A single-nucleotide resolution sequencing method of <i>N</i> <sup>6</sup>-adenine methylation sites in DNA and RNA is described. Using sodium nitrite under acidic conditions, chemoselective deamination of unmethylated adenines readily occurs, without competing deamination of <i>N</i> <sup>6</sup>-adenine sites.
