Highly Sensitive and Selective Fluorescence "Turn-On" Detection of Pb (II) Based on Fe3O4@Au-FITC Nanocomposite.

ABSTRACT: New nanocomposites, Fe3O4@Au-FITC, were prepared and explored to develop a fluorescent detection of Pb2+. The Fe3O4@AuNPs-FITC nanocomposites could be etched by Pb2+ in the presence of Na2S2O3, leading to fluorescence recovery of FITC quenched by Fe3O4@Au nanocomposites. With the increase of Pb2+ concentration, the fluorescence recovery of Fe3O4@AuNPs-FITC increased gradually. Under optimized conditions, a detection limit of 5.2 nmol/L of Pb2+ with a linear range of 0.02-2.0 µmol/L were obtained. The assay demonstrated negligible response to common metal ions. Recoveries of 98.2-106.4% were obtained when this fluorescent method was applied in detecting Pb2+ spiked in a lake-water sample. The above results demonstrated the high potential of ion-induced nanomaterial etching in developing robust fluorescent assays.

Project description:A polymer-based fluorescence "turn-on" chemosensor is demonstrated with high sensitivity and selectivity toward Fe2+ cations. This system is an inorganic/organic hybrid system in which Cu2+ is coordinated at the receptor site of a conjugated polymer sensor. The unique quenching behavior of the Cu2+ toward this conjugated polymer was used to lower the initial fluorescence intensity of the sensory system. This results in a highly sensitive and also selective sensory system toward Fe2+ as compared to other related divalent transition-metal cations in solution.

Metal-organic framework based highly selective fluorescence turn-on probe for hydrogen sulphide.

Project description:Hydrogen sulphide (H2S) is known to play a vital role in human physiology and pathology which stimulated interest in understanding complex behaviour of H2S. Discerning the pathways of H2S production and its mode of action is still a challenge owing to its volatile and reactive nature. Herein we report azide functionalized metal-organic framework (MOF) as a selective turn-on fluorescent probe for H2S detection. The MOF shows highly selective and fast response towards H2S even in presence of other relevant biomolecules. Low cytotoxicity and H2S detection in live cells, demonstrate the potential of MOF towards monitoring H2S chemistry in biological system. To the best of our knowledge this is the first example of MOF that exhibit fast and highly selective fluorescence turn-on response towards H2S under physiological conditions.

A Highly Selective and Sensitive Fluorescent Turn-Off Probe for Cu2+ Based on a Guanidine Derivative.

Project description:A new highly selective and sensitive fluorescent probe for Cu2+, N-n-butyl-4-(1'-cyclooctene-1',3',6'-triazole)-1,8-naphthalimide (L), was synthesized and evaluated. The structure of compound L was characterized via IR, ¹H-NMR, 13C-NMR and HRMS. The fluorescent probe was quenched by Cu2+ with a 1:1 binding ratio and behaved as a "turn-off" sensor. An efficient and sensitive spectrofluorometric method was developed for detecting and estimating trace levels of Cu2+ in EtOH/H₂O. The ligand exhibited excitation and emission maxima at 447 and 518 nm, respectively. The equilibrium binding constant of the ligand with Cu2+ was 1.57 × 10⁴ M-1, as calculated using the Stern.

A Highly Selective and Sensitive Turn-On Fluorescent Chemosensor Based on Rhodamine 6G for Iron(III).

Project description:Recently, more and more rhodamine derivatives have been used as fluorophores to construct sensors due to their excellent spectroscopic properties. A rhodamine-based fluorescent and colorimetric Fe(3+) chemosensor 3',6'-bis(ethylamino)-2-acetoxyl-2',7'-dimethyl-spiro[1H-isoindole-1,9'-[9H]xanthen]-3(2H)-one (RAE) was designed and synthesized. Upon the addition of Fe(3+), the dramatic enhancement of both fluorescence and absorbance intensity, as well as the color change of the solution, could be observed. The detection limit of RAE for Fe(3+) was around 7.98 ppb. Common coexistent metal ions showed little or no interference in the detection of Fe(3+). Moreover, the addition of CN(-) could quench the fluorescence of the acetonitrile solution of RAE and Fe(3+), indicating the regeneration of the chemosensor RAE. The robust nature of the sensor was shown by the detection of Fe(3+) even after repeated rounds of quenching. As iron is a ubiquitous metal in cells and plays vital roles in many biological processes, this chemosensor could be developed to have applications in biological studies.

Anthracene-Based Highly Selective and Sensitive Fluorescent "Turn-on" Chemodosimeter for Hg2.

Project description:Three π-extended anthracene-bearing thioacetals (1-3) have been synthesized, and their fluorescence "turn-on" responses to Hg2+ ions are studied. The chemodosimetric fluorescence-sensing behavior and their resulting hydrolysis via a desulfurization reaction mechanism leads to the formation of highly fluorescent respective aldehyde substitutions. Furthermore, this mechanism was supported by increase in the quantum yields of their resulting aldehydes and is correlated to their molecular substitution. The chemosensors 1-3 have exhibited to be promising receptors toward Hg2+ ions in the presence of other competitive metal ions. Moreover, the detection limits of 1-3 have been found to be in the nanomolar range (94, 59, and 235, respectively). Fluorescence microscopic imaging studies show that 1-2 have been found to be effective for fluorescence imaging in live cells. Moreover, compounds 1-3 act as potential candidates for the detection of Hg2+ in environmental and biological systems as well as real samples.

Boronic Acid-Modified Magnetic Fe3O4@mTiO2 Microspheres for Highly Sensitive and Selective Enrichment of N-Glycopeptides in Amniotic Fluid.

Project description:Although mesoporous materials and magnetic materials are used to enrich glycopeptides, materials sharing both mesoporous structures and magnetic properties have not been reported for glycopeptide analyses. Here we prepared boronic acid-modified magnetic Fe3O4@mTiO2 microspheres by covalent binding of boronic acid molecules onto the surfaces of silanized Fe3O4@mTiO2 microspheres. The final particles (denoted as B-Fe3O4@mTiO2) showed a typical magnetic hysteresis curve, indicating superparamagnetic behavior; meanwhile, their mesoporous sizes did not change in spite of the reduction in surface area and pore volume. By using these particles together with conventional poly(methyl methacrylate) (PMMA) nanobeads, we then developed a synergistic approach for highly specific and efficient enrichment of N-glycopeptides/glycoproteins. Owing to the introduction of PMMA nanobeads that have strong adsorption towards nonglycopeptides, the number of N-glycopeptides detected and the signal-to-noise ratio in analyzing standard proteins mixture both increased appreciably. The recovery of N-glycopeptides by the synergistic method reached 92.1%, much improved than from B-Fe3O4@mTiO2 alone that was 75.3%. Finally, we tested this approach in the analysis of amniotic fluid, obtaining the maximum number and ratio of N-glycopeptides compared to the use of B-Fe3O4@mTiO2 alone and commercial SiMAG-boronic acid particles. This ensemble provides an interesting and efficient enrichment platform for glycoproteomics research.

Highly sensitive and selective fluorescence assays for rapid screening of endothelin-converting enzyme inhibitors.

Project description:The highly potent vasoconstrictor peptide endothelin (ET) is generated from an inactive precursor, big endothelin (bET), by endothelin-converting enzyme (ECE). ECE is a phosphoramidon-sensitive zinc metallopeptidase, which is closely related to neprilysin (neutral endopeptidase). It is possible that compounds which inhibit the formation of ET may be used as new drugs for the treatment of cardiovascular diseases. Such an approach requires a fast, simple and selective assay to measure ECE activity, allowing rapid screening of inhibitors. We describe here two new ECE substrates based on the concept of 'intramolecularly quenched fluorescence' which may fulfill this aim. One, S(1) [Pya(21)-Nop(22)-bET-1(19--35)], is the (19--35) fragment of the natural peptide big-ET-1(1--38), which is modified by introducing the fluorescent amino acid, pyrenylalanine (Pya), in position 21 and a quencher, p-nitrophenylalanine (Nop), in position 22. The second substrate (S(2)) is a small peptide, Ac-Ser-Gly-Pya-Lys-Ala-Phe-Ala-Nop-Gly-Lys-NH(2), from a biased substrate peptide library. The recombinant, hECE-1c, cleaved both Pya(21)-Nop(22)-bET-1(19--35) and the natural substrate selectively between residues 21 and 22, whereas cleavage occurred between alanine and phenylalanine in the small peptide. In both cases, this generated intense fluorescence emission. The synthesis and kinetic parameters of these substrates are described. These assays, which can be used directly on tissue homogenates, are the most sensitive and selective described to date for ECE, and are easily automated for a high-throughput screening of inhibitors.

A highly selective fluorescence "turn-on" sensor for Ca2+ based on diarylethene with a triazozoyl hydrazine unit.

Project description:A new photochromic diarylethene derivative with a triazozoyl hydrazine unit has been designed and synthesized. Its photochromism and photoswitchable fluorescence behaviors were studied systematically by the stimuli of lights and chemical substances in acetonitrile solution. With the addition of Ca2+, the emission intensity enhanced 6.7 fold, accompanied by an obvious fluorescent color change from dark to light blue. The complexation between the derivative and Ca2+ is reversible with the 1 : 1 stoichiometry, which was verified by Job's plot and MS. The limit of detection (LOD) for Ca2+ was determined to be 2.49 × 10-8 mol L-1. Based on this unimolecular platform, a logic circuit was designed with fluorescence intensity at 482 nm as the output and the combined stimuli of UV/vis and Ca2+/EDTA as four inputs.

Fabrication of Fe3O4@PVA-Cu Nanocomposite and Its Application for Facile and Selective Oxidation of Alcohols.

Project description:Fe3O4@PVA-Cu nanocomposite was introduced as an affordable catalyst for selective oxidation of alcohols into various aldehydes and ketones. The synthesized nanocomposite was characterized by applying essential analyses. The peaks that are appeared in FT-IR spectroscopy confirmed the production of the Fe3O4@PVA-Cu nanocomposite. In addition, EDX analysis proved the presence of oxygen, carbon, iron, and copper elements in the catalyst. Further, TGA analysis showed high thermal stability of the nanocomposite. VSM technique was applied to examine the magnetic property of the nanocomposite. The results demonstrated a high magnetic property in the catalyst, which enables easy separation of it from the reaction solution. TEM and SEM imaging showed the nanoscale size of the particles (~20 nm) in the catalyst. Additionally, XRD data was compatible with that of Fe3O4 nanoparticles. The application of the nanocomposite has been studied in the selective oxidation of alcohols in the presence of acetonitrile as solvent, and hydrogen peroxide as a supplementary oxidizing agent. This technique is remarkably facile and inexpensive. Further, the products showed high yields. In addition, the calculated TON and TOF values indicated the phenomenal efficiency of the nanocomposite in preparation of targeted products.

Microbial synthesis of Pd/Fe3O4, Au/Fe3O4 and PdAu/Fe3O4 nanocomposites for catalytic reduction of nitroaromatic compounds.

Project description:Magnetically recoverable noble metal nanoparticles are promising catalysts for chemical reactions. However, the chemical synthesis of these nanocatalysts generally causes environmental concern due to usage of toxic chemicals under extreme conditions. Here, Pd/Fe3O4, Au/Fe3O4 and PdAu/Fe3O4 nanocomposites are biosynthesized under ambient and physiological conditions by Shewanella oneidensis MR-1. Microbial cells firstly transform akaganeite into magnetite, which then serves as support for the further synthesis of Pd, Au and PdAu nanoparticles from respective precursor salts. Surface-bound cellular components and exopolysaccharides not only function as shape-directing agent to convert some Fe3O4 nanoparticles to nanorods, but also participate in the formation of PdAu alloy nanoparticles on magnetite. All these three kinds of magnetic nanocomposites can catalyze the reduction of 4-nitrophenol and some other nitroaromatic compounds by NaBH4. PdAu/Fe3O4 demonstrates higher catalytic activity than Pd/Fe3O4 and Au/Fe3O4. Moreover, the magnetic nanocomposites can be easily recovered through magnetic decantation after catalysis reaction. PdAu/Fe3O4 can be reused in at least eight successive cycles of 4-nitrophenol reduction. The biosynthesis approach presented here does not require harmful agents or rigorous conditions and thus provides facile and environmentally benign choice for the preparation of magnetic noble metal nanocatalysts.
