Variability and Randomness of the Instantaneous Firing Rate.

ABSTRACT: The apparent stochastic nature of neuronal activity significantly affects the reliability of neuronal coding. To quantify the encountered fluctuations, both in neural data and simulations, the notions of variability and randomness of inter-spike intervals have been proposed and studied. In this article we focus on the concept of the instantaneous firing rate, which is also based on the spike timing. We use several classical statistical models of neuronal activity and we study the corresponding probability distributions of the instantaneous firing rate. To characterize the firing rate variability and randomness under different spiking regimes, we use different indices of statistical dispersion. We find that the relationship between the variability of interspike intervals and the instantaneous firing rate is not straightforward in general. Counter-intuitively, an increase in the randomness (based on entropy) of spike times may either decrease or increase the randomness of instantaneous firing rate, in dependence on the neuronal firing model. Finally, we apply our methods to experimental data, establishing that instantaneous rate analysis can indeed provide additional information about the spiking activity.

Project description:Spontaneous baroreflex sensitivity (BRS) is a widely used tool for the quantification of the cardiovascular regulation. Numerous groups use the xBRS method, which calculates the cross-correlation between the systolic beat-to-beat blood pressure and the R-R interval (resampled at 1 Hz) in a 10 s sliding window, with 0-5 s delays for the interval. The delay with the highest correlation is selected and, if significant, the quotient of the standard deviations of the R-R intervals and the systolic blood pressures is recorded as the corresponding xBRS value. In this paper we test the hypothesis that the xBRS method quantifies the causal interactions of spontaneous BRS from non-invasive measurements at rest. We use the term spontaneous BRS in the sense of the sensitivity curve is calculated from non-interventional, i.e., spontaneous, baroreceptor activity. This study includes retrospective analysis of 1828 measurements containing ECG as well as continues blood pressure under resting conditions. Our results show a high correlation between the heart rate - systolic blood pressure variability (HRV/BPV) quotient and the xBRS (r = 0.94, p < 0.001). For a deeper understanding we conducted two surrogate analyses by substituting the systolic blood pressure by its reversed time series. These showed that the xBRS method was not able to quantify causal relationships between the two signals. It was not possible to distinguish between random and baroreflex controlled sequences. It appears xBRS rather determines the HRV/BPV quotient. We conclude that the xBRS method has a potentially large bias in characterizing the capacity of the arterial baroreflex under resting conditions. During slow breathing, estimates for xBRS are significantly increased, which clearly shows that measurements at rest only involve limited baroreflex activity, but does neither challenge, nor show the full range of the arterial baroreflex regulatory capacity. We show that xBRS is exclusively dominated by the heart rate to systolic blood pressure ratio (r = 0.965, p < 0.001). Further investigations should focus on additional autonomous testing procedures such as slow breathing or orthostatic testing to provide a basis for a non-invasive evaluation of baroreflex sensitivity.

Project description:The firing rate of the mitral/tufted cells in the olfactory bulb is known to undergo significant trial-to-trial variability and is affected by anesthesia. Here we ask whether odorant-elicited changes in firing rate depend on the rate before application of the stimulus in the awake and anesthetized mouse. We find that prestimulus firing rate varies widely on a trial-to-trial basis and that the stimulus-induced change in firing rate decreases with increasing prestimulus firing rate. Interestingly, this prestimulus firing rate dependence was different when the behavioral task did not involve detecting the valence of the stimulus. Finally, when the animal was learning to associate the odor with reward, the prestimulus firing rate was smaller for false alarms compared with correct rejections, suggesting that intrinsic activity reflects the anticipatory status of the animal. Thus, in this sensory modality, changes in behavioral status alter the intrinsic prestimulus activity, leading to a change in the responsiveness of the second-order neurons. We speculate that this trial-to-trial variability in odorant responses reflects sampling of the massive parallel input by subsets of mitral cells.SIGNIFICANCE STATEMENT The olfactory bulb must deal with processing massive parallel input from ∼1200 distinct olfactory receptors. In contrast, the visual system receives input from a small number of photoreceptors and achieves recognition of complex stimuli by allocating processing for distinct spatial locations to different brain areas. Here we find that the change in firing rate elicited by the odorant in second-order mitral cells depends on the intrinsic activity leading to a change of magnitude in the responsiveness of these neurons relative to this prestimulus activity. Further, we find that prestimulus firing rate is influenced by behavioral status. This suggests that there is top-down modulation allowing downstream brain processing areas to perform dynamic readout of olfactory information.
