Project description:Perceiving the sensory consequences of action accurately is essential for appropriate interaction with our physical and social environments. Prediction mechanisms are considered necessary for fine-tuned sensory control of action, yet paradoxically may distort perception. Here, we examine this paradox by addressing how movement influences the perceived duration of sensory outcomes congruent with action. Experiment 1 required participants to make judgments about the duration of vibrations applied to a moving or stationary finger. In Experiments 2 and 3, participants judged observed finger movements that were congruent or incongruent with their own actions. In all experiments, target events were perceived to be longer when congruent with movement. Interestingly, this temporal dilation did not differ as a function of stimulus perspective (1st or 3rd person) or spatial location. We propose that this bias may reflect the operation of an adaptive mechanism for sensorimotor selection and control that preactivates anticipated outcomes of action. The bias itself may have surprising implications for both action control and perception of others: we may be in contact with grasped objects for less time than we realize, and others' reactions to us may be briefer than we believe.

Project description:In the New Look literature of the 1950s, it has been suggested that size judgments are dependent on the affective content of stimuli. This suggestion, however, has been 'discredited' due to contradictory findings and methodological problems. In the present study, we revisited this forgotten issue in two experiments. The first experiment investigated the influence of affective content on size perception by examining judgments of the size of target circles with and without affectively loaded (i.e., positive, neutral, and negative) pictures. Circles with a picture were estimated to be smaller than circles without a picture, and circles with a negative picture were estimated to be larger than circles with a positive or a neutral picture confirming the suggestion from the 1950s that size perception is influenced by affective content, an effect notably confined to negatively loaded stimuli. In a second experiment, we examined whether affective content influenced the Ebbinghaus illusion. Participants judged the size of a target circle whereby target and flanker circles differed in affective loading. The results replicated the first experiment. Additionally, the Ebbinghaus illusion was shown to be weakest for a negatively loaded target with positively loaded and blank flankers. A plausible explanation for both sets of experimental findings is that negatively loaded stimuli are more attention demanding than positively loaded or neutral stimuli.

Project description:We investigated categorical perception of rising and falling pitch contours by tonal and non-tonal listeners. Specifically, we determined minimum durations needed to perceive both contours and compared to those of production, how stimuli duration affects their perception, whether there is an intrinsic F0 effect, and how first language background, duration, directions of pitch and vowel quality interact with each other. Continua of fundamental frequency on different vowels with 9 duration values were created for identification and discrimination tasks. Less time is generally needed to effectively perceive a pitch direction than to produce it. Overall, tonal listeners' perception is more categorical than non-tonal listeners. Stimuli duration plays a critical role for both groups, but tonal listeners showed a stronger duration effect, and may benefit more from the extra time in longer stimuli for context-coding, consistent with the multistore model of categorical perception. Within a certain range of semitones, tonal listeners also required shorter stimulus duration to perceive pitch direction changes than non-tonal listeners. Finally, vowel quality plays a limited role and only interacts with duration in perceiving falling pitch directions. These findings further our understanding on models of categorical perception, the relationship between speech perception and production, and the interaction between the perception of tones and vowel quality.

Project description:In blood-feeding dipterans, olfaction plays a role in finding hosts and, hence, in spreading pathogens. Several pathogens are known to alter olfactory responses and behavior in vectors. As a mosquito-borne pathogen, Rift Valley Fever Virus (RVFV) can affect humans and cause great losses in livestock. We test the influence of RVFV infection on sensory perception, olfactory choice behavior and activity on a non-biting insect, Drosophila melanogaster, using electroantennograms (EAG), Y-maze, and locomotor activity monitor. Flies were injected with RVFV MP12 strain. Replication of RVFV and its persistence for at least seven days was confirmed by quantitative reverse transcription-PCR (RT-qPCR). One day post injection, infected flies showed weaker EAG responses towards 1-hexanol, vinegar, and ethyl acetate. In the Y-maze, infected flies showed a significantly lower response for 1-hexanol compared to uninfected flies. At days six or seven post infection, no significant difference between infected and control flies could be found in EAG or Y-maze anymore. Activity of infected flies was reduced at both time points. We found an upregulation of the immune-response gene, nitric oxide synthase, in infected flies. An infection with RVFV is able to transiently reduce olfactory perception and attraction towards food-related odors in Drosophila, while effects on activity and immune effector gene expression persist. A similar effect in blood-feeding insects could affect vector competence in RVFV transmitting dipterans.

Project description:Recent brain connectome studies have evidenced distinct and overlapping brain regions involved in processing olfactory perception. However, neural correlates of hypo- or anosmia in olfactory disorder patients are poorly known. Furthermore, the bottom-up and top-down processing of olfactory perception have not been well-documented, resulting in difficulty in locating the disease foci of olfactory disorder patients. The primary aim of this study is to characterize the bottom-up process of the neural dynamics across peripheral and central brain regions in anesthetized mice. We particularly focused on the neural oscillations of local field potential (LFP) in olfactory epithelium (OE), olfactory blub (OB), prefrontal cortex (PFC), and hippocampus (HC) during an olfactory oddball paradigm in urethane anesthetized mice. Odorant presentations evoked neural oscillations across slow and fast frequency bands including delta (1-4 Hz), theta (6-10 Hz), beta (15-30 Hz), low gamma (30-50 Hz), and high gamma (70-100 Hz) in both peripheral and central nervous systems, and the increases were more prominent in the infrequently presented odorant. During 5 s odorant exposures, the oscillatory responses in power were persistent in OE, OB, and PFC, whereas neural oscillations of HC increased only for short time at stimulus onset. These oscillatory responses in power were insignificant in both peripheral and central regions of the ZnSO4-treated anosmia model. These results suggest that olfactory stimulation induce LFP oscillations both in the peripheral and central nervous systems and suggest the possibility of linkage of LFP oscillations in the brain to the oscillations in the peripheral olfactory system.

Project description:Various insects and small animals can navigate in turbulent streams to find their mates (or food) from sparse pheromone (odor) detections. Their access to internal space perception and use of cognitive maps still are heavily debated, but for some of them, limited space perception seems to be the rule. However, this poor space perception does not prevent them from impressive search capacities. Here, as an attempt to model these behaviors, we propose a scheme that can perform, even without a detailed internal space map, searches in turbulent streams. The algorithm is based on a standardized projection of the probability of the source position to remove space perception and on the evaluation of a free energy, whose minimization along the path gives direction to the searcher. An internal "temperature" allows active control of the exploration/exploitation balance during the search. We demonstrate the efficiency of the scheme numerically, with a computational model of odor plume propagation, and experimentally, with robotic searches of thermal sources in turbulent streams. In addition to being a model to describe animals' searches, this scheme may be applied to robotic searches in complex varying media without odometry error corrections and in problems in which active control of the exploration/exploitation balance is profitable.

Project description:Neuroscientific models of sensory perception suggest that the brain utilizes predictive codes in advance of a stimulus encounter, enabling organisms to infer forthcoming sensory events. However, it is poorly understood how such mechanisms are implemented in the olfactory system. Combining high-resolution functional magnetic resonance imaging with multivariate (pattern-based) analyses, we examined the spatiotemporal evolution of odor perception in the human brain during an olfactory search task. Ensemble activity patterns in anterior piriform cortex (APC) and orbitofrontal cortex (OFC) reflected the attended odor target both before and after stimulus onset. In contrast, prestimulus ensemble representations of the odor target in posterior piriform cortex (PPC) gave way to poststimulus representations of the odor itself. Critically, the robustness of target-related patterns in PPC predicted subsequent behavioral performance. Our findings directly show that the brain generates predictive templates or "search images" in PPC, with physical correspondence to odor-specific pattern representations, to augment olfactory perception.

Project description:Two-component signal-transducing systems are ubiquitously distributed communication interfaces in bacteria. They consist of a histidine kinase that senses a specific environmental stimulus and a cognate response regulator that mediates the cellular response, mostly through differential expression of target genes. Histidine kinases are typically transmembrane proteins harboring at least two domains: an input (or sensor) domain and a cytoplasmic transmitter (or kinase) domain. They can be identified and classified by virtue of their conserved cytoplasmic kinase domains. In contrast, the sensor domains are highly variable, reflecting the plethora of different signals and modes of sensing. In order to gain insight into the mechanisms of stimulus perception by bacterial histidine kinases, we here survey sensor domain architecture and topology within the bacterial membrane, functional aspects related to this topology, and sequence and phylogenetic conservation. Based on these criteria, three groups of histidine kinases can be differentiated. (i) Periplasmic-sensing histidine kinases detect their stimuli (often small solutes) through an extracellular input domain. (ii) Histidine kinases with sensing mechanisms linked to the transmembrane regions detect stimuli (usually membrane-associated stimuli, such as ionic strength, osmolarity, turgor, or functional state of the cell envelope) via their membrane-spanning segments and sometimes via additional short extracellular loops. (iii) Cytoplasmic-sensing histidine kinases (either membrane anchored or soluble) detect cellular or diffusible signals reporting the metabolic or developmental state of the cell. This review provides an overview of mechanisms of stimulus perception for members of all three groups of bacterial signal-transducing histidine kinases.

Project description:Modulation of beta-band neural oscillations during and following movement is a robust marker of brain function. In particular, the post-movement beta rebound (PMBR), which occurs on movement cessation, has been related to inhibition and connectivity in the healthy brain, and is perturbed in disease. However, to realise the potential of the PMBR as a biomarker, its modulation by task parameters must be characterised and its functional role determined. Here, we used MEG to image brain electrophysiology during and after a grip-force task, with the aim to characterise how task duration, in the form of an isometric contraction, modulates beta responses. Fourteen participants exerted a 30% maximum voluntary grip-force for 2, 5 and 10?s. Our results showed that the amplitude of the PMBR is modulated by task duration, with increasing duration significantly reducing PMBR amplitude and increasing its time-to-peak. No variation in the amplitude of the movement related beta decrease (MRBD) with task duration was observed. To gain insight into what may underlie these trial-averaged results, we used a Hidden Markov Model to identify the individual trial dynamics of a brain network encompassing bilateral sensorimotor areas. The rapidly evolving dynamics of this network demonstrated similar variation with task parameters to the 'classical' rebound, and we show that the modulation of the PMBR can be well-described in terms of increased frequency of beta events on a millisecond timescale rather than modulation of beta amplitude during this time period. Our results add to the emerging picture that, in the case of a carefully controlled paradigm, beta modulation can be systematically controlled by task parameters and such control can reveal new information as to the processes that generate the average beta timecourse. These findings will support design of clinically relevant paradigms and analysis pipelines in future use of the PMBR as a marker of neuropathology.

Project description:Inhibitory response occurs throughout the nervous system, including the peripheral olfactory system. While odor-evoked excitation in peripheral olfactory cells is known to encode odor information, the molecular mechanism and functional roles of odor-evoked inhibition remain largely unknown. Here, we examined Drosophila olfactory sensory neurons and found that inhibitory odors triggered outward receptor currents by reducing the constitutive activities of odorant receptors, inhibiting the basal spike firing in olfactory sensory neurons. Remarkably, this odor-evoked inhibition of olfactory sensory neurons elicited by itself a full range of olfactory behaviors from attraction to avoidance, as did odor-evoked olfactory sensory neuron excitation. These results indicated that peripheral inhibition is comparable to excitation in encoding sensory signals rather than merely regulating excitation. Furthermore, we demonstrated that a bidirectional code with both odor-evoked inhibition and excitation in single olfactory sensory neurons increases the odor-coding capacity, providing a means of efficient sensory encoding.
