Confirmatory Analysis of Per and Polyfluoroalkyl Substances in Milk and Infant Formula Using UHPLC-MS/MS.

ABSTRACT: An ultra-high performance liquid chromatography tandem mass spectrometry method was developed and validated for the sensitive determination and unambiguous confirmation of residues of per and polyfluorinated alkyl substances (PFAS) in breastmilk, retail milk and infant formulas following two sample preparation methods. Sample pre-treatment was carried out by a simplified QuEChERS method without requiring dSPE or any further clean-up. The method was validated in accordance with the requirements of Commission Decision 657/2002/EC with slight modifications. The method displayed good linearity with R2 ranging from 0.9843-0.9998 for all target PFAS. The recovery and within-laboratory reproducibility of the method (n = 63) were in the range 60-121% and 5-28%, respectively. The decision limit, detection capability and limit of quantitation ranged from 30-60 ng kg-1 to 40-100 ng kg-1 and 5-50 ng kg-1, respectively. Acceptable matrix effect values in the range -45-29% were obtained with uncertainty of measurement lower than 25% for all target PFAS. The method displays its suitability for the sensitive and high-throughput confirmatory analysis of C4-C14 PFAS in breastmilk, dairy milk and infant formulas.

Project description:BackgroundPrenatal exposures to certain per- and polyfluoroalkyl substances (PFAS) have been linked to lower weight and adiposity at birth but greater weight and adiposity in childhood. We hypothesized that faster growth in early infancy may be associated with maternal PFAS concentrations.MethodsAmong 415 mother-infant pairs in a longitudinal cohort study, we estimated associations between maternal pregnancy serum concentrations of six PFAS and offspring weight and adiposity at ~5 months of age, and growth in early infancy. Linear and logistic regression models were adjusted for potential confounders including maternal pre-pregnancy body mass index. Effect modification by infant sex was evaluated. We evaluated potential confounding by correlated exposures via multipollutant linear regression and elastic net penalized regression.ResultsAssociations between maternal PFAS concentrations and infant weight and adiposity differed by offspring sex. In male infants, maternal perfluorooctanoate and perfluorononanoate were positively associated with adiposity, with percent fat mass increases of 1.5-1.7% per ln-ng/mL increase in PFAS (median adiposity at ~5 months: 24.6%). Maternal perfluorooctane sulfonate (PFOS) and perfluorohexane sulfonate (PFHxS) were associated with lower weight-for-age z-score among female infants only (-0.26 SD per ln-ng/mL PFOS, 95% CI -0.43, -0.10; -0.17 SD per ln-ng/mL PFHxS, 95% CI -0.33, -0.01). In analyses pooled by sex, 2-(N-methyl-perfluorooctane sulfonamido) acetate above vs. below the limit of detection was associated with greater odds of rapid growth in weight-for-age (odds ratio [OR] 2.2, 95% CI 1.1, 4.3) and weight-for-length (OR 3.3, 95% CI 1.8, 6.2). Multipollutant models generally confirmed the results and strengthened some associations.DiscussionWe observed sex- and chemical-specific associations between maternal serum PFAS concentrations and infant weight and adiposity. Multipollutant models suggested confounding by correlated PFAS with opposing effects. Although maternal PFAS concentrations are inversely associated with infant weight and adiposity at birth, rapid gain may occur in infancy, particularly in fat mass.

Project description:PurposePer- and polyfluoroalkyl substances (PFAS) have been found to be widespread, extremely persistent and bioaccumulative with toxicity tendencies. Pre-synthesized nanocomposite-activated carbons, referred to, as physically activated maize tassel silver (PAMTAg) and chemically activated maize tassel silver (CAMTAg) were utilized in the present study. They were used for the removal of 10 PFAS from aqueous solutions.MethodsThe nanocomposite-activated carbons were characterized via scanning electron microscopy (SEM), Fourier transform infrared (FTIR) spectroscopy, Brunauer Emmett Teller (BET) and other techniques. Batch equilibrium experiments were conducted in order to investigate the effects of solution pH, adsorbent dosage, initial PFAS concentration and temperature on the removal of PFAS using PAMTAg and CAMTAg. Langmuir and Freundlich adsorption isotherm models were used to analyse the equilibrium data obtained.ResultsMaximum adsorption capacities of 454.1 mg/g (0.91 mmol/g) and 321.2 mg/g (0.78 mmol/g) were recorded for perfluorooctanesulfonate (PFOS) and perfluorooctanoate (PFOA), respectively using CAMTAg. The values recorded for the Gibbs' free energy (ΔG°) for the adsorption of PFOS and PFOA onto PAMTAg and CAMTAg were negative; PFOS (-9.61, -9.99 and - 10.39), PFOA (-8.77, -9.76 and - 10.21) using PAMTAg; and PFOS (-13.70, -12.70 and - 12.37), PFOA (-12.86, -12.21 and - 11.17) using CAMTAg. Therefore, the adsorption processes were spontaneous and feasible. The values recorded for enthalpy (ΔH°) (kJ/mol) for the adsorption of PFOS (-26.15) and PFOA (-35.86) onto CAMTAg were negative, indicating that the adsorption mechanism is exothermic in nature. Positive values were recorded for ΔH° for the adsorption of PFOS (2.32) and PFOA (12.69) onto PAMTAg, indicative of an endothermic adsorption mechanism. Positive entropy (ΔS°) values (0.04 and 0.07) were recorded for PFOS and PFOA using PAMTAg; whereas negative values (-0.04 and - 0.08) were recorded for ΔS° using CAMTAg. A positive ΔS° indicates an increase in randomness of the adsorbate at the solid-solution interface and the reverse is the case for a negative ΔS°.ConclusionThe interplay of electrostatic attraction and hydrophobic interactions enabled the removal of PFAS using PAMTAg and CAMTAg. Findings suggest that PAMTAg and CAMTAg are effective for the removal of PFAS from aqueous media and are good alternatives to commercially available activated carbons.Graphical abstractSupplementary informationThe online version contains supplementary material available at 10.1007/s40201-020-00597-3.
