Surface-enhanced Raman scattering of water in aqueous dispersions of silver nanoparticles.

ABSTRACT: The resonance Raman effect (RRE) is a phenomenon which results in a strong selective enhancement of Raman signals from the samples. Previous studies showed that the RRE in liquid water directly corresponds to its supramolecular structure. It was also reported that the electric-field-induced orientation of water molecules on the electrode surface results in the surface-enhanced Raman scattering (SERS) effect. In this work, we show the SERS effect for water molecules in the dispersion of silver nanoparticles (AgNPs) without any external electrical field. An enhancement factor was estimated to be (4.8 ± 0.8) × 10⁶ for an excitation wavelength of 514.5 nm and for AgNPs with an average size of 34 ± 14 nm. The temperature experiment results showed a higher enhancement with temperature increase. Performed simulation studies revealed a slowdown of the mobility of the water molecules close to the surface of AgNPs.

Detection of Silver Nanoparticles in Seawater Using Surface-Enhanced Raman Scattering.

Project description:Nanomaterials significantly contribute to the development of new solutions to improve consumer products properties. Silver nanoparticles (AgNPs) are one of the most used, and as human exposure to such NPs increases, there is a growing need for analytical methods to identify and quantify nanoparticles present in the environment. Here we designed a detection strategy for AgNPs in seawater using surface-enhanced Raman Scattering (SERS). Three commercial AgNPs coated with polyvinylpyrrolidone (PVP) were used to determine the relative impact of size (PVP-15nmAgNPs and PVP-100nmAgNPs) and aggregation degree (predefined Ag aggregates, PVP-50-80nmAgNPs) on the SERS-based detection method. The study of colloidal stability and dissolution of selected AgNPs into seawater was carried out by dynamic light scattering and UV-vis spectroscopy. We showed that PVP-15nmAgNPs and PVP-100nmAgNPs remained colloidally stable, while PVP-50-80nmAgNPs formed bigger aggregates. We demonstrated that the SERS-based method developed here have the capacity to detect and quantify single and aggregates of AgNPs in seawater. The size had almost no effect on the detection limit (2.15 ± 1.22 mg/L for PVP-15nmAgNPs vs. 1.51 ± 0.71 mg/L for PVP-100nmAgNPs), while aggregation caused an increase of 2.9-fold (6.08 ± 1.21 mg/L). Our results demonstrate the importance of understanding NPs transformation in seawater since this can influence the detection method performance.

Multimeric Rhodamine Dye-Induced Aggregation of Silver Nanoparticles for Surface-Enhanced Raman Scattering.

Project description:Isotopic variants of Rhodamine 6G (R6G) have previously been used as a method of multiplexed detection for Surface Enhanced Raman Spectroscopy (SERS), including protein detection and quantification. Challenges exist, however, with producing long-term stable SERS signals with exposure to silver or gold metal surfaces without the use of additional protective coatings of nanomaterials. Here, novel rhodamine "dimers" and "trimers" have been created that demonstrate a higher avidity for metal nanoparticles and induce aggregation to create plasmonic "hotspots" as indicated by enhanced Raman scattering in situ. These aggregates can be formed in a colloid, on surfaces, or membrane substrates such as poly(vinylidene fluoride) for applications in biosciences. The integrity of the materials and Raman signals are maintained for months of time on different substrates. These dye materials should provide avenues for simplified in situ generation of sensors for Raman-based assays especially in settings requiring highly robust performance.

Arrayed nanopore silver thin films for surface-enhanced Raman scattering.

Project description:Active substrates are crucial for surface-enhanced Raman scattering (SERS). Among these substrates, large uniform area arrayed nanoporous silver thin films have been developed as active substrates. Arrayed nanoporous silver thin films with unique anisotropic morphologies and nanoporous structures can be fabricated onto the nanoporous anodic aluminum oxide (AAO) of controlled pore size and interspacing by precisely tuning the sputtering parameters. These thin films preserve locally enhanced electromagnetic fields by exciting the surface plasmon resonance, which is beneficial for SERS. In this study, nanoporous silver thin films were transferred into polymethylmethacrylate (PMMA) and polydimethylsiloxane (PDMS) substrates using our recently invented template-assisted sol-gel phase inverse-imprinting process to form two different nanopore thin films. The as-formed Ag nanoporous thin films on PMMA and PDMS exhibited intensively enhanced SERS signals using Rhodamine 6G (R6G) as the model molecule. The two nanopore thin films exhibited opposite pore size-dependent SERS tendencies, which were elucidated by the different enhancement tendencies of the electric field around pores of different diameters. In particular, the Ag nanoporous thin film on PMMA exhibited an R6G detection limit of as low as 10-6 mol L-1, and the SERS enhancement factor (EF) was more than 106. The low detection limit and large EF demonstrated the high sensitivity of the as-prepared SERS substrates for label-free detection of biomolecules. Compared with conventional smooth films, this nanopore structure can facilitate future application in biomolecular sensors, which allows the detection of single molecules via an electronic readout without requirement for amplification or labels.

Designing Silver Nanoparticles for Detecting Levodopa (3,4-Dihydroxyphenylalanine, L-Dopa) Using Surface-Enhanced Raman Scattering (SERS).

Project description:Detection of the drug Levodopa (3,4-dihydroxyphenylalanine, L-Dopa) is essential for the medical treatment of several neural disorders, including Parkinson's disease. In this paper, we employed surface-enhanced Raman scattering (SERS) with three shapes of silver nanoparticles (nanostars, AgNS; nanospheres, AgNP; and nanoplates, AgNPL) to detect L-Dopa in the nanoparticle dispersions. The sensitivity of the L-Dopa SERS signal depended on both nanoparticle shape and L-Dopa concentration. The adsorption mechanisms of L-Dopa on the nanoparticles inferred from a detailed analysis of the Raman spectra allowed us to determine the chemical groups involved. For instance, at concentrations below/equivalent to the limit found in human plasma (between 10-7-10-8 mol/L), L-Dopa adsorbs on AgNP through its ring, while at 10-5-10-6 mol/L adsorption is driven by the amino group. At even higher concentrations, above 10-4 mol/L, L-Dopa polymerization predominates. Therefore, our results show that adsorption depends on both the type of Ag nanoparticles (shape and chemical groups surrounding the Ag surface) and the L-Dopa concentration. The overall strategy based on SERS is a step forward to the design of nanostructures to detect analytes of clinical interest with high specificity and at varied concentration ranges.

A DFT Approach to the Surface-Enhanced Raman Scattering of 4-Cyanopyridine Adsorbed on Silver Nanoparticles.

Project description:A Surface-Enhanced Raman Scattering (SERS) spectrum of 4-cyanopyridine (4CNPy) was recorded on silver plasmonic nanoparticles and analyzed by using Density Functional Theory (DFT) calculations. Two simple molecular models of the metal-4CNPy surface complex with a single silver cation or with a neutral dimer (Ag+-4CNPy, Ag2-4CNPy), linked through the two possible interacting sites of 4CNPy (aromatic nitrogen, N, and nitrile group, CN), were considered. The calculated vibrational wavenumbers and intensities of the adsorbate and the isolated species are compared with the experimental Raman and SERS results. The analysis of the DFT predictions and the experimental data indicates that 4CNPy adsorbs preferentially on neutral/charged active sites of the silver nanoparticles through the nitrogen atom of the aromatic ring with a perpendicular orientation.

Electrospun Nanofibers Made of Silver Nanoparticles, Cellulose Nanocrystals, and Polyacrylonitrile as Substrates for Surface-Enhanced Raman Scattering.

Project description:Nanofibers with excellent activities in surface-enhanced Raman scattering (SERS) were developed through electrospinning precursor suspensions consisting of polyacrylonitrile (PAN), silver nanoparticles (AgNPs), silicon nanoparticles (SiNPs), and cellulose nanocrystals (CNCs). Rheology of the precursor suspensions, and morphology, thermal properties, chemical structures, and SERS sensitivity of the nanofibers were investigated. The electrospun nanofibers showed uniform diameters with a smooth surface. Hydrofluoric (HF) acid treatment of the PAN/CNC/Ag composite nanofibers (defined as p-PAN/CNC/Ag) led to rougher fiber surfaces with certain pores and increased mean fiber diameters. X-ray diffraction (XRD) and X-ray photoelectron spectroscopy (XPS) results confirmed the existence of AgNPs that were formed during heat and HF acid treatment processes. In addition, thermal stability of the electrospun nanofibers increased due to the incorporation of CNCs and AgNPs. The p-PAN/CNC/Ag nanofibers were used as a SERS substrate to detect p-aminothiophenol (p-ATP) probe molecule. The results show that this substrate exhibited high sensitivity for the p-ATP probe detection.

Capture of Single Silver Nanoparticles in Nanopore Arrays Detected by Simultaneous Amperometry and Surface-Enhanced Raman Scattering.

Project description:The attoliter volumes and confinement abilities of zero-dimensional nanopore-electrode arrays (NEAs) hold considerable promise for examining the behavior of single nanoparticles. In this work, we use surface-enhanced Raman scattering (SERS) in tandem with amperometry in order to monitor single Ag Raman-sentinel nanoparticles transported to and captured in single nanopores. To that end, highly ordered solid-state NEAs were fabricated to contain periodic arrays of nanopores, each housing a single recessed Au-ring electrode. These were used to electrostatically capture and trap single silver nanoparticles (AgNPs) functionalized with the electrochemically stable Raman reporter, 1,4-bis(2-methylstyryl)benzene (bis-MSB). Transport and capture of the bis-MSB-tagged AgNPs in the nanopores was followed by simultaneous amperometry and SERS signals characteristic of AgNP oxidation and enhanced Raman scattering by bis-MSB at silver-gold hot spots, respectively. The frequency and magnitude of oxidation-current spikes increased with stepwise increases in DC voltage, and characteristic bis-MSB SERS spectra were observed. Under AC excitation, on the other hand, two distinctly different types of SERS signals were observed, independent of frequency and amplitude: (1) strong, transient (<10 s) spectra and (2) slow (>100 s) monotonically diminishing spectra. We hypothesize that the former behavior results from AgNP aggregates, whereas the latter occurs as a result of multiple incomplete AgNP-oxidation events in succession. These results show that attoliter-volume NEAs are competent for acquiring concurrent SERS spectra and for amperometry of single nanoparticles and that together these measurements can illuminate the collision dynamics of nanoparticles in confined environments.

Surface-Enhanced Resonance Raman Scattering of Rhodamine 6G in Dispersions and on Films of Confeito-Like Au Nanoparticles.

Project description:Surface-enhanced resonance Raman scattering (SERRS) of rhodamine 6G was measured on confeito-like Au nanoparticles (CAuNPs). The large CAuNPs (100 nm in diameter) in aqueous dispersion systems showed stronger enhancing effect (analytical enhancement factor: over 10?) of SERRS than the small CAuNPs (50 nm in diameter), while the spherical Au nanoparticles (20 nm in diameter) displayed rather weak intensities. Especially, minor bands in 1400-1600 cm-1 were uniquely enhanced by the resonance effect of CAuNPs. The enhancement factors revealed a concentration dependence of the enhancing effect at low concentration of rhodamine 6G. This dependency was due to a large capacity of hot-spots on CAuNPs, which were formed without agglomeration. The surface-enhancing behaviour in the film systems was similar to that in the dispersions, although the large CAuNPs had lower enhancing effect in the films, and the small CAuNPs and the spherical Au nanoparticles were more effective in their films. These results suggest that the CAuNPs have an advantage in ultrasensitive devices both in dispersions and films, compared to the agglomerate of spherical Au nanoparticles.

Imaging of Liver Tumors Using Surface-Enhanced Raman Scattering Nanoparticles.

Project description:Complete surgical resection is the ideal first-line treatment for most liver malignancies. This goal would be facilitated by an intraoperative imaging method that enables more precise visualization of tumor margins and detection of otherwise invisible microscopic lesions. To this end, we synthesized silica-encapsulated surface-enhanced Raman scattering (SERS) nanoparticles (NPs) that act as a molecular imaging agent for liver malignancies. We hypothesized that, after intravenous administration, SERS NPs would avidly home to healthy liver tissue but not to intrahepatic malignancies. We tested these SERS NPs in genetically engineered mouse models of hepatocellular carcinoma and histiocytic sarcoma. After intravenous injection, liver tumors in both models were readily identifiable with Raman imaging. In addition, Raman imaging using SERS NPs enabled detection of microscopic lesions in liver and spleen. We compared the performance of SERS NPs to fluorescence imaging using indocyanine green (ICG). We found that SERS NPs delineate tumors more accurately and are less susceptible to photobleaching. Given the known advantages of SERS imaging, namely, high sensitivity and specific spectroscopic detection, these findings hold promise for improved resection of liver cancer.

Spontaneous versus Stimulated Surface-Enhanced Raman Scattering of Liquid Water.

Project description:We have observed for the first time the surface-enhanced (SE) signal of water in an aqueous dispersion of silver nanoparticles in spontaneous (SERS) and femtosecond stimulated Raman (SE-FSRS) processes with different wavelengths of the Raman pump (515, 715, and 755 nm). By estimating the fraction of water molecules that interact with the metal surface, we have calculated enhancement factors (EF): 4.8 × 106 for SERS and (3.6-3.7) × 106 for SE-FSRS. Furthermore, we have tested the role of simultaneous plasmon resonance and Raman resonance conditions for the aν 1 + bν3 overtone mode of water (755 nm) in SE-FSRS signal amplification. When the wavelength of the Raman pump is within the plasmon resonance of the metal nanoparticles, the Raman resonance has a negligible effect on the EF. However, the Raman resonance with the aν 1 + bν3 mode strongly enhances the signal of the fundamental OH stretching mode of water.
