Spatio-temporal models of ambient air pollution can be used to predict pollutant levels across a geographical region. These predictions may then be used as estimates of exposure for individuals in analyses of the health effects of air pollution. Integrated nested Laplace approximations is a method for Bayesian inference, and a fast alternative to Markov chain Monte Carlo methods. Project description:Estimates of daily air pollution concentrations with complete spatial and temporal coverage are important for supporting epidemiologic studies and health impact assessments. While numerous approaches have been developed for modeling air pollution, they typically only consider each pollutant separately. We describe a spatial multipollutant data fusion model that combines monitoring measurements and chemical transport model simulations that leverages dependence between pollutants to improve spatial prediction. For the contiguous United States, we created a data product of daily concentration for 12 pollutants (CO, NOx, NO2, SO2, O3, PM10, and PM2.5 species EC, OC, NO3, NH4, SO4) during the period 2005 to 2014. Out-of-sample prediction showed good performance, particularly for daily PM2.5 species EC (R2 = 0.64), OC (R2 = 0.75), NH4 (R2 = 0.84), NO3 (R2 = 0.73), and SO4 (R2 = 0.80). By employing the integrated nested Laplace approximation (INLA) for Bayesian inference, our approach also provides model-based prediction error estimates. The daily data product at 12km spatial resolution will be publicly available immediately upon publication. To our knowledge this is the first publicly available data product for major PM2.5 species and several gases at this spatial and temporal resolution.

Project description:Studies have linked exposure to air pollutants to short-term and sub-chronic health outcomes. However, individual-level air pollution exposure is difficult to measure at a high spatial and temporal resolution and for larger populations due to limitations in sampling techniques. We presented a hierarchical model to capture spatiotemporal variability of nitrogen dioxide (NO2) and nitrogen oxides (NOx) concentrations in Southern California by combining high temporal resolution data from routine monitoring stations with high spatial resolution data from investigator-initiated episodic measurements. In this model, the spatiotemporal field of concentrations was first decomposed into a mean and residual and the mean representing the seasonal trend was further decomposed into a constant and varying temporal basis functions. The mean of the spatially varying coefficients of temporal basis functions were modeled by local covariates using non-linear generalized additive model and least square fitting using measurements from both routine monitoring and additional episodic sampling locations, while the spatially-correlated residuals of the coefficients were co-kriged. We found traffic, land-use and wind accounted for a large portion of the variance (beyond 35%) for the long-term average trend of concentrations. Spatial residuals accounted for a large portion of the variance of the temporal components (about 30% for NO2 and 20% for NOx). Leave-one-out cross validation produced an R2 of 0.84 for NO2 and 0.81 for NOx when comparing the modeled weekly concentration with the observed trends at all routine monitoring stations.

Project description:Most published epidemiology studies of long-term air pollution health effects have relied on central site monitoring to investigate regional-scale differences in exposure. Few cohort studies have had sufficient data to characterize localized variations in pollution, despite the fact that large gradients can exist over small spatial scales. Similarly, previous data have generally been limited to measurements of particle mass or several of the criteria gases. The Multi-Ethnic Study of Atherosclerosis and Air Pollution (MESA Air) is an innovative investigation undertaken to link subclinical and clinical cardiovascular health effects with individual-level estimates of personal exposure to ambient-origin pollution. This project improves on prior work by implementing an extensive exposure assessment program to characterize long-term average concentrations of ambient-generated PM2.5, specific PM2.5 chemical components, and copollutants, with particular emphasis on capturing concentration gradients within cities. This paper describes exposure assessment in MESA Air, including questionnaires, community sampling, home monitoring, and personal sampling. Summary statistics describing the performance of the sampling methods are presented along with descriptive statistics of the air pollution concentrations by city.

Project description:Exposure to air pollutants is known to exacerbate asthma, with prior studies focused on associations between single pollutant exposure and asthma exacerbations. As air pollutants often exist as a complex mixture, there is a gap in understanding the association between complex air pollutant mixtures and asthma exacerbations. We evaluated the association between the air pollutant mixture (52 pollutants) and pediatric asthma exacerbations.MethodThis study focused on children (age ≤ 19 years) who lived in Douglas County, Nebraska, during 2016-2019. A seasonal-scale joint association between the outdoor air pollutant mixture adjusting for potential confounders (temperature, precipitation, wind speed, and wind direction) in relation to pediatric asthma exacerbation-related emergency department (ED) visits was evaluated using the generalized weighted quantile sum (qWQS) regression with repeated holdout validation.ResultsWe observed associations between air pollutant mixture and pediatric asthma exacerbations during spring (lagged by 5 days), summer (lag 0-5 days), and fall (lag 1-3 days) seasons. The estimate of the joint outdoor air pollutant mixture effect was higher during the summer season (adjusted-βWQS = 1.11, 95% confidence interval [CI]: 0.66, 1.55), followed by spring (adjusted-βWQS = 0.40, 95% CI: 0.16, 0.62) and fall (adjusted-βWQS = 0.20, 95% CI: 0.06, 0.33) seasons. Among the air pollutants, PM2.5, pollen, and mold contributed higher weight to the air pollutant mixture.ConclusionThere were associations between outdoor air pollutant mixture and pediatric asthma exacerbations during the spring, summer, and fall seasons. Among the 52 outdoor air pollutant metrics investigated, PM2.5, pollen (sycamore, grass, cedar), and mold (Helminthosporium, Peronospora, and Erysiphe) contributed the highest weight to the air pollutant mixture.

Project description:Research linking prenatal ambient air pollution with childhood lung function has largely considered one pollutant at a time. Real-life exposure is to mixtures of pollutants and their chemical components; not considering joint effects/effect modification by co-exposures contributes to misleading results.MethodsAnalyses included 198 mother-child dyads recruited from two hospitals and affiliated community health centers in Boston, Massachusetts, USA. Daily prenatal pollutant exposures were estimated using satellite-based hybrid chemical-transport models, including nitrogen dioxide(NO2), ozone(O3), and fine particle constituents (elemental carbon [EC], organic carbon [OC], nitrate [NO3 -], sulfate [SO4 2-], and ammonium [NH4 +]). Spirometry was performed at age 6.99 ± 0.89 years; forced expiratory volume in 1s (FEV1), forced vital capacity (FVC), and forced mid-expiratory flow (FEF25-75) z-scores accounted for age, sex, height, and race/ethnicity. We examined associations between weekly-averaged prenatal pollution mixture levels and outcomes using Bayesian Kernel Machine Regression-Distributed Lag Models (BKMR-DLMs) to identify susceptibility windows for each component and estimate a potentially complex mixture exposure-response relationship including nonlinear effects and interactions among exposures. We also performed linear regression models using time-weighted-mixture component levels derived by BKMR-DLMs adjusting for maternal age, education, perinatal smoking, and temperature.ResultsMost mothers were Hispanic (63%) or Black (21%) with ≤12 years of education (67%). BKMR-DLMs identified a significant effect for O3 exposure at 18-22 weeks gestation predicting lower FEV1/FVC. Linear regression identified significant associations for O3, NH4 +, and OC with decreased FEV1/FVC, FEV1, and FEF25-75, respectively. There was no evidence of interactions among pollutants.ConclusionsIn this multi-pollutant model, prenatal O3, OC, and NH4 + were most strongly associated with reduced early childhood lung function.

Project description:IntroductionWe investigated the extent to which associations of ambient air pollutant concentrations and birth weight varied across birth weight quantiles.MethodsWe analyzed singleton births ≥27 weeks of gestation from 20-county metropolitan Atlanta with conception dates between January 1, 2002 and February 28, 2006 (N = 273,711). Trimester-specific and total pregnancy average concentrations for 10 pollutants, obtained from ground observations that were interpolated using 12-km Community Multiscale Air Quality model outputs, were assigned using maternal residence at delivery. We estimated associations between interquartile range width (IQRw) increases in pollutant concentrations and changes in birth weight using quantile regression.ResultsGestational age-adjusted associations were of greater magnitude at higher percentiles of the birth weight distribution. Pollutants with large vehicle source contributions (carbon monoxide, nitrogen dioxide, PM2.5 elemental carbon, and total PM2.5 mass), as well as PM2.5 sulfate and PM2.5 ammonium, were associated with birth weight decreases for the higher birth weight percentiles. For example, whereas the decrease in mean birthweight per IQRw increase in PM2.5 averaged over pregnancy was -7.8 g (95% confidence interval = -13.6, -2.0 g), the quantile-specific associations were: 10th percentile -2.4 g (-11.5, 6.7 g); 50th percentile -8.9 g (-15.7, -2.0g); and 90th percentile -19.3 g (-30.6, -7.9 g). Associations for the intermediate and high birth weight quantiles were not sensitive to gestational age adjustment. For some pollutants, we saw associations at the lowest quantile (10th percentile) when not adjusting for gestational age.ConclusionsAssociations between air pollution and reduced birth weight were of greater magnitude for newborns at relatively heavy birth weights.

Project description:We introduce spatial (DLfuse) and spatiotemporal (DLfuseST) distributed lag data fusion methods for predicting point-level ambient air pollution concentrations, using, as input, gridded average pollution estimates from a deterministic numerical air quality model. The methods incorporate predictive information from grid cells surrounding the prediction location of interest and are shown to collapse to existing downscaling approaches when this information adds no benefit. The spatial lagged parameters are allowed to vary spatially/spatiotemporally to accommodate the setting where surrounding geographic information is useful in one area/time but not in another. We apply the new methods to predict ambient concentrations of eight-hour maximum ozone and 24-hour average PM2.5 at unobserved spatial locations and times, and compare the predictions with those from several state-of-the-art data fusion approaches. Results show that DLfuse and DLfuseST often provide improved model fit and predictive accuracy when the lagged information is shown to be beneficial. Code to apply the methods is available in the R package DLfuse.

Project description:Background:Limited epidemiological studies have investigated the relationship between prenatal exposure to ambient particulate matter and risk of abnormal fetal growth, and have reached inconclusive results. No study has been conducted in areas with very high air pollution levels. We investigated the hypothesis that exposure to high levels of particulate matter with aerodynamic diameter no larger than 10 µm (PM10) during pregnancy increases the risk of abnormal fetal growth. Methods:A birth cohort study was performed in Lanzhou, China, 2010-12, including 8877 pregnant women with 18 583 ultrasound measurements of four fetal growth parameters during pregnancy, including biparietal diameter (BPD), femur length (FL), head circumference (HC) and abdominal circumference (AC). Mixed-effects modelling was used to examine the associations between PM10 exposure and risk of abnormal fetal growth. Results:When average PM10 exposure from conception until the ultrasound examination exceeded 150 µg/m3, there were significant increases in standardized FL (β = 0.095, P = 0.0012) and HC (β = 0.090, P = 0.0078) measures. When average PM10 exposure was treated as continuous variable, we found a significant decrease in standardized BPD (β = -0.018, P = 0.0016) as per 10 µg/m3 increase in PM10. After examining the associations by various exposure windows, positive associations between higher levels of PM10 and fetal overgrowth were consistently seen for HC measures. Conclusions:Our study suggested that prenatal exposure to high levels of ambient PM10 increased the risk of abnormal fetal growth. The findings from our study have important public health implications and also call for future studies to explore the underlying mechanisms and post-natal consequences of these findings.

Project description:Outdoor air pollution, a spatially and temporally complex mixture, is a human carcinogen. However, ambient measurements may not reflect subject-level exposures, personal monitors do not assess internal dose, and spot assessments of urinary biomarkers may not recapitulate chronic exposures. Nucleophilic sites in serum albumin-particularly the free thiol at Cys34-form adducts with electrophiles. Due to the 4-week lifetime of albumin in circulation, accumulating adducts can serve as intermediate- to long-residence biomarkers of chronic exposure and implicate potential biological effects. Employing nanoflow liquid chromatography-high-resolution mass spectrometry (nLC-HRMS) and parallel reaction monitoring (PRM), we have developed and validated a novel targeted albumin adductomics platform capable of simultaneously monitoring dozens of Cys34 adducts per sample in only 2.5 μL of serum, with on-column limits of detection in the low-femtomolar range. Using this platform, we characterized the magnitude and impact of ambient outdoor air pollution exposures with three repeated measurements over 84 days in n = 26 nonsmoking women (n = 78 total samples) from Qidong, China, an area with a rising burden of lung cancer incidence. In concordance with seasonally rising ambient concentrations of NO2, SO2, and PM10 measured at stationary monitors, we observed elevations in concentrations of Cys34 adducts of benzoquinone (p < 0.05), benzene diol epoxide (BDE; p < 0.05), crotonaldehyde (p < 0.01), and oxidation (p < 0.001). Regression analysis revealed significant elevations in oxidation and BDE adduct concentrations of 300% to nearly 700% per doubling of ambient airborne pollutant levels (p < 0.05). Notably, the ratio of irreversibly oxidized to reduced Cys34 rose more than 3-fold during the 84-day period, revealing a dramatic perturbation of serum redox balance and potentially serving as a portent of increased pollution-related mortality risk. Our targeted albumin adductomics assay represents a novel and flexible approach for sensitive and multiplexed internal dosimetry of environmental exposures, providing a new strategy for personalized biomonitoring and prevention.

Project description:BackgroundStudies of prenatal air pollution (AP) exposure on child neurodevelopment have mostly focused on a single pollutant. We leveraged daily exposure data and implemented novel data-driven statistical approaches to assess effects of prenatal exposure to a mixture of seven air pollutants on cognitive functioning in school-age children from an urban pregnancy cohort.MethodsAnalyses included 236 children born at ≥37 weeks gestation. Maternal prenatal daily exposure levels for nitrogen dioxide (NO2), ozone (O3), and constituents of fine particles [elemental carbon (EC), organic carbon (OC), nitrate (NO3-), sulfate (SO42-), ammonium (NH4+)] were estimated based on residential addresses using validated satellite-based hybrid models or global 3-D chemical-transport models. Children completed Wide Range Assessment of Memory and Learning (WRAML-2) and Conners' Continuous Performance Test (CPT-II) at 6.5 ± 0.9 years of age. Time-weighted levels for mixture pollutants were estimated using Bayesian Kernel Machine Regression Distributed Lag Models (BKMR-DLMs), with which we also explored the interactions in the exposure-response functions among pollutants. Resulting time-weighted exposure levels were used in Weighted Quantile Sum (WQS) regressions to examine AP mixture effects on outcomes, adjusted for maternal age, education, child sex, and prenatal temperature.ResultsMothers were primarily ethnic minorities (81% Hispanic and/or black) reporting ≤12 years of education (68%). Prenatal AP mixture (per unit increase in WQS estimated AP index) was associated with decreased WRAML-2 general memory (GM; β = -0.64, 95%CI = -1.40, 0.00) and memory-related attention/concentration (AC; β = -1.03, 95%CI = -1.78, -0.27) indices, indicating poorer memory functioning, as well as increased CPT-II omission errors (OE; β = 1.55, 95%CI = 0.34, 2.77), indicating increased attention problems. When stratified by sex, association with AC index was significant among girls, while association with OE was significant among boys. Traffic-related pollutants (NO2, OC, EC) and SO42- were major contributors to these associations. There was no significant evidence of interactions among mixture components.ConclusionsPrenatal exposure to an AP mixture was associated with child neurocognitive outcomes in a sex- and domain-specific manner.
