The complete mitochondrial genome of Osmanthus fragrans (Lamiales, Oleaceae) from China.

ABSTRACT: Osmanthus fragrans is a well-known ornamental tree with high medicinal and edible values. In this study, the complete mitochondrial genome sequence of O. fragrans was assembled, annotated, and analyzed using phylogenomic methods. The complete mitochondrial genome of O. fragrans was 563,202 bp in length and displays an overall GC content of 44.58%. Sixteen chloroplast-derived segments with an average length of 1260 bp were identified. The complete mitochondrial genome contained 74 genes in total, including 44 protein-coding, three rRNA, and 27 tRNA genes, among which seven protein-coding and six tRNA genes were chloroplast-derived. Phylogenetic analysis showed that O. fragrans was closely related to Chionanthus rupicola within the Oleaceae. This study could provide genomic resources for a better understanding of O. fragrans and further studies on the evolution of Oleaceae.

Genetic diversity of androdioecious Osmanthus fragrans (Oleaceae) cultivars using microsatellite markers.

Project description:For cultivar classification, identification, and genetic improvement, microsatellite markers were developed to analyze the genetic diversity of androdioecious Osmanthus fragrans cultivars. • Fifteen microsatellite markers were developed from sequences downloaded from the National Center for Biotechnology Information, which included two with null alleles. These primers were screened on 62 typical androdioecious O. fragrans cultivars belonging to four groups (Asiaticus, Albus, Luteus, and Aurantiacus). The number of alleles ranged from two to six, with a mean of 3.7 per locus. The observed and expected heterozygosities ranged from 0.1000 to 0.9091 and from 0.1287 to 0.9167, respectively. Results from structure analyses indicated that Asiaticus and Albus were genetically mixed, and Luteus and Aurantiacus were partially genetically differentiated. • These markers will be useful for genetic study of androdioecious O. fragrans cultivars and facilitate cultivar classification, particularly for the cultivar groups Luteus and Aurantiacus.

Complete plastome sequence of Osmanthus armatus and phylogenetic implications in Oleaceae.

Project description:Osmanthus armatus (Oleaceae) is mainly distributed in the southwest of China and also planted in arboretums as an ornamental plant. In the present study, the plastome of O. armatus was reconstructed using genome skimming sequencing, and the phylogeny analysis was inferred based on whole plastome data. The plastome of O. armatus is 155,259?bp in length, comprising two copies of inverted regions (IR, 25,680?bp) separated by the large single copy (LSC, 86,534?bp) and small single copy (SSC, 17,365?bp) regions. The genome encodes 114 unique genes, including 80 different protein-coding genes, 30 tRNA genes, and four rRNA genes, with 20 duplicated genes in the IR regions. Phylogenetic analysis suggests that the representative species from Osmanthus is monophyletic, and O. armatus is sister to O. fragrans within this genus.

Whole-genome resequencing of Osmanthus fragrans provides insights into flower color evolution.

Project description:Osmanthus fragrans is a well-known ornamental plant that has been domesticated in China for 2500 years. More than 160 cultivars have been found during this long period of domestication, and they have subsequently been divided into four cultivar groups, including the Yingui, Jingui, Dangui, and Sijigui groups. These groups provide a set of materials to study genetic evolution and variability. Here, we constructed a reference genome of O. fragrans 'Liuyejingui' in the Jingui group and investigated its floral color traits and domestication history by resequencing a total of 122 samples, including 119 O. fragrans accessions and three other Osmanthus species, at an average sequencing depth of 15×. The population structure analysis showed that these 119 accessions formed an apparent regional cluster. The results of linkage disequilibrium (LD) decay analysis suggested that varieties with orange/red flower color in the Dangui group had undergone more artificial directional selection; these varieties had the highest LD values among the four groups, followed by the Sijigui, Jingui, and Yingui groups. Through a genome-wide association study, we further identified significant quantitative trait loci and genomic regions containing several genes, such as ethylene-responsive transcription factor 2 and Arabidopsis pseudoresponse regulator 2, that are positively associated with petal color. Moreover, we found a frameshift mutation with a 34-bp deletion in the first coding region of the carotenoid cleavage dioxygenase 4 gene. This frameshift mutation existed in at least one site on both alleles in all varieties of the Dangui group. The results from this study shed light on the genetic basis of domestication in woody plants, such as O. fragrans.

Genome-wide miRNA analysis and integrated network for flavonoid biosynthesis in Osmanthus fragrans.

Project description:BackgroundOsmanthus fragrans is an important economical plant containing multiple secondary metabolites including flavonoids and anthocyanins. During the past years, the roles of miRNAs in regulating the biosynthesis of secondary metabolites in plants have been widely investigated. However, few studies on miRNA expression profiles and the potential roles in regulating flavonoid biosynthesis have been reported in O. fragrans.ResultsIn this study, we used high-throughput sequencing technology to analyze the expression profiles of miRNAs in leaf and flower tissues of O. fragrans. As a result, 106 conserved miRNAs distributed in 47 families and 88 novel miRNAs were identified. Further analysis showed there were 133 miRNAs differentially expressed in leaves and flowers. Additionally, the potential target genes of miRNAs as well as the related metabolic pathways were predicted. In the end, flavonoid content was measured in flower and leaf tissues and potential role of miR858 in regulating flavonoid synthesis was illustrated in O. fragrans.ConclusionsThis study not only provided the genome-wide miRNA profiles in the flower and leaf tissue of O. fragrans, but also investigated the potential regulatory role of miR858a in flavonoid synthesis in O. fragrans. The results specifically indicated the connection of miRNAs to the regulation of secondary metabolite biosynthesis in non-model economical plant.

The odor of Osmanthus fragrans attenuates food intake.

Project description:Odors have been shown to exert an influence on various physiological and behavioral activities. However, little is known whether or not odor stimulation directly affects the levels of feeding-related neuropeptides. Here we show that the neural transmission by Osmanthus fragrans (OSM) decreased the mRNA expression of orexigenic neuropeptides, such as agouti-related protein, neuropeptide Y, melanin-concentrating hormone and prepro-orexin, while increased anorexigenic neuropeptides, such as cocaine- and amphetamine-regulated transcript and proopiomelanocortin in rats. The decreased number of orexin-immunoreactive neurons in the hypothalamus coincided well with the OSM-induced decreases in the expression of prepro-orexin mRNA. This study demonstrates that the OSM odor, which is known to have a mild sedative effect, decreases the motivation to eat, food intake and body weight, accompanied by sluggish masticatory movements. The data suggest that these effects are due to suppression of orexigenic neuropeptides and activation of anorexigenic neuropeptides in the hypothalamus.
