Project description:The neural systems supporting scene-perception and spatial-memory systems of the human brain are well-described. But how do these neural systems interact? Here, using fine-grained individual-subject fMRI, we report three cortical areas of the human brain, each lying immediately anterior to a region of the scene perception network in posterior cerebral cortex, that selectively activate when recalling familiar real-world locations. Despite their close proximity to the scene-perception areas, network analyses show that these regions constitute a distinct functional network that interfaces with spatial memory systems during naturalistic scene understanding. These "place-memory areas" offer a new framework for understanding how the brain implements memory-guided visual behaviors, including navigation.

Project description:Human retrosplenial complex (RSC), located in medial parietal cortex, has been implicated in numerous cognitive functions, including scene perception, spatial navigation, and autobiographical memory retrieval. Recently, a posterior-anterior distinction within RSC was proposed, such that posterior aspects process scene-related visual information (constituting a medial place area [MPA]), whereas anterior aspects process information that is vividly retrieved from memory, thereby supporting remembering and potentially navigation. Here, we tested this proposed distinction in a single group of participants (both male and female) using fMRI with both perceptual and mnemonic tasks. After completing a resting-state scan, participants performed a task that required constructing scenes from memory and completed a scene selectivity localizer task. We tested directly perceptual and mnemonic responses in MPA and an anterior, connectivity-defined region (CON), which showed strong functional connectivity with anterior parahippocampal place area. A double dissociation was observed, such that CON was more strongly activated during scene construction than was MPA, whereas MPA was more perceptually responsive than CON. Further, peak responses from the scene construction task were anterior to perceptual peaks in all but 1 participant and hemisphere. Finally, through analyses of the posterior-anterior response profiles, we identify the fundus of the parieto-occipital sulcus as a potential location for the crossover from perceptual to mnemonic representations and highlight a potential left-hemisphere advantage for mnemonic representations. Collectively, our results support a distinction between posterior and anterior aspects of the RSC, suggesting that more specific functional-anatomic terms should be used in its place in future work.SIGNIFICANCE STATEMENT The retrosplenial complex (RSC) has been implicated in vision, spatial cognition, and memory. We previously speculated on a potential posterior-anterior distinction within RSC for scene perception and memory-based scene construction/navigation. Here, we tested this distinction through a combination of resting-state, perceptual, and mnemonic task data. Consistent with our predictions, we demonstrate that perceptual responses peak consistently posterior of those elicited by memory-based scene construction within the broader RSC. Further, we highlight (1) the fundus of the parieto-occipital sulcus as a landmark for the transition between these representations, (2) the anterior bank of parieto-occipital sulcus as the point of maximal separation between these representations, and (3) identify a potential hemispheric asymmetry in mnemonic representations. These data support functional dissociations within RSC.

Project description:In navigating our environment, we rapidly process and extract meaning from visual cues. However, the relationship between visual features and categorical representations in natural scene perception is still not well understood. Here, we used natural scene stimuli from different categories and filtered at different spatial frequencies to address this question in a passive viewing paradigm. Using representational similarity analysis (RSA) and cross-decoding of magnetoencephalography (MEG) data, we show that categorical representations emerge in human visual cortex at ∼180 ms and are linked to spatial frequency processing. Furthermore, dorsal and ventral stream areas reveal temporally and spatially overlapping representations of low and high-level layer activations extracted from a feedforward neural network. Our results suggest that neural patterns from extrastriate visual cortex switch from low-level to categorical representations within 200 ms, highlighting the rapid cascade of processing stages essential in human visual perception.

Project description:Reactivation of earlier perceptual activity is thought to underlie long-term memory recall. Despite evidence for this view, it is unclear whether mnemonic activity exhibits the same tuning properties as feedforward perceptual activity. Here, we leverage population receptive field models to parameterize fMRI activity in human visual cortex during spatial memory retrieval. Though retinotopic organization is present during both perception and memory, large systematic differences in tuning are also evident. Whereas there is a three-fold decline in spatial precision from early to late visual areas during perception, this pattern is not observed during memory retrieval. This difference cannot be explained by reduced signal-to-noise or poor performance on memory trials. Instead, by simulating top-down activity in a network model of cortex, we demonstrate that this property is well explained by the hierarchical structure of the visual system. Together, modeling and empirical results suggest that computational constraints imposed by visual system architecture limit the fidelity of memory reactivation in sensory cortex. Neural activity from perception is thought to be reactivated during memory recall. Here, the authors show that memory reactivation in visual cortex exhibits different tuning properties from those observed during perception.

Project description:Apolipoprotein E (APOE) ε4 is a major genetic risk factor for Alzheimer's disease (AD), yet the mechanisms by which APOE-ε4 influences early-life brain function, and hence, in turn, risk for later-life AD, are poorly understood. Here, we report a novel, and selective, pattern of functional brain activity alteration in healthy young adult human APOE-ε4 carriers. Our findings suggest that APOE-ε4 may influence vulnerability to poorer later life cognitive health via its effect on posteromedial cortex (PMC), a hub region within a brain network involved in spatial processing, and necessary for episodic memory. In two neuroimaging tasks, APOE-ε4 carriers showed an inability to effectively modulate PMC during scene, but not face and object, working memory and perception. This striking pattern overlaps both functionally and topographically, with the earliest cognitive deficits seen in clinical AD, as well as reported alterations in the default network in amyloid-positive individuals at increased risk of AD.

Project description:In complex real-world scenes, image content is conveyed by a large collection of intertwined visual features. The visual system disentangles these features in order to extract information about image content. Here, we investigate the role of one integral component: the content of spatial frequencies in an image. Specifically, we measure the amount of image content carried by low versus high spatial frequencies for the representation of real-world scenes in scene-selective regions of human visual cortex. To this end, we attempted to decode scene categories from the brain activity patterns of participants viewing scene images that contained the full spatial frequency spectrum, only low spatial frequencies, or only high spatial frequencies, all carefully controlled for contrast and luminance. Contrary to the findings from numerous behavioral studies and computational models that have highlighted how low spatial frequencies preferentially encode image content, decoding of scene categories from the scene-selective brain regions, including the parahippocampal place area (PPA), was significantly more accurate for high than low spatial frequency images. In fact, decoding accuracy was just as high for high spatial frequency images as for images containing the full spatial frequency spectrum in scene-selective areas PPA, RSC, OPA and object selective area LOC. We also found an interesting dissociation between the posterior and anterior subdivisions of PPA: categories were decodable from both high and low spatial frequency scenes in posterior PPA but only from high spatial frequency scenes in anterior PPA; and spatial frequency was explicitly decodable from posterior but not anterior PPA. Our results are consistent with recent findings that line drawings, which consist almost entirely of high spatial frequencies, elicit a neural representation of scene categories that is equivalent to that of full-spectrum color photographs. Collectively, these findings demonstrate the importance of high spatial frequencies for conveying the content of complex real-world scenes.

Project description:Three cortical areas (Retro-Splenial Cortex (RSC), Transverse Occipital Sulcus (TOS) and Parahippocampal Place Area (PPA)) respond selectively to scenes. However, their wider role in spatial encoding and their functional connectivity remain unclear. Using fMRI, first we tested the responses of these areas during spatial comparison tasks using dot targets on white noise. Activity increased during task performance in both RSC and TOS, but not in PPA. However, the amplitude of task-driven activity and behavioral measures of task demand were correlated only in RSC. A control experiment showed that none of these areas were activated during a comparable shape comparison task. Secondly, we analyzed functional connectivity of these areas during the resting state. Results revealed a significant connection between RSC and frontal association areas (known to be involved in perceptual decision-making). In contrast, TOS showed functional connections dorsally with the Inferior Parietal Sulcus, and ventrally with the Lateral Occipital Complex--but not with RSC and/or frontal association areas. Moreover, RSC and TOS showed differentiable functional connections with the anterior-medial and posterior-lateral parts of PPA, respectively. These results suggest two parallel pathways for spatial encoding, including RSC and TOS respectively. Only the RSC network was involved in active spatial comparisons.

Project description:Summary Real-world scene perception unfolds remarkably quickly, yet the underlying visual processes are poorly understood. Space-centered theory maintains that a scene’s spatial structure (e.g., openness, mean depth) can be rapidly recovered from low-level image statistics. In turn, the statistical relationship between a scene’s spatial properties and semantic content allows for semantic identity to be inferred from its layout. We tested this theory by investigating (1) the temporal dynamics of spatial and semantic perception in real-world scenes, and (2) dependencies between spatial and semantic judgments. Participants viewed backward-masked images for 13.3 to 106.7 ms, and identified the semantic (e.g., beach, road) or spatial structure (e.g., open, closed-off) category. We found no temporal precedence of spatial discrimination relative to semantic discrimination. Computational analyses further suggest that, instead of using spatial layout to infer semantic categories, humans exploit semantic information to discriminate spatial structure categories. These findings challenge traditional ‘bottom-up’ views of scene perception. Project description:Working memory (WM) is the key process linking perception to action. Several lines of research have, accordingly, highlighted WM's engagement in sensori-motor associations between retrospective stimuli and future behavior. Using human fMRI we investigated whether prior information about the effector used to respond in a WM task would have an impact on the way the same sensory stimulus is maintained in memory despite a behavioral response could not be readily planned. We focused on WM-related activity in posterior parietal cortex during the maintenance of spatial items for a subsequent match-to-sample comparison, which was reported either with a verbal or with a manual response. We expected WM activity to be higher for manual response trials, because of posterior parietal cortex's engagement in both spatial WM and hand movement preparation. Increased fMRI activity for manual response trials in bilateral anterior intraparietal sulcus confirmed our expectations. These results imply that the maintenance of sensory material in WM is optimized for motor context, i.e. for the effector that will be relevant in the upcoming behavioral responses.
