Synthesis, Characterization, and Nanodisc Formation of Non-ionic Polymers*.

ABSTRACT: Although lipid nanodiscs are increasingly used in the structural studies of membrane proteins, drug delivery and other applications, the interaction between the nanodisc belt and the protein to be reconstituted is a major limitation. To overcome this limitation and to further broaden the scope of nanodiscs, a family of non-ionic amphiphilic polymers synthesized by hydrophobic functionalization of fructo-oligosaccharides/inulin is reported. We show the stability of lipid nanodiscs formed by these polymers against pH and divalent metal ions, and their magnetic-alignment properties. The reported results also demonstrate that the non-ionic polymers extract membrane proteins with unprecedented efficiency.

Ionothermal Synthesis of Cadmium Coordination Polymers: Ionic Liquid Effects on the Synthesis, Structural, and Thermal Characterization.

Project description:Ionothermal synthesis is a little used method for the preparation of coordination polymers. By this method, two cadmium compounds were synthesized, 1, with formula Cd3(ox)F2(Ina)2 (Ina = isonicotinate) and 2, Cd(NO3)2(4,4'-Bpy) (4,4'-Bpy = 4,4'-Bipyridine). The modification of the reaction conditions has allowed to obtain 2 as a pure phase. The structure of both compounds was determined by a single-crystal X-ray diffraction. Compound 1 is isostructural to the previously reported Cd2Zn(ox)(OH)2(Ina)2. It crystallizes in the monoclinic space group P21/n and present a three-dimensional (3D) network, built-up from [Cd3(ox)F2]n2n+ layers, linked by isonicotinate ligands. Crystals of 2 are formed by twins of two components which are rotated ca. 180° to each other. This compound crystallizes in the triclinic P-1 space group and its structure can be describe as a two-dimensional (2D) 4 connected 'sql' net. The layers are composed by [Cd(NO3)2]n chains linked through 4,4'-Bpy ligands, and are pillared along the [011] direction. The thermal decomposition of 2 was studied by thermogravimetric and thermodiffractiometric techniques. The compound decomposes gradually starting from 160 °C, and due to heating, the structure suffers slight reversible changes in the bond distances and angles.

Porphyrinic Ionic Liquid Dyes: Synthesis and Characterization.

Project description:Four porphyrinic ionic liquids and four higher melting salts (>100 °C) were synthesized as potential photosensitizers from highly symmetric porphyrins by introducing alkyl chains and exchanging anions to tune their solubility and singlet oxygen generation capability. Among the synthesized compounds was 5,10,15,20-tetra(4-dodecylpyridinum)porphyrin tetrakis-bis(trifluoromethylsulfonyl)-amide, a room-temperature ionic liquid that could be crystallized as a solvate with nitrobenzene.

Synthesis through 3D printing: formation of 3D coordination polymers.

Project description:Coordination polymers (CPs) and coordination network solids such as metal-organic frameworks (MOFs) have gained increasing interest during recent years due to their unique properties and potential applications. Preparing 3D printed structures using CP would provide many advantages towards utilization in fields such as catalysis and sensing. So far, functional 3D structures were printed mostly by dispersing pre-synthesized particles of CPs and MOFs within a polymerizable carrier. This resulted in a CP active material dispersed within a 3D polymeric object, which may obstruct or impede the intrinsic properties of the CP. Here, we present a new concept for obtaining 3D free-standing objects solely composed of CP material, starting from coordination metal complexes as the monomeric building blocks, and utilizing the 3D printer itself as a tool to in situ synthesize a coordination polymer during printing, and to shape it into a 3D object, simultaneously. To demonstrate this, a 3D-shaped nickel tetra-acrylamide monomeric complex composed solely of the CP without a binder was successfully prepared using our direct print-and-form approach. We expect that this work will open new directions and unlimited potential in additive manufacturing and utilization of CPs.

Eco-friendly synthesis of ionic helical polymers and their chemical properties and reactivity.

Project description:Reaction of N-(2,4-dinitrophenyl)pyridinium chloride (salt(Cl-)) with sodium dicyanamide (Na(CN)2N) resulted in anion exchange between Cl- and (CN)2N- to yield a new Zincke salt, salt((CN)2N-). Reactions of salt((CN)2N-) with piperazine, specifically (R)-(-)- or (S)-(+)-2-methylpiperazine under eco-friendly conditions, such as in aqueous solution, in the absence of a catalyst, and at room temperature, resulted in pyridinium ring opening to yield ionic high-molecular-weight polymers with 5-2,4-dienylideneammonium dicyanamide units or chiral 5-(2-methylpiperazinium)-2,4-dienylideneammonium dicyanamide units, namely, polymer(H;(CN)2N-), polymer(R-Me;(CN)2N-), and polymer(S-Me;(CN)2N-). UV-Vis measurements revealed that the π-conjugation system expanded along the polymer chain due to the orbital interaction between the electrons on the two nitrogen atoms of the piperazinium ring. Circular dichroism (CD) measurements revealed a helical conformation of the main chain in polymer(R-Me;(CN)2N-) and polymer(S-Me;(CN)2N-). The reaction of polymer(H;(CN)2N-) with p-phenylenediamine (PDA) caused recyclization of the 2,4-dienylideneammonium unit and resulted in depolymerization to yield N-(4-aminophenyl)pyridinium dicyanamide. Cyclic voltammetry analysis suggested that the polymers obtained in this study undergo electrochemical oxidation and reduction.

Synthesis and characterization of bioactive tamoxifen-conjugated polymers.

Project description:Macromolecular conjugates of tamoxifen could perhaps be used to circumvent some of the limitations of the extensively used breast cancer drug. To test the feasibility of these conjugates, a 4-hydroxytamoxifen analogue was conjugated to a diaminoalkyl linker and then conjugated to activated esters of a poly(methacrylic acid) polymer synthesized by atom transfer radical polymerization. A polymer conjugated to the 4-hydroxytamoxifen analogue with a six-carbon linker showed high affinity for both estrogen receptor alpha and estrogen receptor beta and potent antagonism of the estrogen receptor in cell-based transcriptional reporter assays. These results suggest that the conjugation of 4-hydroxytamoxifen to a polymer results in a macromolecular conjugate that can display ligand in a manner that can be recognized by estrogen receptor and still act as a potent antiestrogen in cells.

Factors influencing the detergent-free membrane protein isolation using synthetic nanodisc-forming polymers.

Project description:The detergent-free isolation of membrane proteins using synthetic polymers is becoming the desired approach for functional and structural studies of membrane proteins. Since the expression levels for many membrane proteins are low and a high yield of functionalized reconstituted membrane proteins is essential for in vitro studies, it is crucial to optimize the experimental conditions for a given polymer to effectively solubilize target membranes/proteins. The factors that affect membrane solubilization and subsequently the isolation of a target membrane protein include polymer concentration, polymer charge, temperature, pH, and concentration of divalent metal ions. Therefore, it is important to have knowledge about the efficacy of different types of polymers in solubilizing cell membranes. In this study, we evaluate the efficacy of inulin-based non-ionic polymers in solubilizing E. coli membranes enriched with rat flavin mononucleotide binding-domain (FBD) of cytochrome-P450-reductase (CPR) and rabbit cytochrome-b5 (Cyt-b5) under various solubilization conditions. Our results show that a 1:1 (w/w) membrane:polymer ratio, low temperature, high pH and sub-millimolar concentration of metal ions favor the solubilization of E. coli membrane enriched with FBD or Cyt-b5. Conversely, the presence of excess divalent metal ions affected the final protein levels in the polymer-solubilized samples. We believe that the results from this study provides knowledge to assess and plan the use of non-ionic polymers in membrane protein studies.

Synthesis, characterization, and electrospinning of novel polyaniline-peptide polymers.

Project description:Aniline-peptide (FLDQV, FLDQVC, Dansyl-FLDQV, Dansyl-FLDQVC, and FLDQV-AMC) mixtures underwent oxidative chemical and electrochemical polymerization in excess of aniline. The products of the chemical polymerization were low molecular weight polymers containing more than 70% peptide. Electrochemically polymerized species polyaniline-FLDQV (PANI-FLDQV) consisted mainly of polyaniline units containing about 10% peptide. The solubility of the latter in 1,1,1,3,3,3-hexafluoro-2-propanol (HFP) was similar to the camphorsulfonic acid (CSA) doped emeraldine base (PANI-CSA) solubility, however the weight composition of the electrospun fibers produced from the two polymers was significantly different. 2D 1H-13C HSQC analyses were employed to analyze the binding between the aniline and peptide moieties. Binding of peptide to polyaniline is reflected by the appearance of extra cross-peaks which display line broadening between the free polyaniline and the free pentapeptide. Peptides may be chemically bonded to the polymer molecules, but they may also act as doping agents to the nitrogen atoms via hydrogen bonding.

Non-ionic, thermo-responsive DEA/DMA nanogels: synthesis, characterization, and use for DNA separations by microchip electrophoresis.

Project description:Thermo-responsive polymer "nanogels" (crosslinked hydrogel particles with sub-100 nm diameters) are intriguing for many potential applications in biotechnology and medicine. There have been relatively few reports of electrostatically neutral, thermosensitive nanogels comprising a high fraction of hydrophilic co-monomer. Here we demonstrate the syntheses and characterization of novel, non-ionic nanogels based on random N,N-diethylacrylamide (DEA)/N,N-dimethylacrylamide (DMA) copolymers, made by free-radical, surfactant-free dispersion polymerization. The volume-phase transition temperatures of these DEA/DMA nanogels are strongly affected by co-monomer composition, providing a way to "tune" the phase transition temperature of these non-ionic nanogels. While DEA nanogels (comprising no DMA) can be obtained at 70 °C by standard emulsion precipitation, DEA/DMA random co-polymer nanogels can be obtained only in a particular range of temperatures, above the initial phase transition temperature and below the critical precipitation temperature of the DEA/DMA copolymer, controlled by co-monomer composition. Increasing percentages of DMA in the nanogels raises the phase transition temperature, and attenuates and broadens it as well. We find that concentrated DEA/DMA nanogel dispersions are optically clear at room temperature. This good optical clarity was exploited for their use in a novel DNA sieving matrix for microfluidic chip electrophoresis. An ultrafast, high-efficiency dsDNA separation was achieved in less than 120 s for dsDNA ranging from 75 bp to 15,000 bp.

Synthesis and Characterization of Biodegradable Polymers Based on Glucose Derivatives.

Project description:Syntheses of two new monomers, namely the glucose derivatives 2,3,4,6-tetra-O-acetyl-1 methacryloyl-glucopyranose (MGlc) and 2,3,4,6 tetra-O-acetyl-1-acryloylglucopyranose (AGlc), are presented. Their chemical structures were determined by the FTIR, 1H and 13C NMR spectroscopies, the single-crystal X-ray analysis, supported by the powder X-ray diffraction, and the DSC analyses. Molecules of both monomers exist in the β-anomeric form in the solid state. The variable temperature X-ray diffraction studies, supported by the DSC analyses, revealed AGlc's propensity for polymorphism and temperature-induced phase transitions. MGlc and AGlc crystallised from methanol were polymerized or copolymerized with methyl methacrylate and N-vinylpyrrolidone. The biodegradabilities of polymers as well as thermal and optical properties were studied. The results show that some properties of the obtained homopolymers and copolymers resemble those of PMMA. The main difference is that the AGlc and MGlc homopolymers are biodegradable while PMMA is not. The ternary copolymers, i.e., MGlc/AGlc-MMA-NVP lose more than 10% of their weight after six months.

Synthesis and Characterization of Metallo-Supramolecular Polymers Based on Benzodipyrrolidone.

Project description:A simple route to the preparation of benzodipyrrolidone (BDP) based monomeric building blocks containing 2,2':6',2″-terpyridines is reported from a common precursor 4'-(4-pinacolatoboronphenyl)-2,2':6',2″-terpyridine via Suzuki coupling reaction. Self-assembly polymerization with ruthenium (II) ions under mild conditions yielded a series of novel metallo-supramolecular polymers with weak donor-acceptor (D-A) structures based on benzodipyrrolidone. The structure of the bridge connected BDP with terpyridine have a significant impact on the wavelength and intensity of the intramolecular charge transfer (ICT) absorption peak. The resulting metallo-polymers exhibited strong double absorption bands around 315 nm and 510 nm involved in π-π* transitions and ICT or metal to ligand charge transfer (MLCT) absorption bands. The forming of D-A structure and coordination with ruthenium (II) ions is favorable to narrow the energy gap and the energy gaps of the resulting metallo-supramolecular polymers are 2.01 and 1.62 eV, respectively.
