Horizontal spin of ratchet motor by vertical agitation.

ABSTRACT: The horizontal spin of a ratchet motor by vertical vibration is reported. A macroscopic ratchet gear is placed on a granular bed, where nearly half of the gear is penetrated in the bed. The gear and granular bed are mechanically vibrated. The gear shows a random motion or one-way spin that depend on the diameter of the granules, vibration frequency, and degree of vertical motion allowed for the gear. Even when one-way spin is observed, the spin direction depends on the abovementioned factors. Although the dependency is complicated, it is deterministic because the motion or flows of granular matter determines it. The characteristics observed in the experiments are explained by a simple model that accounts for the statistical variance in the motion of the granular matter. Extraction of systematic motion from small and non-useful motions such as mechanical agitation will be developed into energy harvest technology and may facilitate the science of a spontaneously moving system in a uniform potential field.

Project description:A ratchet gear on a vibrating water bed exhibits a one-way spin. However, the spinning direction is opposite to that of the gear placed on the granular bed. The one-way spin is caused by the surface waves of water. Surface deformation causes transportation of the water element to rotate the gear. The spatial symmetry of the surface wave and gear geometry regulates the rotational torque. In this study, the same ratchet shows reversed motion between the granular and water beds, and the direction is not determined only by the ratchet geometry. The self-organization of the fluid medium caused by small agitation induces a nontrivial inversion of the spinning direction.

A DNA origami rotary ratchet motor.

Project description:To impart directionality to the motions of a molecular mechanism, one must overcome the random thermal forces that are ubiquitous on such small scales and in liquid solution at ambient temperature. In equilibrium without energy supply, directional motion cannot be sustained without violating the laws of thermodynamics. Under conditions away from thermodynamic equilibrium, directional motion may be achieved within the framework of Brownian ratchets, which are diffusive mechanisms that have broken inversion symmetry1-5. Ratcheting is thought to underpin the function of many natural biological motors, such as the F1F0-ATPase6-8, and it has been demonstrated experimentally in synthetic microscale systems (for example, to our knowledge, first in ref. 3) and also in artificial molecular motors created by organic chemical synthesis9-12. DNA nanotechnology13 has yielded a variety of nanoscale mechanisms, including pivots, hinges, crank sliders and rotary systems14-17, which can adopt different configurations, for example, triggered by strand-displacement reactions18,19 or by changing environmental parameters such as pH, ionic strength, temperature, external fields and by coupling their motions to those of natural motor proteins20-26. This previous work and considering low-Reynolds-number dynamics and inherent stochasticity27,28 led us to develop a nanoscale rotary motor built from DNA origami that is driven by ratcheting and whose mechanical capabilities approach those of biological motors such as F1F0-ATPase.

Vertical and horizontal gene transfer tradeoffs direct plasmid fitness

Project description:Plasmid fitness is directed by two orthogonal processes—vertical transfer through cell division and horizontal transfer through conjugation. When considered individually, improvements in either mode of transfer can promote how well a plasmid spreads and persists. Together, however, the metabolic cost of conjugation could create a tradeoff that constrains plasmid evolution. Here we present evidence for the presence, consequences, and molecular basis of a conjugation-growth tradeoff across 40 plasmids derived from clinical E. coli pathogens. We discover that most plasmids operate below a conjugation efficiency threshold for major growth effects, indicating strong natural selection for vertical transfer. Below this threshold, E. coli demonstrates a remarkable growth tolerance to over four orders of magnitude change in conjugation efficiency. This tolerance fades as nutrients become scarce and horizontal transfer attracts a greater share of host resources. Our results provide insight into evolutionary constraints directing plasmid fitness and strategies to combat the spread of antibiotic resistance.

Does Changing Vertical Disparity Induce Horizontal Head Movement?

Project description:Theoretically, one can estimate the direction of an object that is relative to the head using vertical disparity if the distance from the head to the object is known. However, several reports describe vertical disparity as having little or no effect on the perception of visual direction. It has been suggested, however, that the visual processes involved in action are different from those involved in perception, and the effect of visual disparity on action has not been investigated in previous studies. This study investigated the influence of vertical disparity on the stability of head direction as a motor response to visual information. We presented a stimulus consisting of horizontal lines with vertical size-disparity oscillation, and examined whether the stimulus affected the subject's head movement. The results showed that the head movement in the condition of vertical size-disparity oscillation was not significantly different from that in the condition of no disparity oscillation. Our results suggest that, despite theoretical validity, vertical disparity is not used for controlling head movement.

Spin-Dependent Transport in Fe/GaAs(100)/Fe Vertical Spin-Valves.

Project description:The integration of magnetic materials with semiconductors will lead to the development of the next spintronics devices such as spin field effect transistor (SFET), which is capable of both data storage and processing. While the fabrication and transport studies of lateral SFET have attracted greatly attentions, there are only few studies of vertical devices, which may offer the opportunity for the future three-dimensional integration. Here, we provide evidence of two-terminal electrical spin injection and detection in Fe/GaAs/Fe vertical spin-valves (SVs) with the GaAs layer of 50 nanometers thick and top and bottom Fe electrodes deposited by molecular beam epitaxy. The spin-valve effect, which corresponds to the individual switching of the top and bottom Fe layers, is bias dependent and observed up to 20 K. We propose that the strongly bias- and temperature-dependent MR is associated with spin transport at the interfacial Fe/GaAs Schottky contacts and in the GaAs membranes, where balance between the barrier profiles as well as the dwell time to spin lifetime ratio are crucial factors for determining the device operations. The demonstration of the fabrication and spin injection in the vertical SV with a semiconductor interlayer is expected to open a new avenue in exploring the SFET.

Vertical versus horizontal Spatial-Numerical Associations (SNA): A processing advantage for the vertical dimension.

Project description:Humans have associations between numbers and physical space on both horizontal and vertical dimensions, called Spatial-Numerical Associations (SNAs). Several studies have considered the hypothesis of there being a dominant orientation by examining on which dimension people are more accurate and efficient at responding during various directional SNA tasks. However, these studies have difficulty differentiating between a person's efficiency at accessing mental representations of numbers in space, and the efficiency at which they exercise motor control functions, particularly bilateral ones, when manifesting a response during an explicit directional SNA task. In this study we use a conflict test employing combined explicit magnitude and spatial directional processing in which pairs of numbers are placed along the diagonal axes and response accuracy/efficiency are considered across the horizontal and vertical dimensions simultaneously. Participants indicated which number in each pair was largest using a joystick that only required unilateral input. The experiment was run in English using Arabic numerals. Results showed that directional SNAs have a vertical rather than horizontal dominance. A moderating factor was also found during post-hoc analysis, where response efficiency, but not accuracy, is conditional on a person's native language being oriented the same as the language of the experiment, left to right. The dominance of the vertical orientation suggests adopting more vertical display formats for numbers may provide situational advantages, particularly for explicit magnitude comparisons, with some domains like flight controls and the stock market already using these in some cases.

Contextual spatial modelling in the horizontal and vertical domains.

Project description:Multi-scale contextual modelling is an important toolset for environmental mapping. It accounts for spatial dependence by using covariates on multiple spatial scales and incorporates spatial context and structural dependence to environmental properties into machine learning models. For spatial soil modelling, three relevant scales or ranges of scale exist: quasi-local soil formation processes that are independent of the spatial context, short-range catenary processes, and long-range processes related to climate and large-scale terrain settings. Recent studies investigated the spatial dependence of topsoil properties only. We hypothesize that soil properties within a soil profile were formed due to specific interactions between different features and scales of the spatial context, and that there are depth gradients in spatial and structural dependencies. The results showed that for topsoil, features at small to intermediate scales do not increase model accuracy, whereas large scales increase model accuracy. In contrast, subsoil models benefit from all scales-small, intermediate, and large. Based on the differences in relevance, we conclude that the relevant ranges of scales do not only differ in the horizontal domain, but also in the vertical domain across the soil profile. This clearly demonstrates the impact of contextual spatial modelling on 3D soil mapping.

Horizontal and vertical growth of S. cerevisiae metabolic network.

Project description:BACKGROUND: The growth and development of a biological organism is reflected by its metabolic network, the evolution of which relies on the essential gene duplication mechanism. There are two current views about the evolution of metabolic networks. The retrograde model hypothesizes that a pathway evolves by recruiting novel enzymes in a direction opposite to the metabolic flow. The patchwork model is instead based on the assumption that the evolution is based on the exploitation of broad-specificity enzymes capable of catalysing a variety of metabolic reactions. RESULTS: We analysed a well-studied unicellular eukaryotic organism, S. cerevisiae, and studied the effect of the removal of paralogous gene products on its metabolic network. Our results, obtained using different paralog and network definitions, show that, after an initial period when gene duplication was indeed instrumental in expanding the metabolic space, the latter reached an equilibrium and subsequent gene duplications were used as a source of more specialized enzymes rather than as a source of novel reactions. We also show that the switch between the two evolutionary strategies in S. cerevisiae can be dated to about 350 million years ago. CONCLUSIONS: Our data, obtained through a novel analysis methodology, strongly supports the hypothesis that the patchwork model better explains the more recent evolution of the S. cerevisiae metabolic network. Interestingly, the effects of a patchwork strategy acting before the Euascomycete-Hemiascomycete divergence are still detectable today.

Vertical Lasso and Horizontal Lasso Sutures for Repair of Horizontal Cleavage and Horizontal Oblique Meniscal Tears: Surgical Technique and Indications.

Project description:Horizontal cleavage and horizontal oblique meniscal tears have traditionally been treated with partial meniscectomy. Recent research has shown the deleterious long-term effects of meniscectomy in these patients regarding the development of osteoarthritis. Meniscal preservation is thus the preferred method of surgical treatment in patients with these tears. However, traditional repair techniques using inside-out sutures or suture anchor-based devices do not address the horizontally aligned nature of these particular tears and thus do not compress the meniscal tissue in the correct plane. The recent development of an arthroscopic suture-passing system now allows surgeons to place arthroscopic sutures in any pattern or direction. This allows surgeons to treat tears of any type with sutures ideally placed to repair the given tears. This Technical Note describes and demonstrates 2 all-suture, all-inside, arthroscopic meniscus repair techniques to treat these challenging horizontal tear types. The repair techniques are named the vertical lasso and horizontal lasso.
