Tree diversity and functional leaf traits drive herbivore-associated microbiomes in subtropical China.

ABSTRACT: Herbivorous insects acquire microorganisms from host plants or soil, but it remains unclear how the diversity and functional composition of host plants contribute to structuring herbivore microbiomes. Within a controlled tree diversity setting, we used DNA metabarcoding of 16S rRNA to assess the contribution of Lepidoptera species and their local environment (particularly, tree diversity, host tree species, and leaf traits) to the composition of associated bacterial communities. In total, we obtained 7,909 bacterial OTUs from 634 caterpillar individuals comprising 146 species. Tree diversity was found to drive the diversity of caterpillar-associated bacteria both directly and indirectly via effects on caterpillar communities, and tree diversity was a stronger predictor of bacterial diversity than diversity of caterpillars. Leaf toughness and dry matter content were important traits of the host plant determining bacterial species composition, while leaf calcium and potassium concentration influenced bacterial richness. Our study reveals previously unknown linkages between trees and their characteristics, herbivore insects, and their associated microbes, which contributes to developing a more nuanced understanding of functional dependencies between herbivores and their environment, and has implications for the consequences of plant diversity loss for trophic interactions.

Project description:Herbivores and fungal pathogens are key drivers of plant community composition and functioning. The effects of herbivores and pathogens are mediated by the diversity and functional characteristics of their host plants. However, the combined effects of herbivory and pathogen damage, and their consequences for plant performance, have not yet been addressed in the context of biodiversity-ecosystem functioning research. We analyzed the relationships between herbivory, fungal pathogen damage and their effects on tree growth in a large-scale forest-biodiversity experiment. Moreover, we tested whether variation in leaf trait and climatic niche characteristics among tree species influenced these relationships. We found significant positive effects of herbivory on pathogen damage, and vice versa. These effects were attenuated by tree species richness-because herbivory increased and pathogen damage decreased with increasing richness-and were most pronounced for species with soft leaves and narrow climatic niches. However, herbivory and pathogens had contrasting, independent effects on tree growth, with pathogens decreasing and herbivory increasing growth. The positive herbivory effects indicate that trees might be able to (over-)compensate for local damage at the level of the whole tree. Nevertheless, we found a dependence of these effects on richness, leaf traits and climatic niche characteristics of the tree species. This could mean that the ability for compensation is influenced by both biodiversity loss and tree species identity-including effects of larger-scale climatic adaptations that have been rarely considered in this context. Our results suggest that herbivory and pathogens have additive but contrasting effects on tree growth. Considering effects of both herbivory and pathogens may thus help to better understand the net effects of damage on tree performance in communities differing in diversity. Moreover, our study shows how species richness and species characteristics (leaf traits and climatic niches) can modify tree growth responses to leaf damage under real-world conditions.

Project description:Research about biodiversity-productivity relationships has focused on herbaceous ecosystems, with results from tree field studies only recently beginning to emerge. Also, the latter are concentrated largely in the temperate zone. Tree species diversity generally is much higher in subtropical and tropical than in temperate or boreal forests, with reasons not fully understood. Niche overlap and thus complementarity in the use of resources that support productivity may be lower in forests than in herbaceous ecosystems, suggesting weaker productivity responses to diversity change in forests. We studied stand basal area, vertical structure, leaf area, and their relationship with tree species richness in a subtropical forest in south-east China. Permanent forest plots of 30 x 30 m were selected to span largely independent gradients in tree species richness and secondary successional age. Plots with higher tree species richness had a higher stand basal area. Also, stand basal area increases over a 4-year census interval were larger at high than at low diversity. These effects translated into increased carbon stocks in aboveground phytomass (estimated using allometric equations). A higher variability in tree height in more diverse plots suggested that these effects were facilitated by denser canopy packing due to architectural complementarity between species. In contrast, leaf area was not or even negatively affected by tree diversity, indicating a decoupling of carbon accumulation from leaf area. Alternatively, the same community leaf area might have assimilated more C per time interval in more than in less diverse plots because of differences in leaf turnover and productivity or because of differences in the display of leaves in vertical and horizontal space. Overall, our study suggests that in species-rich forests niche-based processes support a positive diversity-productivity relationship and that this translates into increased carbon storage in long-lived woody structures. Given the high growth rates of these forests during secondary succession, our results further indicate that a forest management promoting tree diversity after disturbance may accelerate CO2 sequestration from the atmosphere and thus be relevant in a climate-change context.

Project description:1.Insect herbivory can strongly affect ecosystem processes, and its relationship with plant diversity is a central topic in biodiversity-functioning research. However, very little is known about this relationship from complex ecosystems dominated by long-lived individuals, such as forests, especially over gradients of high plant diversity.2.We analysed insect herbivory on saplings of 10 tree and shrub species across 27 forest stands differing in age and tree species richness in an extraordinarily diverse subtropical forest ecosystem in China. We tested whether plant species richness significantly influences folivory in these highly diverse forests or whether other factors play a more important role at such high levels of phytodiversity.3.Leaf damage was assessed on 58 297 leaves of 1284 saplings at the end of the rainy season in 2008, together with structural and abiotic stand characteristics.4.Species-specific mean damage of leaf area ranged from 3% to 16%. Herbivory increased with plant species richness even after accounting for potentially confounding effects of stand characteristics, of which stand age-related aspects most clearly covaried with herbivory. Intraspecific density dependence or other abiotic factors did not significantly influence overall herbivory across forest stands.5.Synthesis.The positive herbivory-plant diversity relationship indicates that effects related to hypotheses of resource concentration, according to which a reduction in damage by specialized herbivores might be expected as host plant concentration decreases with increasing plant diversity, do not seem to be major determinants for overall herbivory levels in our phytodiverse subtropical forest ecosystem. We discuss the potential role of host specificity of dominant herbivores, which are often expected to show a high degree of specialization in many (sub)tropical forests. In the forest system we studied, a much higher impact of polyphagous species than traditionally assumed might explain the observed patterns, as these species can profit from a broad dietary mix provided by high plant diversity. Further testing is needed to experimentally verify this assumption.

Project description:Subtropical tree species may experience severe drought stress due to variable rainfall under future climates. However, the capacity to restore hydraulic function post-drought might differ among co-occurring species with contrasting leaf habits (e.g., evergreen and deciduous) and have implications for future forest composition. Moreover, the links between hydraulic recovery and physiological and morphological traits related to water-carbon availability are still not well understood. Here, potted seedlings of six tree species (four evergreen and two deciduous) were grown outdoors under a rainout shelter. They grew under favorable water conditions until they were experimentally subjected to a soil water deficit leading to losses of ca. 50% of hydraulic conductivity, and then soils were re-watered to field capacity. Traits related to carbon and water relations were measured. There were differences in drought responses and recovery between species, but not as a function of evergreen or deciduous groups. Sapindus mukorossi exhibited the most rapid drought response, which was associated with a suite of physiological and morphological traits (larger plant size, the lowest hydraulic capacitance (C branch), higher minimum conductance (g min) and lower HV (Huber value)). Upon re-watering, xylem water potential exhibited fast recovery in 1-3 days among species, while photosynthesis at saturating light (A sat) and stomatal conductance (g s) recovery lagged behind water potential recovery depending on species, with g s recovery being more delayed than A sat in most species. Furthermore, none of the six species exhibited significant hydraulic recovery during the 7 days re-watering period, indicating that xylem refilling was apparently limited; in addition, NSC availability had a minimal role in facilitating hydraulic recovery during this short-term period. Collectively, if water supply is limited by insignificant hydraulic recovery post-drought, the observed carbon assimilation recovery of seedlings may not be sustained over the longer term, potentially altering seedling regeneration and shifting forest species composition in subtropical China under climate change.

Project description:Biodiversity loss in riparian forests has the potential to alter rates of leaf litter decomposition in stream ecosystems. However, studies have reported the full range of positive, negative and no effects of plant diversity loss on decomposition, and there is currently no explanation for such inconsistent results. Furthermore, it is uncertain whether plant diversity loss affects other ecological processes related to decomposition, such as fine particulate organic matter production or detritivore growth, which precludes a thorough understanding of how detrital stream food webs are impacted by plant diversity loss. We used a microcosm experiment to examine the effects of plant diversity loss on litter decomposition, fine particulate organic matter production, and growth of a dominant leaf-shredding detritivore, using litter mixtures varying in species composition. We hypothesized that plant diversity loss would decrease the rates of all studied processes, but such effects would depend on the leaf traits present in litter mixtures (both their average values and their variability). Our findings partly supported our hypotheses, showing that plant diversity loss had a consistently negative effect on litter decomposition and fine particulate organic matter production (but not on detritivore growth) across litter mixtures, which was mediated by detritivores. Importantly, the magnitude of the diversity effect and the relative importance of different mechanisms underlying this effect (i.e., complementarity vs. selection) varied depending on the species composition of litter mixtures, mainly because of differences in litter nutritional quality and trait variability. Complementarity was prevalent but varied in size, with positive selection effects also occurring in some mixtures. Our results support the notion that loss of riparian plant species is detrimental to key stream ecosystem processes that drive detrital food webs, but that the magnitude of such effects largely depends on the the order of species loss.
