Ascophyllum nodosum Based Extracts Counteract Salinity Stress in Tomato by Remodeling Leaf Nitrogen Metabolism.

ABSTRACT: Biostimulants have rapidly and widely been adopted as growth enhancers and stress protectants in agriculture, however, due to the complex nature of these products, their mechanism of action is not clearly understood. By using two algal based commercial biostimulants in combination with the Solanum lycopersicum cv. MicroTom model system, we assessed how the modulation of nitrogen metabolites and potassium levels could contribute to mediate physiological mechanisms that are known to occur in response to salt/and or osmotic stress. Here we provide evidence that the reshaping of amino acid metabolism can work as a functional effector, coordinating ion homeostasis, osmotic adjustment and scavenging of reactive oxygen species under increased osmotic stress in MicroTom plant cells. The Superfifty biostimulant is responsible for a minor amino acid rich-phenotype and could represent an interesting instrument to untangle nitrogen metabolism dynamics in response to salinity and/or osmotic stress.

Project description:This study evaluated the effectiveness of a commercially available Ascophyllum nodosum alkaline extract as a plant growth stimulant and defense elicitor against foliar diseases of tomato and sweet pepper caused by Xanthomonas campestris pv. vesicatoria and Alternaria solani in a tropical environment. Foliar applications of 0.5% A. nodosum extract (AN) at 10-day intervals resulted in significant (P < 0.05) increase in plant growth parameters, including plant height (40%), leaf number (50%), plant dry biomass (52%), root length (59%) and chlorophyll content (20%) compared to control. Treated plants also had a significantly higher number of flower clusters, flower numbers, fruits per cluster and total harvested fruit yield. The Ascophyllum extract significantly (P < 0.05) reduced disease incidence by the pathogens in both crops under greenhouse and field conditions. The combinatory treatment of seaweed extract and a minimum dose of contact fungicide in field trials, recorded the overall lowest disease levels (60% reduction) and highest yield (57% increase). Investigations into the mechanisms of disease suppression revealed the effects of the extract in inducing the activities of defense-related enzymes including phenylalanine ammonia lyase, peroxidase, polyphenol oxidase, chitinase and β-1,3-glucanase, as well as the levels of total phenolic compounds. The effect on SA, JA and ET-mediated signalling defense pathways was examined by quantifying expression levels of marker genes including PR1-a, PinII and ETR-1, for the above pathways respectively. Both crop plants treated with A. nodosum extract had significantly higher expression levels of the PinII and ETR-1 marker genes than controls. This was coupled with a marked increase in gene transcripts involved in auxin (IAA), gibberellin (Ga2Ox) and cytokinin (IPT) biosynthesis, which provides possible evidence for induced growth in plants treated with AN extract. Cross-talks between growth and defense responses as a result of seaweed extract application could evidently implicate the benefits of seaweed extract usage in sustainable crop production.

Project description:BackgroundRosa damascena is extensively cultivated in various regions of Iran due to its aesthetic attributes, medicinal qualities, and essential oil production. This study investigated the efficacy of Ascophyllum nodosum extract (AnE) at concentrations of 0, 2, and 3 g L- 1 and Nano-silicon (nSiO2) at concentrations of 0, 50, and 100 mg L- 1 in ameliorating the impact of salinity on two genotypes of Damask rose ('Chaharfasl' and 'Kashan') under in vitro culture conditions. Additionally, various physio-chemical characteristics of R. damascena explants were assessed.ResultsThe findings revealed that exposure to 100 mM NaCl resulted in a substantial reduction in the Relative Water Content (RWC), Membrane Stability Index (MSI), leaf pigments (Chlorophyll b, Chlorophyll a, total Chlorophyll, and carotenoids), chlorophyll fluorescence parameters, and protein content in both genotypes when compared to control conditions. Salinity induced a significant increase in the parameter F0 and a decrease in the parameter Fv/Fm compared to the control conditions in both genotypes. Nonetheless, the genotype Kashan treated with 3 g L- 1 AnE + 100 mg L- 1 nSiO2 exhibited the maximum Fm value under control conditions, with a significant difference compared to other treatments. Furthermore, salinity caused a considerable reduction in Fm in both 'Kashan' and 'Chaharfasl' by 22% and 17%, respectively, when compared to the control condition. 'Kashan' displayed the maximum Fv/Fm compared to the other genotype. The maximum levels of Malondialdehyde (MAD) and hydrogen peroxide (H2O2) were also observed in explants affected by salinity. The combination of 3 g L- 1 AnE + 100 mg L- 1 nSiO2, followed by 2 g L- 1 AnE + 100 mg L- 1 nSiO2, exhibited substantial positive effects. Salinity also led to an increase in proline content and the activity of peroxidase (POD), superoxide dismutase (SOD), guaiacol peroxidase (GPX), and catalase (CAT) in both genotypes. The activity of these enzymes was further enhanced when AnE was applied at concentrations of 2 and 3 g L- 1 in combination with 100 mg L- 1 nSiO2.ConclusionsThe 'Kashan' genotype displayed greater tolerance to salinity by enhancing water balance, maintaining membrane integrity, and augmenting the activity of antioxidant enzymes compared to 'Chaharfasl'. The utilization of nSiO2 and AnE biostimulants demonstrated potential benefits for R. damascena, both under salinity and control conditions. These findings hold substantial importance for researchers, policymakers, and farmers, offering valuable insights into the development of salinity-tolerant crop varieties.

Project description:Intensive agricultural production utilizes large amounts of nitrogen (N) mineral fertilizers that are applied to the soil to secure high crop yields. Unfortunately, up to 65% of this N fertilizer is not taken up by crops and is lost to the environment. To compensate these issues, growers usually apply more fertilizer than crops actually need, contributing significantly to N pollution and to GHG emissions. In order to combat the need for such large N inputs, a better understanding of nitrogen use efficiency (NUE) and agronomic solutions that increase NUE within crops is required. The application of biostimulants derived from extracts of the brown seaweed Ascophyllum nodosum has long been accepted by growers as a sustainable crop production input. However, little is known on how Ascophyllum nodosum extracts (ANEs) can influence mechanisms of N uptake and assimilation in crops to allow reduced N application. In this work, a significant increase in nitrate accumulation in Arabidopsis thaliana 6 days after applying the novel proprietary biostimulant PSI-362 was observed. Follow-up studies in barley crops revealed that PSI-362 increases NUE by 29.85-60.26% under 75% N input in multi-year field trials. When PSI-362 was incorporated as a coating to the granular N fertilizer calcium ammonium nitrate and applied to barley crop, a coordinated stimulation of N uptake and assimilation markers was observed. A key indicator of biostimulant performance was increased nitrate content in barley shoot tissue 22 days after N fertilizer application (+17.9-72.2%), that was associated with gene upregulation of root nitrate transporters (NRT1.1, NRT2.1, and NRT1.5). Simultaneously, PSI-362 coated fertilizer enhanced nitrate reductase and glutamine synthase activities, while higher content of free amino acids, soluble protein and photosynthetic pigments was measured. These biological changes at stem elongation stage were later translated into enhanced NUE traits in harvested grain. Overall, our results support the agronomic use of this engineered ANE that allowed a reduction in N fertilizer usage while maintaining or increasing crop yield. The data suggests that it can be part of the solution for the successful implementation of mitigation policies for water quality and GHG emissions from N fertilizer usage.
