Project description:Studies using zebrafish (Danio rerio) in neuro-behavioural research are growing. Measuring fish behavior by computational methods is one of the most efficient ways to avoid human bias in experimental analyses, extending them to various approaches. Sometimes, thorough analyses are difficult to do, as fish can behave unpredictably during an experimental strategy. However, the analyses can be implemented in an automated way, using an online strategy and video processing for a complete assessment of the zebrafish behavior, based on the detection and tracking of fish during an activity. Here, a fully automatic conditioning and detailed analysis of zebrafish behavior is presented. Microcontrolled components were used to control the delivery of visual and sound stimuli, in addition to the concise amounts of food after conditioned stimuli for adult zebrafish groups in a conventional tank. The images were captured and processed for automatic detection of the fish, and the training of the fish was done in two evaluation strategies: simple and complex. In simple conditioning, the zebrafish showed significant responses from the second attempt, learning that the conditioned stimulus was a predictor of food presentation in a specific space of the tank, where the food was dumped. When the fish were subjected to two stimuli for decision-making in the food reward, the zebrafish obtained better responses to red light stimuli in relation to vibration. The behavior change was clear in stimulated fish in relation to the control group, thus, the distances traveled and the speed were greater, while the polarization was lower in stimulated fish. This automated system allows for the conditioning and assessment of zebrafish behavior online, with greater stability in experiments, and in the analysis of the behavior of individual fish or fish schools, including learning and memory studies.

Project description:Monitoring accurately temperature is a key issue in biological studies involving living experimental material. It is especially true for insects which body temperature is mostly controlled by environmental temperature, with profound consequences of a few degrees variation on most physiological processes such as survival, development, fecundity, and mobility. If programmable rearing units can be purchased, it remains important to monitor and store temperature information acquired inside the rearing unit to ensure that observed phenomena are not the result of unintended and not scarily noticeable changes in temperature, and to account for the effect of temperature variation in statistical analysis. As most laboratories involved in insect rearing dispose of a large number of rearing units, the technical solution should meet the monitoring needs while being affordable and adaptable to various experimental designs. For that purpose, we designed a low cost (below 100€) and open source automatic temperature monitoring system for rearing units in laboratory. Project description:From learning environments to battlefields to marketing teams, the desire to measure cognition and cognitive fatigue in real time has been a grand challenge in optimizing human performance. Near-infrared spectroscopy (NIRS) is an effective optical technique for measuring changes in subdermal hemodynamics, and it has been championed as a more practical method for monitoring brain function compared to MRI. This study reports on an innovative functional NIRS (fNIRS) sensor that integrates the entire system into a compact and wearable device, enabling long-term monitoring of patients. The device provides unrestricted mobility to the user with a Bluetooth connection for settings configuration and data transmission. A connected device, such as a smartphone or laptop equipped with the appropriate interface software, collects raw data, then stores and generates real-time analyses. Tests confirm the sensor is sensitive to oxy- and deoxy-hemoglobin changes on the forehead region, which indicate neuronal activity and provide information for brain activity monitoring studies.

Project description:In humans, infants respond positively to slow, gentle stroking-processed by C-tactile (CT) nerve fibers-by showing reductions in stress and increases in eye contact, smiling, and positive vocalizations. More frequent maternal touch is linked to greater activity and connectivity strength in social brain regions, and increases children's attention to and learning of faces. It has been theorized that touch may prime children for social interactions and set them on a path towards healthy social cognitive development. However, less is known about the effects of touch on young infants' psychological development, especially in the newborn period, a highly sensitive period of transition with rapid growth in sensory and social processing. It remains untested whether newborns can distinguish CT-targeted touch from other types of touch, or whether there are benefits of touch for newborns' social, emotional, or cognitive development. In the present study, we experimentally investigated the acute effects of touch in newborn monkeys, a common model for human social development. Rhesus macaques (Macaca mulatta), like humans, are highly social, have complex mother-infant interactions with frequent body contact for the first weeks of life, making them an excellent model of infant sociality. Infant monkeys in the present study were reared in a neonatal nursery, enabling control over their early environment, including all caregiver interactions. One-week-old macaque infants (N = 27) participated in three 5-minute counter-balanced caregiver interactions, all with mutual gaze: stroking head and shoulders (CT-targeted touch), stroking palms of hands and soles of feet (Non-CT touch), or no stroking (No-touch). Immediately following the interaction, infants watched social and nonsocial videos and picture arrays including faces and objects, while we tracked their visual attention with remote eye tracking. We found that, during the caregiver interactions, infants behaved differently while being touched compared to the no-touch condition, irrespective of the body part touched. Most notably, in both touch conditions, infants exhibited fewer stress-related behaviors-self-scratching, locomotion, and contact time with a comfort object-compared to when they were not touched. Following CT-targeted touch, infants were faster to orient to the picture arrays compared to the other interaction conditions, suggesting CT-targeted touch may activate or prime infants' attentional orienting system. In the No-touch condition infants attended longer to the nonsocial compared to the social video, possibly reflecting a baseline preference for nonsocial stimuli. In contrast, in both touch conditions, infants' looked equally to the social and nonsocial videos, suggesting that touch may influence the types of visual stimuli that hold infants' attention. Collectively, our results reveal that newborn macaques responded positively to touch, and touch appeared to influence some aspects of their subsequent attention, although we found limited evidence that these effects are mediated by CT fibers. These findings suggest that newborn touch may broadly support infants' psychological development, and may have early evolutionary roots, shared across primates. This study illustrates the unique insight offered by nonhuman primates for exploring early infant social touch, revealing that touch may positively affect emotional and attentional development as early as the newborn period.

Project description:In order to provide the real-time monitoring for identifying the sources of pollution and improving the irrigation water quality management, the integration of continuous automatic sampling techniques and cloud technologies is essential. In this study, we have established an automatic real-time monitoring system for improving the irrigation water quality management, especially for heavy metals such as Cd, Pb, Cu, Ni, Zn, and Cr. As a part of this work, we have first provided several examples on the basic water quality parameters (e.g., pH and electrical conductance) to demonstrate the capacity of data correction by the smart monitoring system, and then evaluated the trend and variance of water quality parameters for different types of monitoring stations. By doing so, the threshold (to initiate early warming) of different water quality parameters could be dynamically determined by the system, and the authorities could be immediately notified for follow-up actions. We have also provided and discussed the representative results from the real-time automatic monitoring system of heavy metals from different monitoring stations. Finally, we have illustrated the implications of the developed smart monitoring system for ensuring the safety of irrigation water in the near future, including integration with automatic sampling for establishing information exchange platform, estimating fluxes of heavy metals to paddy fields, and combining with green technologies for nonpoint source pollution control.

Project description:Airborne pollen monitoring has been an arduous task, making ecological applications and allergy management virtually disconnected from everyday practice. Over the last decade, intensive research has been conducted worldwide to automate this task and to obtain real-time measurements. The aim of this study was to evaluate such an automated biomonitoring system vs. the conventional 'gold-standard' Hirst-type technique, attempting to assess which may more accurately provide the genuine exposure to airborne pollen. Airborne pollen was monitored in Augsburg since 2015 with two different methods, a novel automatic Bio-Aerosol Analyser, and with the conventional 7-day recording Hirst-type volumetric trap, in two different sites. The reliability, performance, accuracy, and comparability of the BAA500 Pollen Monitor (PoMo) vs. the conventional device were investigated, by use of approximately 2.5 million particles sampled during the study period. The observations made by the automated PoMo showed an average accuracy of approximately 85%. However, it also exhibited reliability problems, with information gaps within the main pollen season of between 17 to 19 days. The PoMo automated algorithm had identification issues, mainly confusing the taxa of Populus, Salix and Tilia. Hirst-type measurements consistently exhibited lower pollen abundances (median of annual pollen integral: 2080), however, seasonal traits were more comparable, with the PoMo pollen season starting slightly later (median: 3 days), peaking later (median: 5 days) but also ending later (median: 14 days). Daily pollen concentrations reported by Hirst-type traps vs. PoMo were significantly, but not closely, correlated (r = 0.53-0.55), even after manual classification. Automatic pollen monitoring has already shown signs of efficiency and accuracy, despite its young age; here it is suggested that automatic pollen monitoring systems may be more effective in capturing a larger proportion of the airborne pollen diversity. Even though reliability issues still exist, we expect that this new generation of automated bioaerosol monitoring will eventually change the aerobiological era, as known for almost 70 years now.

Project description:Animals can perform complex and purposeful behaviors by executing simpler movements in flexible sequences. It is particularly challenging to analyze behavior sequences when they are highly variable, as is the case in language production, certain types of birdsong and, as in our experiments, flies grooming. High sequence variability necessitates rigorous quantification of large amounts of data to identify organizational principles and temporal structure of such behavior. To cope with large amounts of data, and minimize human effort and subjective bias, researchers often use automatic behavior recognition software. Our standard grooming assay involves coating flies in dust and videotaping them as they groom to remove it. The flies move freely and so perform the same movements in various orientations. As the dust is removed, their appearance changes. These conditions make it difficult to rely on precise body alignment and anatomical landmarks such as eyes or legs and thus present challenges to existing behavior classification software. Human observers use speed, location, and shape of the movements as the diagnostic features of particular grooming actions. We applied this intuition to design a new automatic behavior recognition system (ABRS) based on spatiotemporal features in the video data, heavily weighted for temporal dynamics and invariant to the animal's position and orientation in the scene. We use these spatiotemporal features in two steps of supervised classification that reflect two time-scales at which the behavior is structured. As a proof of principle, we show results from quantification and analysis of a large data set of stimulus-induced fly grooming behaviors that would have been difficult to assess in a smaller dataset of human-annotated ethograms. While we developed and validated this approach to analyze fly grooming behavior, we propose that the strategy of combining alignment-invariant features and multi-timescale analysis may be generally useful for movement-based classification of behavior from video data.

Project description:Collecting a myriad of prototype data through various types of monitoring sensors plays a virtual important role in many aspects of dam safety such as real-time grasp of safety state, exposure of hidden dangers, and inspection design and construction. However, the current methods of prediction are weak in the long-term sequence of nodes with missing and abnormal error value. Moreover, the limitation caused by the apparatus, environmental factors, and network transmission can lead to the deviation and inconsistency of diagnosis and evaluation of local region. In this paper, we consider the correlation of data on nodes in the entire monitoring network. To avoid the deviation caused by noise and missing value in the single-node data sequence, we calculate the correlation between the multiple sequences. A single-node assessment model based on multiple relevant sequence (SAM) is proposed to improve the accuracy of single node assessment. Given the different nodes of a local region have varying impacts on the evaluation results, a local region evaluation algorithm based on node credibility (LREA) is presented to model the credibility of nodes in order to alleviate inconsistent evaluation results in the local region of dam. LREA can assess the dam's operation state by considering the variations in credibility and multiple nodes coordination. The experimental results illustrate the LREA can reveal the trends of the monitoring values change in a timely and accurate way, which can elevate the accuracy of evaluation results of dam safety.

Project description:The usage of automatic feeding systems (AFS) in cattle offers multiple advantages, mostly due to the possibility of an increased feeding frequency. While it is gaining more and more importance in dairy farming, there is still a lack of experience and scientific knowledge regarding its use in fattening cattle. The aim of this study was to describe the behavior of 56 Simmental bulls fed with an AFS six times daily a total mixed ration. The animals arrived at the farm with an average age of 148 ± 11 days. They were housed in four straw-bedded pens in groups of 14 animals each. Their average slaughter age was 558 ± 20 days. Behavioral observations were made during three observation periods (OP) at an average of 11, 14, and 16 months of age. Using scan sampling, feeding, and lying behavior of all animals and the order of bulls feeding after feed delivery were recorded. Furthermore, body condition and health status were monitored and complemented with the carcass weights. Body condition, health status, and carcass weights of the bulls were found to be satisfactory: Mean body condition score increased from 2.8 ± 0.3 in OP1 to 3.0 ± 0.1 in OP3 and mean carcass weight was 432.71 ± 40.82 kg. No severe health problems occurred. The feeding activity of the bulls was spread out over the course of the day with peaks in the afternoon and evening. Percentages of bulls feeding per pen never exceeded 20%, animals feeding mostly alone (during 28.04 ± 2.15% of total observation time) or in groups of two to three (16.61 ± 2.00% and 6.74 ± 1.90%). The order of bulls feeding after feed delivery varied indicating that all animals had similar access to fresh feed. These results emphasize the importance of constant feed availability and quality at any time of the day, thus indicating the ability of an AFS with six daily feedings to ensure such a consistency.

Project description:Objective evaluations of cough frequency are considered important for assessing the clinical state of patients with respiratory diseases. However, cough monitors with audio recordings are rarely used in clinical settings. Issues regarding privacy and background noise with audio recordings are barriers to the wide use of these monitors; to solve these problems, we developed a novel automatic cough frequency monitoring system combining a triaxial accelerator and a stretchable strain sensor. Eleven healthy adult volunteers and 10 adult patients with cough were enrolled. The participants wore two devices for 30 min for the cough measurements. An accelerator was attached to the epigastric region, and a stretchable strain sensor was worn around their neck. When the subjects coughed, these devices displayed specific waveforms. The data from all the participants were categorized into a training dataset and a test dataset. Using a variational autoencoder, a machine learning algorithm with deep learning, the components of the test dataset were automatically judged as being a "cough unit" or "non-cough unit". The sensitivity and specificity in detecting coughs were 92% and 96%, respectively. Our cough monitoring system has the potential to be widely used in clinical settings without any concerns regarding privacy or background noise.
