Project description:The NIR excitation and large anti-Stokes shift of upconversion nanoparticles (UCNPs) have made them an ideal choice as biological nanoprobes. A key challenge in the field is to confer biorecognition units to UCNPs so that they can be used to probe specific targets in biological systems. While various agents have been combined with UCNPs to meet such a challenge, most studies are limited to small molecules, while biomolecules such as metalloproteins that possess much higher affinity and selectivity for metal ions have not been explored. Herein we demonstrate that fusion of zwitterion-coated UCNPs with azurin, a member of a family of redox-active copper proteins called cupredoxins that play important roles in diverse biological functions, can serve as an ideal platform for the label-free upconversion luminescence sensing of Cu2+ with a ratiometric response. The selectively binding of apo-azurin with Cu2+ induce a significant absorbance at about 625 nm, and hence decreases the red emission of the UCNPs. In contrast, the green emission of the UCNPs remains constant and acts as an internal standard reference for the ratiometric sensing of Cu2+. This approach opens a new window for the development of assays for biosensing based on a combination of specific metalloproteins with UCNPs.

Project description:Despite intense efforts on surface functionalization to generate hydrophilic upconversion nanoparticles (UCNPs), long-term colloidal stability in physiological buffers remains a major concern. Here we quantitatively investigate the competitive adsorption of phosphate, carboxylic acid and sulphonic acid onto the surface of UCNPs and study their binding strength to identify the best conjugation strategy. To achieve this, we designed and synthesized three di-block copolymers composed of poly(ethylene glycol) methyl ether acrylate and a polymer block bearing phosphate, carboxylic or sulphonic acid anchoring groups prepared by an advanced polymerization technique, Reversible Addition Fragmentation Chain Transfer (RAFT). Analytical tools provide the evidence that phosphate ligands completely replaced all the oleic acid capping molecules on the surface of the UCNPs compared with incomplete ligand exchange by carboxylic and sulphonic acid groups. Meanwhile, simulated quantitative adsorption energy measurements confirmed that among the three functional groups, the calculated adsorption strength for phosphate anchoring ligands is higher which is in good agreement with experimental results regarding the best colloidal stability, especially in phosphate buffer solution. This finding suggests that polymers with multiple anchoring negatively charged phosphate moieties provide excellent colloidal stability for lanthanide ion-doped luminescent nanoparticles for various potential applications.

Project description:Multicolor upconversion luminescence of Er3+ was successfully enhanced by optimizing the interface in dye-coordinated nanoparticles with a core/shell structure. Red and green upconversion emissions of Er3+ were obtained at the interface of oxide nanoparticles via the intramolecular energy transfer from the coordinating squaraine dye with high light-absorption ability, which was more efficient than emissions through the energy transfer from metal ions such as Yb3+. Additionally, CaF2 nanoparticles as a core material minimized the energy loss with nonradiative downward relaxations in Er3+, resulting in the observation of unusual blue upconversion emissions from the upper energy level of Er3+ by nonlaser excitation using a continuous-wave (CW) Xe lamp at an excitation power of 1.2 mW/cm2.

Project description:The emerging optical multiplexing within nanoscale shows super-capacity in encoding information by using lifetime fingerprints from luminescent nanoparticles. However, the optical diffraction limit compromises the decoding accuracy and throughput of the nanoparticles during conventional widefield imaging. This, in turn, challenges the quality of nanoparticles to afford the modulated excitation condition and further retain the multiplexed optical fingerprints for super-resolution multiplexing. Here we report a tailor-made multiplexed super-resolution imaging method using the lifetime-engineered upconversion nanoparticles. We demonstrate that the nanoparticles are bright, uniform, and stable under structured illumination, which supports a lateral resolution of 185 nm, less than 1/4th of the excitation wavelength. We further develop a deep learning algorithm to coordinate with super-resolution images for more accurate decoding compared to a numeric algorithm. We demonstrate a three-channel super-resolution imaging based optical multiplexing with decoding accuracies above 93% for each channel and larger than 60% accuracy for potential seven-channel multiplexing. The improved resolution provides high throughput by resolving the particles within the diffraction-limited spots, which enables higher multiplexing capacity in space. This lifetime multiplexing super-resolution method opens a new horizon for handling the growing amount of information content, disease source, and security risk in modern society.

Project description:Photon upconversion (UC) based on triplet-triplet annihilation is a very promising phenomenon with potential application in several areas, though, due to the intrinsic mechanism, the achievement of diffusion-limited solid materials with air-stable UC is still a challenge. Herein, we report UC coordination polymer nanoparticles (CPNs) combining sensitizer and emitter molecules especially designed with alkyl spacers that promote the amorphous character. Beyond the characteristic constraints of crystalline MOFs, amorphous CPNs facilitate high dye density and flexible ratio tunability. To show the universality of the approach, two types of UC-CPNs are reported, exhibiting highly photostable UC in two different visible spectral regions. Given their nanoscale, narrow size distribution, and good chemical/colloidal stability in water, the CPNs were also successfully printed as anticounterfeiting patterns and used to make highly transparent and photostable films for luminescent solar concentrators, both showing air-stable UC.

Project description:Optical coherence tomography (OCT) is an emerging 3D imaging technique that allows quantification of intrinsic optical properties such as scattering coefficient and back-scattering coefficient, and has proved useful in distinguishing delicate microstructures in the human brain. The origins of scattering in brain tissues are contributed by the myelin content, neuron size and density primarily; however, no quantitative relationships between them have been reported, which hampers the use of OCT in fundamental studies of architectonic areas in the human brain and the pathological evaluations of diseases. Here, we built a generalized linear model based on Mie scattering theory that quantitatively links tissue scattering to myelin content and neuron density in the human brain. We report a strong linear relationship between scattering coefficient and the myelin content that is retained across different regions of the brain. Neuronal cell body turns out to be a secondary contribution to the overall scattering. The optical property of OCT provides a label-free solution for quantifying volumetric myelin content and neuron cells in the human brain.

Project description:Multifunctional integration on single upconversion nanoparticles (UCNPs), such as the simultaneous achievement of imaging, sensing, and therapy, will be extremely attractive in various application fields. Herein, we demonstrated that single core/shell NaGdF4:Yb/Er-based UCNPs (<10 nm) with a highly Yb3+ or Nd3+ doped shell simultaneously exhibited good upconversion luminescence (UCL), temperature sensing, and photothermal conversion properties under 980 or 808 nm excitation, respectively. The spatial separation between the emission/sensing core and the heating shell was able to tailor the competition between the light and heat generation processes, and hence higher UCL efficiency and enhanced heating capability were achieved by introducing the rational core/shell design. Especially, Nd3+-sensitized core/shell nanoparticles were excitable to the laser at a more biocompatible wavelength of 808 nm, and hence the heating effect of water was greatly minimized. The heating and sensing capabilities of Nd3+-sensitized core/shell UCNPs with smaller sizes (<10 nm) were confirmed in aqueous environment under single 808 nm laser excitation, implying their promising applications in imaging-guided and temperature-monitored photothermal treatments.

Project description:Three-dimensional (3D) rapid prototyping technology based on near-infrared light-induced polymerization of photocurable compositions containing upconversion nanomaterials has been explored. For this aim, the rationally-designed core/shell upconversion nanoparticles NaYF4:Yb3+,Tm3+/NaYF4, with the distinct ultraviolet-emitting lines and unprecedentedly high near-infrared to ultraviolet conversion efficiency of [Formula: see text] have been used. The upconverted ultraviolet photons were capable to efficiently activate photoinitiators contained in light-sensitive resins under moderate intensities of NIR excitation below 10 W cm-2 and induce generation of radicals and photopolymerization in situ. Near infrared-activated polymerization process, both at the millimeter and sub-micron scales, was investigated. Polymeric macro- and microstructures were fabricated by means of near infrared laser scanning photolithography in the volume of liquid photocurable compositions with focused laser light at 975 nm wavelength. Examination of the polymerization process in the vicinity of the nanoparticles shows strong differences in the rate of polymer shell growth on flat and edge nanoparticle sides. This phenomenon mainly defines the resolution of the demonstrated near infrared - ultraviolet 3D printing technology at the micrometer scale level.

Project description:Lanthanide upconversion nanophosphor (UCNP) has attracted increasing attention for potential applications in bioimaging due to its excellence in deep and high contrast imaging. To date, most upconversion imaging applications were demonstrated in dark surroundings without ambient light for higher signal-to-noise ratio, which hindered the application of optical imaging guided surgery. Herein, the new established NaLuF?-based UCNP (NaLuF?:Yb,Tm, ~17 nm) with bright upconversion emission around 800 nm as imaging signal was used to realize imaging under ambient light to provide more convenient for clinician. Moreover, due to the existance of heavy element lutetium (Lu) in the host lattice, the NaLuF?:Yb,Tm nanoparticles can also be used as an X-ray CT imaging agent to enhance the imaging depth and in vivo imaging resolution.
