Characterization of the complete chloroplast genome of Hippophae salicifolia D. Don (Elaeagnaceae).

ABSTRACT: Seabuckthorn (Hippophae L.) is a pioneer species widely distributed in Eurasia. We assembled and annotated the chloroplast genome of Hippophae salicifolia from Illumina pair-end data, which was 155,420 bp in length with 36.74% GC content; this plastome featured a quadripartite structure with two copies of a large inverted repeat (IR) of 26,528 bp separated by large single copy (LSC) region of 83,504 bp and small single copy region (SSC) of 18,860 bp. In total, 131 complete genes were annotated, including 38 tRNA, eight rRNA, and 85 protein-coding genes. Phylogenetic analysis recovered H. salicifolia and H. gyantsensis as monophyletic and sister to all other Hippophae species for which complete plastome sequences have been published.

Project description:Elaeagnus pungens is an evergreen shrub with high medicinal values. In this study, the complete chloroplast (cp) genome of E. pungens was characterized. The chloroplast genome of E. pungens is 152,218 bp in length, consisting of two 25,876 bp inverted repeats, 18,231 bp small single copy region, and 82,235 bp large single copy region. The chloroplast genome contains 113 unique genes, including 79 protein-coding genes, 30 tRNA genes, and 4 rRNA genes. Phylogenomic analysis revealed that E. pungens and species from Elaeagnus formed a monophyletic clade sister to the clade consisting of species from Hippophae.

Characterization of the complete chloroplast genome sequence of Elaeagnus henryi Warb. ex Diels (Elaeagnaceae).

Project description:Elaeagnus henryi Warb. ex Diels belongs to the Elaeagnaceae. Here, we reveal the complete chloroplast genome of Elaeagnus henryi. The complete chloroplast genome is 152,244 bp in length and contains a large single-copy (LSC) region of 82,235 bp, a small single-copy (SSC) region of 18,279 bp and a pair of inverted repeats (IRs) of 25,865 bp. There are 126 genes, including 81 protein-coding, 37 transfer RNA (tRNA), and eight ribosomal RNA (rRNA) genes. The total GC content of the chloroplast genome sequence is 37.1%. The maximum-likelihood phylogenetic analysis indicated that E. henryi was sister to Elaeagnus pungens (MW145133). The result may be because the species are advanced and developed from the same ancestor.

Molecular docking studies of anti-cancerous candidates in Hippophae rhamnoides and Hippophae salicifolia.

Project description:Actinorhizal plants contain numerous antioxidants that may play a crucial role in preventing the formation of tumors. H-Ras p21, a member of the Ras-GTPase family, is a promising target to treat various kinds of cancers. An in silico docking study was carried out to identify the inhibitory potential of compounds of these plants against H-Ras by using Discovery Studio 3.5 and by using Autodock 4.2. Docking studies revealed that four compounds, isorhamnetin-7-rhamnoside, quercetin-3-glucoside-7-rhamnoside (present in H. rhamnoides), zeaxanthin, and translutein (present in H. salicifolia) significantly bind with binding energies -17.1534, -14.7936, -10.2105 and -17.2217 Kcal/mol, respectively, even though they slightly deviate from Lipinski's rule. Absorption, distribution, metabolism, excretion and toxicity (ADME/tox) analyses of these compounds and their stereoisomers showed that they were less toxic and non-mutagenic. Amongst them, isorhamntein-7-rhamnoside showed hepatotoxicity. Hence, these compounds can be further investigated in vivo to optimize their formulation and concentration and to develop potential chemical entities for the prevention and treatment of cancers.

Amelioration of antioxidant potential, toxicity, and antihyperglycemic activity of Hippophae salicifolia D. Don leaf extracts in alloxan-induced diabetic rats.

Project description:Efficacy of several plant extracts in the clinical research for modulating oxidative stress correlated with diabetes mellitus (DM) is well documented. In the present study, we investigated the in vitro antioxidant activity, toxicity, and anti-diabetic activity of methanolic extract of Hippophae salicifolia leaves in normal and alloxan-induced diabetic wistar rats. H. salicifolia leaves were found to be rich in antioxidants. The acute toxicity test of methanolic extract of H. salicifolia leaves revealed that the median lethal dose (LD50) was found to be 3.92 g/kg body weight in mice. Administration of H. salicifolia leaves at 200 mg/kg and 400 mg/kg in alloxan-induced diabetic rats illustrated significant reduction (22% and 39%, respectively) in fasting blood glucose compared to diabetic control. Both the doses were found to be effective when compared to diabetic rats. The Hippophae-treated diabetic rats showed significant increase in the endogenous antioxidant enzymes, superoxide dismutase (50% and 74%, respectively), glutathione peroxidase (57% and 41%, respectively) and decrease in malondialdehyde (33% and 15%, respectively) levels. These results suggested that the methanolic leaf extract of H. salicifolia enhanced the antioxidant defence against reactive oxygen species produced under hyperglycaemic conditions.

Dietary and bioactive properties of the berries and leaves from the underutilized Hippophae salicifolia D. Don grown in Northeast India.

Project description:The physico-chemical, polyphenols, antioxidant and antibacterial properties of berries and mixture of male and female leaves of Hippophae salicifolia were investigated. The mineral, vitamin C, sugar, total protein, and total tannin contents of the berries and the leaves were evaluated. Further, the extracts of berries and mixture of leaves samples obtained by successive solvent extraction were investigated for their polyphenols, antioxidant and antibacterial properties. Total phenolic content was highest in leaves-methanol extract (157.97 ± 2.09 mg GAE/g) followed by berries-aqueous extract (48.45 ± 1.94 mg GAE/g), while total flavonoid was predominant in leaves-acetone extract (75.64 ± 3.21 mg QE/g) and berries-methanol extract (28.93 ± 2.08 mg QE/g). Gallic acid, caffeic acid, and rutin were the major polyphenols confirmed by HPLC analysis. Berries-aqueous and leaves-methanol extracts showed excellent global antioxidant score. Best antibacterial activity was observed by methanol extracts against eight different strains. Overall, the leaves and berries of Hippophae salicifolia collected from Northeast India exhibited good antioxidant and antibacterial activity and can be utilized by food and pharmaceutical sectors.Supplementary informationThe online version contains supplementary material available at 10.1007/s10068-021-00988-8.

Optimization of polyphenol extraction from Hippophae salicifolia D. Don leaf using supercritical CO2 by response surface methodology.

Project description:In this study, an eco-friendly supercritical carbon dioxide (SC-CO2) extraction of polyphenolic compounds from Hippophae salicifolia leaf was optimized to achieve the highest extraction yield with maximum total phenolic content (TPC) and minimum IC50. The central composite design was used to establish an experimental design for RSM. The effect of the pressure, temperature, carbon dioxide flow rate, and co-solvent amount was scrutinized using variance analysis (ANOVA). Under optimized condition (25.13 MPa, 47.53 °C, 14.47 g/min, and 2.43%), the experimental data (yield of extraction: 4.38%, TPC: 84.31 mg GAE/g, and IC50: 41.94 µg/mL) showed good agreement with the predicted values (yield of extraction: 4.53%, TPC: 83.37 mg GAE/g, and IC50: 40.2 µg/mL). Nine polyphenolic compounds: gallic acid, caffeic acid, ferulic acid, vanillic acid, p-coumaric acid, quercetin, myricetin, kaempferol, and rutin were analyzed in SC-CO2 extract using HPLC. SC-CO2 extraction was more selective for ferulic acid, myricetin, and quercetin extraction. Project description:The complete chloroplast genome sequence of Hippophae rahmnoides subsp. sinensis was characterized from Illumina pair-end sequencing. The chloroplast genome of H. rahmnoides subsp. sinensis was 156,355 bp in length, containing a large single-copy region (LSC) of 84,002 bp, a small single-copy region (SSC) of 19,055 bp, and two inverted repeat (IR) regions of 26,649 bp. The overall GC content is 36.6%, while the corresponding values of the LSC, SSC, and IR regions are 64.5%, 69.2%, and 60.1%, respectively. The genome contains 131 complete genes, including 88 protein-coding genes, 38 tRNA genes (29 tRNA species), and 8 rRNA genes (4 rRNA species). The neighbour-joining phylogenetic analysis showed that H. rahmnoides subsp. sinensis and H. rahmnoides clustered together as sisters to other H. rahmnoides species.

Here, we reported and characterized the complete chloroplast (cp) genome sequence of S. blumei (GenBank accession number: MN418904) to provide genomic resources for promoting its conservation. The total chloroplast genome is 155,957 bp in length and contains the typical chloroplast structure, including two inverted repeat (IR) regions of 26,343 bp, a large single-copy (LSC) region of 84,384 bp, and a small single-copy (SSC) region of 18,887 bp. The overall guanine-cytosine (GC) content of the S. blumei chloroplast genome is 36.8%. The cp genome encodes 133 unique genes, including 85 protein-coding genes (PCGs), 40 tRNA genes, and 8 rRNA genes. We used the cp genome of S. blumei and 26 other cp genomes to perform a phylogenetic analysis, which indicated that S. blumei was closely related to S. martini in Rosaceae.

Characterization of chloroplast genome and phylogenetic analysis of the Elaeagnus hybrid variety E. 'viridis' (Elaeagnaceae).

Project description:Elaeagnus 'viridis', an artificial hybrid of E. macrophylla (♂) Thunb. (1784) × E. pungens (♀) Thunb. (1784), is known for its economic and ecological value. In this study, we sequenced and assembled the whole chloroplast (cp) genome of E. 'viridis'. The results showed that its cp genome was 152,284 bp long, showing a typical quadripartite structure and containing a large single-copy region (LSC, 82,299 bp), a small single-copy region (SSC, 18,239 bp), and a pair of inverted repeats (IRs, 51,746 bp). The cp genome contains 132 genes, including 86 protein-coding genes (PCGs), 38 tRNA genes, and 8 rRNA genes. Phylogenetic analysis based on 66 common PCGs revealed that E. 'viridis' is most closely related to its maternal parent E. pungens. The chloroplast genomic information reported in this study will shed some useful light for further genetic studies in the genus Elaeagnus.

However, the complete chloroplast genome of E. oldhamii has never been studied at present. We obtained the complete chloroplast genome of E. oldhamii, which was 152,283 bp in length, with a typical quadripartite structure that includes a large single-copy region of 82,229 bp, a small single-copy region of 18,256 bp, and 2 inverted repeat (IR) regions of 25,899 bp. The genome contained 128 unique genes with a GC content of 37%, including 83 protein-coding genes, 37 tRNAs, and 8 rRNAs. Phylogenetic analysis suggested that E. oldhamii was closely related to E. glabra and E. macrophylla.
