Project description:Although healthy aging is accompanied by a general decline in memory functioning, environmental support at retrieval can improve older adults' (+65 years) episodic remembering. Despite those over the age of 65 years representing a growing proportion of the population, few environmental retrieval support methods have been empirically evaluated for use with older witnesses and victims of crime. Here, the efficacy of a novel retrieval technique, the Sketch Mental Reinstatement of Context, is compared with a standard Mental Reinstatement of Context and a no support control (Control). Fifty-one participants witnessed an unexpected live event, and 48 hours later were interviewed using one of three aforementioned techniques. In line with predictions emanating from cognitive theories of aging and the environmental support hypothesis, participants in the Sketch Mental Reinstatement of Context condition recalled significantly more correct information and fewer inaccurate items. The Sketch Mental Reinstatement of Context technique appears to scaffold memory retrieval in an age-appropriate manner during a post-event interview, possibly by encouraging more effortful retrieval and reducing dual-task load. As such, this procedure offers an effective alternative to current approaches, adding to the toolbox of techniques available to forensic and other interviewers.
Project description:Dementia is one among the consequences of aging, and amnesia is often one of the most common symptoms. The lack of memory, as a consequence of both "healthy" aging or neurodegenerative conditions, such as in Alzheimer's disease, has a dramatic impact on the patient's lifestyle. In fact, the inability to recall information made by a previous experience could not only alter the interaction with the environment, but also lead to a loss of identity. Mitochondria are key regulators of brain's activity; thanks to their "dynamic organelles" nature they constantly rearrange in the cell body and move along axons and dendrites, changing in dimension, shape, and location, accordingly to the cell's energy requirements. Indeed, the energy they can provide is essential to maintain synaptic plasticity and to ensure transmission through presynaptic terminals and postsynaptic spines. Stressful conditions, like the ones found in neurodegenerative diseases, seriously impair mitochondria bioenergetic, leading to both loss of proper neuronal interaction and of neuron themselves. Here, we highlighted the current knowledge about the role of mitochondria and mitochondrial dynamics in relation to neurodegenerative disorders linked to aging. Furthermore, we discuss the obstacles as well as the future perspectives aimed to enlarge our knowledge about mitochondria as target for new therapeutic strategies to slow down aging and neurodegenerative disease's symptoms.
Project description:PurposeTo determine the incidence and density of Demodex species on the eyelashes of subjects with normal eyelids, anterior blepharitis (AB), meibomian-gland dysfunction (MGD), and mixed blepharitis (MB).Materials and methodsOne hundred and fifty consecutive patients diagnosed with AB, MGD, and MB were recruited. An additional 50 individuals were recruited who were free of lid and margin disease to serve as a control group. All patients underwent a standard eye examination. Data on ocular symptomatology were gathered. Digital photography was performed of the lid margins. Lash sampling was performed by epilating the lashes and the lashes were checked for Demodex based on morphology using light microscopy. The total number of mites were tabulated for each eye. Comparison between the affected eyes and the control eyes was performed. Data were analyzed using the Chi-square test.ResultsA total of 200 patients were included. Twenty percenth had AB, 40% had MGD, and 40% had MB. The incidence of Demodex infestation was 90% in cases of AB, 60% in MGD cases, and 90% in MB cases. The incidence of Demodex in control subjects was 18%.ConclusionsThe incidence and density of Demodex infestation was highest in patients with AB and MB. Lid irritation and presence of cylindrical dandruff were indicative of a high-density infestation. These signs should alert the clinician to treat concomitant Demodex infestation.
Project description:Aging is a major risk factor for many diseases including metabolic syndrome, cancer, inflammation, and neurodegeneration. Identifying mechanistic common denominators underlying the impact of aging is essential for our fundamental understanding of age-related diseases and the possibility to propose new ways to fight them. One can define aging biochemically as prolonged metabolic stress, the innate cellular and molecular programs responding to it, and the new stable or unstable state of equilibrium between the two. A candidate to play a role in the process is protein kinase R (PKR), first identified as a cellular protector against viral infection and today known as a major regulator of central cellular processes including mRNA translation, transcriptional control, regulation of apoptosis, and cell proliferation. Prolonged imbalance in PKR activation is both affected by biochemical and metabolic parameters and affects them in turn to create a feedforward loop. Here, we portray the central role of PKR in transferring metabolic information and regulating cellular function with a focus on cancer, inflammation, and brain function. Later, we integrate information from open data sources and discuss current knowledge and gaps in the literature about the signaling cascades upstream and downstream of PKR in different cell types and function. Finally, we summarize current major points and biological means to manipulate PKR expression and/or activation and propose PKR as a therapeutic target to shift age/metabolic-dependent undesired steady states.
Project description:As advancements in science and technology, such as the Internet of things, smart home systems, and automobile displays, become increasingly embedded in daily life, there is a growing demand for displays with customized sizes and shapes. This study proposes a pen drawing display technology that can realize a boardless display in any form based on the user's preferences, without the usual restrictions of conventional frame manufacturing techniques. An advantage of the pen drawing method is that the entire complex fabrication process for the display is encapsulated in a pen. The display components, light-emitting layers, and electrodes are formed using felt-tip drawing pens that contain the required solutions and light-emitting materials. The morphology and thickness of each layer is manipulated by adjusting the drawing speed, number of drawing cycles, and substrate temperature. This study is expected to usher in the upcoming era of customized displays that can reflect individual user needs.
Project description:Escherichia coli (E. coli) bacteria govern their trajectories by switching between running and tumbling modes as a function of the nutrient concentration they experienced in the past. At short time one observes a drift of the bacterial population, while at long time one observes accumulation in high-nutrient regions. Recent work has viewed chemotaxis as a compromise between drift toward favorable regions and accumulation in favorable regions. A number of earlier studies assume that a bacterium resets its memory at tumbles - a fact not borne out by experiment - and make use of approximate coarse-grained descriptions. Here, we revisit the problem of chemotaxis without resorting to any memory resets. We find that when bacteria respond to the environment in a non-adaptive manner, chemotaxis is generally dominated by diffusion, whereas when bacteria respond in an adaptive manner, chemotaxis is dominated by a bias in the motion. In the adaptive case, favorable drift occurs together with favorable accumulation. We derive our results from detailed simulations and a variety of analytical arguments. In particular, we introduce a new coarse-grained description of chemotaxis as biased diffusion, and we discuss the way it departs from older coarse-grained descriptions.
Project description:In 1969, social psychologist Milgram and his colleagues conducted an experiment on a busy city street where passers-by witnessed a set of actors spontaneously looking up towards a building. The experiment showed that the crowd's propensity to mimic the actor's gaze increased with the number of actors that looked up. This form of behavioural contagion is found in many social organisms and is central to how information travels through large groups. With the advancement of virtual reality and its continued application towards understanding human response to crowd behaviour, it remains to be verified if behavioural contagion occurs in walkable virtual environments, and how it compares with results from real-world experiments. In this study, we adapt Milgram's experiment for virtual environments and use it to reproduce behavioural contagion. Specifically, we construct a replica of an indoor location and combine two established pedestrian motion models to create an interactive crowd of 60 virtual characters that walk through the indoor location. The stimulus group comprised a subset of the characters who look up at a random time as the participants explore the virtual environment. Our results show that the probability of looking up by a participant is dependent on the size of the stimulus group saturating to near certainty when three or more characters look up. The role of stimulus size was also evident when participant actions were compared with survey responses which showed that more participants selected to not look up even though they saw characters redirect their gaze upwards when the size of the stimulus group was small. Participants also spent more time looking up and exhibited frequent head turns with a larger stimulus group. Results from this study provide evidence that behavioural contagion can be triggered in the virtual environment, and can be used to build and test complex hypotheses for understanding human behaviour in a variety of crowd scenarios.
Project description:This article presents the outcomes from a mixed-methods study of drawing practitioners (e.g., professional illustrators, fine artists, and art students) that was conducted in Autumn 2018 as a preliminary investigation for the development of a physical human-AI co-creative drawing system. The aim of the study was to discover possible roles that technology could play in observing, modeling, and possibly assisting an artist with their drawing. The study had three components: a paper survey of artists' drawing practises, technology usage and attitudes, video recorded drawing exercises and a follow-up semi-structured interview which included a co-design discussion on how AI might contribute to their drawing workflow. Key themes identified from the interviews were (1) drawing with physical mediums is a traditional and primary way of creation; (2) artists' views on AI varied, where co-creative AI is preferable to didactic AI; and (3) artists have a critical and skeptical view on the automation of creative work with AI. Participants' input provided the basis for the design and technical specifications of a co-creative drawing prototype, for which details are presented in this article. In addition, lessons learned from conducting the user study are presented with a reflection on future studies with drawing practitioners.
Project description:Vivid episodic memories in people have been characterized as the replay of unique events in sequential order [1-3]. Animal models of episodic memory have successfully documented episodic memory of a single event (e.g., [4-8]). However, a fundamental feature of episodic memory in people is that it involves multiple events, and notably, episodic memory impairments in human diseases are not limited to a single event. Critically, it is not known whether animals remember many unique events using episodic memory. Here, we show that rats remember many unique events and the contexts in which the events occurred using episodic memory. We used an olfactory memory assessment in which new (but not old) odors were rewarded using 32 items. Rats were presented with 16 odors in one context and the same odors in a second context. To attain high accuracy, the rats needed to remember item in context because each odor was rewarded as a new item in each context. The demands on item-in-context memory were varied by assessing memory with 2, 3, 5, or 15 unpredictable transitions between contexts, and item-in-context memory survived a 45 min retention interval challenge. When the memory of item in context was put in conflict with non-episodic familiarity cues, rats relied on item in context using episodic memory. Our findings suggest that rats remember multiple unique events and the contexts in which these events occurred using episodic memory and support the view that rats may be used to model fundamental aspects of human cognition.
