Project description:Limonene is one of the most abundant naturally occurring cyclic monoterpenes and has recently emerged as a sustainable alternative to petroleum-based solvents as well as a chemical platform for the production of value-added compounds. The biocatalytic epoxidation of both enantiomers of limonene was carried out in the presence of a peroxygenase-containing preparation from oat (Avena sativa) flour. Different reaction profiles were observed depending on the starting enantiomer of limonene, but in both cases the 1,2-monoepoxide was obtained as the main product with excellent diastereoselectivity. Trans-1,2-monoepoxide and cis-1,2-monoepoxide were isolated from the reaction of (R)-limonene and (S)-limonene, respectively, and the reactions were scaled-up to 0.17 M substrate concentration. The process is valuable for operational simplicity, lack of toxic metal catalysts, and cost-effectiveness of the enzymatic source. Pure stereoisomers of 1,2-monoepoxides of limonene constitute a useful starting material for biorenewable polymers, but can be also converted into other chiral derivatives by epoxide ring opening with nucleophiles. As a proof of concept, a tandem protocol for the preparation of enantiopure (1S,2S,4R)-1,2-diol from (R)-limonene and (1R,2R,4S)-1,2-diol from (S)-limonene was developed.

Project description:Peroxygenases are heme-dependent enzymes that use peroxide-borne oxygen to catalyze a wide range of oxyfunctionalization reactions. Herein, we report the engineering of an unusual cofactor-independent peroxygenase based on a promiscuous tautomerase that accepts different hydroperoxides (t-BuOOH and H2 O2 ) to accomplish enantiocomplementary epoxidations of various α,β-unsaturated aldehydes (citral and substituted cinnamaldehydes), providing access to both enantiomers of the corresponding α,β-epoxy-aldehydes. High conversions (up to 98 %), high enantioselectivity (up to 98 % ee), and good product yields (50-80 %) were achieved. The reactions likely proceed via a reactive enzyme-bound iminium ion intermediate, allowing tweaking of the enzyme's activity and selectivity by protein engineering. Our results underscore the potential of catalytic promiscuity for the engineering of new cofactor-independent oxidative enzymes.

Project description:Oleate ion was intercalated into a hydrotalcite obtained using a conventional precipitation method and an alternative, novel method involving microwave-assisted ion exchange. The latter method gave a more crystalline hydrotalcite with better textural properties than the former. The spacing between layers in the hydrotalcites was used to epoxidize the double bond in oleate ion. X-ray diffraction patterns revealed that the layered structure of the hydrotalcites was not altered by the reaction. The epoxidation of oleate ion with m-chloroperbenzoic acid (mCPBA) was monitored using Raman spectroscopy. The results of this work testify to the high potential of hydrotalcites as molecular reactors for reactions in confined spaces.

Project description:The styrene monooxygenase (SMO) system from Pseudomonas sp. consists of two enzymes (StyA and StyB). StyB catalyses the reduction of FAD at the expense of NADH. After the transfer of FADH2 from StyB to StyA, reaction with O2 generates FAD-OOH, which is the epoxidising agent. The wastage of redox equivalents due to partial diffusive transfer of FADH2 , the insolubility of recombinant StyB and the impossibility of expressing StyA and StyB in a 1:1 molar ratio reduce the catalytic efficiency of the natural system. Herein we present a chimeric SMO (Fus-SMO) that was obtained by genetic fusion of StyA and StyB through a flexible linker. Thanks to a combination of: 1) balanced and improved expression levels of reductase and epoxidase units, and 2) intrinsically higher specific epoxidation activity of Fus-SMO in some cases, Escherichia coli cells expressing Fus-SMO possess about 50 % higher activity for the epoxidation of styrene derivatives than E. coli cells coexpressing StyA and StyB as discrete enzymes. The epoxidation activity of purified Fus-SMO was up to three times higher than that of the two-component StyA/StyB (1:1, molar ratio) system and up to 110 times higher than that of the natural fused SMO. Determination of coupling efficiency and study of the influence of O2 pressure were also performed. Finally, Fus-SMO and formate dehydrogenase were coexpressed in E. coli and applied as a self-sufficient biocatalytic system for epoxidation on greater than 500 mg scale.

Project description:The application of whole cells as biocatalysts is often limited by the toxicity of organic solvents, which constitute interesting substrates/products or can be used as a second phase for in situ product removal and as tools to control multistep biocatalysis. Solvent-tolerant bacteria, especially Pseudomonas strains, are proposed as promising hosts to overcome such limitations due to their inherent solvent tolerance mechanisms. However, potential industrial applications suffer from tedious, unproductive adaptation processes, phenotypic variability, and instable solvent-tolerant phenotypes. In this study, genes described to be involved in solvent tolerance were identified in Pseudomonas taiwanensis VLB120, and adaptive solvent tolerance was proven by cultivation in the presence of 1% (vol/vol) toluene. Deletion of ttgV, coding for the specific transcriptional repressor of solvent efflux pump TtgGHI gene expression, led to constitutively solvent-tolerant mutants of P. taiwanensis VLB120 and VLB120ΔC. Interestingly, the increased amount of solvent efflux pumps enhanced not only growth in the presence of toluene and styrene but also the biocatalytic performance in terms of stereospecific styrene epoxidation, although proton-driven solvent efflux is expected to compete with the styrene monooxygenase for metabolic energy. Compared to that of the P. taiwanensis VLB120ΔC parent strain, the maximum specific epoxidation activity of P. taiwanensis VLB120ΔCΔttgV doubled to 67 U/g of cells (dry weight). This study shows that solvent tolerance mechanisms, e.g., the solvent efflux pump TtgGHI, not only allow for growth in the presence of organic compounds but can also be used as tools to improve redox biocatalysis involving organic solvents.

Project description:Unlike the excellent (S)-enantioselective epoxidation of styrene performed by natural styrene monooxygenases (ee > 99%), the (R)-enantioselective epoxidation of styrene has not yet achieved a comparable efficiency using natural or engineered oxidative enzymes. This report describes the H2O2-dependent (R)-enantioselective epoxidation of unfunctionalized styrene and its derivatives by site-mutated variants of a unique non-natural P450BM3 peroxygenase, working in tandem with a dual-functional small molecule (DFSM). The observed (R)-enantiomeric excess (ee) of styrene epoxidation is up to 99% with a turnover number (TON) of 918 by the best enantioselective mutant F87A/T268I/L181Q, while the best active mutant F87A/T268I/V78A/A184L (with 98% ee) gave a catalytic TON of 4350, representing the best activity of a P450 peroxygenase towards styrene epoxidation to date. Following this approach, a set of styrene derivatives, such as o-, m-, p-chlorostyrenes and fluorostyrenes, could also be epoxidized with modest to very good TONs (362-3480) and high (R)-enantioselectivities (95-99% ee). The semi-preparative scale synthesis of (R)-styrene oxide performed at 0 °C with high conversion, maintaining enantioselectivity, and moderate isolated yields, further suggests the potential application of the current P450 enzymatic system in styrene epoxidation. This study indicates that the synergistic use of protein engineering and an exogenous DFSM constitutes an efficient strategy to control the enantioselectivity of styrene epoxidation, thus substantially expanding the chemical scope of P450 enzymes as useful bio-oxidative catalysts.

Project description:The controlled generation of reactive oxygen species (ROS) to selectively epoxidize styrene is a grand challenge. Herein, cyano-group-modified carbon nitrides (CNCY x and CN-T y ) are prepared, and the catalysts show better performance in regulating ROS and producing styrene oxide than the cyano-free sample. The in situ diffuse reflectance infrared and density functional theory calculation results reveal that the cyano group acts as the adsorption and activation site of oxygen. X-ray photoelectron spectroscopy and NMR spectrum results confirm that the cyano group bonds with the intact heptazine ring. This unique structure could inhibit H2O2 and ⋅OH formation, resulting in high selectivity of styrene oxide. Furthermore, high catalytic activity is still achieved when the system scales up to 2.7 L with 100 g styrene under solar light irradiation. The strategy of cyano group modification gives a new insight into regulating spatial configuration for tuning the utilization of oxygen-active species and shows potential applications in industry.

Project description:Single-atom catalysts provide a pathway to elucidate the nature of catalytically active sites. However, keeping them stabilized during operation proves to be challenging. Herein, we employ cryptomelane-type octahedral molecular sieve nanorods featuring abundant manganese vacancy defects as a support, to periodically anchor single-atom Ag. The doped Ag atoms with tetrahedral coordination are found to locate at cation substitution sites rather than being supported on the catalyst surface, thus effectively tuning the electronic structure of adjacent manganese atoms. The resulting unique Ag-O-MnO x unit functions as the active site. Its turnover frequency reaches 1038 h-1, one order of magnitude higher than for previously reported catalysts, with 90% selectivity for anti-Markovnikov phenylacetaldehyde. Mechanistic studies reveal that the activation of styrene on the ensemble site of Ag-O-MnO x is significantly promoted, which can accelerate the oxidation of styrene and, in particular, the rate-determining step of forming the epoxide intermediate. Such an extraordinary electronic promotion can be extended to other single-atom catalysts and paves the way for their practical applications.

Project description:NiAl Layered Double Hydroxide (LDH) alginate bionanocomposites were synthesized by confined coprecipitation within alginate beads. The NiAl based bionanocomposites were prepared either by impregnation by divalent and trivalent metal cations of pre-formed calcium cross-linked alginate beads or by using the metal cations (Ni2+, Al3+) as crosslinking cationic agents for the biopolymer network. The impregnation step was systematically followed by a soaking in NaOH solution to induce the LDH coprecipitation. Powder x-ray diffraction (PXRD), infrared spectroscopy (FTIR), energy dispersive X-ray analysis (EDX), thermogravimetry analysis (TGA), electron microscopies (SEM and TEM) confirmed the biotemplated coprecipitation of LDH nanoparticles ranging from 75 to 150 nm for both strategies. The drying of the LDH@alginate beads by supercritical CO2 drying process led to porous bionanocomposite aerogels when Ca2+ cross-linked alginate beads were used. Such confined preparation of NiAl LDH was extended to bionanocomposite films leading to similar results. The permeability and the electrochemical behavior of these NiAl@alginate bionanocomposites, as thin films coated on indium tin oxide (ITO) electrodes, were investigated by cyclic voltammetry, demonstrating an efficient diffusion of the K4Fe(CN)6 redox probe through the LDH@alginate based films and the improvement of the electrochemical accessibility of the Ni sites.
