A Cyaphide Transfer Reagent.

ABSTRACT: The cyanide ion plays a key role in a number of industrially relevant chemical processes, such as the extraction of gold and silver from low grade ores. Metal cyanide compounds were arguably some of the earliest coordination complexes studied and can be traced back to the serendipitous discovery of Prussian blue by Diesbach in 1706. By contrast, heavier cyanide analogues, such as the cyaphide ion, C≡P^-, are virtually unexplored despite the enormous potential of such ions as ligands in coordination compounds and extended solids. This is ultimately due to the lack of a suitable synthesis of cyaphide salts. Herein we report the synthesis and isolation of several magnesium-cyaphido complexes by reduction of ⁱPr₃SiOCP with a magnesium(I) reagent. By analogy with Grignard reagents, these compounds can be used for the incorporation of the cyaphide ion into the coordination sphere of metals using a simple salt-metathesis protocol.

Rapid heteroatom transfer to arylmetals utilizing multifunctional reagent scaffolds.

Project description:Arylmetals are highly valuable carbon nucleophiles that are readily and inexpensively prepared from aryl halides or arenes and widely used on both laboratory and industrial scales to react directly with a wide range of electrophiles. Although C-C bond formation has been a staple of organic synthesis, the direct transfer of primary amino (-NH2) and hydroxyl (-OH) groups to arylmetals in a scalable and environmentally friendly fashion remains a formidable synthetic challenge because of the absence of suitable heteroatom-transfer reagents. Here, we demonstrate the use of bench-stable N-H and N-alkyl oxaziridines derived from readily available terpenoid scaffolds as efficient multifunctional reagents for the direct primary amination and hydroxylation of structurally diverse aryl- and heteroarylmetals. This practical and scalable method provides one-step synthetic access to primary anilines and phenols at low temperature and avoids the use of transition-metal catalysts, ligands and additives, nitrogen-protecting groups, excess reagents and harsh workup conditions.

Diazomethane umpolung atop anthracene: an electrophilic methylene transfer reagent.

Project description:Formal addition of diazomethane's terminal nitrogen atom to the 9,10-positions of anthracene yields H2CN2 A (1, A = C14H10 or anthracene). The synthesis of this hydrazone is reported from Carpino's hydrazine H2N2 A through treatment with paraformaldehyde. Compound 1 has been found to be an easy-to-handle solid that does not exhibit dangerous heat or shock sensitivity. Effective umpolung of the diazomethane unit imbues 1 with electrophilicity at the methylene carbon center. Its reactivity with nucleophiles such as H2CPPh3 and N-heterocyclic carbenes is exploited for C[double bond, length as m-dash]C bond formation with elimination of dinitrogen and anthracene. Similarly, 1 is demonstrated to deliver methylene to a nucleophilic singlet d2 transition metal center, W(ODipp)4 (2), to generate the robust methylidene complex [2[double bond, length as m-dash]CH2]. This behavior is contrasted with that of the Wittig reagent H2CPPh3, a more traditional and Brønsted basic methylene source that upon exposure to 2 contrastingly forms the methylidyne salt [MePPh3][2[triple bond, length as m-dash]CH].

Benzothiazole Sulfinate: A Sulfinic Acid Transfer Reagent under Oxidation-Free Conditions.

Project description:Sulfinic acids are commonly encountered intermediates found in natural product synthesis and medicinal chemistry. However, because of high reactivity, instability, and harsh reaction conditions, they are difficult to synthesize. Herein we have developed an oxidation-free method to produce sulfinic acids and sulfinate salts using 2-sulfinyl benzothiazole (BTS). We have also demonstrated the synthetic usefulness by developing one-pot syntheses of sulfones and sulfonamides.

Sulfur Pentafluoride is a Preferred Reagent Cation for Negative Electron Transfer Dissociation.

Project description:Negative mode proteome analysis offers access to unique portions of the proteome and several acidic post-translational modifications; however, traditional collision-based fragmentation methods fail to reliably provide sequence information for peptide anions. Negative electron transfer dissociation (NETD), on the other hand, can sequence precursor anions in a high-throughput manner. Similar to other ion-ion methods, NETD is most efficient with peptides of higher charge state because of the increased electrostatic interaction between reacting molecules. Here we demonstrate that NETD performance for lower charge state precursors can be improved by altering the reagent cation. Specifically, the recombination energy of the NETD reaction-largely dictated by the ionization energy (IE) of the reagent cation-can affect the extent of fragmentation. We compare the NETD reagent cations of C16H10?+ (IE?=?7.9 eV) and SF5?+ (IE?=?9.6 eV) on a set of standard peptides, concluding that SF5?+ yields greater sequence ion generation. Subsequent proteome-scale nLC-MS/MS experiments comparing C16H10?+ and SF5?+ further supported this outcome: analyses using SF5?+ yielded 4637 peptide spectral matches (PSMs) and 2900 unique peptides, whereas C16H10?+ produced 3563 PSMs and 2231 peptides. The substantive gain in identification power with SF5?+ was largely driven by improved identification of doubly deprotonated precursors, indicating that increased NETD recombination energy can increase product ion yield for low charge density precursors. This work demonstrates that SF5?+ is a viable, if not favorable, reagent cation for NETD, and provides improved fragmentation over the commonly used fluoranthene reagent. Graphical Abstract ?.

A versatile snap chip for high-density sub-nanoliter chip-to-chip reagent transfer.

Project description:The coordinated delivery of minute amounts of different reagents is important for microfluidics and microarrays, but is dependent on advanced equipment such as microarrayers. Previously, we developed the snap chip for the direct transfer of reagents, thus realizing fluidic operations by only manipulating microscope slides. However, owing to the misalignment between arrays spotted on different slides, millimeter spacing was needed between spots and the array density was limited. In this work, we have developed a novel double transfer method and have transferred 625 spots cm(-2), corresponding to >10000 spots for a standard microscope slide. A user-friendly snapping system was manufactured to make liquid handling straightforward. Misalignment, which for direct transfer ranged from 150-250 ?m, was reduced to <40 ?m for double transfer. The snap chip was used to quantify 50 proteins in 16 samples simultaneously, yielding limits of detection in the pg/mL range for 35 proteins. The versatility of the snap chip is illustrated with a 4-plex homogenous enzyme inhibition assay analyzing 128 conditions with precise timing. The versatility and high density of the snap chip with double transfer allows for the development of high throughput reagent transfer protocols compatible with a variety of applications.

Tellurolate: an effective Te-atom transfer reagent to prepare the triad of group 5 metal bis(tellurides).

Project description:We show in this work how lithium tellurolate Li(X)nTeCH2SiMe3 (X = THF, n = 1, 1; X = 12-crown-4, n = 2, 2), can serve as an effective Te-atom transfer reagent to all group 5 transition metal halide precursors irrespective of the oxidation state. Mononuclear and bis(telluride) complexes, namely (PNP)M(Te)2 (M = V; Nb, 3; Ta, 4; PNP- = N[2-PiPr2-4-methylphenyl]2), are reported herein including structural and spectroscopic data. Whereas the known complex (PNP)V(Te)2 can be readily prepared from the trivalent precursor (PNP)VCl2, two equiv. of tellurolate, and elemental Te partially solubilized with PMe3, complex 3 can also be similarly obtained following the same procedure but with or without a reductant, Na/NaCl. Complex 4 on the other hand is formed from the addition of four equiv. of tellurolate to (PNP)TaF4. Having access to a triad of (PNP)M(Te)2 systems for group 5 metals has allowed us to compare them using a combination of theory and spectroscopy including Te-L1 edge XANES data.

Negishi's Reagent Versus Rosenthal's Reagent in the Formation of Zirconacyclopentadienes.

Project description:Zirconacyclopentadienes are versatile precursors for a large number of heteroles, which are accessible by Zr-element exchange reactions. The vast majority of reports describe their preparation by the use of Negishi's reagent, which is a species that is formed in situ. The zirconacyclopentadiene is then formed by the addition of one equivalent of a diyne or two equivalents of a monoyne moiety to this Negishi species. Another route involves Rosenthal's reagent (Cp2 Zr(py)Me3 SiC≡CSiMe3 ), which then reacts with a diyne or monoyne moiety. In this work, the efficiency of both routes was compared in terms of reaction time, stability of the product in the reaction mixture, and yield. The synthetic implications of using both routes are evaluated. Novel zirconacyclopentadienes were synthesized, characterized directly from the reaction mixture, and crystal structures could be obtained in most cases.

Renewable Reagent for Nucleophilic Fluorination.

Project description:Herein, we report a study on the reactivity of three 1,3-diarylimidazolium-based fluoride reagents, with a general formula of [IPrH][F(HF)n] (n = 0, 1, or 2), that tackle the challenges of limited solubility, hygroscopicity, instability, and laborious preparation procedures of nucleophilic fluoride reagents. Fluorination of 4-tert-butylbenzyl bromide reveals that trifluoride [IPrH][F(HF)2] is the most selective reagent. Microwave-assisted activation coupled with the addition of sterically hindered amine DIPEA or alkali metal fluorides increases the rate of fluorination with [IPrH][F(HF)2], making it an excellent reagent for the fluorination of various organic substrates. The scope of substrates includes benzyl bromides, iodides, chlorides, aliphatic halides, tosylates, mesylates, α-haloketones, a silyl chloride, acyl and sulfuryl chlorides, and a nitroarene. The exceptional stability of the air-stable and nonhygroscopic [IPrH][F(HF)2] reagent is illustrated by its convenient synthesis and detailed experimental regeneration protocol using hydrofluoric acid without organic solvents.

Metal nitrite: a powerful oxidizing reagent.

The combination of a Lewis acid and a metal nitrite was applied to the oxidation of silyl enol ethers. Project description:When ketones flanked on both sides by nucleophilic atoms react with Seebach's nitropropenyl pivaloate reagent, direct couplings take place to give two new ring systems and three bonds. Cis-ring fusions are observed in unions leading to 5,5-, 5,6-, and 6,6-bicycles. Densely functionalized and rigid frameworks may be rapidly formed by the chemistry described herein.
