of age except of ducks of both sexes at 14 d. At 49 d of age, significant differences were observed between the tested ducks in all the body measurements. Duck genotype had a significant effect on preslaughter body weight, carcass weight and breast muscle, neck and remainders contents, caeca length, liver weight and gizzard percentage. The results show that the tested ducks were significantly different and unique, mainly in terms of the body biometric characteristics.

Project description:The objective was to determine effects of old and new rubber matting in a slatted, indoor cattle feeding facility on cattle growth performance, locomotion, and carcass characteristics. In experiment 1, fall-born Angus × Simmental steers (N = 207; body weight = 222 ± 38 kg) were blocked by weight and assigned to 32 pens. Pens were randomly assigned to 1 of 4 treatments: no matting/concrete (CONC1), 12-yr-old Animat Pebble matting (OLD1), new Animat Maxgrip matting (MG), and new Animat Pebble matting (PEB1). Steers were fed a common diet for 209 d with a minimum stocking density of 3.40 m2 per animal. Final body weight (BW) and average daily gain (ADG) were affected (P = 0.02 and P < 0.01, respectively) by treatment with steers on PEB1 finishing heaviest with the greatest growth, MG and CONC1 intermediate, and OLD1 finishing at the lightest final BW with the least growth. Flooring treatment did not affect overall dry matter intake (DMI; P = 0.16) or gain to feed ratio (G:F; P = 0.94). Flooring treatment did not affect (P ≥ 0.19) any carcass traits. Locomotion scores (LS) were affected (P < 0.01) by flooring treatment with CONC1 having the worst mobility while OLD1, MG, and PEB1 were similar (P ≥ 0.24). Locomotion score had a day effect (P < 0.01) where cattle gait and mobility worsened as days on feed increased. In experiment 2, fall-born Angus × Simmental steers (N = 189; BW = 352 ± 43 kg) were blocked by weight and assigned to 21 pens. Pens were randomly assigned to 1 of 3 treatments: no matting/concrete (CONC2), 15-yr-old Animat Pebble matting (OLD2), and new Animat Pebble matting (PEB2). Steers were fed a common diet for 152 d with a stocking density of 2.65 m2 per steer. After 152 d on feed, flooring treatment did not affect (P ≥ 0.30) BW, ADG, or DMI nor did treatment affect (P ≥ 0.17) carcass traits. However, steers housed on OLD2 or PEB2 had improved locomotion scores (P = 0.02) compared with steers housed on CONC2. Locomotion score had a day effect (P < 0.01) as cattle gait and mobility worsened with greater number of days on feed, regardless of treatment. Overall, results suggest that new rubber matting increased ADG and HCW during a 209-d trial when cattle were stocked at 3.4 m2 in small pens and that rubber matting regardless of age improved cattle locomotion scores in slatted indoor feeding facilities.

Project description:Little to no research has been conducted on the gut microbiome of the Pekin duck, yet over 24.5 million ducks are raised for human consumption each year in the United States alone. Knowledge of the microbiome could lead to an understanding of the effects of growing conditions such as the use of prebiotics, probiotics, and enzymes in feeding practices, the use of antibiotics, and the sources of pathogenic bacteria in diseased ducks. In order to characterize changes in the caecal microbiome that occur as ducks develop through a typical industry grow-out period, a 16S rRNA community analysis of caecal contents collected over a 6-week period was conducted using a next generation sequencing approach. Transitions in the composition of the caecal microbiome occurred throughout the lifespan, with a large shift during days 4 through 10 posthatch. Two major phyla of bacteria were found to be present within the caeca of aviary raised ducks, with the relative abundance of each phylum varying by age of the duck. Proteobacteria is dominant for the first 3 days of age, and Firmicutes increases and dominates beginning at day 4. Barn raised ducks contained a significant population of Bacteroidetes in addition to Proteobacteria and Firmicutes at later developmental time points, though this phylum was absent in aviary raised ducks. Genera containing pathogens of anseriformes most often found in industry settings were either absent or found as normal parts of the caecal microbial populations. The high level differences in phylum abundance highlight the importance of well-designed sampling strategies for microbiome based studies. Results showed clear distinctions between Pekin Duck caecal contents and those of Broiler Chickens and Turkey in a qualitative comparison. These data provide a reference point for studies of the Pekin Duck through industry grow-out ages, provide a foundation for understanding the types of bacteria that promote health, and may lead to improved methods to increase yields and decrease instances of disease in agricultural production processes.

Project description:Real-time and automatic detection of chickens (e.g., laying hens and broilers) is the cornerstone of precision poultry farming based on image recognition. However, such identification becomes more challenging under cage-free conditions comparing to caged hens. In this study, we developed a deep learning model (YOLOv5x-hens) based on YOLOv5, an advanced convolutional neural network (CNN), to monitor hens' behaviors in cage-free facilities. More than 1000 images were used to train the model and an additional 200 images were adopted to test it. One-way ANOVA and Tukey HSD analyses were conducted using JMP software (JMP Pro 16 for Mac, SAS Institute, Cary, North Caronia) to determine whether there are significant differences between the predicted number of hens and the actual number of hens under various situations (i.e., age, light intensity, and observational angles). The difference was considered significant at p < 0.05. Our results show that the evaluation metrics (Precision, Recall, F1 and mAP@0.5) of the YOLOv5x-hens model were 0.96, 0.96, 0.96 and 0.95, respectively, in detecting hens on the litter floor. The newly developed YOLOv5x-hens was tested with stable performances in detecting birds under different lighting intensities, angles, and ages over 8 weeks (i.e., birds were 8-16 weeks old). For instance, the model was tested with 95% accuracy after the birds were 8 weeks old. However, younger chicks such as one-week old birds were harder to be tracked (e.g., only 25% accuracy) due to interferences of equipment such as feeders, drink lines, and perches. According to further data analysis, the model performed efficiently in real-time detection with an overall accuracy more than 95%, which is the key step for the tracking of individual birds for evaluation of production and welfare. However, there are some limitations of the current version of the model. Error detections came from highly overlapped stock, uneven light intensity, and images occluded by equipment (i.e., drinking line and feeder). Future research is needed to address those issues for a higher detection. The current study established a novel CNN deep learning model in research cage-free facilities for the detection of hens, which provides a technical basis for developing a machine vision system for tracking individual birds for evaluation of the animals' behaviors and welfare status in commercial cage-free houses.

Project description:The reasons for differences in lipid depositions between fatty-type (F-T) and lean-type (L-T) ducks remain unknown. The present study aimed to compare the growth performance, lipid deposition, and gene expression related to lipid droplet formation in F-T and L-T Pekin ducks. One-day-old, 140 each L-T and F-T male ducks were selected and distributed separately into 20 replicate cages. All ducks were fed commercial diets up to 35 d of age. F-T ducks had a higher average daily gain from 21 to 28 d of age. On 35-day-old, F-T ducks had higher serum levels of high- and low-density lipoprotein cholesterol, cholesterol, albumin, and hydroxybutyrate dehydrogenase activity than L-T ducks. F-T ducks had higher abdominal fat and subcutaneous fat percentages than those in L-T ducks. Liver histological examination showed that L-T ducks contained more lipid droplets in the liver, which gradually decreased with increasing age. The average adipocyte area and diameter of abdominal fat and subcutaneous fat in the F-T and L-T ducks increased with age and were higher in F-T ducks than those in L-T ducks. Furthermore, the gene expression of perilipin 1, perilipin 2, angiopoietin-like protein 4, adipose triglyceride lipase, alpha/beta-hydrolase domain-containing protein 5 (ABHD5), and serine/threonine kinase 17a in the liver, abdominal fat, and subcutaneous fat of F-T ducks was higher than that in L-T ducks, and it increased with age. Compared to L-T ducks, F-T ducks had higher expression of ABHD5 in the abdominal fat and subcutaneous fat and lower expression in the liver. Thus, F-T ducks displayed lower hepatic lipid deposition and a higher percentage of abdominal fat and subcutaneous fat, suggesting that F-T ducks had higher lipid storage capacity due to increased gene expression related to lipid droplets.

Project description:Muscle development and lipid deposition are complex processes regulated by a coordinated expression profile of genes. Pekin ducks displayed a significant difference from Cherry Valley duck in muscle fiber development and IMF contents. Genetic comparisons between these two breeds would contribute to the exploration of mechanisms underlying the phenotypic differences. In the present study, breast tissues of Pekin (BD) and Cherry Valley (CD) ducks were used for RNA-seq in two different time points (3 and 6-week) to investigate the transcriptome basis. A total of 16705 genes from duck breast muscle were detected as positively expressed genes (RPKM>0.1) and 2273 genes were regarded as novel genes. Differentially expressed genes (DEGs) in BD3 VS CD3 were regarded as muscle development-related genes as CD3 grown faster than BD3 in muscle fibers. DEGs in BD6 VS CD6 were regarded as lipid-related genes as a much higher IMF contents in BD6 than CD6 while no differences observed in 3-week. DEGs obtained from BD3 VS BD6 and CD3 VS CD6 were also analyzed as relative genes here. In all, 9 genes (MAT1A, KAZALD1, SHROOM3, KLHL6, MYH13, ALDOA, TNNT2, PDLIM3 and PFKM) were validated as muscle development correlated candidate genes, and another 11 genes (DHCR24, CNTFR, ANGPTL4, BCL6, HIP1, TRIB1, ADORA1, C1QTNF2, CD36, PRKAG2 and ACSL1) were regarded as lipid correlated genes in duck. What’s more, functional analysis confirmed that the immune reaction, ECM and energy metabolism also correlated with muscle development and lipid deposition closely. The present study not only enlarged the duck genetic information pool but also provided a dozen of candidate genes related to muscle development and lipid deposition, which laid a firm foundation for the further studies on molecular mechanisms in duck.

Project description:Aquatic bird bornavirus 1 (ABBV-1) is a neurotropic virus that causes persistent infection in the nervous system of wild waterfowl. This study evaluated whether Pekin ducks, the most common waterfowl raised worldwide, are susceptible to ABBV-1 infection and associated disease. Groups of Pekin ducks were inoculated with ABBV-1 through the intracranial (IC; n, 32), intramuscular (IM; n, 30), and choanal (CH; n, 30) routes. Controls (CO; n, 29) received carrier only. At 1, 12, and 21 weeks postinfection (wpi), 7-14 birds were euthanized to assess virus distribution and lesions. Infection rates in the IC and IM groups were over 70%, while only 4 ducks in the CH group became infected. Neurological signs were observed in 8 ducks only, while over 25% of IC and IM birds had encephalitis and/or myelitis. Seroconversion was highest in the IC and IM groups, and mucosal ABBV-1 RNA shedding was most frequent in the IC group (53%). None of the fertile eggs laid during the experiment tested positive for ABBV-1 RNA. This study shows that Pekin ducks are permissive to ABBV-1 infection and partly susceptible to associated disease. While mucosal shedding may be an important route of transmission, congenital infection appears unlikely.

Project description:Marine plastic pollution is a pressing global issue nowadays. To address this problem, automated image analysis techniques that can identify plastic litter are necessary for scientific research and coastal management purposes. The Beach Plastic Litter Dataset version 1 (BePLi Dataset v1) comprises 3709 original images taken in various coastal environments, along with instance-based and pixel-level annotations for all plastic litter objects visible in the images. The annotations were compiled in the Microsoft Common Objects in Context (MS COCO) format, which was partially modified from the original format. The dataset enables the development of machine-learning models for instance-level and/or pixel-wise identification of beach plastic litter. All original images in the dataset were extracted from beach litter monitoring records operated by the local government of Yamagata Prefecture in Japan. Litter images were taken in different backgrounds, such as sand beaches, rocky beaches, and tetrapods. The annotations for instance segmentation of beach plastic litter were made manually, and were given for all plastics objects, including PET bottles, containers, fishing gear, and styrene foams,all of which were categorized in a single class "plastic litter". Technologies developed using this dataset have the potential to enable further scalability for the estimation of plastic litter volume. This would help researchers, including individuals, and the the government to monitor or analyze beach litter and the corresponding pollution levels.

Project description:This study was conducted to investigate the influence of zinc (Zn) supplementation on growth performance, intestinal development and intestinal barrier function in Pekin ducks. A total of 480, one-day-old male Pekin ducks were divided into 6 groups with 8 replicates: 0 mg/kg Zn, 0 mg/kg Zn +0.5 mg/kg lipopolysaccharide (LPS), 30 mg/kg Zn, 30 mg/kg Zn +0.5 mg/kg LPS, 120 mg/kg Zn, 120 mg/kg Zn +0.5 mg/kg LPS. The duck primary intestinal epithelial cells (DIECs) were divided into 6 groups: D-Zn (Zinc deficiency, treated with 2 µmol/L zinc Chelator TPEN), A-Zn (Adequate Zinc, basal medium), H-Zn (High level of Zn, supplemented with 20 µmol/L Zn), D-Zn + 20 µg/mL LPS, A-Zn + 20 µg/mL LPS, H-Zn + 20 µg/mL LPS. The results were as follows: in vivo, with Zn supplementation of 120 mg/kg reduced LPS-induced decrease of growth performance and intestine damage (P < 0.05), and increased intestinal digestive enzyme activity of Pekin ducks (P < 0.05). In addition, Zn supplementation also attenuated LPS-induced intestinal epithelium permeability (P < 0.05), inhibited LPS-induced the expression of proinflammatory cytokines and apoptosis-related genes (P < 0.05), as well as reduced LPS-induced the intestinal stem cells mobilization of Pekin ducks (P < 0.05). In vitro, 20 µmol/L Zn inhibited LPS-induced expression of inflammatory factors and apoptosis-related genes (P < 0.05), promoted the expression of cytoprotection-related genes, and attenuated LPS-induced intestinal epithelium permeability in DIECs (P < 0.05). Mechanistically, 20 µmol/L Zn enhanced tight junction protein markers including CLDN-1, OCLD, and ZO-1 both at protein and mRNA levels (P < 0.05), and also increased the level of phosphorylation of TOR protein (P < 0.05) and activated the TOR signaling pathway. In conclusion, Zn improves growth performance, digestive enzyme activity, and intestinal barrier function of Pekin ducks. Importantly, Zn also reverses LPS-induced intestinal barrier damage via enhancing the expression of tight junction proteins and activating the TOR signaling pathway.
