Project description:Amylosucrase from Neisseria polysaccharea (NpAS) synthesizes α-1,4 glucan polymer from sucrose. In this study, we coated various botanical sources of raw starch with an α-glucan layer generated by NpAS to improve physicochemical properties. Field-emission scanning electron microscopy demonstrated that all surfaces of the starch granules were successfully coated via the NpAS reaction. X-ray diffraction analysis revealed that the crystallinity decreased and the crystal pattern changed to C-type as an amylose layer formed around the surface of the starch granules. Based on rapid viscosity and differential scanning calorimetry analyses, the gelatinization resistance of the α-glucan-coated starch increased owing to decreased viscosity and increased melting temperature. Therefore, the α-glucans coated the starches by enzymatic reactions of various botanical sources; these have applicability in the food and starch industries owing to various physicochemical properties such as enhanced thermostability.Supplementary informationThe online version contains supplementary material available at 10.1007/s10068-022-01113-z.

Project description:Influences of allelic variations in starch synthesis-related genes (SSRGs) on rice grain quality were examined. A total of 187 nonglutinous Korean rice varieties, consisting of 170 Japonica and 17 Tongil-type varieties, were grown in the field and in two greenhouse conditions. The percentages of head rice and chalky grains, amylose content, alkali digestion value, and rapid visco-analysis characteristics were evaluated in the three different environments. Among the 10 previously reported SSRG markers used in this study, seven were polymorphic, and four of those showed subspecies-specific allele distributions. Six out of the seven polymorphic SSRG markers were significantly associated with at least one grain quality trait (R (2) > 0.1) across the three different environments. However, the association level and significance were markedly lower when the analysis was repeated using only the 170 Japonica varieties. Similarly, the significant associations between SSRG allelic variations and changes in grain quality traits under increased temperature were largely attributable to the biased allele frequency between the two subpopulations. Our results suggest that within Korean Japonica varieties, these 10 major SSRG loci have been highly fixed during breeding history and variations in grain quality traits might be influenced by other genetic factors.

Project description:Green synthesis of nanoparticles by using starch has recently attracted considerable attention due to their biodegradability, non-toxicity, and cost effectiveness. The data presented in this article are related to the article entitled "Evaluation of antibacterial activity of Ag/Fe3O4 nanocomposites synthesized by using starch" (Ghaseminezhad and Shojaosadati, 2016) [1]. Here, Fe3O4 nanoparticles and silver nanoparticles were synthesized by using starch under alkaline condition. Hydrodynamic diameter of starch and starch coated silver nanoparticles were determined under heat treatment and different pH. This data also display absorbance peak of silver nanoparticles synthesized by starch under different pH conditions (6.5, 8, and 10). Iodometric titration confirmed that both components of starch (amylose and amylopectin) can adsorb on the surface of Fe3O4 nanoparticles.

Project description:Cherry tomato (Lycopersicon esculentum M.) is considered a healthy fruit worldwide due to its wide range of nutrients. Flavonol, one of the major nutrients in cherry tomato, has antioxidant and cell-modulating properties. In this study, we showed a correlation between the expression of SlMYB12 and flavonol content (R2 = 0.922). To characterize the function of SlMYB12, SlMYB12-overexpressing transgenic tomato plants were generated in three different cherry tomato varieties. Significant increases in flavonol content and flavonol biosynthetic gene expression were identified in SlMYB12-overexpressing plants. Therefore, we suggest that SlMYB12 plays a positive role in the flavonol biosynthesis pathway in cherry tomatoes, which further indicates a potential role as a marker in analyzing flavonol content in different cherry tomato varieties.

Project description:Cannabis sativa L. (hemp) is a plant used in the textile industry and green building material industry, as well as for the phytoremediation of soil, medical treatments, and supplementary food products. The synergistic effect of terpenes, flavonoids, and cannabinoids in hemp extracts may mediate the biogenic synthesis of metal nanoparticles. In this study, the chemical composition of aqueous leaf extracts of three varieties of Romanian hemp (two monoecious, and one dioecious) have been determined by Fourier-Transformed Infrared spectroscopy (FT-IR), high-performance liquid chromatography, and mass spectrometry (UHPLC-DAD-MS). Then, their capability to mediate the green synthesis of silver nanoparticles (AgNPs) and their pottential antibacterial applications were evaluated. The average antioxidant capacity of the extracts had 18.4 ± 3.9% inhibition determined by 2,2-diphenyl-1-picrylhydrazyl (DPPH•) and 78.2 ± 4.1% determined by 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS™) assays. The total polyphenolic content of the extracts was 1642 ± 32 mg gallic acid equivalent (GAE) L-1. After this, these extracts were reacted with an aqueous solution of AgNO3 resulting in AgNPs, which were characterized by UV-VIS spectroscopy, FT-IR, scanning electron microscopy (SEM-EDX), and dynamic light scattering (DLS). The results demonstrated obtaining spherical, stable AgNPs with a diameter of less than 69 nm and an absorbance peak at 435 nm. The mixture of extracts and AgNPs showed a superior antioxidant capacity of 2.3 ± 0.4% inhibition determined by the DPPH• assay, 88.5 ± 0.9% inhibition as determined by the ABTS•+ assay, and a good antibacterial activity against several human pathogens: Escherichia coli, Klebsiella pneumoniae, Pseudomonas fluorescens, and Staphylococcus aureus.

Project description:In order to investigate starch accumulation, and the enzymes activity changes and the expression levels of genes and their relationships among them at different developmental stages of wheat grain. We choose Annong9912 and E28 were used in the study. During starch accumulating rate and grain filling rate, and there were obvious genotype difference between Annong9912 and E28. Whether low or high starch content of starch content, the accumulation courses of amylopectin, amylose and total starch were well fitted to the logistic equation by relating starch contents against DAP. The simulation parameters revealed that the higher contents of amylopectin and amylose resulted from earlier initiating accumulation time and greater accumulation rate. And amylose, amylopectin and total starch accumulation rate of two wheat cultures were significantly and positively correlated with activities of SBE, SSS and GBSS, but amylose accumulation rate of E28 had no correlation with the activities of SBE. In addition, there were significant correlations among activities of SBE, SSS and GBSS in two wheat cultivars. We speculated that these enzymes proteins may have a coordinating action in starch biosynthesis within the amyloplast, operating as functional multiprotein complexes. And expression levels of enzyme genes demonstrated a single-peak curve, and 12-18 DAP reached their peaks and then began to drop, and all had high expression level in earlier stage of endosperm development, but in E28 were higher than in Annong9912. The GBSS-I transcripts on average were expressed over 60 times more than GBSS-II transcript in E28. SBE, SSS, DBE may control starch synthesis at the transcriptional level, and GBSS-I may control starch synthesis at the post transcriptional level. The expression level of DBE on average was lower than SS-1 and SBE-IIa genes, and similar to SS-III and SBE-IIb genes, but higher than GBSS-I and GBSS-II genes.

Project description:A polyphasic approach was applied to characterize 35 G. diazotrophicus isolates obtained from sugarcane varieties cultivated in Brazil. The isolates were analyzed by phenotypic (use of different carbon sources) and genotypic tests (ARDRA and RISA-RFLP techniques). Variability among the isolates was observed in relation to the carbon source use preference. Glucose and sucrose were used by all isolates in contrast to myo-inositol, galactose and ribose that were not metabolized. The results of the analysis showed the presence of two groups clustered at 68% of similarity. The genetic distance was higher when RISA-RFLP analysis was used. Analysis of 16S rDNA sequences from isolates showed that all of them belonged to the G. diazotrophicus species. Neither effect of the plant part nor sugarcane variety was observed during the cluster analysis. The observed metabolic and genetic variability will be helpful during the strain selection studies for sugarcane inoculation in association with sugarcane breeding programs.

Project description:Soluble starch synthase (SS) is one of the key enzymes involved in amylopectin biosynthesis in plants. However, no information is currently available about this gene family in the important fruit crop banana. Herein, we characterized the function of MaSSIII-1 in amylopectin metabolism of banana fruit and described the putative role of the other MaSS family members. Firstly, starch granules, starch and amylopectin content were found to increase during banana fruit development, but decline during storage. The SS activity started to increase later than amylopectin and starch content. Secondly, four putative SS genes were cloned and characterized from banana fruit. Among them, MaSSIII-1 showed the highest expression in banana pulp during fruit development at transcriptional levels. Further Western blot analysis suggested that the protein was gradually increased during banana fruit development, but drastically reduced during storage. This expression pattern was highly consistent with changes in starch granules, amylopectin content, and SS activity at the late phase of banana fruit development. Lastly, overexpression of MaSSIII-1 in tomato plants distinctly changed the morphology of starch granules and significantly increased the total starch accumulation, amylopectin content, and SS activity at mature-green stage in comparison to wild-type. The findings demonstrated that MaSSIII-1 is a key gene expressed in banana fruit and responsible for the active amylopectin biosynthesis, this is the first report in a fresh fruit species. Such a finding may enable the development of molecular markers for banana breeding and genetic improvement of nutritional value and functional properties of banana fruit.

Project description:The starch-stabilized Ag nanoparticles were successfully synthesized via a reduction approach and characterized with SPR UV/Vis spectroscopy, TEM, and HRTEM. By utilizing the redox reaction between Ag nanoparticles and Hg(2+), and the resulted decrease in UV/Vis signal, we develop a colorimetric method for detection of Hg(2+) ion. A linear relationship stands between the absorbance intensity of the Ag nanoparticles and the concentration of Hg(2+) ion over the range from 10 ppb to 1 ppm at the absorption of 390 nm. The detection limit for Hg(2+) ions in homogeneous aqueous solutions is estimated to be ~5 ppb. This system shows excellent selectivity for Hg(2+) over other metal ions including Na(+), K(+), Ba(2+), Mg(2+), Ca(2+), Fe(3+), and Cd(2+). The results shown herein have potential implications in the development of new colorimetric sensors for easy and selective detection and monitoring of mercuric ions in aqueous solutions. Project description:A microwave reaction to convert 99 ± 1% of Ag+ to silver nanoparticles (AgNPs) of size <10 nm within 4.5 min with a specific production rate and energy input of 5.75 mg AgNP L-1 min-1 and 5.45 W mL-1 reaction volume was developed. The glucose reduced and food grade starch stabilized particles remained colloidally stable with less than a 4% change in the surface plasmon resonance band at 425-430 nm at t > 300 days. TEM determined the size of AgNPs, while TEM-EDS and XRD verified elemental composition. The conversion was determined by inductively coupled plasma atomic emission spectroscopy (ICP-AES) and thermal gravimetric analysis (TGA). Additionally, the required silver to starch input mass ratio, 1.0:1.3, to produce colloidally stabilized AgNPs is significantly reduced compared to previous studies. The antibacterial activity of freshly prepared AgNPs and AgNPs aged >300 days was demonstrated against E. coli as determined by agar diffusion assays. This result, corroborated by spectrophotometric and TEM measurements, indicates long-term colloidal stability of the product. Thus, this study sustainably produced antibacterial AgNPs from minimal inputs. In the broader context, the current work has quantified a sustainable platform technology to produce sphere-like inorganic nanoparticles with antimicrobial properties.
